| 1 | Mitochondrial dysfunction activates the AMPK signaling and autophagy to promote cell survival显示文摘Autophagy is a cellular self-eating process essential for stress response and maintainingtissue homeostasis by lysosomal degradation of unwanted or damaged proteins and organelles.Here, we show that cells with defective mitochondria induce autophagy to promotecell survival through activating the AMPK pathway. Loss of mitochondrial complex III proteincytochrome b activates the AMPK signaling and induced autophagy. Inhibiting mitochondria energeticsby mitochondria-targeted agents activates the AMPK signaling and induced autophagy.Genetic inhibition of AMPK inhibits autophagy induction in cells with defective mitochondria,while genetic inhibition of autophagy has no effect on AMPK activation. Mitochondria dysfunctionhas no effect of DNA repair of UV-induced DNA damage. However, mitochondria dysfunctionsensitizes cells to apoptosis induced by UV radiation. Genetic inhibition of autophagy orAMPK sensitized cells to apoptosis in cells with defective mitochondria. Our results demonstratethat AMPK and autophagy senses mitochondria dysfunction and serves as a mechanismfor survival. | 2 | Tomato lycopene attenuates myocardial infarction induced by isoproterenol:Electrocardiographic,biochemical and anti-apoptotic study显示文摘Objective:To assess the protective effects of lycopene on electrocardiographic,hemodynamic, biochemical and apoptotic changes in isoproterenol induced myocardial infarction.Methods: Myocardial infarction was induced in rats by subcutaneous injection of isoproterenol(200 mg/kg) for two consecutive days at an interval of 24 h.Rats were treated with lycopene(10 mg/kg/day, p.o.) for a period of 30 days and isoproterenol(ISO) was injected on the 29th and 30th day.At the end of experiment i.e.on the 31st day electrocardiographic,hemodynamic,biochemical and apoptotic changes were monitored from control and experimental groups.Results:ISO injected ruts showed a significant alteration in electrocardiograph pattern and hemodynamic changes(i.e. systolic,diastolic and mean arterial pressure).It also showed significant increase in C-reactive protein,myeloperoxidase,nitrite levels and Caspase-3 protease activity.In addition,it also exhibited alteration in the levels of electrolytes(Na^+,K^+ and Ca^(2+),vitamin E,uric acid and serum protein.Cel electrophoresis of ISO injected rats showed increase in DNA fragmentation.Triphenyl tetrazolium chloride staining of the heart section shows increase area of infarction in ISO injected rats.Pre-co-treatment with lycopene significantly prevented the ISO induced ullerution in ECC, haemodynamic,biochemical and apoptotic changes.Conclusions:The present result shows that treatment of lycopene in ISO injected rats significantly attenuates induced myocardial infarction. | Aman Upaganlawar Vaibhav Patel Balaraman R | 2012 | Asian Pacific Journal of Tropical Biomedicine2012,2,5: | 6 |
| 3 | Implications of the presence of an aberrant right hepatic artery in patients undergoing pancreaticoduodenectomy显示文摘AIM:To analyze the differences in outcomes and the clinical impact following pancreatoduodenectomy(PD)in patients with and without aberrant right hepatic artery(aRHA).METHODS:All patients undergoing PD between January 2008 and December 2012 were divided into two groups,one with aRHA and the other without.These groups were compared to identify differences in the intraoperative variables,the oncological clearance and the postoperative morbidity,mortality and hospital stay.RESULTS:A total of 225 patients underwent PD,of which 43(19.1%)patients were found to have eitheraccessory or replaced right hepatic arteries(aRHA group).The aRHA was preserved in 79%of the patients.There was no significant difference in the intraoperative blood loss but operative time was prolonged,reflecting the complexity of the procedure[420±44(240-540)min vs 480±45(300-600)min,P<0.05)].There were no differences in the incidence of postoperative complications(pancreatic leak,pancreatic fistula,delayed gastric emptying and mortality)and hospital stay.Oncological clearance in the form of positive resection margins[13(7.1%)vs 3(6.9%)]and lymph node yield were also similar in the two groups.CONCLUSION:An aRHA is found in approximately one fifth of patients undergoing PD.Preservation is technically possible in most patients and can increase the operative complexity but does not negatively affect the safety or oncological outcomes of the procedure. | Ashwin Rammohan Ravichandran Palaniappan Anbalagan Pitchaimuthu Kamalakannan Rajendran Senthil Kumar Perumal Kesavan Balaraman Ravi Ramasamy Jeswanth Sathyanesan Manoharan Govindan | 2014 | World Journal of Gastrointestinal Surgery2014,6,1: | 3 |
| 4 | Carvacrol Promotes Cell Cycle Arrest and Apoptosis through PI3K/AKT Signaling Pathway in MCF-7 Breast Cancer Cells显示文摘Objective:To examine the role of carvacrol in modulating PI3 K/AKT signaling involved in human breast cancer pathogenesis using in vitro experimental model MCF-7 cells.Methods:MTT and lactate dehydrogenase assays were performed with cells treated with different doses of carvacrol(0–250μmol/L)at different time points(24 and 48 h).The nuclear morphology was assessed in MCF-7 cells with propidium iodide(PI)and acridine orange/ethidium bromide(AO/EB)staining and analyzed by fluorescence microscopy.Events like cell cycle arrest and apoptosis were observed by flow cytometric analysis and expressions of p-Rb,cyclin D1,cyclin-dependent kinase 4(CDK4),CDK6,Bax,Bcl-2,PI3 K/p-AKT were analyzed by immunoblot.Results:Carvacrol significantly reduced cell viability with the half maximal inhibitory concentration value of 200μmol/L at 24 and 48 h(P<0.05).importantly,there was a significant increase in the accumulation of the G0/G1 phase upon treatment with carvacrol in MCF-7 cells(P<0.05 or P<0.01).A remarkable decrease in protein expressions of p-Rb,cyclin D1,CDK4 and CDK6 denoted cell cycle arrest(P<0.05 or P<0.01).In addition,carvacrol treatment significantly inhibited PI3 K/p-AKT protein expressions leading to induction of apoptosis mediated by decreased Bcl2 and increased Bax protein expressions.Further,Annexin V/PI staining by FACS analysis,dual staining by AO/EB and PI staining studies suggested induction of apoptosis by carvacrol through PI3 K/Akt signaling pathway in MCF-7 cells.Conclusion:Carvacrol significantly inhibited the breast cancer MCF-7 cell proliferation and induced apoptosis via suppressing PI3/AKT signaling pathway. | Ashok Mari Gopikrishnan Mani Sirpu Natesh Nagabhishek Gopaiakrishnan Balaraman Nirmala Subramanian Fathima Bushra Mirza Jagan Sundaram Devaki Thiruvengadam | 2021 | Chinese Journal of Integrative Medicine2021,27,9: | 3 |
| 5 | Antibacterial potential of sponge endosymbiont marine Enterobacter sp at Kavaratti Island,Lakshadweep archipelago显示文摘Objective:To isolate antibacterial potential of sponge endosymbiotic bacteria from marine sponges at Lakshadweep archipelago.Also to identify the potent bacteria by 16s rDNA sequencing and determine the antibacterial activity against clinical pathogens by MIC.Methods: Sponge samples was collected from sub-tidal habitats at Kavaratti Island and identified.The endosymbiotic bacteria were isolated and selected potential bacteria which show antibacterial activity in preliminary screening against clinical pathogens Staphylococcus aureus(S.aureus), Escherichia coli(E.coli),Salmonella typhi(S.typhi),Klebsiella pneumoniea(K.pneumoniea) and Streptococcus sp.by disc diffusion assay.The crude extracts of potential bacteria LB3 was tested against clinical pathogens by MIC.The LB3 strain was identified by 16s rDNA sequencing, 1111 bp was submitted in NCBI(HQ589912) and constructed phylogenetic tree.Results:Sponge sample was identified as Dysidea granulosa(D.granulosa) and potential bacteria LB3 identified as Enterobacter sp TTAG.Preliminary screening of sponge isolates against clinical pathogens,LB3 strain was selected as potential producer of secondary metabolites and crude extract was implies on MIC of LB3 have confirmed with lowest concentration of 5.0 mg/mL in broth medium influence of crude extract on growth inhibitory activity after 5 h of incubation period and completed the inhibitory activity at 15 h.Conclusions:The present study concluded that phylogenetic analysis of endosymbiotic bacteria Enterobacter sp from sponge D.granulosa of Lakshadweep islands showed significant antibacterial activity against clinical bacterial pathogens. | Mohan Gopi Subramanian Kumaran Thipparamalai Thangappanpillai Ajith Kumar Balaraman Deivasigamani Kumarappan Alagappan Srinivasan Guru Prasad | 2012 | Asian Pacific Journal of Tropical Medicine2012,5,2: | 2 |

| 7 | Antibiotic resistant Esherichia coli strains from seafood and its susceptibility to seaweed extracts显示文摘Objective:To determine the prevalence and antibiotic resistance of Escherichia coli(E. coli),in seafood obtained from Cuddalore and Parangipettai fish landing centres.Also,to identify the susceptibility of E.coli against predominant seaweeds red alga Kappaphycus alvarezii(K.alvarezii) and brown alga Padina boergessenii(P.boergessenii) extracts as sulfated polysaccharides and polyphenols respectively.Methods:A total of 48 samples(Two stations Cuddalore and Parangipettai,Tamil Nadu,India).Sampling area are fish landing centre where fishes caught from sea and estuary,seafood processing plants(packing and ice packed fishes) and local fish markets(fish samples).After isolation totally 80 strains were analyzed for its antimicrobial resistance and sensitivity against commercially 10 antibiotics.The ampicillin resistant E.coli CE21 was identified through molecular techniques as I6S rDNA sequencing. Two seaweeds K.alvarezii and P.boergessenii were screened lor antibacterial activity against 12 antibiotic resistant E.coli strains.Results:Totally 48 swabbed samples from two different fish handling area were characterized for total bacterial and E.coli count.Mostly,the E.coli strains were isolated from fish local market and seafood processing plants before and after packaging process.In that maximum 56.25%strains were resistant to ampicillin and the minimum 2.5%strains were resistant to chloramphenicol.Therefore,the E.coli CE21 was identified through molecular techniques E.coli(GenBank accession number GU065251),The MIC value for polyphenol extract was slightly less than sulfated polysaccharides.E.coli strain isolated from Parangipettai was considerably increased MIC value that Cuddalore.Conclusions: The polyphenol and sulfated polysaccharides showed promising inhibitory response against all antimicrobial resistant E.coli strains and in particular the inhibitory response of ampicillin resistant E.coli. | Subramanian Kumaran Balaraman Deivasiganiani Kumarappan Alagappan Mannikam Sakthivel Rajamani Karthikeyan | 2010 | Asian Pacific Journal of Tropical Medicine2010,3,12: | 1 |
