1Bacillus cereus AR156 primes induced systemic resistance by suppressing miR825/825 and activating defense-related genes in Arabidopsis显示文摘Small RNAs play an important role in plant immune responses. However, their regulatory function in induced systemic resistance(ISR) is nascent. Bacillus cereus AR156 is a plant growth-promoting rhizobacterium that induces ISR in Arabidopsis against bacterial infection. Here,by comparing small RNA profiles of Pseudomonas syringae pv. tomato(Pst) DC3000-infected Arabidopsis with and without AR156 pretreatment, we identified a group of Arabidopsis micro RNAs(mi RNAs) that are differentially regulated by AR156 pretreatment. mi R825 and mi R825 are two mi RNA generated from a single mi RNA gene.Northern blot analysis indicated that they were significantly downregulated in Pst DC3000-infected plants pretreated with AR156, in contrast to the plants without AR156 pretreatment. mi R825 targets two ubiquitin-protein ligases,while mi R825 targets toll-interleukin-like receptor(TIR)-nucleotide binding site(NBS) and leucine-rich repeat(LRR)type resistance(R) genes. The expression of these target genes negatively correlated with the expression of mi R825 and mi R825. Moreover, transgenic plants showing reduced expression of mi R825 and mi R825 displayed enhanced resistance to Pst DC3000 infection, whereas transgenic plants overexpressing mi R825 and mi R825 were more susceptible. 2Reconstructing Spatial Distribution of Historical Cropland in China′s Traditional Cultivated Region: Methods and Case Study显示文摘As an important part of land use/cover change(LUCC), historical LUCC in long time series attracts much more attention from scholars. Currently, based on the view of combining the overall control of cropland area and ′top-down′ decision-making behaviors, here are two global historical land-use datasets, generally referred as the Sustainability and the Global Environment datasets(SAGE datasets) and History Database of the Global Environment datasets(HYDE datasets). However, at the regional level, these global datasets have coarse resolutions and inevitable errors. Considering various factors that influenced cropland distribution, including cropland connectivity and the limitation of natural and human factors, this study developed a reconstruction model of historical cropland based on constrained Cellular Automaton(CA) of ′bottom-up′. Then, an available labor force index is used as a proxy for the amount of cropland to inspect and calibrate these spatial patterns. Applied the reconstruction model to Shandong Province, we reconstructed its spatial distribution of cropland during 8 periods. The reconstructed results show that: 1) it is properly suitable for constrained CA to simulate and reconstruct the spatial distribution of cropland in traditional cultivated region of China; 2) compared with ′SAGE datasets′ and ′HYDE datasets′, this study have formed higher-resolution Boolean spatial distribution datasets of historical cropland with a more definitive concept of spatial pattern in terms of fractional format.YANG Xuhong GUO Beibei JIN Xiaobin LONG Ying ZHOU Yinkang 2015Chinese Geographical Science2015,25,5:4
3Mapping a leaf senescence gene els1 by BSR-Seq in common wheat显示文摘Leaf senescence is normally the last stage of plant development. Early senescence of functional leaves significantly reduces the photosynthetic time and efficiency, seriously affecting grain yield and quality in wheat. Discovering genes responsible for early leaf senescence(els) are necessary for developing novel germplasms and cultivars with delayed leaf-senescence through molecular manipulation and marker assisted selection. In this study, we identified an early leaf senescence line M114 in a derivative of a wheat breeding population. Genetic analysis indicated that early leaf senescence in M114 is controlled by a single recessive gene, provisionally designated els1. By applying bulked segregant analysis and RNA-Seq(BSR-Seq), seven polymorphic markers linked to els1 were developed and the gene was located on chromosome arm 2 BS in a 1.5 c M genetic interval between markers WGGB303 and WGGB305. 4Polymersome-mediated cytosolic delivery of cyclic dinucleotide STING agonist enhances tumor immunotherapy显示文摘Cyclic dinucleotides(CDNs)as stimulator of interferon genes(STING)agonists capable of inducing strong antitumor innate immune response are highly promising for tumor immunotherapy.The efficacy of these CDNs is,however,reduced greatly by their fast clearance,poor cell uptake and inefficient cytosolic transportation.Here,we report that reduction-responsive biodegradable chimaeric polymersomes(CPs)markedly enhance tumor retention and cytosolic delivery of a synthetic CDN,ADU-S100,and bolster STING pathway activation in the tumor microenvironment and tumor draining lymph nodes,giving significantly better tumor repression and survival of B16F10 melanoma-bearing mice compared with free CDN control.The superiority of CPs-mediated CDN delivery is further verified in combination therapy with low-dose fractionated radiation,which brings about clearly stronger and longer-term immunotherapeutic effects and protection against tumor re-challenge.The development of nano-STING agonists that are able to overcome the delivery barriers of CDNs represents an effective strategy to potentiate cancer immunotherapy.Huan Zheng Beibei Guo Xinyun Qiu Yifeng Xia Yan Qu Liang Cheng Fenghua Meng Zhiyuan Zhong 2022Bioactive Materials2022,7,10:2

6Mesoporous Ternary Nitrides of Earth?Abundant Metals as Oxygen Evolution Electrocatalyst显示文摘As sustainable energy becomes a major concern for modern society,renewable and clean energy systems need highly active,stable,and low-cost catalysts for the oxygen evolution reaction(OER).Mesoporous materials offer an attractive route for generating efficient electrocatalysts with high mass transport capabilities.Herein,we report an efficient hard templating pathway to design and synthesize three-dimensional(3-D)mesoporous ternary nickel iron nitride(Ni3FeN).The as-synthesized electrocatalyst shows good OER performance in an alkaline solution with low overpotential(259 mV)and a small Tafel slope(54 mV dec?1),giving superior performance to IrO2 and RuO2 catalysts.The highly active contact area,the hierarchical porosity,and the synergistic effect of bimetal atoms contributed to the improved electrocatalytic performance toward OER.In a practical rechargeable Zn–air battery,mesoporous Ni3FeN is also shown to deliver a lower charging voltage and longer lifetime than RuO2.This work opens up a new promising approach to synthesize active OER electrocatalysts for energy-related devices.Ali Saad Hangjia Shen Zhixing Cheng Ramis Arbi Beibei Guo Lok Shu Hui Kunyu Liang Siqi Liu John Paul Attfield Ayse Turak Jiacheng Wang Minghui Yang 2020Nano-Micro Letters2020,12,6:2

10Genome-wide signatures of the geographic expansion and breeding of soybean显示文摘Soybean is a leguminous crop that provides oil and protein. Exploring the genomic signatures of soybean evolution is crucial for breeding varieties with improved adaptability to environmental extremes. We analyzed the genome sequences of 2,214 soybeans and proposed a soybean evolutionary route, i.e., the expansion of annual wild soybean(Glycine soja Sieb. & Zucc.) from southern China and its domestication in central China, followed by the expansion and local breeding selection of its landraces(G. max(L.) Merr.). We observed that the genetic introgression in soybean landraces was mostly derived from sympatric rather than allopatric wild populations during the geographic expansion. Soybean expansion and breeding were accompanied by the positive selection of flowering time genes, including GmSPA3c. 14Precisely quantifying bulk transition metal valence evolution in conventional battery electrode by inverse partial fluorescence yield显示文摘Precisely quantifying transition metal(TM) redox in bulk is a key to understand the fundamental of optimizing cathode materials in secondary batteries. At present, the commonly used methods to probe TM redox are hard X-ray absorption spectroscopy(hXAS) and soft X-ray absorption spectroscopy(sXAS).However, they are both facing challenges to precisely quantify the valence states of some transition metals such as Mn. In this paper, Mn-L iPFY(inverse partial fluorescence yield) spectra extracted from Mn-L m RIXS(mapping of resonant inelastic X-ray scattering) is adopted to quantify Mn valence states. Mn-L i PFY spectra has been considered as a bulk-sensitive, non-distorted probe of TM valence states.However, the exact precision of this method is still unclear in quantifying practical battery electrodes.Herein, a series of LiMn_(2)O_(4) electrodes with different charge and discharge states are prepared. Based on their electrochemical capacity(generally considered to be very precise), the precision of Mn iPFY in quantifying bulk Mn valence state is confirmed, and the error range is unraveled. 17Hierarchical N-Doped Porous Carbons for Zn–Air Batteries and Supercapacitors显示文摘Nitrogen-doped carbon materials with a large specific surface area,high conductivity,and adjustable microstructures have many prospects for energy-related applications.This is especially true for N-doped nanocarbons used in the electrocatalytic oxygen reduction reaction(ORR)and supercapacitors.Here,we report a low-cost,environmentally friendly,large-scale mechanochemical method of preparing N-doped porous carbons(NPCs)with hierarchical micro-mesopores and a large surface area via ball-milling polymerization followed by pyrolysis.The optimized NPC prepared at 1000°C(NPC-1000)offers excellent ORR activity with an onset potential(Eonset)and half-wave potential(E1/2)of 0.9 and 0.82 V,respectively(vs.a reversible hydrogen electrode),which are only approximately 30 mV lower than that of Pt/C.The rechargeable Zn–air battery assembled using NPC-1000 and the NiFe-layered double hydroxide as bifunctional ORR and oxygen evolution reaction electrodes offered superior cycling stability and comparable discharge performance to RuO2 and Pt/C.Moreover,the supercapacitor electrode equipped with NPC prepared at 800℃ exhibited a high specific capacity(431 F g^−1 at 10 mV s^−1),outstanding rate,performance,and excellent cycling stability in an aqueous 6-M KOH solution.This work demonstrates the potential of the mechanochemical preparation method of porous carbons,which are important for energy conversion and storage.Beibei Guo Ruguang Ma Zichuang Li Shaokui Guo Jun Luo Minghui Yang Qian Liu Tiju Thomas Jiacheng Wang 2020Nano-Micro Letters2020,12,2:1
18Degradation of SARS-CoV-2 receptor ACE2 by the E3 ubiquitin ligase Skp2 in lung epithelial cells显示文摘An unexpected observation among the COVID-19 pandemic is that smokers constituted only 1.4%–18.5%of hospitalized adults,calling for an urgent investigation to determine the role of smoking in SARS-CoV-2 infection.Here,we show that cigarette smoke extract(CSE)and carcinogen benzo(a)pyrene(BaP)increase ACE2 mRNA but trigger ACE2 protein catabolism.BaP induces an aryl hydrocarbon receptor(AhR)-dependent upregulation of the ubiquitin E3 ligase Skp2 for ACE2 ubiquitination.ACE2 in lung tissues of non-smokers is higher than in smokers,consistent with the findings that tobacco carcinogens downregulate ACE2 in mice.Tobacco carcinogens inhibit SARS-CoV-2 spike protein pseudovirions infection of the cells.Given that tobacco smoke accounts for 8 million deaths including 2.1 million cancer deaths annually and Skp2 is an oncoprotein,tobacco use should not be recommended and cessation plan should be prepared for smokers in COVID-19 pandemic.Guizhen Wang Qun Zhao Hui Zhang Fan Liang Chen Zhang Jun Wang Zhenyin Chen Ran Wu Hong Yu Beibei Sun Hua Guo Ruie Feng Kaifeng Xu Guangbiao Zhou 2021Frontiers of Medicine2021,15,2:1
19HITAC-seq enables high-throughput cost-effective sequencing of plasmids and DNA fragments with identity显示文摘DNA sequencing is vital for many aspects of biological research and diagnostics. Despite the development of second and third generation sequencing technologies, Sanger sequencing has long been the only choice when required to precisely track each sequenced plasmids or DNA fragments. Here, we report a complete set of novel barcoding and assembling system, Highly-parallel Indexed Tagmentation-reads Assembled Consensus sequencing(HITAC-seq), that could massively sequence and track the identities of each individual sequencing sample. With the cost of much less than that of single read of Sanger sequencing,HITAC-seq can generate high-quality contiguous sequences of up to 10 kilobases or longer. The capability of HITAC-seq was confirmed through large-scale sequencing of thousands of plasmid clones and hundreds of amplicon fragments using approximately 100 pg of input DNAs. Due to its long synthetic length, HITACseq was effective in detecting relatively large structural variations, as demonstrated by the identification of a~1.3 kb Copia retrotransposon insertion in the upstream of a likely maize domestication gene. 20High frequency of alternative splicing variants of the oncogene Focal Adhesion Kinase in neuroendocrine tumors of the pancreas and breast显示文摘The characteristic genetic abnormality of neuroendocrine neoplasms(NENs),a heterogeneous group of tumors found in various organs,remains to be identified.Here,based on the analysis of the splicing variants of an oncogene Focal Adhesion Kinase(FAK)in The Cancer Genome Atlas datasets that contain 9193 patients of 33 cancer subtypes,we found that Box 6/Box 7-containing FAK variants(FAK^(6/7))were observed in 7(87.5%)of 8 pancreatic neuroendocrine carcinomas and 20(11.76%)of 170 pancreatic ductal adenocarcinomas(PDACs).We tested FAK variants in 157 tumor samples collected from Chinese patients with pancreatic tumors,and found that FAK^(6/7)was positive in 34(75.6%)of 45 pancreatic NENs,19(47.5%)of 40 pancreatic solid pseudopapillary neoplasms,and 2(2.9%)of 69 PDACs.We further tested FAK splicing variants in breast neuroendocrine carcinoma(BrNECs),and found that FAK^(6/7)was positive in 14(93.3%)of 15 BrNECs but 0 in 23 non-NEC breast cancers.We explored the underlying mechanisms and found that a splicing factor serine/arginine repetitive matrix protein 4(SRRM4)was overexpressed in FAK^(6/7)-positive pancreatic tumors and breast tumors,which promoted the formation of FAK^(6/7)in cells.These results suggested that FAK^(6/7)could be a biomarker of NENs and represent a potential therapeutic target for these orphan diseases.Dawei Xie Zheng Wang Beibei Sun Liwei Qu Musheng Zeng Lin Feng Mingzhou Guo Guizhen Wang Jihui Hao Guangbiao Zhou 2023Frontiers of Medicine2023,17,5:0
