Implementation of regional Acute Stroke Care Map increases thrombolysis rates for acute ischaemic stroke in Chinese urban area in only 3 months显示文摘Background The rate of intravenous thrombolysis for acute ischaemic stroke remains low in China. We investigated whether the implementation of a citywide Acute Stroke Care Map(ASCaM)is associated with an improvement of acute stroke care quality in a Chinese urban area. Methods The ASCaM comprises 10 improvement strategies and has been implemented through a network consisting of 20 tertiary hospitals. We identified 7827 patients with ischaemic stroke admitted from April to October 2017,and 506 patients underwent thrombolysis were finally included for analysis. Results Compared with‘pre-ASCaM period’,we observed an increased rate of administration of tissue plasminogen activator within 4.5 hours(65.4% vs 54.5%;adjusted OR,1.724;95% CI 1.21 to 2.45;p=0.003)during‘ASCaM period’.In multivariate analysis models,‘ASCaM period’was associated with a significant reduction in onset-to door time(114.1±55.7 vs 135.7±58.4 min,p=0.0002)and onset-to needle time(ONT)(169.2±58.1 vs 195.6±59.3 min,p<0.0001).Yet no change was found in door-to needle time.Clinical outcomes such as symptomatic intracranial haemorrhage, favourable functional outcome(modified Rankin Scale≤2)and in-hospital mortality remained unchanged. Conclusion The implementation of ASCaM was significantly associated with increased rates of intravenous thrombolysis and shorter ONT.The ASCaM may,in proof-of principle, serve as a model to reduce treatment delay and increase thrombolysis rates in Chinese urban areas and possibly other highly populated Asian regions. | Yi Sui Jianfeng Luo Chunyao Dong Liqiang Zheng Weijin Zhao Yao Zhang Ying Xian Huaguang Zheng Bernard Yan Mark Parsons Li Ren Ying Xiao Haoyue Zhu Lijie Ren Qi Fang Yi Yang Weidong Liu Bing Xu | 2021 | Stroke & Vascular Neurology2021,6,1: | 4
Potential dual expansion of domesticated donkeys revealed by worldwide analysis on mitochondrial sequences显示文摘Molecular studies on donkey mitochondrial sequences have clearly defined two distinct maternal lineages involved in domestication. However, domestication histories of these two lineages remain enigmatic. We therefore compared several population characteristics between these two lineages based on global sampling, which included 171 sequences obtained in this study(including Middle Asian,East Asian, and African samples)plus 536 published sequences(including European,Asian, and African samples). The two lineages were clearly separated from each other based on whole mitochondrial genomes and partial non-coding displacement loop(D-loop)sequences,respectively.The Clade I lineage experienced an increase in population size more than 8000 years ago and shows a complex haplotype network. In contrast, the population size of the Clade II lineage has remained relatively constant, with a simpler haplotype network.Although the distribution of the two lineages was almost equal across the Eurasian mainland, they still presented discernible but complex geographic bias in most parts of Africa, which are known as their domestication sites.Donkeys from sub-Saharan Africa tended to descend from the Clade I lineage, whereas the Clade II lineage was dominant along the East and North coasts of Africa.Furthermore, the migration routes inferred from diversity decay suggested different expansion across China between the two lineages.Altogether, these differences indicated non-simultaneous domestication of the two lineages, which was possibly influenced by the response of pastoralists to the desertification of the Sahara and by the social expansion and trade of ancient humans in Northeast Africa, respectively. | Xi-Yao Ma Tiao Ning Adeniyi CAdeola Jie Li Ali Esmailizadeh Jacqueline KLichoti Bernard RAgwanda Jainagul Isakova Almaz AAldashev Shi-Fang Wu He-Qun Liu Najmudinov Tojiddin Abdulloevich Manilova Elena Afanasevna Khudoidodov Behruz Ibrohimovich Rahamon Akinyele Moshood Adedokun Sunday Charles Olaogun Oscar JSanke Godwin FMangbon Xi Chen Wei-Kang Yang Zhe Wang Min-Sheng Peng Sheila COmmeh Yan Li Ya-Ping Zhang | 2020 | Zoological Research2020,41,1: | 3

Genetic Evidence of Middle East Respiratory Syndrome Coronavirus(MERS-Cov) and Widespread Seroprevalence among Camels in Kenya显示文摘We describe the first genome isolation of Middle East respiratory syndrome coronavirus(MERS-CoV) in Kenya. This fatal zoonotic pathogen was first described in the Kingdom of Saudi Arabia in 2012. Epidemiological and molecular evidence revealed zoonotic transmission from camels to humans and between humans. Currently, MERS-CoV is classified by the WHO as having high pandemic potential requiring greater surveillance. Previous studies of MERS-CoV in Kenya mainly focused on site-specific and archived camel and human serum samples for antibodies. We conducted active nationwide cross-sectional surveillance of camels and humans in Kenya, targeting both nasal swabs and plasma samples from 1,163 camels and 486 humans collected from January 2016 to June 2018. A total of 792 camel plasma samples were positive by ELISA. Seroprevalence increased with age, and the highest prevalence was observed in adult camels(82.37%, 95%confidence interval(CI) 79.50–84.91). More female camels were significantly seropositive(74.28%, 95% CI 71.14–77.19)than male camels(P \ 0.001)(53.74%, 95% CI 48.48–58.90). Only 11 camel nasal swabs were positive for MERS-CoV by reverse transcription-quantitative PCR. Phylogenetic analysis of whole genome sequences showed that Kenyan MERSCoV clustered within sub-clade C2, which is associated with the African clade, but did not contain signature deletions of orf4 b in African viruses. None of the human plasma screened contained neutralizing antibodies against MERS-CoV. This study confirms the geographically widespread occurrence of MERS-CoV in Kenyan camels. Further one-health surveillance approaches in camels, wildlife, and human populations are needed. | Sheila Ommeh Wei Zhang Ali Zohaib Jing Chen Huajun Zhang Ben Hu Xing-Yi Ge Xing-Lou Yang Moses Masika Vincent Obanda Yun Luo Shan Li Cecilia Waruhiu Bei Li Yan Zhu Desterio Ouma Vincent Odendo Lin-Fa Wang Danielle E.Anderson Jacqueline Lichoti Erick Mungube Francis Gakuya Peng Zhou Kisa-Juma Ngeiywa Bing Yan Bernard Agwanda Zheng-Li Shi | 2018 | Virologica Sinica2018,33,6: | 1
Tranexamic acid for intracerebral haemorrhage within 2 hours of onset: protocol of a phase Ⅱ randomised placebo-controlled double-blind multicentre trial显示文摘Rationale Haematoma growth is common early after intracerebral haemorrhage(ICH),and is a key determinant of outcome.Tranexamic acid,a widely available antifibrinolytic agent with an excellent safety profile, may reduce haematoma growth. Methods and design Stopping intracerebral haemorrhage with tranexamic acid for hyperacute onset presentation including mobile stroke units(STOP-MSU)is a phase Ⅱ double-blind,randomised,placebo-controlled,multicentre,international investigator-led clinical trial, conducted within the estimand statistical framework.Hypothesis In patients with spontaneous ICH,treatment with tranexamic acid within 2 hours of onset will reduce haematoma expansion compared with placebo.Sample size estimates A sample size of 180 patients(90 in each arm)would be required to detect an absolute difference in the primary outcome of 20%(placebo 39%vs treatment 19%)under a two-tailed significance level of 0.05.An adaptive sample size re-estimation based on the outcomes of 144 patients will allow a possible increase to a prespecified maximum of 326 patients.Intervention Participants will receive 1 g intravenous tranexamic acid over 10 min, followed by 1 g intravenous tranexamic acid over 8 hours;or matching placebo.Primary efficacy measure The primary efficacy measure is the proportion of patients with haematoma growth by 24±6 hours, defined as either≥33%relative increase or≥6 mL absolute increase in haematoma volume between baseline and follow-up CT scan.Discussion We describe the rationale and protocol of STOP-MSU,a phase Ⅱ trial of tranexamic acid in patients with ICH within 2 hours from onset, based in participating mobile stroke units and emergency departments. | Nawaf Yassi Henry Zhao Leonid Churilov Bruce C V Campbell Teddy Wu Henry Ma Andrew Cheung Timothy Kleinig Helen Brown Philip Choi Jiann-Shing Jeng Annemarei Ranta Hao-Kuang Wang Geoffrey C Cloud Rohan Grimley Darshan Shah Neil Spratt Der-Yang Cho Karim Mahawish Lauren Sanders John Worthington Ben Clissold Atte Meretoja Vignan Yogendrakumar Mai Duy Ton Duc Phuc Dang Nguyen Thai My Phuong Huy-Thang Nguyen Chung Y Hsu Gagan Sharma Peter J Mitchell Bernard Yan Mark W Parsons Christopher Levi Geoffrey A Donnan Stephen M Davis | 2022 | Stroke & Vascular Neurology2022,7,2: | 1

Genome-wide identification and expression profiling of MYB transcription factor genes in radish(Raphanus sativus L.)显示文摘Radish(Raphanus sativus L.), an important root vegetable crop of the Brassicaceae family, has a high level of anthocyanin accumulation in its pigment root tissues. It was reported that MYB transcription factors(TFs) play vital roles in plant development and anthocyanin metabolism, and the PAP1/2 could promote expression of anthocyanin biosynthesis genes. In this study, a total of 187 radish MYB genes(Rs MYBs) were identified in the radish genome and clustered into 32 subfamilies. Among them, 159 Rs MYBs were localized on nine radish chromosomes. Interestingly, 14 Rs MYBs exhibited differential expression profiles in different taproot developmental stages among four differently colored radish lines. A number of Rs MYBs were highly expressed in the pigmented root tissues at the maturity stage, several genes including Rs MYB41, Rs MYB117, and Rs MYB132 being homologous to PAP1/2, showed high expression levels in the red skin of NAU-YH(red skin-white flesh) taproot, while Rs MYB65 and Rs MYB159 were highly expressed in the purple root skin of NAU-YZH(purple skin-red flesh), indicating that these Rs MYBs might positively regulate the process of anthocyanin accumulation in radish taproot. These results would provide valuable information for further functional characterization of Rs MYBs, and facilitate clarifying the molecular mechanism underlying anthocyanin biosynthesis in radish. | Everlyne M'mbone WIULEKE WANG Yan ZHANG Wan-ting XU Liang YING Jia-li Bernard K.KARANJA ZHU Xian-wen FAN Lian-xue Zarwali AHMADZAI LIU Li-wang | 2021 | Journal of Integrative Agriculture2021,20,1: | 1

Macrophages in Ischemic Heart Failure:Yesterday,Today, and Tomorrow显示文摘With continually improving reperfusion strategies and patient care, the overall mortality of acute myocardial infarction(AMI)has been significantly reduced during the past two decades. However, this success is a double-edged sword,as many patients surviving an AMI will progress towards ischemic heart failure(HF)over time. The pathologic causes of ischemic HF are undoubtedly multifactorial. However, the inflammatory response is considered one of the most important causes of pathological remodeling because it spans the whole process of HF development. The macrophage-mediated inflammatory response was once considered a purely harmful factor leading to pathological remodeling and HF.However, growing evidence demonstrates that multiple subgroups of macrophage exist and contribute differently to ischemic HF development.Understanding macrophage populations and how they contribute to post-MI remodeling and consequent ischemic HF is,therefore, critical to understanding and treating the disease. This review focuses on different macrophage populations that regulate post-MI cardiac injury and how immunoregulation therapy may benefit patients with ischemic HF. | Demin Liu Wenjun Yan Jingwen Huang Jianli Zhao Houston Kilby Theodore AChristopher Bernard Lopez Ling Tao Xinliang Ma Guoqiang Gu Yajing Wang | 2021 | Cardiology Discovery2021,1,2: | 0
