| 3 | CD 147 receptor is essential for TFF3-mediated signaling regulating colorectal cancer progression显示文摘Major gaps in understanding the molecular mechanisms of colorectal cancer(CRC)progression and intestinal mucosal repair have hampered therapeutic development for gastrointestinal disorders.Trefoil factor 3(TFF3)has been reported to be involved in CRC progression and intestinal mucosal repair;however,how TFF3 drives tumors to become more aggressive or metastatic and how TFF3 promotes intestinal mucosal repair are still poorly understood.Here,we found that the upregulated TFF3 in CRC predicted a worse overall survival rate.TFF3 deficiency impaired mucosal restitution and adenocarcinogenesis.CD147,a membrane protein,was identified as a binding partner for TFF3.Via binding to CD147,TFF3 enhanced CD147-CD44s interaction,resulting in signal transducer and activator of transcription 3(STAT3)activation and prostaglandin G/H synthase 2(PTGS2)expression,which were indispensable for TFF3-induced migration,proliferation,and invasion.PTGS2-derived PGE2 bound to prostaglandin E2 receptor EP4 subtype(PTGER4)and contributed to TFF3-stimulated CRC progression.Solution NMR studies of the TFF3-CD147 interaction revealed the key residues critical for TFF3 binding and the induction of PTGS2 expression.The ability of TFF3 to enhance mucosal restitution was weakened by a PTGS2 inhibitor.Blockade of TFF3-CD147 signaling using competitive inhibitory antibodies or a PTGS2 inhibitor reduced CRC lung metastasis in mice.Our findings bring strong evidence that CD147 is a novel receptor for TFF3 and PTGS2 signaling is critical for TFF3-induced mucosal restitution and CRC progression,which widens and deepens the understanding of the molecular function of trefoil factors. | Hong-Yong Cui Shi-Jie Wang Fei Song Xu Cheng Gang Nan Yu Zhao Mei-Rui Qian Xi Chen Jia-Yue Li Fen-Ling Liu Yu-Meng Zhu Ruo-Fei Tian Bin Wang Bin Wu Yang Zhang Xiu-Xuan Sun Ting Guo Xiang-Min Yang Hai Zhang Ling Li Jing Xu Hui-Jie Bian Jian-Li Jiang Zhi-Nan Chen | 2021 | Signal Transduction and Targeted Therapy2021,6,8: | 3 |
| 4 | Transcriptome analysis reveals the differential regulatory effects of red and blue light on nitrate metabolism in pakchoi(Brassica campestris L.)显示文摘Pakchoi(Brassica campestris L. ssp. chinensis) is an important leafy vegetable. Various light spectra, especially red and blue light, play vital roles in the regulation of nitrate metabolism. Information on the effects of red and blue light on nitrate metabolism at the transcriptome level in pakchoi is still limited, so this study used RNA sequencing technology to explore this molecular mechanism. Through pairwise comparisons with white LED light, 3 939 and 5 534 differentially expressed genes(DEGs) were identified under red and blue light, respectively. By Kyoto Encyclopedia of Genes and Genomes(KEGG) and Gene Ontology(GO) analyses, these unigenes were found to be involved in nitrate assimilation, plant-pathogen interaction, biosynthesis of secondary metabolites, and phenylpropanoid biosynthesis. The differential effects of light spectra on the nitrate concentration and metabolism-related enzyme activities were also confirmed at the physiological level. Several signal transduction modules, including Crys/Phys-COP1-HY5/HY5-like, were found to be involved in red and blue light-induced nitrate metabolism, and the transcript levels for this complex were consistent with the observed degree of nitrate assimilation. The expression patterns of 15 randomly selected DEGs were further validated using qPCR. | 9 | Distribution of Avian Influenza A Viruses in Poultry-Related Environment and Its Association with Human Infection in Henan, 2016 to 2017显示文摘Objective To survey avian influenza A viruses(AIVs) in the environment and explore the reasons for the surge in human H7 N9 cases.Methods A total of 1,045 samples were collected from routine surveillance on poultry-related environments and 307 samples from human H7 N9 cases-exposed environments in Henan from 2016 to2017. The nucleic acids of influenza A(Flu A), H5, H7, and H9 subtypes were detected by real-time polymerase chain reaction.Results A total of 27 H7 N9 cases were confirmed in Henan from 2016 to 2017, 24 had a history of live poultry exposure, and 15 had H7 N9 virus detected in the related live poultry markets(LPMs). About 96%(264/275) Flu A positive-environmental samples were from LPMs. H9 was the main AIV subtype(10.05%) from routine surveillance sites with only 1 H7-positive sample, whereas 21.17% samples were H7-positive in H7 N9 cases-exposed environments. Samples from H7 N9 cases-exposed LPMs(47.56%)had much higher AIVs positive rates than those from routine surveillance sites(12.34%). The H7+H9 combination of mixed infection was 78.18%(43/55) of H7-positive samples and 41.34%(43/104) of H9-positive samples.Conclusion The contamination status of AIVs in poultry-related environments is closely associated with the incidence of human infection caused by AIVs. | 18 | NRN1 and CAT Gene Polymorphisms, Complex Noise, and Lifestyles interactively Affect the Risk of Noise-induced Hearing Loss显示文摘Objective The effects of interactions between genetic and environmental factors on the noise-induced hearing loss(NIHL)are still unclear.This study aimed to assess interactions among gene polymorphisms,noise metrics,and lifestyles on the risk of NIHL.Methods A case-control study was conducted using 307 patients with NIHL and 307 matched healthy individuals from five manufacturing industries.General demographic data,lifestyle details,and noise exposure levels were recorded.The Kompetitive allele-specific polymerase chain reaction(KASP)was used to analyze the genotypes of 18 SNPs.Results GMDR model demonstrated a relevant interaction between NRN1 rs3805789 and CAT rs7943316(P=0.0107).Subjects with T allele of rs3805789 or T allele of rs7943316 had higher risks of NIHL than those with the SNP pair of rs3805789-CC and rs7943316-AA(P<0.05).There was an interaction among rs3805789,rs7943316,and kurtosis(P=0.0010).Subjects exposed to complex noise and carrying both rs3805789-CT and rs7943316-TT or rs3805789-CT/TT and rs7943316-AA had higher risks of NIHL than those exposed to steady noise and carrying both rs3805789-CC and rs7943316-AA(P<0.05).The best six-locus model involving NRN1 rs3805789,CAT rs7943316,smoking,video volume,physical exercise,and working pressure for the risk of NIHL was found to be the interaction(P=0.0010).An interaction was also found among smoking,video volume,physical exercise,working pressure,and kurtosis(P=0.0107).Conclusion Concurrence of NRN1 and CAT constitutes a genetic risk factor for NIHL.Complex noise exposure significantly increases the risk of NIHL in subjects with a high genetic risk score.Interactions between genes and lifestyles as well as noise metrics and lifestyles affect the risk of NIHL. | LIU Shuang Yan SONG Wei Qin XIN Jia Rui LI Zheng LEI Song CHEN Ying Qi ZHAO Tian Yu WANG Hai Yan XU Liang Wen ZHANG Mei Bian HONG Yu YANG Lei | 2021 | Biomedical and Environmental Sciences2021,34,9: | 0 |
