2Extrapolation Model of High Temperature Creep for 12Cr1MoV Steel显示文摘The creep curves of heat resistant steel 12Cr1MoV was investigated. by constant stress creep tests. The creep curves of constant stress creep are quite different from that of constant load creep. The original θ- Concept Project procedure can be validated to successfully describe the constant stress creep curves but not constant load creep. A modified θprocedure was developed. This approach can successfully described both constant stress creep and constant load creep curves. 3Quantum interference effect in the charge transport through single-molecule benzene dithiol junction at room temperature:An experimental investigation显示文摘The quantum interference effect in the charge transport through single-phenyl molecules received intensive interests from theory but remained as an experimental challenge. In this paper, we investigated the charge transport through single-molecule benzene dithiol(BDT) junction with different connectivities using mechanically controllable break junction(MCBJ) technique. By further improving the mechanical stability and the electronic measuring component of the MCBJ set-up, we obtained the conductance histograms of BDT molecules(BDTs) from the statistical analysis of conductance-distance traces without data selection. By tuning the connectivity, the conductance of BDTs is determined to be 10^(-1.2)G_0, 10^(-2.2) G_0 and 10^(-1.0)G_0 for para, meta, and ortho connectivity, following the trend that ortho-BDT >para-BDT >meta-BDT. Furthermore, the displacements of the junctions followed the trend that para > meta > ortho, suggesting the charge transport through the molecules via the gold-thiol bond.The different trends between conductance and displacement for different connectivities suggests the presence of destructive quantum interference effect on meta-BDT, which provides the experimental evidence for the quantum interference effect through single-phenyl molecular junctions.Guogang Yang Hao Wu Junying Wei Jueting Zheng Zhaobin Chen Junyang Liu Jia Shi Yang Yang Wenjing Hong 2018Chinese Chemical Letters2018,29,1:2
4Tyrosine hydroxylase-positive cells and dopaminergic neuronal function in human embryonic stem cells: An electrophysiological validation显示文摘BACKGROUND: Induced differentiation strategies and cytochemical properties of human embryonic stem cells (hESCs) have been investigated. However, the electrophysiological functions of tyrosine hydroxylase (TH)-positive cells derived from hESCs remain unclear. OBJECTIVE: To investigate the differentiation efficiency of TH-positive cells from hESCs in vitro using modified four-step culture methods, including embryoid body formation, and to examine the functional characteristics of the differentiated TH-positive cells using electrophysiological techniques. DESIGN, TIME AND SETTING: Neuroelectrophysiology was performed at the Reproductive Medicine Center and Stem Cell Research Center, Peking University Third Hospital, and the Neuroscience Research Institute and Department of Neurobiology, Peking University, from September 2004 to August 2008. MATERIALS: The hESC line, PKU-1.1, a monoclonal cell line derived from a pre-implantation human blastocyst in the Reproductive Medical Center of Peking University Third Hospital. The patch clamp recording system was provided by the Neuroscience Research Institute and Department of Neurobiology, Peking University. METHODS: The hESC line was induced to differentiate into TH-positive cells in vitro using a modified four-step culture method, including the formation of embryoid body, as well as the presence of sonic hedgehog and fibroblast growth factor 8. The cell karyotype was assessed by G-banding karyotype analysis techniques and specific markers were detected immunocytochemically. Whole-cell configuration was obtained after obtaining a tight seal of over 1 GΩ. Ionic currents were detected by holding the cells at -70 mV and stepping to test voltages between -80 and 40 mV in 10-mV increments in voltage-clamp configuration. MAIN OUTCOME MEASURES: We measured the cell karyotype, specific cell markers, and the electrophysiological properties of the voltage-gated ion channels on the cell membrane of TH-positive dopaminergic cells differentiated from our hESCs line in vitro. RESULTS: The differentiated cells had a consistent appearance, and the majority of cells (> 90%) expressed TH and β-tubulion, as well as the neural progenitor marker, nestin. Cell karyotype analysis demonstrated that all of the hESCs had a stable and normal karyotype (46, XX) after dif-ferentiation. In addition, patch clamp recording showed that the 10 recorded TH-positive cells exhibited a fast inward current when the test voltage depolarized to -30 mV, and a delayed outward current when the test voltage depolarized to -10 mV. The peak of inward current was obtained at voltage between -10 mV and 0 mV, while the peak of outward current was obtained at 40 mV. The average peak of inward current density was (-50.05 ± 15.50) pA/pF, and the average peak of outward current density was (41.98 ± 13.55) pA/pF. CONCLUSION: More than 90% of the differentiated hESC-derived cells induced by the modified four-step culture method exhibit dopaminergic neuronal properties, including general electrophysiological functional properties, such as functional potassium and sodium channels.Tianran Song 2010Neural Regeneration Research2010,5,3:1
5Revisiting an ancient inorganic aggregation-induced emission system:An enlightenment to clusteroluminescence显示文摘Organic and inorganic clusteroluminescence have attracted great attention while the underlying mechanisms is still not well understood.Here,we employed a series of ancient inorganic complexes platinocyanides with aggregation-induced emission property to elucidate the mechanism of clusteroluminescence including how does the chromophore form and how does the solid structures influence the luminescence behaviors.The results indicate that the isolated platinocyanide cannot work as a chromophore to emit visible light,while their clusterization at aggregate state can trigger the d-orbitals coupling of the platinum atoms to facilitate the electron exchange and delocalization to form a new chromophore to emit visible light.Furthermore,the counter ions and H2O ligands help to rigidify the three-dimensional network structure of the platinocyanides to suppress the excited state nonradiative decay,resulting in the high quantum yield of up to 96%.This work fundamentally helps understanding both the organic and inorganic clusteroluminescence phenomenon.Zheng Zhao Zaiyu Wang Javad Tavakoli Guogang Shan Jianyu Zhang Chen Peng Yu Xiong Xuepeng Zhang Tsz Shing Cheung Youhong Tang Bolong Huang Zhaoxun Yu Jacky W.Y.Lam Ben Zhong Tang 2021Aggregate2021,2,2:1

8Construction of Forest and Grass Industry Economic Belt Using Land Resources in Road Side Slope显示文摘The construction of expressway and high-speed rail is at the sacrifice of the soil ecological environment. It brings about much damaged land and bare slope. It is necessary to restore vegetation and rebuild landscape. In the design of target plant community and configuration plants,it is required to select as far as possible plants that can blossom and bear fruit or pods. Then seeds of these plants can be harvested,processed and sold,and applied for other projects,so as to develop into a complete forest and grass seed industry chain. This not only reasonably utilizes land resources,but also provides a new approach to the problem of insufficient fine forest seed sources in China. This paper discussed the use of land resources in road side slope and forest and grass seed selection and breeding technology,and elaborated the industrialized development paths for forest and grass seed industries in road side slope. Finally,it arrived at following recommendations.( i) The construction of forest and grass industry economic belt using land resources in road side slope can realize maximum land value. 13Identification of a Long-Chain Fatty Acid Elongase from Nannochloropsis sp. Involved in the Biosynthesis of Fatty Acids by Heterologous Expression in Saccharomyces cerevisiae显示文摘The marine microalga Nannochloropsis sp.contains various elongases and desaturases that are critical for biosynthesis of polyunsaturated fatty acids.A full-length cDNA encoding a long-chain fatty acid elongase,named Ns FAE,was cloned from Nannochloropsis sp..The open reading frame of Ns FAE(GenBank accession no.MF680548)consisted of 1068 bp and encoded a predicted protein of 355 amino acids with molecular mass 38.8 k Da.The deduced polypeptide showed 43%–44%identity to fatty acyl elongases from other algae.RT-PCR experiments indicated that the Ns FAE gene exhibited the highest expression in Nannochloropsis sp.at 72 h(i.e.,during the third growth stage)and the expression was significantly lower in the other four growth stages.Plasmid p Ns FAE-CRISPR and a recombinant DNA fragment(ADH1p-Ns FAE-CYCt)were transformed into Saccharomyces cerevisiae strain BY4742 using the CRISPR-Cas system.Yeast transformants containing Ns FAE produced three fatty acids not normally present in wild-type BY4742-linoleic acid,linolenic acid and eicosadienoic acid-indicating that Ns FAE encodes a functional elongase enzyme.GUO Minrui CHEN Guogang CHEN Jiluan ZHENG Minggang 2019Journal of Ocean University of China2019,18,5:1

16Migration routes of the endangered Oriental Stork(Ciconia boyciana)from Xingkai Lake,China,and their repeatability as revealed by GPS tracking显示文摘The Oriental Stork(Ciconia boyciana)is listed as'Endangered'on the International Union for the Conservation of Nature(IUCN)Red List of Threatened Species and is classified as a first category nationally protected bird species in China.Understanding this species'seasonal movements and migration will facilitate effective conservation to promote its population.We tagged 27 Oriental Stork nestlings at Xingkai Lake on the Sanjiang Plain in Heilongjiang Province,China,used GPS tracking to follow them over the periods of 2014-2017 and 2019-2022,and confirmed their detailed migratory routes using the spatial analysis function of ArcGIS 10.7.We discovered four migration routes during autumn migration:one common long-distance migration route in which the storks migrated along the coastline of Bohai Bay to the middle and lower reaches of the Yangtze River for wintering,one short-distance migration route in which the storks wintered in Bohai Bay and two other migration routes in which the storks crossed the Bohai Strait around the Yellow River and wintered in South Korea.There were no significant differences in the number of migration days,residence days,migration distances,number of stopovers and average number of days spent at stopover sites between the autumn and spring migrations(P>0.05).However,the storks migrated significantly faster in spring than in autumn(P=0.03).The same individuals did not exhibit a high degree of repetition in their migration timing and route selection in either autumn or spring migration.Even storks from the same nest exhibited considerable between-individual variation in their migration routes.Some important stopover sites were identified,especially in the Bohai Rim Region and on the Songnen Plain,and we further explored the current conservation status at these two important sites.Overall,our results contribute to the understanding of the annual migration,dispersal and protection status of the endangered Oriental Stork and provide a scientific basis for conservation decisions and the development of action plans for this species.Zeyu Yang Lixia Chen Ru Jia Hongying Xu Yihua Wang Xuelei Wei Dongping Liu Huajin Liu Yulin Liu Peiyu Yang Guogang Zhang 2023Avian Research2023,14,2:0

18Insights from coastline survey in Shandong Province,China显示文摘The coastline,as the boundary between land and sea,is shaped by both natural factors and human activities.Based on concrete practice from coastline survey in Shandong Province,this study discusses the applicability of the local standard Specification for Coastline Survey(DB 37/T 3588-2019).The coastline classification system takes into account the coastline characteristics and marine management requirements in Shandong Province.To be specific,the coastline with natural beach morphology and ecological function is involved,and its delimitation method is innovated.The coastline delimitation of sandy beach,aquaculture pond and salt pan follows the trace line of the mean high water springs(MHWS),but the coastline location of sandy beach can also be at the base of cliffs or at the foot of seawalls and roads considering the dynamic changes of the beach in different seasons and the integrity of the beach topography.The delineation of estuarial coastline is to achieve the complete coastline,and should not be used for the statistics of natural coastline retention rate.At present,the review of survey results has been completed.With the promulgation of survey results next,the formulation of relevant mechanisms and policies should promptly be put on the agenda,including regular investigation and data updating,natural coastline assessment,natural coastline requisition-compensation,and coastline protection planning.XIU Chun HUO Suxia WANG Guogang DUAN Haiqin CHEN Shengtao YAO Haiyan 2022Global Geology2022,25,2:0
19Enhancing the deep-red/near-infrared fluorescence of higher rylene diimides via the chalcogen-annulation strategy显示文摘Deep-red/near-infrared fluorescence is highly suitable for bioimaging owing to its ability to deeply penetrate tissues,organs,and live animals.However,developing organic fluorophores with high deep-red/near-infrared fluorescence quantum yield(Φ_(FL))and fluorescent brightness remain a significant challenge owing to the energy gap law.Herein,we developed a straightforward and effective chalcogen-annulation strategy by introducing O,S and Se into the bay region of TDI and QDI fluorophores,realizing the increase ofΦFLand fluorescent brightness up to 10 times.To our best knowledge,this study potentially stands as the pioneering instance showcasing the anti-heavy-atom effect of chalcogens,and the absoluteΦFL(93%)and fluorescent brightness(128,200 cm^(-1)mol^(-1)L)of Se-TDI is among top deep-red/near-infrared organic fluorophores currently available.The femtosecond transient absorption(fs-TA)measurements show the absence of obvious changes of the excited state lifetime after the introduction of chalcogens in TDI and QDI fluorophores,indicating that intersystem crossing(ISC)can be neglected in TDI and QDI fluorophores.Theoretical calculations further reveal the chalcogen-annulation strategy increase the radiative rates and reduce the reorganization energy of several accepting modes at the ground state in TDI fluorophores,leading to the suppression of internal conversion(IC)processes.Our chalcogen-annulation strategy,which effectively increases the Φ_(FL)and restricts the IC processes,while remaining unaffected by the heavy-atom effect,offers novel insights and theoretical support for the design and synthesis of deep-red/near-infrared organic fluorophores with high Φ_(FL)and fluorescent brightness.Kai Chen Xiao Chen Ke Hu Yilun Zhao Yujian Liu Guogang Liu Jinquan Chen Wei Jiang Zhigang Shuai Da-Hui Qu Zhaohui Wang 2024Science China Chemistry2024,67,4:0
