1Endoscopic Surgery versus External Ventricular Drainage Surgery for Severe Intraventricular Hemorrhage显示文摘The efficacy and applied value of endoscopic hematoma evacuation vs. external ventricular drainage (EVD) in the treatment of severe ventficular hemorrhage (IVH) were explored and compared.From Jan.2015 to Dec.2016,the clinical data of 42 cases of IVH were retrospectively analyzed,including 18 patients undergoing endoscopic hematoma evacuation (group A),and 24patients receiving EVD (group B).The hematoma clearance rate was calculated by 3D Slicer software,and complications and outcomes were compared between the two groups.There were no significant differences in age,sex and Graeb score between groups A and B (P>0.05).The hematoma clearance rate was 70.81%±27.64% in group A and 48.72%±36.58% in group B with a statistically significant difference (P<0.05).The operative time in groups A and B was 72.45±25.26 min and 28.54±15.27min,respectively (P<0.05).The Glasgow Coma Scale (GCS) score increased from 9.28±2.72 at baseline to 11.83±2.91 at 1 week postoperatively in group A,and from 8.25±2.62 at baseline to 10.79±4.12 at 1 week postoperatively in group B (P<0.05).The length of hospital stay was 12.67±5.97 days in group A and 17.33±8.91 days in group B with a statistically significant difference (P<0.05).The GOS scores at 6 months after surgery were 3.83±1.12 in group A,and 2.75±1.23 in group B (P<0.05). 4Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. 5The Intervention Effect of Rosiglitozone in Ovarian Fibrosis of PCOS Rats显示文摘Objective To explore the Intervention effect of Rosiglitozone in ovarian fibrosis of PCOS rats.Methods 60 female SD rats were randomly divided into 3 groups:control group,model group and treatment group.The model and treatment groups were established by subcutaneous injection of DHEA,while the treatment group was given RGZ.The serum hormone values,pathohistology of ovarian structure of rats,ovarian ultrastructure and the expressions of TGF-β 1 and CTGF were detected.Results The PCOS model was established successfully.The expression intensity of TGF-β 1 and CTGF in Oocytes of the PCOS groups was 9.545±2.954 and 9.665±2.400,respectively and was significantly higher than that of the control group 6.636±2.264 and 7.036±2.133;after treatment with rosiglitazone,the expression was significantly decreased 6.980±2.421 and 6.642±2.721 as compared with that of the model group (P<0.05,P<0.001).The values in serum of the PCOS groups were 3.749±2.054 and 0.265±0.129,and 1.914±1.801 and 0.096±0.088 in the control group which had statistically significant difference (P<0.05,P<0.001).After treatment with rosiglitazone,the values were 2.3100±1.825 and 0.112±0.187 and were significantly different with those of the model group (P<0.05,P<0.001).Conclusion TGF-β 1 and CTGF play an important role in the development of ovary fibrosis in PCOS.However,RGZ may postpone the development of fibrosis by decreasing the levels of TGF-β 1 and CTGF.MIAO Zhu Lin GUO Liang WANG Yong Xia CUI Rong YANG Ning HUANG Mi Qiong QIN Wei Bing CHEN Jin LI Hong Mei WANG Zi Neng WEI Xiang Cai 2012Biomedical and Environmental Sciences2012,25,1:14

7Orthogonal Test Design for Optimization of the Extraction of Flavonid from the Fructus Gardeniae显示文摘Objective It is imperative to provide some consistent experimental results for the extraction of flavonid from Fructus Gardeniae.Methods The key extraction parameters that influenced the yield of flavonid from Fructus Gardeniae were optimized by employing an orthogonal experiment [L9(3)4],including the ratio of buffer solution (Na2B4O7·10H2O) to raw material,concentration of Fructus Gardeniae in extracting solution,extraction time and pH of buffer solution.An UV/Vis detector was used to perform the qualitative and quantitative analyses of the extracted flavonid with the using of the standard sample.Results The maximum extraction yield of the crude extract was 5.0533 (mg/g) after 20 min when the mass ratio of Na2B4O7·10H2O to raw material was 0.4%,the concentration of Fructus Gardeniae in the extraction solution was 1/12 (g/mL),and pH of buffer solution was 4.5.The positive reactions to the Molish and HCl‐Mg tests suggested that the extracted compound was flavonoid,and FTIR measurements also identified the presence of flavonoid in the extracts.Conclusion This work is expected to provide a basis for further research,development,and utilization of Fructus gardenia in flavonid extraction.CAI Ding Jian SHU Qing XU Bao Quan PENG Li Mei HE Yan 2011Biomedical and Environmental Sciences2011,24,6:12
8Grey matter volume abnormalities in patients with bipolar I depressive disorder and unipolar depressive disorder:a voxelbased morphometry study显示文摘Bipolar disorder and unipolar depressive disorder(UD) may be different in brain structure. In the present study,we performed voxel-based morphometry(VBM) to quantify the grey matter volumes in 23 patients with bipolar I depressive disorder(BP1) and 23 patients with UD,and 23 age-,gender-,and educationmatched healthy controls(HCs) using magnetic resonance imaging. We found that compared with the HC and UD groups,the BP1 group showed reduced grey matter volumes in the right inferior frontal gyrus and middle cingulate gyrus,while the UD group showed reduced volume in the right inferior frontal gyrus compared to HCs. In addition,correlation analyses revealed that the grey matter volumes of these regions were negatively correlated with the Hamilton depression rating scores. 11Psychological status and diabetes-related distress of Chinese type 1 diabetes patients in Jiangsu province, China显示文摘We sought to assess the psychological status and diabetes-related distress of Chinese type 1 diabetes patients in Jiangsu province,China.According to the World Health Organization criteria,42 patients with type 1 diabetes were enrolled in the study and assessed with the Self-Rating Anxiety Scale(SAS),Self-Rating Depression Scale(SDS)and Diabetes Distress Scale(DDS).All data were tabulated and statistical analyses were performed.The study summarized cases of 42 patients with type 1 diabetes,including 17 males and 25 females with a mean age of 23±12 years and a mean duration of diabetes of 7±6 years.Compared to the Chinese normative data,the SAS standard score was significantly higher,whereas SDS standard score had no statistical significance.The SAS standard score was most highly correlated with diabetes duration(γ=0.547,P=0.011).Additionally,19.5%of the patients had moderate or even severe diabetes-related distress and 21.4%had moderate or even severe emotional burden while 26.2%had regimen-related distress.Multiple stepwise regression analysis showed that the mean correlation between DDS and the four domains was high,particularly the emotional burden domain(estimatedβ=0.363,P<0.001)and regimen-related distress domain(estimatedβ=0.356,P<0.001).The correlation between SAS and DDS was positive(estimatedβ=0.039,P=0.027).In conclusion,the results showed the importance of psychological aspects in Chinese individuals with type 1 diabetes.Screening and treatment of psychological aspects may result in better adherence and increased quality of life for patients with diabetes.Lulu Kong Yun Cai Gang Mei Rong Gu Xiaojiao Zhang Yao Qin Ya Cai Yan Li Hongwen Zhou Mei Zhang Tao Yang 2013The Journal of Biomedical Research2013,27,5:9

13Concurrent chemoradiotherapy with weekly docetaxel versus cisplatin in the treatment of locoregionally advanced nasopharyngeal carcinoma:a propensity score-matched analysis显示文摘Background:Promising efficacy and manageable toxicity of docetaxel-based concurrent chemoradiotherapy(CCRT)were reported in head and neck cancer.In addition,the effect of CCRT in combination with cisplatin and/or 5-fluorouracil on both locoregionally advanced and metastatic/recurrent nasopharyngeal carcinoma(NPC)was verified.However,CCRT with docetaxel for locoregionally advanced NPC are not well studied.This study aimed to compare effectiveness and toxicities of CCRT with weekly docetaxel versus tri-weekly cisplatin for locoregionally advanced NPC.Methods:Clinical data of patients with locoregionally advanced NPC newly diagnosed between January 2010 and December 2014 receiving CCRT with either weekly docetaxel(15 mg/m2)or tri-weekly cisplatin(80-100 mg/m2)were reviewed.Propensity score matching at a 1:1 ratio was performed to balance baseline characteristics.Adverse events and survival were compared between the two groups.Results:A total of 962 patients were included as the whole cohort,and 448 patients were matched and were regarded as the matched cohort.The median follow-up duration was 48 months for the whole cohort.The 3-year nodal recurrence-free survival rate was significantly increased for patients treated with docetaxel in both the whole(hazard ratio[HR]=0.37,95%confidence interval[CI]0.19-0.72,P=0.030)and matched cohorts(HR=0.33,95%CI 0.14-0.79,P=0.023).However,no significant differences were observed in overall survival,local recurrence-free survival,and distant metastasis-free survival between the two groups in both cohorts.Significantly higher rates of grade 3 radiodermatitis(6.7%vs.1.8%,P=0.001),mucositis(74.5%vs.37.9%,P<0.001),and leucopenia(2.2%vs.11.6%,P<0.001)were observed in the docetaxel group,but any grade of renal injury(1.8%vs.15.1%,P<0.001),vomiting(18.8%vs.88.3%,P<0.001),and ALT elevation(19.2%vs.31.3%,P=0.027)were more common in the cisplatin group.Conclusions:CCRT with weekly low-dose docetaxel is an effective and tolerable therapeutic regimen for locally advanced NPC.It provides a survival benefit mainly by improving the control of regional lymph node metastases,especially for patients with low pretreatment EBV DNA levels.Jun-Fang Liao Qun Zhang Xiao-Jing Du Mei Lan Shan Liu Yun-Fei Xia Xiu-Yu Cai Wei Luo 2019Cancer Communications2019,39,1:9
14Exosome-based small RNA delivery:Progress and prospects显示文摘RNA interfering(RNAi), mediated by small interfering RNAs and microRNAs, is currently one of the most promising tools of gene therapy. Small RNAs are capable of inducing specific post-transcriptional gene silencing, providing a potentially effective platform for the treatment of a wide array of diseases. However, similar to other nucleic acid-based drugs,the major hurdle of RNAi therapy is lack of efficient and non-immunogenic delivery vehicles. Currently, viruses, synthetic polymers, and lipid-based carriers are among the most widely studied vehicles for small RNA delivery. However, many drawbacks are reported to be associated with these delivery vehicles. There is a pressing need to replace them with more efficient and better-tolerated approaches. Exosomes secreted from the endocytic compartment of live cells, are a subtype of endogenous extracellular vesicles that transfer genetic and biochemical information among different cells, thus playing an important role in cellcell communication. Recently, accumulating attention has been focused on harnessing exosomes as nanaocarriers for small RNAs delivery. Due to their natural role in shuttling endogenous nucleic acid in our body, exosomes may exhibit higher delivery efficiency, lower immunogenicity, and better compatibility than existing foreign RNA carriers. Importantly,exosomes own intrinsic homing capacity that can guide small RNAs across natural membranous barriers. Moreover, such a capacity can be further improved by adding appropriate targeting moieties. 16The role of transcriptional factor D-site-binding protein in circadian CCL2 gene expression in anti-Thy1 nephritis显示文摘Mesangial proliferative glomerulonephritis(MsPGN)is an inflammatory disease,but both the nature of disease progression and its regulation remain unclear.In the present study,we monitored the course of anti-Thy1 nephritis from days 1 to 5 and established gene expression profiles at each time point using microarrays to explore the development of inflammation.According to the gene expression profiles,macrophage infiltration(triggered by CCL2 activation)was evident on day 1 and enhanced inflammation over the next few days.We screened for genes with expression levels similar to CCL2 and found that the upregulation of the circadian gene albumin D-site-binding protein(DBP)was involved in CCL2 activation in mesangial cells.More importantly,CCL2 expression showed oscillatory changes similar to DBP,and DBP induced peak CCL2 expression at 16:00 a clock on day 1 in the anti-Thy1 nephritis model.We knocked down DBP through transfection with a small interfering RNA(siRNA)and used RNA sequencing to identify the DBP-regulated TNF-α-CCL2 pathway.We performed chromatin immunoprecipitation sequencing(ChIP-Seq)and the dual luciferase assay to show that DBP bound to the TRIM55 promoter,regulating gene expression and in turn controlling the TNF-α-CCL2 pathway.In conclusion,DBP-regulated circadian CCL2 expression by the TRIM55-TNF pathway in injured mesangial cells at an early stage,which promoted macrophage recruitment and in turn triggered infiltration and inflammation in a model of anti-Thy1 nephritis.Yang Lu Yan Mei Lei Chen Lingling Wu Xu Wang Yingjie Zhang Bo Fu Xizhao Chen Yuansheng Xie Guangyan Cai Xueyuan Bai Qinggang Li Xiangmei Chen 2019Cellular & Molecular Immunology2019,16,9:7
