2Comparative effect of allopolyploidy on transposable element composition and gene expression between Gossypium hirsutum and its two diploid progenitors显示文摘An allopolyploidization event formed allotetraploid Gossypium species from an A-genome diploid species and a D-genome diploid species. To explore the responses of transposable elements(TEs) to allopolyploidy, we assembled parallel TE datasets from G. hirsutum, G. arboreum and G. raimondii and analyzed the TE types and the effects of TEs on orthologous gene expression in the three Gossypium genomes.Gypsy was the most abundant TE type and most TEs were located $500 bp from genes in all three genomes. In G. hirsutum, 35.6% of genes harbored TE insertions, whereas insertions were more frequent in G. arboreum and G. raimondii. G. hirsutum had the highest proportion of uniquely matching 24-nt small interfering RNAs(siRNAs) that targeted TEs. TEs,particularly those targeted by 24-nt siRNAs, were associated with reduced gene expression, but the effect of TEs on orthologous gene expression varied substantially among species. Orthologous gene expression levels in G. hirsutum were intermediate between those of G. arboreum and G. raimondii, which did not experience TE expansion or reduction resulting from allopolyploidization. 9A Novel Chinese Polar Knowledge Repository Based on Polar Data-Sharing Ontology显示文摘In order to archive and utilize the information from Chinese polar expeditions to the greatest extent, we design a novel knowledge repository, in which an automatic query model based on neural networks is proposed and a data call trigger is established to keep data consistent between polar data-sharing platforms. And in this repository, anybody can make contributions to the repository by creating or updating entries with version control and an authority control mechanism. In this paper, the data sources,data processes and network structure of this repository are described, and the keywords extraction and decision support operation are detailed. 13A novel inactivated whole-cell Pseudomonas aeruginosa vaccine that acts through the cGAS-STING pathway显示文摘Pseudomonas aeruginosa infection continues to be a major threat to global public health,and new safe and efficacious vaccines are needed for prevention of infections caused by P.aeruginosa.X-ray irradiation has been used to prepare whole-cell inactivated vaccines against P.aeruginosa infection.However,the immunological mechanisms of X-ray-inactivated vaccines are still unclear and require further investigation.Our previous study found that an X-ray-inactivated whole-cell vaccine could provide protection against P.aeruginosa by boosting T cells.The aim of the present study was to further explore the immunological mechanisms of the vaccine.Herein,P.aeruginosa PAO1,a widely used laboratory strain,was utilized to prepare the vaccine,and we found nucleic acids and 8-hydroxyguanosine in the supernatant of X-ray-inactivated PAO1(XPa).By detecting CD86,CD80,and MHCII expression,we found that XPa fostered dentritic cell(DC)maturation by detecting.XPa stimulated the cGAS-STING pathway as well as Toll-like receptors in DCs in vitro,and DC finally underwent apoptosis and pyroptosis after XPa stimulation.In addition,DC stimulated by XPa induced CD8+T-cell proliferation in vitro and generated immunologic memory in vivo.Moreover,XPa vaccination induced both Th1 and Th2 cytokine responses in mice and reduced the level of inflammatory factors during infection.XPa protected mice in pneumonia models from infection with PAO1 or multidrug-resistant clinical isolate W9.Chronic obstructive pulmonary disease(COPD)mice immunized with XPa could resist PAO1 infection.Therefore,a new mechanism of an X-ray-inactivated whole-cell vaccine against P.aeruginosa infection was discovered in this study.Cuicui Ma Xiao Ma Boguang Jiang Hailong Pan Xueyuan Liao Li Zhang Wenfang Li Yingjie Luo Zhixue Shen Xingjun Cheng Mao Lian Zhenling Wang 2021Signal Transduction and Targeted Therapy2021,6,11:0
14Inactivated SARS-CoV-2 booster vaccine enhanced immune responses in patients with chronic liver diseases显示文摘Chronic liver disease(CLD)entails elevated risk of COVID-19 severity and mortality.The effectiveness of the booster dose of inactivated SARS-CoV-2 vaccine in stimulating antibody response in CLD patients is unclear.Therefore,we conducted a cross-sectional study involving 237 adult CLD patients and 170 healthy controls(HC)to analyze neutralizing antibodies(NAbs)against SARS-CoV-2 prototype and BA.4/5 variant,anti-receptor binding domain(RBD)IgG,and total anti-SARS-CoV-2 antibodies.Serum levels of the total anti-SARS-CoV-2 antibodies,anti-RBD IgG and inhibition efficacy of NAbs were significantly elevated in CLD patients after the booster dose compared with the pre-booster dose,but were relatively lower than those of HCs.Induced humoral responses decreased over time after booster vaccination.The neutralization efficiency of the serum against BA.4/5 increased but remained below the inhibition threshold.All four SARS-CoV-2 antibodies,including total anti-SARS-CoV-2 antibodies,anti-RBD IgG and NAbs against prototype and BA.4/5,were lower in patients with severe CLD than those with non-severe CLD.After booster shot,age and time after the last vaccine were the risk factors for seropositivity of NAb against BA.4/5 in CLD patients.Additionally,white blood cell counts and hepatitis B core antibodies were the protective factors,and severe liver disease was the risk factor associated with seropositivity of total anti-SARS-CoV-2 antibodies.Overall,our data uncovered that antibody responses were improved in CLD patients and peaked at 120 days after the booster vaccines.All antibodies excepting total anti-SARS-CoV-2 antibodies declined after peak.CLD patients exhibited impaired immunologic responses to vaccination and weakened NAbs against BA.4/5,which hindered the protective effect of the booster shot against Omicron prevalence.Cellular immune responses should be further evaluated to determine the optimal vaccine regimen for CLD patients.Yongmei Liu Jianhua Lu Haoting Zhan Wenfang Yuan Xiaomeng Li Haiyan Kang Haolong Li Yongliang Chen Linlin Cheng Xingli Sun Haojie Zheng Wei Wang Erhei Dai Yongzhe Li 2023Virologica Sinica2023,38,5:0
