1A sparsity adaptive compressed signal reconstruction based on sensing dictionary显示文摘Signal reconstruction is a significantly important theoretical issue for compressed sensing.Considering the situation of signal reconstruction with unknown sparsity,the conventional signal reconstruction algorithms usually perform low accuracy.In this work,a sparsity adaptive signal reconstruction algorithm using sensing dictionary is proposed to achieve a lower reconstruction error.The sparsity estimation method is combined with the construction of the support set based on sensing dictionary.Using the adaptive sparsity method,an iterative signal reconstruction algorithm is proposed.The sufficient conditions for the exact signal reconstruction of the algorithm also is proved by theory.According to a series of simulations,the results show that the proposed method has higher precision compared with other state-of-the-art signal reconstruction algorithms especially in a high compression ratio scenarios.SHEN Zhiyuan WANG Qianqian CHENG Xinmiao 2021Journal of Systems Engineering and Electronics2021,32,6:1
2Novel phthalimides regulating PD-1/PD-L1 interaction as potential immunotherapy agents显示文摘Programmed cell death 1(PD-1)/programmed cell death ligand 1(PD-L1)have emerged as one of the most promising immune checkpoint targets for cancer immunotherapy.Despite the inherent advantages of small-molecule inhibitors over antibodies,the discovery of small-molecule inhibitors has fallen behind that of antibody drugs.Based on docking studies between small molecule inhibitor and PD-L1 protein,changing the chemical linker of inhibitor from a flexible chain to an aromatic ring may improve its binding capacity to PD-L1 protein,which was not reported before.A series of novel phthalimide derivatives from structure-based rational design was synthesized.P39 was identified as the best inhibitor with promising activity,which not only inhibited PD-1/PD-L1 interaction(IC_(50)=8.9 nmol/L),but also enhanced killing efficacy of immune cells on cancer cells.Co-crystal data demonstrated that P39 induced the dimerization of PD-L1 proteins,thereby blocking the binding of PD-1/PD-L1.Moreover,P39 exhibited a favorable safety profile with a LD_(50)>5000 mg/kg and showed significant in vivo antitumor activity through promoting CD8^(+)T cell activation.All these data suggest that P39 acts as a promising small chemical inhibitor against the PD-1/PD-L1 axis and has the potential to improve the immunotherapy efficacy of T-cells.Chengliang Sun Yao Cheng Xiaojia Liu Gefei Wang Wenjian Min Xiao Wang Kai Yuan Yi Hou Jiaxing Li Haolin Zhang Haojie Dong Liping Wang Chenguang Lou Yanze Sun Xinmiao Yu Hongbin Deng Yibei Xiao Peng Yang 2022Acta Pharmaceutica Sinica B2022,12,12:0
3Response of Salmonella enterica serovar Typhimurium to alginate oligosaccharides fermented with fecal inoculum:integrated transcriptomic and metabolomic analyses显示文摘Alginate oligosaccharides(AOS),extracted from marine brown algae,are a common functional feed additive;however,it remains unclear whether they modulate the gut microbiota and microbial metabolites.The response of Salmonella enterica serovar Typhimurium,a common poultry pathogen,to AOS fermented with chicken fecal inocula was investigated using metabolomic and transcriptomic analyses.Single-strain cultivation tests showed that AOS did not directly inhibit the growth of S.Typhimurium.However,when AOS were fermented by chicken fecal microbiota,the supernatant of fermented AOS(F-AOS)exhibited remarkable antibacterial activity against S.Typhimurium,decreasing the abundance ratio of S.Typhimurium in the fecal microbiota from 18.94 to 2.94%.Transcriptomic analyses showed that the 855 diferentially expressed genes induced by F-AOS were mainly enriched in porphyrin and chlorophyll metabolism,oxidative phosphorylation,and Salmonella infection-related pathways.RT-qPCR confrmed that F-AOS downregulated key genes involved in fagellar assembly and the type III secretory system of S.Typhimurium,indicating metabolites in F-AOS can infuence the growth and metabolism of S.Typhimurium.Metabolomic analyses showed that 205 microbial metabolites were signifcantly altered in F-AOS.Among them,the increase in indolelactic acid and 3-indolepropionic acid levels were further confrmed using HPLC.This study provides a new perspective for the application of AOS as a feed additive against pathogenic intestinal bacteria.Jiaying Cheng Mengshi Xiao Xinmiao Ren Francesco Secundo Ying Yu Shihao Nan Weimiao Chen Changliang Zhu Qing Kong Youtao Huang Xiaodan Fu Haijin Mou 2023Marine Life Science & Technology2023,5,2:0
