1Rice SPL10 positively regulates trichome development through expression of HL6 and auxin-related genes显示文摘Trichomes function in plant defenses against biotic and abiotic stresses;examination of glabrous lines,which lack trichomes,has revealed key aspects of trichome development and function.Tests of allelism in 51 glabrous rice(Oryza sativa)accessions collected worldwide identified OsSPL10 and OsWOX3B as regulators of trichome development in rice.Here,we report that OsSPL10 acts as a transcriptional regulator controlling trichome development.Haplotype and transient expression analyses revealed that variation in the approximately 700-bp OsSPL10 promoter region is the primary cause of the glabrous phenotype in the indica cultivar WD-17993.Disruption of OsSPL10 by genome editing decreased leaf trichome density and length in the NIL-HL6 background.Plants with genotype OsSPL10^(WD-17993)/HL6 generated by crossing WD-17993 with NIL-HL6 also had fewer trichomes in the glumes.HAIRY LEAF6(HL6)encodes another transcription factor that regulates trichome initiation and elongation,and OsSPL10 directly binds to the HL6 promoter to regulate its expression.Moreover,the transcript levels of auxin-related genes,such as OsYUCCA5 and OsPIN-FORMED1b,were altered in OsSPL10 overexpression and RNAi transgenic lines.Feeding tests using locusts(Locusta migratoria)demonstrated that non-glandular trichomes affect feeding by this herbivore.Our findings provide a molecular framework for trichome development and an ecological perspective on trichome functions.Jinjie Li Bo Tang Yingxiu Li Chenguang Li Minjie Guo Haiyang Chen Shichen Han Jin Li Qijin Lou Wenqiang Sun Peng Wang Haifeng Guo Wei Ye Zhanying Zhang Hongliang Zhang Sibin Yu Long Zhang Zichao Li 2021Journal of Integrative Plant Biology2021,63,8:2
2Causes of Recurrence of Rice Chilo suppressalis( Walker) in Longyou County and Prevention and Control Measures显示文摘In recent years,the occurrence of rice Chilo suppressalis( Walker) in Longyou County of Zhejiang Province has increased year by year and has become aggravated. This poses a great threat to the safety of rice production. 3Enhanced monopole transition strength from the cluster decay of ^(13)C显示文摘The inelastic excitations and cluster decay of ^(13)C have been measured using the reaction,9Be(^(13)C, ^(13)C* →~9Be + α)~9Be. We observe strong excitation to the 14.3-MeV(1/2-) resonant state from the cluster-decay channel, leading to an enhanced monopole matrix element of(6.3 ± 0.6) fm^2. This large cluster-related monopole strength is a clear indication of the cluster-structure domination of this state and is consistent with the recent prediction of the orthogonality condition model(OCM). 4Novel phthalimides regulating PD-1/PD-L1 interaction as potential immunotherapy agents显示文摘Programmed cell death 1(PD-1)/programmed cell death ligand 1(PD-L1)have emerged as one of the most promising immune checkpoint targets for cancer immunotherapy.Despite the inherent advantages of small-molecule inhibitors over antibodies,the discovery of small-molecule inhibitors has fallen behind that of antibody drugs.Based on docking studies between small molecule inhibitor and PD-L1 protein,changing the chemical linker of inhibitor from a flexible chain to an aromatic ring may improve its binding capacity to PD-L1 protein,which was not reported before.A series of novel phthalimide derivatives from structure-based rational design was synthesized.P39 was identified as the best inhibitor with promising activity,which not only inhibited PD-1/PD-L1 interaction(IC_(50)=8.9 nmol/L),but also enhanced killing efficacy of immune cells on cancer cells.Co-crystal data demonstrated that P39 induced the dimerization of PD-L1 proteins,thereby blocking the binding of PD-1/PD-L1.Moreover,P39 exhibited a favorable safety profile with a LD_(50)>5000 mg/kg and showed significant in vivo antitumor activity through promoting CD8^(+)T cell activation.All these data suggest that P39 acts as a promising small chemical inhibitor against the PD-1/PD-L1 axis and has the potential to improve the immunotherapy efficacy of T-cells.Chengliang Sun Yao Cheng Xiaojia Liu Gefei Wang Wenjian Min Xiao Wang Kai Yuan Yi Hou Jiaxing Li Haolin Zhang Haojie Dong Liping Wang Chenguang Lou Yanze Sun Xinmiao Yu Hongbin Deng Yibei Xiao Peng Yang 2022Acta Pharmaceutica Sinica B2022,12,12:0
