1Anti-diabetic effects and mechanisms of action of a Chinese herbal medicine preparation JQ-R in vitro and in diabetic KKAy mice显示文摘Refined-JQ(JQ-R) is a mixture of refined extracts from Coptis chinensis(Ranunculaceae),Astragalus membranaceus(Leguminosae) and Lonicera japonica(Caprifoliaceae), the three major herbs of Jin Qi-Jiang Tang tablet, a traditional Chinese medicine(TCM) formula. The mechanisms by which JQ-R regulates glucose metabolism and improves insulin sensitivity were studied in type 2 diabetic KK^(Ay)mice and insulin-resistant L6 myotubes. To investigate the mechanisms by which JQ-R improves insulin sensitivity, a model of insulin-resistant cells induced with palmitic acid(PA) was established in L6 myotubes. Glucose uptake and expression of factors involved in insulin signaling, stress, and inflammatory pathways were detected by immunoblotting. JQ-R showed beneficial effects on glucose homeostasis and insulin resistance in a euglycemic clamp experiment and decreased fasting insulin levels in diabetic KK^(Ay)mice. JQ-R also improved the plasma lipid profiles. JQ-R directly increased the activity of superoxide dismutase(SOD) and decreased malondialdehyde(MDA) as well as inducible nitric oxide synthase(iNOS) levels in insulin-resistant L6 cells, and elevated the insulin-stimulated glucose uptake with upregulated phosphorylation of AKT. The phosphorylation levels of nuclear factor kappa B(NF-κB p65), inhibitor of NF-κB(IκBα), c-Jun N-terminal kinase(JNK1/2) and extracellular-signal-regulated kinases(ERK1/2) were also changed after JQ-R treatment compared with the control group. Together these findings suggest that JQ-R improved glucose and lipid metabolism in diabetic KK^(Ay)mice. JQ-R directly enhanced insulin-stimulated glucose uptake in insulin-resistant myotubes with improved insulin signalling and inflammatory response and oxidative stress. 6Depositional environment of shale in Wufeng and Longmaxi Formations, Sichuan Basin显示文摘Formation of organic-rich shale was controlled by depositional processes and environment condition.According to petrology,mineralogy,fossil composition and sedimentary structure,seven lithofacies of shale were identified in the Wufeng Formation to Longmaxi Formation,e.g.,siliceous shale,silty shale,argillaceous shale,calcareous shale,shell-bearing argillaceous limestone/calcareous mudstone,siltstonefine sandstone and bentonite.For shale in the Wufeng and Longmaxi Formations,the depositional mode was not only the suspension deposition under the low-energy hydrodynamic condition,but also the biogenic deposition,the storm deposition and the bottom current deposition indicated by large amount of siliceous biological fossils,biological detritus with size graded structure,silt laminations,lenticular bedding,ripple cross-bedding and other sedimentary structures.Trace element analysis suggested that the siliceous shale in theWufeng Formation and the lower part of Longmaxi Formation was developed in the oxygen-dysoxic/anoxic depositional environment,which was rich in organic matters;then due to influence of the sea level gradually decline and bottom current,the silty and argillaceous shales were formed,and the oxygen-dysoxic/anoxic depositional environment was destroyed,gradually leading to the worse preservation condition of organic matter.Jianhua Zhao Zhijun Jin Zhenkui Jin Xin Wen Yikai Geng Caina Yan Haikuan Nie 2017Petroleum Research2017,2,3:2

17Delay-dependent passive control of linear systems with nonlinear perturbation显示文摘The problem of delay-dependent passive control of a class of linear systems with nonlinear perturbation and time-varying delay in states is studied. The main idea aims at designing a state-feedback controller such that for a time-varying delay in states, the linear system with nonlinear perturbation remains robustly stable and passive. In the system, the delay is time-varying. And the derivation of delay has the maximum and minimum value. The time-varying nonlinear perturbation is allowed to be norm-bounded. Using the effective linear matrix inequality methodology, the sufficient condition is primarily obtained for the system to have robust stability and passivity. Subsequently the existent condition of a state feedback controller is given, and the explicit expression of the controller is obtained by means of the solution of linear matrix inequalities (LMIs). 18Establishment of a selective evaluation method for DPP4 inhibitors based on recombinant human DPP8 and DPP9 proteins显示文摘Dipeptidyl peptidase 4(DPP4)is recognised as an attractive anti-diabetic drug target,and several DPP4 inhibitors are already on the market.As members of the same gene family,dipeptidyl peptidase 8(DPP8)and dipeptidyl peptidase 9(DPP9)share high sequence and structural homology as well as functional activity with DPP4.However,the inhibition of their activities was reported to cause severe toxicities.Thus,the development of DPP4 inhibitors that do not have DPP8 and DPP9 inhibitory activity is critical for safe anti-diabetic therapy.To achieve this goal,we established a selective evaluation method for DPP4 inhibitors based on recombinant human DPP8 and DPP9 proteins expressed by Rosetta cells.In this method,we used purified recombinant 120 kDa DPP8 or DPP9 protein from the Rosetta expression system.The optimum concentrations of the recombinant DPP8 and DPP9 proteins were 30 ng/mL and 20 ng/mL,respectively,and the corresponding concentrations of their substrates were both 0.2 mmol/L.This method was highly reproducible and reliable for the evaluation of the DPP8 and DPP9 selectivity for DPP4 inhibitor candidates,which would provide valuable guidance in the development of safe DPP4 inhibitors.Jinglong Liu Yi Huan Caina Li Minzhi Liu Zhufang Shen 2014Acta Pharmaceutica Sinica B2014,4,2:1
19FoxJ1 inhibits African swine fever virus replication and viral S273R protein decreases the expression of FoxJ1 to impair its antiviral effect显示文摘African swine fever(ASF)is a highly pathogenic swine infectious disease that affects domestic pigs and wild boar,which is caused by the African swine fever virus(ASFV).ASF has caused huge economic losses to the pig industry and seriously threatens global food security and livestock health.To date,there is no safe and effective commercial vaccine against ASF.Unveiling the underlying mechanisms of ASFV-host interplay is critical for developing effective vaccines and drugs against ASFV.In the present study,RNA-sequencing,RT-qPCR and Western blotting analysis revealed that the transcriptional and protein levels of the host factor FoxJ1 were significantly down-regulated in primary porcine alveolar macrophages(PAMs)infected by ASFV.RT-qPCR analysis showed that overexpression of FoxJ1 upregulated the transcription of type I interferon and interferon stimulating genes(ISGs)induced by poly(dA:dT).FoxJ1 revealed a function to positively regulate innate immune response,therefore,suppressing the replication of ASFV.In addition,Western blotting analysis indicated that FoxJ1 degraded ASFV MGF505-2R and E165R proteins through autophagy pathway.Meanwhile,RT-qPCR and Western blotting analysis showed that ASFV S273R inhibited the expression of FoxJ1.Altogether,we determined that FoxJ1 plays an antiviral role against ASFV replication,and ASFV protein impairs FoxJ1-mediated antiviral effect by degradation of FoxJ1.Our findings provide new insights into the antiviral function of FoxJ1,which might help design antiviral drugs or vaccines against ASFV infection.Caina Ma Shasha Li Fan Yang Weijun Cao Huisheng Liu Tao Feng Keshan Zhang Zixiang Zhu Xiangtao Liu Yonghao Hu Haixue Zheng 2022Virologica Sinica2022,37,3:1
