1The internalization pathway,metabolic fate and biological effect of superparamagnetic iron oxide nanoparticles in the macrophage-like RAW264.7 cell显示文摘The potential applications of superparamagnetic iron oxide nanoparticles (SPIONs) in several nanomedical fields have attracted intense interest based on the cell-nano interaction. However, the mechanisms underlying cell uptake, the intracellular trail, final fate and the biological effects of SPIONs have not yet been clearly elucidated. Here, we showed that multiple endocytic pathways were involved in the internalization process of SPIONs in the RAW264.7 macrophage. The internalized SPIONs were biocompatible and used three different metabolic pathways: The SPIONs were distributed to daughter cells during mitosis; they were degraded in the lysosome and free iron was released into the intracellular iron metabolic pool; and, the intact SPIONs were potentially exocytosed out of the cells. The internalized SPIONs did not induce cell damage but affected iron metabolism, inducing the upregulation of ferritin light chain at both the mRNA and protein levels and ferroportin 1 at the mRNA level. 2Kv1.3 channel blockade enhances the phagocytic function of RAW264.7 macrophages显示文摘This study aimed to comprehend the largely unknown role of voltage-gated potassium channel 1.3(Kv1.3) in the phagocytic function of macrophages. We found that blocking of the Kv1.3 channel with 100 pmol L-1 Stichodactyla helianthus neurotoxin(Sh K) enhanced the phagocytic capacities of both resting and lipopolysaccharide(LPS)-stimulated RAW264.7 macrophages in the chicken erythrocyte system. In the fluorescein isothiocyanate(FITC)-labeled Escherichia coli k-12 system, Sh K increased the phagocytic capacities of resting RAW264.7 cells, but not of the LPS-stimulated cells, as LPS alone stimulated almost saturated phagocytosis of the macrophages. Sh K increased the nitric oxide(NO) production in LPS-activated cells, but not in resting RAW264.7 cells. There was no effect of Sh K alone on the cytokine secretions in resting RAW264.7 cells, but it suppressed IL-1β secretion in LPS-stimulated RAW264.7 cells. At a concentration of 100 pmol L-1, Sh K did not affect the viability of the tested cells. Kv1.3 was expressed in RAW264.7 cells; this expression was downregulated by LPS, but significantly upregulated by disrupting caveolin-dependent endocytosis with filipin III. In addition, cytochalasin D, an inhibitor of actin polymerization, did not affect the Kv1.3 expression. Thus, blocking of the Kv1.3 channel enhances the phagocytic capacity and NO production of this cell line. 3Development and Application of a Regional Land Subsidence Model for the Plain of Tianjin显示文摘In this paper, a coupled numerical groundwater and land subsidence model was developed for the Tianjin Plain. The model was employed to investigate the groundwater resources and their changes over the last decade, and to further predict the changing patterns of the groundwater level and associated land subsidence in future. First, according to the regional hydrogeology, the simulation area was defined with an area of 10.6×103 km2, which was divided into six aquifer units. A coupled groundwater and land subsidence numerical model was built by using Modflow2005 and the land subsidence simulation module SUB(subsidence and aquifer-system compaction), in which the groundwater flow was modeled as three-dimensional unsteady flow and the land subsidence simulation was based on one-dimensional consolidation theory. The model was then calibrated by using the groundwater level contour lines, hydrographs, and land subsidence hydrographs over the period of 1998–2008. In addition, groundwater balance analysis of the simulation period indicated that under multi-year groundwater withdrawal condition the cross-flow recharge, compression release, and lateral boundary inflow contributed 44.43%, 32.14%, and 21.88% to the deep aquifer recharge, respectively. Finally, the model was applied to predict the changing patterns of the groundwater levels and the associated variations in land subsidence under the control of groundwater exploitation after implementation of the south-to-north water diversion project. The simulation results demonstrated that the groundwater level may gradually increase year by year with an decrease in the groundwater withdrawal; and the land in dominated land subsidence regions including the urban area, Dagang, Hangu, Jinghai, Wuqing, and Jinnan, may rebound at an average rate of 2–3 mm/a, and the land subsidence rate in the other regions may decrease.Yali Cui Chen Su Jingli Shao Yabin Wang Xiaoyuan Cao 2014Journal of Earth Science2014,25,3:2
4Influence of solid retention time on sludge characteristics and effluent quality in immersed membrane bioreactor显示文摘Here the effect of solid retention time(SRT) on the concentration of the mixed liquor suspend solid(MLSS),the sludge characteristics,the content of extra-cellular polymeric substances(EPS),the viscosity of mixed liquor and effluent quality in the immersed membrane bioreactor(IMBR) was investi-gated.The results indicate that the increase of the EPS content is the main reason for the increase of mixed liquor viscosity,the former is positively correlated with the latter(R2 = 0.9751).The size distribution profile of particles in the mixed liquor presents double-peak shape at SRT more than 30 days.The filtration resistance of membrane in IMBR is mainly caused by the tiny particles and the viscosity of the mixed liquor.In this study,the extension of SRT can hardly affect the removal efficiency of Chemical Oxygen Demand(COD) and NH+4―N,and when SRT is below 30 days,silt density index(SDI15) is less than 3,the effluent can be deeply treated by using reverse osmosis system(RO) or nano-filtration system(NF).The method of controlling SRT is put forward by analyzing the relationship between SRT and the minimum generation-time of dominant bacteria(at the maximum specific growth rate under the operation temperature).CAO ZhanPing ZHANG JingLi ZHANG HongWei 2008Chinese Science Bulletin2008,53,24:2

6Pertussis toxin–induced inflammatory response exacerbates intracerebral haemorrhage and ischaemic stroke in mice显示文摘Background Stroke is a devastating disease,including intracerebral haemorrhage(ICH)and ischaemic stroke.Emerging evidences indicate that systemic inflammatory cascades after stroke contribute to brain damage.However,the direct effects and features of systemic inflammation on brain injury,especially comparing between ischaemic and haemorrhagic stroke,are still obscure.Methods Pertussis toxin(PT)was used to build a pro-inflammatory milieu after ICH and ischaemic stroke in mouse model.The neurodeficits,stroke lesion,immune response and blood–brain barrier(BBB)destruction were assessed.Results In ICH mouse model,PT-induced systemic inflammation exacerbated neurological deficits,and enlarged haemorrhage lesion and perihaematomal oedema.We also found promoted leucocyte infiltration and inflammatory cytokine release into the brain after PT treatment.Moreover,the integrity of the BBB was further disrupted after receiving PT.Furthermore,we demonstrated that PT enhanced brain inflammation and aggravated stroke severity in middle cerebral artery occlusion mouse model.Conclusions Our results suggest that PT increases inflammatory response that exacerbates brain injury after ICH or ischaemic stroke in mouse model.Ming Zou Yan Feng Yuwhen Xiu Yan Li Ying Zhang Junwan Fan Haowen Li Jingli Cao Wenyan He Wei-Na Jin 2022Stroke & Vascular Neurology2022,7,1:1

8Design of an Index System for Deep Groundwater Management Efficiency Evaluation: A Case Study in Tianjin City, China显示文摘An effective evaluation system can provide specific and practical suggestions to the deep groundwater management. But such kind of evaluation system has not been proposed in China. In this study, an evaluation index system is specifically developed to evaluate deep groundwater management efficiency. It is composed of three first-level indicators(law enforcement capability, management ability, and management effectiveness) and eleven second-level indicators. The second-level indicators include seven mandatory indicators and four optional indicators. Piecewise linear function is used to normalize the quantitative indicators, and expert scoring method and questionnaire survey method are used to normalize the qualitative indicators. Then a comprehensive indicator weighting evaluation method is used to evaluate the first-level indicators and the target topic. A case study is carried out to evaluate deep groundwater management efficiency in Tianjin City. According to the evaluation score in each period, the management efficiency of every district in Tianjin City gradually improved. The overall evaluation score in the early deep groundwater extraction period is 0.12. After a series of deep groundwater protection efforts, this score reached to 0.61 in 2007, and met the regulation criteria. The evaluation results also showed that the further groundwater management efforts in Tianjin City should be focused on building a dynamic database to collect comprehensive deep well-log data; and on a reasonable design and distribution of the groundwater monitoring network. It demonstrated that the index system is suitable to locate the deficiencies of current groundwater management systems and to guide further improvements. 9Intellectual disability-related chromatin regulator Brpfl affects electrophysiological feature of hippocampal neurons and related pathways显示文摘Intellectual disability(ID)is a prevalent neurodevelopmental disorder,affecting 1%-2%of children.Recent researches revealed that haploinsufficiency of Brpf1 could result in ID.Hippocampus plays important roles in learning and memory which are impaired in people with ID.To study the roles of Brpf1 in hippocampus,we investigated the morphology,electrophysiology and molecular mechanism in both cultured mouse hippocampal neurons by infecting with AAV-shBrpf1 virus and in mouse model by injecting into hippocampal CA1 area with AAV-shBrpf1 virus.The results showed that knockdown of Brpf1(at about 50%)reduced the mEPSC frequency and amplitude of cultured hippocampal neurons,though it had no effects on the morphology of these neurons;reduced expression of Brpf1 also downregulated the mRNA level of Syt4,Grik3 and Neurod6,which involved in neuronal excitability;knockdown of Brpf1 in mouse hippocampal CA1 region led to increased expression of Cxcl10,which was related with the death and apoptosis of hippocampal neurons.Conclusively,our study found that reduced expression of Brpf1 affected the mEPSC frequency and ampltude of hippocampal neurons as well as dysregulate molecular pathways,which shed new light on Brpfl's role in ID.Xian Weirvei Yuan Xiangshan Cao Jingli Zhou Gruomin You Linya 2021解剖学杂志2021,44,S01:0
10Brpf1 regulates the function of MGE-derived GABAergic interneurons显示文摘Intellectual disability is a broad-spectrum neurodevelopmental disorder characterized by marked impairment of intelligence and adaptive function.Impaired GABA neurotransmission is associated with mental disorders such as epilepsy and mental retardation,and patients with BRPF1 mutations were mentally retarded and often accompanied by epilepsy.Our research is to investigate the role of Brpf1 in the development and function of GABAergic interneurons.We used mouse primary in vitro cultured GABAergic interneurons derived from medial ganglionic eminence(MGE)and utilized AAV-shBrpf1 virus to knockdown Brpf1.The results showed that Brpf1 knockdown led to increased threshold and decreased number of evoked action potentials,and a downward trend of the amplitude and frequency of mIPSC.However,Brpf1 knockdown had lttle effect on the differentiation of GABAergic interneurons as revealed by parvalbumin and somatostatin staining.Moreover,little influence on the morphology was observed as shown by MAP2 and Golgi stain.Finally,it was found that most of the up-regulated genes are involved in protein binding,ubiquitination,and inflammatory response by RNA Seq.Jingli Cao Weiwei Xian Guomin Zhou Linya You 2021解剖学杂志2021,44,S01:0
