1Wx^lv、the Ancestral Allele of Rice Waxy Gene显示文摘In rice grains,the Waxy (Wx) gene is responsible for the synthesis of amylose,the most important determinant for eating and cooking quality.The effects of several Wx alleles on amylose content and the taste of cooked rice have been elucidated.However,the relationship between artificial selection and the evolution of various Wx alleles as well as their distribution remain unclear.Here we report the identification of an ancestral allele,Wx^lv,which dramatically affects the mouthfeel of rice grains by modulating the size of amylose molecules.We demonstrated that WF originated directly from wild rice,and the three major Wx alleles in cultivated rice (Wx^b,Wx^a,and Wx^in) differentiated after the substitution of one base pair at the functional sites.These data indicate that the Wx^lv allele played an important role in artificial selection and domestication.The findings also shed light on the evolution of various Wx alleles,which have greatly contributed to improving the eating and cooking quality of rice.Changquan Zhang Jihui Zhu Shengjie Chen Xiaolei Fan Qianfeng Li Yan Lu Min Wang Hengxiu Yu Chuandeng Yi Shuzhu Tang Minghong Gu Qiaoquan Liu 2019Molecular Plant2019,12,8:22
2A rare Waxy allele coordinately improves rice eating and cooking quality and grain transparency显示文摘In rice(Oryza sativa), amylose content(AC) is the major factor that determines eating and cooking quality(ECQ). The diversity in AC is largely attributed to natural allelic variation at the Waxy(Wx)locus. Here we identified a rare Wx allele, Wx^(mw) ,which combines a favorable AC, improved ECQ and grain transparency. Based on a phylogenetic analysis of Wx genomic sequences from 370 rice accessions, we speculated that Wx^(mw) may have derived from recombination between two important natural Wx alleles, Wx^(in) and Wx^(b). We validated the effects of Wx^(mw) on rice grain quality using both transgenic lines and near-isogenic lines(NILs). When introgressed into the japonica Nipponbare(NIP) background, Wx^(mw) resulted in a moderate AC that was intermediate between that of NILs carrying the Wx^(b)allele and NILs with the Wx^(mp) allele. Notably, mature grains of NILs fixed for Wx^(mw) had an improved transparent endosperm relative to soft rice. Further, we introduced Wx^(mw) into a high-yielding japonica cultivar via molecular marker-assisted selection: the introgressed lines exhibited clear improvements in ECQ and endosperm transparency. 3Genetic analysis and gene cloning of a triangular hull 1 (tri1) mutant in rice (Oryza sativa L.)显示文摘Grain shape and size are two key factors that determine rice yield and quality. In the present study, a rice triangular hull mutant (tri1) was obtained from the progeny of japonica rice variety Taipei 309 treated with 60Co γ-rays. Compared to the wild type, the tri1 mutant presents a triangular hull, and exhibits an increase in grain thickness and protein content, but with a slight decrease in plant height and grain weight. Genetic analysis indicated that the mutant phenotype was controlled by a recessive nuclear gene which is stably inherited. Using a map-based cloning strategy, we fine-mapped tri1 to a 47-kb region between the molecular markers CHR0122 and CHR0127 on the long arm of chromosome 1, and showed that it co-segregates with the molecular marker CHR0119. According to the rice genome sequence annotation there are six predicated genes within the mapped region. Sequencing analysis of the mutant and the wild type indicated that there was a deletion of an A nucleotide in exon 3 of the OsMADS32 gene, which could result in a downstream frameshift mutation and premature termination of the predicted polypeptide. Both semi-quantitative and real-time RT-PCR analyses showed that this gene expressed highly in young inflorescences, while expressed at very low levels in other tissues. These results implied that the OsMADS32 gene could be a candidate of TRI1. 4Generation of selectable marker-free soft transgenic rice with transparent kernels by downregulation of SSSⅡ-2显示文摘The amylose content(AC)of rice endosperm starch varies from 0 to 35%,and is associated with rice cooking and eating quality.Soft rice has low AC,generally between 6%and 15%,and its eating quality is high whether it is consumed hot or cold.However,the appearance quality of current soft rice cultivars needs to be improved,especially opaque endosperm.Conventional genetic engineering has improved some agronomic traits of soft rice varieties,but not endosperm appearance.In the present study,a RNAi construct of the soluble starch synthase II-2(SSSII-2)and the hygromycin phosphotransferase(HPT)gene were introduced into an elite japonica rice variety,Kangtiaowuyunjing(KWY8)by co-transformation.Several selectable marker-free(SMF)transgenic lines were obtained,and SSSII-2 expression was significantly downregulated in selected transgenic lines,resulting in lower AC of the endosperm.The physicochemical properties of the transgenic rice kernels,including gel consistency(GC)and rapid visco analyzer(RVA)profile,differed significantly from those of wild-type rice and were similar to those of a soft rice variety,Nanjing 46(NJ46).These findings indicate that the cooking,eating,and processing qualities of transgenic rice are comparable to those of NJ46.However,the transgenic rice endosperm retained a transparent appearance under low-moisture conditions.Thus,SMF SSSII-2 RNAi rice provides a resource for breeding soft rice with transparent endosperm.Zhihao Xu Meimei Yu Youwei Yin Cenwen Zhu Wen Ji Changquan Zhang Qianfeng Li Honggen Zhang Shuzu Tang Hengxiu Yu Qiaoquan Liu 2020The Crop Journal2020,8,1:2

10Impact of regio-isomeric monochlorinated end groups on packing mode,miscibility,and photovoltaic performance of asymmetric selenophene-fused M-series acceptors显示文摘Optimal bulk-heterojunction(BHJ)morphology is crucial for efficient charge transport and good photovoltaic performance in organic solar cells(OSCs).Yet,the correlation between chemical structures of nonfullerene acceptors(NFAs)and molecular interaction in the BHJ blends remains opaque.Herein,we study three isomeric NFAs referred to as MQ1-x(x=β,y,or 8)that shared an asymmetric selenophene-fused heteroheptacene backbone end-capped by two monochlorinated end groups.Remarkably,miscibility between the polymer donor of PM6 and MQ1-x successively elevates as the chlorine atoms move fromβ-,to y-,to 8-position of terminals.Combined with the varied molecular crystallinity of these NFAs,diverse BHJ morphologies are observed in their blend films.As a result,the MQ1-8-based devices present the highest PCE of 12.08%owing to the efficient charge dissociation and transport induced by the compact molecular packing and optimal BHJ morphology.Our investigation provides a new insight in the material design that has a good balance in molecular packing and film morphology for high-performance OSCs.Ruochuan Liao Changquan Tang Yunlong Ma Qingdong Zheng 2023Chinese Chemical Letters2023,34,12:0
11Enhancing Photovoltaic Performance of Ladder-Type Heteroarene-Based Electron Acceptors by Modulating Molecular Packing显示文摘Comprehensive Summary,The development of novel building blocks with sp3-hybridized-carbon-free conjugated skeletons is important to further advance and enrich nonfullerene acceptors(NFAs),but this remains a challenge due to the lack of strategies to effectively modulate the aggregation behavior of resulting NFAs.Herein,two novel nitrogen-bridged octacyclic ladder-type heteroarenes end-capped with thiophene rings(BTPS)or selenophene rings(BTPSe)are designed and synthesized as the donor cores for constructing NFAs(MQX-2 and MQX-4).It is found that replacing the sulfur atoms(MQX-2)at the outer positions of the heteroarene core with selenium atoms(MQX-4)can effectively modulate the molecular packing mode of the NFAs.The incorporation of selenium atoms induces stronger O···Se noncovalent interaction than O···S,thus promoting the formation of mixed H/J-type aggregates in MQX-4.Benefiting from more electron hopping channels,MQX-4 exhibits higher electron transport(more than 1-fold enhancement)and photovoltaic properties compared to MQX-2,which forms only H-type aggregates.Hongxin Tao Changquan Tang Yunlong Ma Shu-Quan Zhang Ruochuan Liao Qisheng Tu Qingdong Zheng 2023Chinese Journal of Chemistry2023,41,17:0
