1Quality Markers of Traditional Chinese Medicine:Concept,Progress,and Perspective显示文摘As the most important complementary medication against a variety of diseases,traditional Chinese medicines(TCMs)have been extensively applied over thousands of years.Current quality control of herbal medicines,however,is in great dispute.Unlike chemical drugs,which have clear and validated quality standards,the content of only one(or a few)compounds of many herbs and preparations is currently assessed as an indicator of quality,even though the assessed compound(s)is neither closely associated with the efficacy nor representative of the medicine as a whole.Based on the clinical use,compatibility of multiple component prescriptions,and manufacturing process of TCM,the new concept of a TCM quality marker that was proposed in previous work is discussed further here.In addition,practical technological approaches are described for the qualitative analysis and quantification of TCMs including herbs,processed products,and preparations,which lead to the discovery and identification of specific chemicals as quality markers and new quality control patterns.The progress that has been made in TCM quality control is also addressed.This work provides useful information for the quality control of herbal medicines in the future.Yazhuo Li Ying Xie Yufei He Wenbin Hou Maoliang Liao Changxiao Liu 2019Engineering2019,5,5:22
2An Improved Radar Emitter Recognition Method Based on Dezert-Smarandache Theory显示文摘The parameters of radar emitter are fast changing in the current complicated electromagnetic environment, and the radar emitter recognition rate which used single sensor method cannot satisfied. To solve the problem, an improved radar emitter recognition method based on Dezert-Smarandache theory is proposed, which can improve proportional conflict redistribution rule to solve fuzzy and conflicting information from radar emitter. 3Potential synergic mechanism of Wutou-Gancao herb-pair by inhibiting efflux transporter P-glycoprotein显示文摘Wutou-Gancao herb-pair is extensively used to attenuate the toxicity and enhance the efficacy of aconite.In this study,potential synergic mechanism of the herb pair was investigated by utilizing multiple ap-proaches.In silico and in vitro Caco-2 cell models were applied to study the potential binding mode of bioactive ingredients existing in liquorice with P-glycoprotein(P-gp),as well as the inhibition effects on P-gp.Additionally,anti-inflammatory activity of aconitine(AC)combined with active ingredients of liquorice,as well as pharmacokinetic patterns of AC after co-administration was investigated.Anti-inflammatory effect of AC(1 mg/kg)in rats was enhanced in combination with bioactive ingredients of liquorice(10 mg/kg).In the meanwhile,the exposure of AC in vivo was altered,in terms of Cmax and AUC.For instance,the Cmax and AUC were increased to 1.9 and 1.3 folds,respectively,when used in combination with liquiritigenin.The in silico study revealed the potential binding mode with outward facing conformation of P-gp.The resulting data obtained from transport of rhodamine-123(Rh-123)across Caco-2 cell monolayer further indicated that the function of P-gp was inhibited by chemicals in liquorice.The synergic effect was therefore proposed to be attributed to inhibition of P-gp by liquorice since AC has been demonstrated to be the substrate of P-gp.The resuls revealed that potential synergic mechanism of Wutou-Gancao herb-pair by inhibiting function of key efflux transporter P-gp to enhance the exposure of AC in systematic circulation,and further the anti-inflammatory effect,which helps clarify the compatibility rationale of these two herbs.Yufei He Zihong Wei Ying Xie Xiulin Yi Yong Zeng Yazhuo Li Changxiao Liu 2020Journal of Pharmaceutical Analysis2020,10,2:5
4Research and development in drug innovation: reflections from the 2013 bioeconomy conference in China, lessons learned and future perspectives显示文摘The enormous progress biotechnology,bioinformatics and nanotechnology made in recent years provides opportunities and scientific framework for development of biomedicine and constitutes a paradigm shift in pharmaceutical R&D and drug innovation.By analyzing the data and related information at R&D level over the past decades,developmental tendency and R&D patterns were summarized.We found that a growing number of biologics in the pipeline of pharma companies with successful products already in the market though,small molecular entities have primarily dominated drug innovation.Additionally,small/medium size companies will continue to play a key role in the development of small molecule drugs and biologics in a multi-channel integrated process.More importantly,modern and effective R&D strategies in biomedicine development to predict and evaluate efficacy and/or safety of 21st century therapeutics are urgently needed.To face new challenges,developmental strategies were proposed,in terms of molecular targeted medicine,generic drugs,new drug delivery system and protein-based drugs.Under the current circumstances,interdisciplinary cooperation mode and policy related to drug innovation in China were deeply discussed as well.Changxiao Liu Panayiotis P.Constantinides Yazhuo Li 2014Acta Pharmaceutica Sinica B2014,4,2:5
5Application of immune checkpoint targets in the anti-tumor novel drugs and traditional Chinese medicine development显示文摘Immune checkpoints are the crucial regulators of immune system and play essential roles in maintaining self-tolerance,preventing autoimmune responses,and minimizing tissue damage by regulating the duration and intensity of the immune response.Furthermore,immune checkpoints are usually overexpressed in cancer cells or noninvasive cells in tumor tissues and are capable of suppressing the antitumor response.Based on substantial physiological analyses as well as preclinical and clinical studies,checkpoint molecules have been evaluated as potential therapeutic targets for the treatment of multiple types of cancers.In the last few years,extensive evidence has supported the immunoregulatory effects of traditional Chinese medicines(TCMs).The main advantage of TCMs and natural medicine is that they usually contain multiple active components,which can act on multiple targets at the same time,resulting in additive or synergistic effects.The strong immune regulation function of traditional Chinese medicine on immune checkpoints has also been of great interest.For example,Astragalus membranaceus polysaccharides can induce anti-PD-1 antibody responses in animals,and these antibodies can overcome the exhaustion of immune cells under tumor immune evasion.Furthermore,many other TCM molecules could also be novel and effective drug candidates for the treatment of cancers.Therefore,it is essential to assess the application of immune checkpoints in the development of new drugs and TCMs.In this review,we focus on research progress in the field of immune checkpoints based on three topics:(1)immune checkpoint targets and pathways,(2)development of novel immune checkpoint-based drugs,and(3)application of immune checkpoints in the development of TCMs.Yuli Wang Xingyan Zhang Yuyan Wang Wenjing Zhao Huling Li Lixing Zhang Xinping Li Tiejun Zhang Hongbing Zhang He Huang Changxiao Liu 2021Acta Pharmaceutica Sinica B2021,11,10:3
6Outcome of bystander cardiopulmonary resuscitation after out-of-hospital cardiac arrest in Beijing显示文摘Aim:We aimed to investigate the association between bystander cardiopulmonary resuscitation(CPR)and survival of patients with out-of-hospital cardiac arrests(OHCA)in Beiing.Methods:This observational study analyzed adult patients with OHCA treated by the Beiing emergency medical service(EMS)from January 2013 to December 2017.Data were collected in a Utstein style with a 1-year follow-up and a primary outcome 01 survival to hospital discharge.Secondary outcomes were return of spontaneous circulation(ROSC),survival to admission,favorable neurological outcome at hospital discharge,and survival and favorable neurological outcomes of up to 1 year.Results:A total of 5016 patients with OHCA from Beiing's urban area were recorded by EMS,wherein 765 patients(15.25%)underwent bystander CPR.The data were propensity score-matched forage,sex,location,witness,aetiology,initial rhythm,and call to EMS arrival to compare the difference between the occurrence and nonoccurrence of bystander CPR.The survival upon the discharge of patients who experienced bystander CPR was superior to that of patients who did not receive bystander CPR(3.7%vs 1.2%,respectively;P<0.001).Moreover,patients with OHCA resuscitated with bystander CPR achieved better outcomes of ROSC,survival to admission,favorable neurological outcome at hospital discharge,survival and favorable neurological outcome after 1 year compared with those who were not resuscitated with bystander CPR.Conclusion:Survival and neu rological outcome of patients who underwent bystander CPR was better than those who underwent nonbystander CPR in Beiing.However,the rate of bystander CPR was low.Xian Shi Yang Wu Haibin Li Shengkui Ma Dou Li Ding Gao Hao Cui Changxiao Yu Song Yang Ziren Tang Fei Shao 2021Emergency and Critical Care Medicine2021,1,2:2
7Pharmacokinetics and Pharmacodynamics of Subcutaneous Single Doses of Pegylated Human G-CSF Mutant(PEG30-rhG-CSF) in Beagle Dogs显示文摘OBJECTIVE To compare the pharmacokinetics and pharmacodynamics of PEG30-rhG-CSF administered to beagle dogs at three different dosages with PEG20-rhG-CSF administered at one dosage,and to provide an experimental basis for clinical trials.METHODS Beagle dogs received single,subcutaneous doses of PEG30-rhG-CSF at 100,200 and 400 μg/kg or PEG20-rhG-CSF at 200 μg/kg.PEG30-rhG-CSF and PEG20-rhG-CSF concentrations in serum were analyzed using an enzyme-linked immunosorbent assay(ELISA).WBC,ANC and PLT counts of whole blood samples were measured using fully automated analytic instrumentation.Pharmacokinetic and pharmacodynamic parameters were calculated using DAS 2.0 statistical analysis so ware.RESULTS The pharmacokinetic parameters of PEG30-rhG-CSF calculated from the serum concentration data determined by ELISA were as follows:the mean elimination half-life(t1/2ke) was 40.6 h(33.5~45.4 h);the mean time to reach peak concentration(Tmax) was 19.2 h(11.7~24.0 h);the drug clearance from the serum(CL) was decreased with increasing doses;the peak concentration(Cmax) and the area under the serum concentration-time curve(AUC) were increased with increasing doses.For PEG20-rhG-CSF,the half-life was shorter(12 h) and Tmax was achieved much earlier(10 h) relative to PEG30-rhG-CSF.The AUC of PEG30-rhG-CSF was much greater than that of PEG20-rhG-CSF,and the relative bioavailability with a subcutaneous injection was 158.7%.Administration of single doses of PEG30-rhG-CSF resulted in substantial increases in the absolute neutrophil count(ANC).The time to reach ANCmax(ANCTmax) was 72 h.The maximum observed absolute neutrophil counts(ANCmax) and the area over the baseline effect curve(AOBEC) was increased with increasing doses.The effect-elimination half-life(t1/2E) ranged from 60 h to 80 h a er subcutaneous administration.The PLT count was slightly elevated 8~12 h a er s.c.injection,and declined a er 24 h.CONCLUSION The mean elimination half-life of PEG30-rhG-CSF was longer than that of PEG20-rhG-CSF at the same dose,and the other main pharmacokinetic and pharmacodynamic parameters of PEG30-rhG-CSF,including Cmax,ANCmax,AUC and AOBEC were much greater than those following PEG20-rhG-CSF injection.Yongming Cai Zhengmin Chen Ling Jiang Ming Li Changxiao Liu 2008Chinese Journal of Clinical Oncology2008,5,5:2

15Development unit division and favorable area evaluation for joint mining coalbed methane显示文摘Based on the productivity equation of coalbed methane (CBM) well, considering the impact of coal reservoir reformability on gas well productivity, the main production layer optimization index in the “three-step method” of optimal combination of production layers is corrected, and then the CBM production layer potential index is introduced to evaluate favorable areas for commingled multi-coal seam production. Through analysis of the key parameters of coal reservoirs affecting the CBM productivity index, a development unit division method for areas with multi-coal seams is established, and a quantitative grading index system is proposed. On this basis, the evaluation process of CBM development favorable area is developed: the mature 3-D modeling technology is used to characterize the reservoir physical properties of multi-coal seams in full-scale;the production layer potential index of each grid is calculated, and the production layer potential index contour under single-layer or commingled multi-layer production are plotted;according to the distribution of the contour of production layer potential index, the quantitative index of CBM development unit is adopted to outline the grade I, II, III coal reservoir distribution areas, and thus to pick out the favorable development areas. 16Absolute bioavailability,dose proportionality,and tissue distribution of rotundic acid in rats based on validated LC-QqQ-MS/MS method显示文摘Rotundic acid(RA),an ursane-type pentacyclic triterpene acid isolated from the dried barks of Ilex rotunda Thunb.(Aquifoliaceae),possesses diverse bioactivities.To further study its pharmacokinetics,a simple and sensitive liquid chromatography with triple quadrupole mass spectrometry(LC-QqQ-MS/MS)method was developed and validated to quantify RA concentration in rat plasma and tissue using etofesalamide as an internal standard(IS).Plasma and tissue samples were subjected to one-step protein precipitation.Chromatographic separation was achieved on a ZORBAX Eclipse XDB-C_(18) column(4.6mm×50mm,5μm)under gradient conditions with eluents of methanol:acetonitrile(1:1,V/V)and 5mM ammonium formate:methanol(9:1,V/V)at 0.5mL/min.Multiple reaction monitoring transitions were performed at m/z 487.30→437.30 for RA and m/z 256.10→227.10 for IS in the negative mode.The developed LC-QqQ-MS/MS method exhibited good linearity(2-500 ng/mL)and was fully validated in accordance with U.S.Food and Drug Administration bioanalytical guidelines.Dose proportionality and bioavailability in rats were determined by comparing pharmacokinetic data after single oral(10,20,and 40mg/kg)and intravenous(10mg/kg)administration of RA.Tissue distribution was studied following oral administration at 20mg/kg.The results showed that the absolute bioavailability of RA after administration at different doses ranged from 16.1%to 19.4%.RA showed good dose proportionality over a dose range of 10-40 mg/kg.RA was rapidly absorbed in a dose-dependent manner and highly distributed in the liver.In conclusion,this study is the first to systematically elucidate the absorption and distribution characteristics of RA in rats,which can provide additional information for further development and evaluation of RA in drug metabolism and pharmacokinetic studies.Haihua Shang Xiaohan Dai Mi Li Yueyi Kai Zerong Liu Min Wang Quansheng Li Yuan Gu Changxiao Liu Duanyun Si 2022Journal of Pharmaceutical Analysis2022,12,2:0
17Simultaneously enhanced energy storage performance and luminance resistance in (K_(0.5)Na_(0.5))NbO_(3) -based ceramics via synergistic optimization strategy显示文摘The rapidly advancing energy storage performance of dielectric ceramics capacitors has garnered significant interest for applications in fast charge/discharge and high-power electronic techniques.Exploring the exceptional electrical properties in harsh environments can further promote their practical applications.Defect carriers can be excited under luminance irradiation,thereby leading to degradation of energy storage performance.Herein,a synergic optimization strategy is proposed to enhance energy storage properties and luminance resistance of(K_(0.5)Na_(0.5))NbO_(3)-base(KNN)ceramics.First,the introduction of Bi(Zn0.5Ti0.5)O_(3) solid solution and La3+ions disrupts the long-range polar orders and enhances super paraelectric relaxation characteristics.Additionally,doping La3+ions can increase the band gap and reduce oxygen vacancy concentration,resulting in excellent luminance resistance.Finally,the viscous polymer process is employed to suppress the grain growth and promote chemical homogeneity.As a result,ultrahigh recoverable energy storage density(Wrec)of 8.11 J/cm3 and high efficiency(η)of 80.98%are achieved under an electric field of 568 kV/cm.Moreover,the variations in Wrec andηare only 12.45%and 1.75%,respectively,under 500 W xenon lamp irradiation compared to the performance under a dark environment.These findings hold great potential in facilitating the practical application of dielectric ceramic capacitors in luminance irradiation environments.Yu Huan Diyu Gui Changxiao Li Tao Wei Lingzhi Wu Xinjian Wang Xiaozhi Wang Zhenxiang Cheng 2024Journal of Advanced Ceramics2024,13,1:0
18Superior energy storage efficiency through tailoring relaxor behavior and band energy gap in KNN-based ferroelectric ceramic capacitors显示文摘With the increasing demand of high-power and pulsed power electronic devices,environmental-friendly potassium sodium niobate((Na_(0.5)K_(0.5))NbO_(3),KNN)ceramic-based capacitors have attracted much attention in recent years owning to the boosted energy storage density(W_(rec)).Nevertheless,the dielectric loss also increases as the external electric field increases,which will generate much dissipated energy and raise the temperature of ceramic capacitors.Thus,an effective strategy is proposed to enhance the energy storage efficiency(η)via tailoring relaxor behavior and bad gap energy in the ferroelectric 0.9(Na_(0.5)K_(0.5))-NbO_(3)-0.1Bi(Zn_(2/3)(Nb_(x)Ta_(1−x))1/3)O_(3) ceramics.On the one hand,the more diverse ions in the B-sites owing to introducing the Ta could further disturb the long-range ferroelectric polar order to form the short−range polar nanoregions(PNRs),resulting in the highη.On the other hand,the introduction of Ta ions could boost the intrinsic band energy gap and thus improve the Eb.As a result,high Wrec of 3.29 J/cm^(3) and ultrahighηof 90.1%at the high external electric field of 310 kV/cm are achieved in x=0.5 sample.These results reveal that the KNN-based ceramics are promising lead-free candidate for high-power electronic devices.Limin Hou Changxiao Li Xinjian Wang Xiaozhi Wang Ting Wang Yu Huan 2023Journal of Advanced Dielectrics2023,13,1:0
