2The combined genotypes effect of ESR and FSHβ genes on litter size traits in five different pig breeds显示文摘Estrogen receptor (ESR) and Follicular-stimula-ting hormone beta subunit (FSHβ) genes were chosen as candidates to determine whether they control litter size and some other reproductive traits in swine. 269 sows from five different pig breeds were genotyped by an established PCR-RFLPs protocol at both ESR and FSHβ loci. The effects of both ESR and FSHβ on pig reproductive traits, including total number born (TNB) and number born alive (NBA), are analyzed by SAS software (version 6.12). These computation results demonstrated that both ESR locus and FSHβ locus are the major genes influencing litter size in pigs. The sows of BBBB combined genotype of ESR and FSHβ loci generally produce 1.85-3.01 TNB and 2.0-3.0 NBA more than those of ABAA combined genotype. The notable effect of ESR locus and FSHβ locus on litter size of pigs have made it possible to improve the pig reproduction by Marker-assisted selection (MAS). 3Cloned pigs derived from somatic cell nuclear transferembryos cultured in vitro atlow oxygen tension显示文摘Pig cloning has great potential to human xenotransplantation. The present study was designed to establish a more efficient system for producing cloned pigs by somatic cell nuclear transfer (SCNT). Our approach was as follows: SCNT embryos were reconstructed by using fetal fibroblasts of Chinese miniature pig as donors and in vitro matured oocytes of prepubertal gilts as recipients. Reconstructed em- bryos were induced by electrical fusion/activation and cultured in BSA-containing North Carolina State University 23 medium (NCSU-23) or Porcine Zygote Medium (PZM-3) at the gas condition of 5% CO2, 7% O2, 88% N2. A total of 230 cloned embryos were transferred to three surrogate sows, producing three piglets. One of them is apparently healthy. The clonal provenance of the piglet was indicated by its coat color and confirmed by DNA microsatellite analysis. 4Integrative analysis of miRNA and mRNA profiles reveals that gga-miR-106-5p inhibits adipogenesis by targeting the KLF15 gene in chickens显示文摘Background:Excessive abdominal fat deposition in commercial broilers presents an obstacle to profitable meat quality,feed utilization,and reproduction.Abdominal fat deposition depends on the proliferation of preadipocytes and their maturation into adipocytes,which involves a cascade of regulatory molecules.Accumulating evidence has shown that microRNAs(miRNAs)serve as post-transcriptional regulators of adipogenic differentiation in mammals.However,the miRNA-mediated molecular mechanisms underlying abdominal fat deposition in chickens are still poorly understood.This study aimed to investigate the biological functions and regulatory mechanism of miRNAs in chicken abdominal adipogenesis.Results:We established a chicken model of abdominal adipocyte differentiation and analyzed miRNA and mRNA expression in abdominal adipocytes at different stages of differentiation(0,12,48,72,and 120 h).A total of 217 differentially expressed miRNAs(DE-miRNAs)and 3520 differentially expressed genes were identified.Target prediction of DE-miRNAs and functional enrichment analysis revealed that the differentially expressed targets were significantly enriched in lipid metabolism-related signaling pathways,including the PPAR signaling and MAPK signaling pathways.A candidate miRNA,gga-miR-106-5p,exhibited decreased expression during the proliferation and differentiation of abdominal preadipocytes and was downregulated in the abdominal adipose tissues of fat chickens compared to that of lean chickens.gga-miR-106-5p was found to inhibit the proliferation and adipogenic differentiation of chicken abdominal preadipocytes.A dual-luciferase reporter assay suggested that the KLF15 gene,which encodes a transcriptional factor,is a direct target of gga-miR-106-5p.gga-miR-106-5p suppressed the posttranscriptional activity of KLF15,which is an activator of abdominal preadipocyte proliferation and differentiation,as determined with gain-and loss-of-function experiments.Conclusions:gga-miR-106-5p functions as an inhibitor of abdominal adipogenesis by targeting the KLF15 gene in chickens.These findings not only improve our understanding of the specific functions of miRNAs in avian adipogenesis but also provide potential targets for the genetic improvement of excessive abdominal fat deposition in poultry.Weihua Tian Xin Hao Ruixue Nie Yao Ling Bo Zhang Hao Zhang Changxin Wu 2022Journal of Animal Science and Biotechnology2022,13,6:3
5Single nucleotide polymorphism analysis of the encoding region of EX-FABP gene and its association with fattiness trait in chicken显示文摘Fattiness is an important parameter to estimate meat quality, which has high heritability. In this experiment, F2 chickens derived from Broilers crossing to Silky were used to study the effect of extracellular fatty acid inding protein (EX-FABP) gene on abdominal fat accumulation. Five exons and partial introns of the gene were amplified by six pairs of primers, and then single nucleotide polymorphisms (SNPs) were detected by single strand conformation polymorphism (SSCP) and subsequently confirmed by sequencing. There were four nucleotides variations found, T-C at 659, C-T at 924, CA-TG at 968 and C-T at 1985 respectively. The result of least square analysis suggests that the chickens with HH genotype defined by the third pair of primers have a lower abdominal fat weight and abdominal fat percentage than the chickens with other genotypes (GG, GI, GH and II). 6A small scale expression screen identifies tissue specific markers in the Dugesia japonica strain Pek-1显示文摘Freshwater planaria has tremendous capacity to reform the missing part of the body and therefore is considered as one of the most important model organism for regeneration study. At present, Schmidtea mediterranea and Dugesia japonica are the two major species utilized for laboratory manipulations. Dugesia japonica flatworms are widely distributed in the Far East including Cherry Valley region in the northwest area of Beijing, China. We reported here the establishment of an asexual Dugesia japonica strain Pek1, as a suitable system for regeneration study. Using morphological, karyotypical as well as phylogenetic analyses, we confirmed that these flatworms indeed belonged to Dugesia japonica. We went on to show that the commonly used in situ probes and immunohistochemistry reagents and protocols were applicable to the Pek1 strain. Using this strain, we carried out small scale analysis on EST, RNAi and gene expression. We identified 193 unique EST sequences and 65 of them had not been reported in planarian. By RNAi analysis, we showed that 48 genes, when downregulated individually, had no effect on regeneration. Furthermore, we identified 3 groups of tissue specific expressing genes that were useful for cell lineage analysis. 7Comparision of China's volatile organic compound pollution management:a computable general equilibrium approach显示文摘The public health and ecological impacts of volatile organic compound(VOCs) pollution have become a serious problem in China,arousing increasing attention to emissions control.In this context,this paper analyses the effectiveness of VOC reduction policies,namely pollution charges and environmental taxes at the national and industrial sector levels.It uses a computable general equilibrium model,which connects macroeconomic variables with VOC emissions inventory,to simulate the effects of policy scenarios(with 2007 as the reference year).This paper shows that VOC emissions are reduced by 2.2% when a pollution charge equal to the average cost of engineering reduction methods-the traditional approach to regulation in China-is applied.In order to achieve a similar reduction,an 8.9% indirect tax would have to be imposed.It concludes that an environmental tax should be the preferred method of VOC regulation due to its smaller footprint on the macroeconomy.Other policies,such as subsidies,should be used as supplements.Yufei Wang Changxin Liu Tong Wu Zhengping Hao Zheng Wang 2016Chinese Journal of Population,Resources and Environment2016,16,4:2

9Sequencing and alignment of mitochondrial genomes of Tibetan chicken and two lowland chicken breeds显示文摘Tibetan chicken lives in high-altitude area and has adapted well to hypoxia genetically. Shouguang chicken and Silky chicken are both lowland chicken breeds. In the present study, the complete mito-chondrial genome sequences of the three chicken breeds were all sequenced. The results showed that the mitochondrial DNAs (mtDNAs) of Shouguang chicken and Silky chicken consist of 16784 bp and 16785 bp respectively, and Tibetan chicken mitochondrial genome varies from 16784 bp to 16786 bp. After sequence analysis, 120 mutations, including 4 single nucleotide polymorphisms (SNPs) in tRNA genes, 9 SNPs and 1 insertion in rRNA genes, 38 SNPs and 1 deletion in D-LOOP, 66 SNPs in pro-tein-coding genes, were found. 12Effects of different states of sheep fetal fibroblasts as donor cells on the early development in vitro of reconstructed sheep embryos显示文摘To investigate the effects of different states of donor cells on the development of reconstructed sheep embryos, we designed five treatments of donor cells, including cell passage, cell size, serum starvation, colchicine treatment and gene transfection. Results are as follows: (Ⅰ) Compared with 16-18 passage cells, the morula/blastocyst rate of 5-7 passage cells as donor nuclei was significantly higher (17.3% vs. 4.9%, P<0.05), suggesting the advantage of short-time cultured cells in supporting the development of reconstructed embryos. (Ⅱ) The morula/blastocyst rate of reconstructed embryos derived from medium cells (15-25μm) as donor nuclei was higher than that from large cells (25-33μm) and small cells (8-15μm)( 20.0% vs. 8.0%, 9.7%), indicating that reconstructed embryos from medium cells had a greater potentiality to develop into morula/blastocysts than those from small or large ones. (Ⅲ) The morula/blastocyst rate of reconstructed embryos from donor cells of SS (serum starvation) was lower than that from donor cells of NSS (non-serum starvation), but no significant difference was detected between SS and NSS(11.8% vs. 18.6%, P>0.05). (Ⅳ) Fetal fibroblasts treated with 0.05μmol/L colchicine exhibited a higher morula/blastocyst rate of reconstructed embryos than those treated with 0.10 μmol/L colchicine and untreated ones (27.5% vs. 12.1%, 17.1%), however, no significant difference among the three treatments was detected (P>0.05). (Ⅴ) The morula/blastocyst rate of reconstructed embryos from fetal fibroblasts transfected with GFP gene only was 3.1%, significantly lower than that from non-transgenic cells (3.1% vs. 20.4%, P<0.05). In conclusion, our results demonstrated that fetal fibroblasts of fewer passages, medium size could ensure a higher morula/blastocyst rate of reconstructed embryos. Serum starvation of donor cells might be unnecessary to the development of reconstructed embryos. Donor cells treated with 0.05μmol/L colchicine could facilitate the development of reconstructed embryos. Additionally, as cells transfected with GFP gene were used as donor nuclei, adverse effect on the development of reconstructed embryos was observed. 15Djrho2 is involved in regeneration of visual nerves in Dugesia japonica显示文摘The freshwater planarian is a powerful animal model for studying regeneration and stem cell activity in vivo.During regeneration,stem cells(neoblasts in planarian) migrated to the wounding edge to re-build missing parts of the body.However,proteins involved in regulating cell migration during planarian regeneration have not been studied extensively.Here we report two small GTPase genes(Djrho2 and Djrho3) of Dugesia japonica(strain Pek-1).In situ hybridization results indicated that Djrho2 was expressed throughout the body with the exception of the pharynx region while Djrho3 was specifically expressed along the gastro-vascular system.Djrho2 was largely expressed in neoblasts since its expression was sensitive to X-ray irradiation.In Djrho2-RNAi planarians,smaller anterior blastemas were observed in tail fragments during regeneration.Consistently,defective regeneration of visual nerve was detected by immunostainning with VC-1 antibody.These results suggested that Djrho2 is required for proper anterior regeneration in planairan.In contrast,no abnormality was observed after RNAi of Djrho3.We compared protein compositions of control and Djrho2-RNAi planarians using an optimized proteomic approach.Twenty-two up-regulated and 26 de-regulated protein spots were observed in the two-dimensional electrophoresis gels,and 17 proteins were successfully identified by Mass Spectrometry(MS) analysis.Among them,6 actin-binding or cytoskeleton-related proteins were found de-expressed in Djrho2-RNAi animals,suggesting that abnormal cytoskeleton assembling and cell migration were likely reasons of defected regeneration.Changxin Ma Yang Gao Guoliang Chai Hanxia Su Niejun Wang Yigang Yang Chunbo Li Di Miao Wei Wu 2010Journal of Genetics and Genomics2010,37,11:1
16Sequences and polymorphisms of exons 3 and 4 in porcine UCP_2 gene显示文摘Uncoupling proteins are mitochondrial membrane transporters, which regulate metabolic pathways of energy balance, and are associated with biological traits of animal body weight, resting metabolic rates and energy conversion. In this study, a region of the exons 3 and 4 of pig UCP2 gene was cloned and analyzed, and a new single nucleotide polymorphic site was detected by PCR-SSCP in five pig breeds. 18Simplified Numerical Models of the Unsteady Tip Leakage Flow in Compressor显示文摘Tip leakage flow(TLF)in compressors,which can cause flow blockage in blade passage and induce efficiency loss,is also a potential threat to the unsteady flow stability of modern aeroengines.This paper provides an overview of the significance of tip leakage flow research,and introduces relevant previous studies.After calculating by different methods,large eddy simulation(LES)is demonstrated again as a suitable compromise between accuracy and computational cost for the unsteady flow study.Two types of simplified tip leakage flow models using LES are adopted with a focus on the unsteady characteristics of shedding vortices in a cavity plane.This paper applies these models to study the unsteady tip leakage flow which triggers the onset of acoustic resonance in a multistage axial compressor.Compared with the detected acoustic resonance frequency of 5.22 rotational frequency(RF)in the previous experiment,the computed combination frequency in the 2-D model is equal to 5.232RF,and the simplified 3-D unsteady tip leakage flow model results in a combination frequency of 5.316RF.Therefore,based on the small relative error between model results and experimental results,the simplified numerical models are validated to be sufficiently accurate,and theoretically provide a useful basis for the subsequent research of unsteady tip leakage flow in turbomachinery.SI Changxin WU Zihao LIU Xiaohua 2023Journal of Thermal Science2023,32,6:0
19Neuroglobin mutation associated with hypoxia adaptation in Tibet chicken显示文摘Globin protein family plays an important role in storing and transporting oxygen. As a newly reported globin, the revealed function of neuroglobin includes binding and storing oxygen as well as facilitating the utilization of oxygen in neuronal cells. In the present study, mutations in exons of chicken Ngb gene were identified with the method of sequencing and created restriction site PCR in Tibet chicken and other four lowland chicken breeds. The mutations of Lys-2224(E4)-Asn and Ser-2279(E4)-Gly were identified in exon 4 of the gene. The Lys-2224(E4)-Asn mutation existed only in Tibet chicken and the mutation frequencies increased with increasing altitude. Analysis of the haplotype and diplotype of the two mutations in Tibet chicken populations of different altitudes showed that the frequencies of TG haplotype and TTGG diplotype also increased with increasing altitude, while the reverse tendency was found on GGAA diplotype. Under the hypoxic simulation incubation, the main haplotype was TG in living embryos and GA in dead embryos. 20Semantic segmentation via pixel-to-center similarity calculation显示文摘Since the fully convolutional network has achieved great success in semantic segmentation,lots of works have been proposed to extract discriminative pixel representations.However,the authors observe that existing methods still suffer from two typical challenges:(i)The intra-class feature variation between different scenes may be large,leading to the difficulty in maintaining the consistency between same-class pixels from different scenes;(ii)The inter-class feature distinction in the same scene could be small,resulting in the limited performance to distinguish different classes in each scene.The authors first rethink se-mantic segmentation from a perspective of similarity between pixels and class centers.Each weight vector of the segmentation head represents its corresponding semantic class in the whole dataset,which can be regarded as the embedding of the class center.Thus,the pixel-wise classification amounts to computing similarity in the final feature space between pixels and the class centers.Under this novel view,the authors propose a Class Center Similarity(CCS)layer to address the above-mentioned challenges by generating adaptive class centers conditioned on each scenes and supervising the similarities between class centers.The CCS layer utilises the Adaptive Class Center Module to generate class centers conditioned on each scene,which adapt the large intra-class variation between different scenes.Specially designed Class Distance Loss(CD Loss)is introduced to control both inter-class and intra-class distances based on the predicted center-to-center and pixel-to-center similarity.Finally,the CCS layer outputs the processed pixel-to-center similarity as the segmentation prediction.Extensive experiments demonstrate that our model performs favourably against the state-of-the-art methods.Dongyue Wu Zilin Guo Aoyan Li Changqian Yu Nong Sang Changxin Gao 2024CAAI Transactions on Intelligence Technology2024,9,1:0
