| 1 | Rhizospheric microbial communities are driven by Panax ginseng at different growth stages and biocontrol bacteria alleviates replanting mortality显示文摘The cultivation of Panax plants is hindered by replanting problems, which may be caused by plantdriven changes in the soil microbial community. Inoculation with microbial antagonists may efficiently alleviate replanting issues. Through high-throughput sequencing, this study revealed that bacterial diversity decreased,whereas fungal diversity increased, in the rhizosphere soils of adult ginseng plants at the root growth stage under different ages. Few microbial community, such as Luteolibacter, Cytophagaceae, Luteibacter, Sphingomonas,Sphingomonadaceae, and Zygomycota, were observed; the relative abundance of microorganisms, namely,Brevundimonas, Enterobacteriaceae, Pandoraea, Cantharellales, Dendryphion, Fusarium, and Chytridiomycota,increased in the soils of adult ginseng plants compared with those in the soils of 2-year-old seedlings. Bacillus subtilis 50-1, a microbial antagonist against the pathogenic Fusarium oxysporum, was isolated through a dual culture technique. These bacteria acted with a biocontrol efficacy of 67.8%. The ginseng death rate and Fusarium abundance decreased by 63.3% and 46.1%, respectively, after inoculation with B. subtilis 50-1. Data revealed that microecological degradation could result from ginseng-driven changes in rhizospheric microbial communities;these changes are associated with the different ages and developmental stages of ginseng plants. | 2 | Integrated metabolomic and transcriptomic analyses revealed the distribution of saponins in Panax notoginseng显示文摘Panax notoginseng is famous for its important therapeutic effects. Saponins are bioactive compounds found in different parts and developmental stages of P. notoginseng plants. Thus, it is urgently to study saponins distribution in different parts and growth ages of P. notoginseng plants. In this study,potential biomarkers were found, and their chemical characteristic differences were revealed through metabolomic analysis. High-performance liquid chromatography data indicated the higher content of saponins(i.e., Rg1, Re, Rd, and Rb1) in the underground parts than that in the aerial parts. 20(S)-Protopanaxadiol saponins were mainly distributed in the aerial parts. Additionally, the total saponin content in the 3-year-old P. notoginseng plant(188.0 mg/g) was 1.4-fold higher than that in 2-year-old plant(130.5 mg/g). The transcriptomic analysis indicated the tissue-specific transcription expression of genes, namely, PnFPS, PnSS, PnSE1, PnSE2, and PnDS, which encoded critical synthases in saponin biosyntheses. These genes showed similar expression patterns among the parts of P. notoginseng plants.The expression levels of these genes in the flowers and leaves were 5.2 fold higher than that in the roots and fibrils. These results suggested that saponins might be actively synthesized in the aerial parts and transformed to the underground parts. | 3 | Manifestations in Corrosion Prophase of Ultra-high Strength Steel 30CrMnSiNi2 A in Sodium Chloride Solutions显示文摘The corrosion behaviors of ultra-high strength steel 30CrMnSiNi2A in sodium chloride solution were studied by weight loss and electrochemical methods. The morphology of corrosion products was observed using scanning electron microscopy(SEM) and the composition was analyzed using an energy dispersive spectroscopy(EDS) and X-Ray diffraction(XRD). The experimental results showed that the corrosion came from pitting corrosion and the rust layer was composed of outer rust layer γ-FeOOH and inner rust layer Fe2O3 with a little β-FeOOH. The correlation between corrosion rate and test time accorded with exponential rule. The corrosion current measured by polarization methods was higher than that calculated by weight loss method after a long-time immersion, the main reason was that β-FeOOH and γ-Fe2O3 transformed by γ-FeOOH led to overestimating corrosion rate. The processes of corrosion prophase were obtained from XRD and EIS results. | 4 | Distribution of Calcium Carbonate in the Process of Concrete Self-healing显示文摘The complete deposition distribution process of calcium carbonate is summarized in three directions of cracks. Distribution of calcium carbonate in the self-healing process of microbial concrete is studied in detail, with the help of a variety of analytical techniques. The results show that carbonate deposits along the x-axis direction of the cracks. The farther from the crack surfaces of concrete matrix in x-axis direction, the more the content of the substrate, the less content of calcium carbonate. Gradual accumulation of calcium carbonate along the y-axis direction is like building a house with bricks. Different repair points are gradually connected, and ultimately the whole of cracks are completely filled. | 6 | EMMD-Prony approach for dynamic validation of simulation models显示文摘Model validation and updating is critical to model credibility growth. In order to assess model credibility quantitatively and locate model error precisely, a new dynamic validation method based on extremum field mean mode decomposition(EMMD) and the Prony method is proposed in this paper. Firstly, complex dynamic responses from models and real systems are processed into stationary components by EMMD. These components always have definite physical meanings which can be the evidence about rough model error location. Secondly, the Prony method is applied to identify the features of each EMMD component. Amplitude similarity, frequency similarity, damping similarity and phase similarity are defined to describe the similarity of dynamic responses.Then quantitative validation metrics are obtained based on the improved entropy weight and energy proportion. Precise model error location is realized based on the physical meanings of these features. | 11 | Investigation on the genetic-inconsistent paternity cases using the MiSeq FGx system显示文摘Mutations might challenge the paternity index calculation in forensic identification.While many studies have focussed on the autosomal short tandem repeats(A-STR),the mutation status of sex chromosomes and single nucleotide polymorphism(SNP)remain blank.Next generation sequencing(NGS),known as high throughput and large sequence polymorphism,is a promising tool for forensic genetics.To describe the mutation landscapes in the paternity cases with genetic inconsistencies,a total of 63 parentage confirmed paternity cases contained at least one mismatched locus have been collected.The mutations were subsequently evaluated using Verogen’s MPSForenSeqTM DNASignature Kit and a microsatellite instability(MSI)detection kit.The result showed 98.41%(62/63)of the cases had no additional autosomal mutations even when the number of A-STRs increased to 27.As for the sex chromosomes,about 11.11%(7/63)of the cases exhibited either X-STR or Y-STR mutations.D2S1338,FGAand Penta Ewere the most frequent altered STRs,which suggested they might be the mutation hotspots.In addition,a male with sex chromosome abnormality was observed accidently,whose genotype might be 47,XXY,rather than MSI.Nearly 56.90%of the STR loci possessed isoalleles,which might result in higher STR polymorphisms.No Mendelian incompatibility was detected among the SNP markers,which indicated that SNP was a more reliable genetic marker in the genetic-inconsistent paternity cases. | Anqi Chen Ruiyang Tao Chengtao Li Suhua Zhang | 2022 | Forensic Sciences Research2022,7,4: | 0 |

| 20 | Validation studies of the Para DNA^(■) Intelligence System with artificial evidence items显示文摘Short tandem repeat(STR)profiling is one of the mostly used systems for forensic applications.In certain circumstances,STR profiling is time-consuming and costly,which potentially leads to delays in criminal investigations.LGC(Laboratory of the Government Chemist,UK)Forensics has developed a robust STR profiling platform called the ParaDNAVR Intelligence Test System which can provide early tactical intelligence and aid investigators in making informed decisions on sample prioritization for detection.Here,we validated the ParaDNA^(■) intelligence test for its application in forensic cases using a range of mock evidence items following guidelines set by the Scientific Working Group on DNA Analysis Methods(SWGDAM).Specifically,we tested the sensitivity and accuracy of the ParaDNA intelligence test,as well as the success rates for detecting mock samples and for use in case scenarios.Our findings demonstrate that the ParaDNA intelligence test generates useful DNA profiles,especially for samples such as blood,saliva,and semen that contain ample DNA,indicating the benefits of including ParaDNA as a prior step in forensic STR profiling pipelines. | Min Li Ruiyang Tao Wei Zhou Yanan Li Meng Meng Yilun Zhang Linsheng Yu Liqin Chen Yingnan Bian Chengtao Li | 2021 | Forensic Sciences Research2021,6,1: | 0 |
