1Efficacy and advantages of modified Traditional Chinese Medicine treatments based on'kidney reinforcing'for chronic aplastic ane-mia:a randomized controlled clinical trial显示文摘OBJECTIVE:To compare the efficacy of modified treatments based on 'kidney reinforcing' in the management of chronic aplastic anemia(CAA),and explore their advantages and specialties.METHODS:One hundred and eleven patients with CAA were randomly divided into three groups:kidney reinforcing alone(KA),'kidney reinforcing and Qi tonifying'(KQ),and 'kidney reinforcing and blood circulation invigorating'(KC).Normal and positive control groups were also formed.All patients were treated for 6 months(two courses).Hemograms,Traditional Chinese Medicine(TCM) syndrome scores,and therapeutic effects were assessed,and changes in T-lymphocyte subsets,regulatory T cells and cytokines were detected.RESULTS:The KQ and KC groups had lower TCM syndrome scores than the positive control group after 6 months(P < 0.05).The KQ group had a higher overall efficacy than the positive control group after 3 months(P < 0.05),while platelet counts increased in the KC group after 6 months(P < 0.05).CD3+ T-lymphocyte ratios decreased only in the KQ group,while CD3 + CD4 + CD8-Tlymphocytes increased only in the KC group after 6 months(P <0.05).Levels of interferon-γ,tumor necrosis fac-tor-α,interleukin(IL)-2 and IL-6 decreased and levels of IL-4 and IL-10 increased in all treated groups after 6 months.Levels of IL-6 in the KQ and KC groups were lower than those in the positive control group(P < 0.05).CONCLUSION:Treatments based on kidney reinforcing have a rebalancing effect on cytotoxic and T helper cells,and regulate expression of interferon-γ,IL-2,IL-6 and IL-4.KQ may be more effective in treating CAA,and KC may have an advantage in platelet recovery.Wu Dijiong Shen Yiping Ye Baodong Fang Bingmu Lin Shengyun Chen Zhilu Jiang Huifang Feng Changwei He Lüyuan Gao Yanting Liu Yonglin Liu Yonghua Zhu Jiajia Wu Liqiang Shao Keding Zhou Yuhong 2016Journal of Traditional Chinese Medicine2016,36,4:6
2Nitric oxide-generating compound and bio-clickable peptide mimic for synergistically tailoring surface anti-thrombogenic and anti-microbial dual-functions显示文摘Application of extracorporeal circuits and indwelling medical devices has saved many lives.However,it is accompanied with two major complications:thrombosis and infection.To address this issue,we apply therapeutic nitric oxide gas(NO)and antibacterial peptide for synergistically tailoring such devices for surface anti-thrombogenic and antifouling dual functions.Such functional surface is realized by stepwise conjugation of NO-generating compound of 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid(DOTA)chelated copper ions(Cu-DOTA)and dibenzylcyclooctyne-(DBCO-)modified antimicrobial peptide based on carbodiimide and click chemistry respectively.The integration of peptide and Cu-DOTA grants the modified surface the ability to not only efficiently inhibit bacterial growth,but also catalytically generate NO from endogenous s-nitrosothiols(RSNO)to reduce adhesion and activation of platelets,preventing the formation of thrombus.We envision that the stepwise synergistic modification strategy by using anticoagulant NO and antibacterial peptide would facilitate the surface multifunctional engineering of extracorporeal circuits and indwelling medical devices,with reduced clinical complications associated with thrombosis and infection.Han Yu Shaoxing Yu Hua Qiu Peng Gao Yingzhong Chen Xin Zhao Qiufen Tu Minggang Zhou Lin Cai Nan Huang Kaiqin Xiong Zhilu Yang 2021Bioactive Materials2021,6,6:3
3PTX-symmetric metasurfaces for sensing applications显示文摘In this paper,we introduce an ultra-sensitive optical sensing platform based on the parity-time-reciprocal scaling(PT^-symmetric non-Hermitian metasurfaces,which leverage exotic singularities,such as the exceptional point(EP)and the coherent perfect absorber-laser(CPAL)point,to significantly enhance the sensitivity and detectability of photonic sensors.We theoretically studied scattering properties and physical limitations of the PTX-symmetric metasurface sensing systems with an asymmetric,unbalanced gain-loss profile.The PTLY-symmetric metasurfaces can exhibit similar scattering properties as their Pr-symmetric counterparts at singular points,while achieving a higher sensitivity and a larger modulation depth,possible with the reciprocal-scaling factor(i.e.,X transformation).Specifically,with the optimal reciprocalscaling factor or near-zero phase offset,the proposed PTX-symmetric metasurface sensors operating around the EP or CPAL point may achieve an over 100 dB modulation depth,thus paving a promising route toward the detection of small-scale perturbations caused by,for example,molecular,gaseous,and biochemical surface adsorbates.Zhilu YE Minye YANG Liang ZHU Pai-Yen CHEN 2021Frontiers of Optoelectronics2021,14,2:2
4Advances in hydrogel-based vascularized tissues for tissue repair and drug screening显示文摘The construction of biomimetic vasculatures within the artificial tissue models or organs is highly required for conveying nutrients,oxygen,and waste products,for improving the survival of engineered tissues in vitro.In recent times,the remarkable progress in utilizing hydrogels and understanding vascular biology have enabled the creation of three-dimensional(3D)tissues and organs composed of highly complex vascular systems.In this review,we give an emphasis on the utilization of hydrogels and their advantages in the vascularization of tissues.Initially,the significance of vascular elements and the regeneration mechanisms of vascularization,including angiogenesis and vasculogenesis,are briefly introduced.Further,we highlight the importance and advantages of hydrogels as artificial microenvironments in fabricating vascularized tissues or organs,in terms of tunable physical properties,high similarity in physiological environments,and alternative shaping mechanisms,among others.Furthermore,we discuss the utilization of such hydrogels-based vascularized tissues in various applications,including tissue regeneration,drug screening,and organ-on-chips.Finally,we put forward the key challenges,including multifunctionalities of hydrogels,selection of suitable cell phenotype,sophisticated engineering techniques,and clinical translation behind the development of the tissues with complex vasculatures towards their future development.Ying Wang Ranjith Kumar Kankala Caiwen Ou Aizheng Chen Zhilu Yang 2022Bioactive Materials2022,7,3:1
5Slope stability prediction based on a long short-term memory neural network:comparisons with convolutional neural networks,support vector machines and random forest models显示文摘The numerical simulation and slope stability prediction are the focus of slope disaster research.Recently,machine learning models are commonly used in the slope stability prediction.However,these machine learning models have some problems,such as poor nonlinear performance,local optimum and incomplete factors feature extraction.These issues can affect the accuracy of slope stability prediction.Therefore,a deep learning algorithm called Long short-term memory(LSTM)has been innovatively proposed to predict slope stability.Taking the Ganzhou City in China as the study area,the landslide inventory and their characteristics of geotechnical parameters,slope height and slope angle are analyzed.Based on these characteristics,typical soil slopes are constructed using the Geo-Studio software.Five control factors affecting slope stability,including slope height,slope angle,internal friction angle,cohesion and volumetric weight,are selected to form different slope and construct model input variables.Then,the limit equilibrium method is used to calculate the stability coefficients of these typical soil slopes under different control factors.Each slope stability coefficient and its corresponding control factors is a slope sample.As a result,a total of 2160 training samples and 450 testing samples are constructed.These sample sets are imported into LSTM for modelling and compared with the support vector machine(SVM),random forest(RF)and convo-lutional neural network(CNN).The results show that the LSTM overcomes the problem that the commonly used machine learning models have difficulty extracting global features.Furthermore,LSTM has a better prediction performance for slope stability compared to SVM,RF and CNN models.Faming Huang Haowen Xiong Shixuan Chen Zhitao Lv Jinsong Huang Zhilu Chang Filippo Catani 2023International Journal of Coal Science & Technology2023,10,2:1
6Design,synthesis,and evaluation of PD-L1 degraders to enhance T cell killing activity against melanoma显示文摘Inhibitor targeting immune checkpoint is a promising new anticancer therapy.Blocking the interaction between PD-1 and PD-L1 can reverse the immunosuppression state and improve the lethality of immune cells to tumor cells.Here,we report PROTAC-based PD-L1 degraders to enhance T cell killing activity against melanoma.Four series of PD-L1 degraders were designed and synthesized to VHL,CRBN,MDM2 or c IAP E3 ligase system,in which CRBN-ligand-based compound BMS-37-C3 was identified as the most active PROTAC molecule.BMS-37-C3 also significantly enhanced the killing ability of T cells in a co-culture model of A375 and T cells.Furthermore,western blot data and flow cytometry demonstrated that BMS-37-C3 could reduce the protein levels of PD-L1 in dose and time dependent manner,which may provide a new therapeutic method for tumor immunotherapy.Yang Liu Mengzhu Zheng Zhilu Ma Yirong Zhou Junfeng Huo Wenbo Zhang Yu Liu Yuanyuan Guo Xuechen Zhou Hua Li Lixia Chen 2023Chinese Chemical Letters2023,34,5:0
7Nonmuscle myosin IIA promotes the internalization of influenza A virus and regulates viral polymerase activity through interacting with nucleoprotein in human pulmonary cells显示文摘Influenza A virus(IAV),responsible for seasonal epidemics and recurring pandemics,represents a global threat to public health.Given the risk of a potential IAV pandemic,it is increasingly important to better understand virushost interactions and develop new anti-viral strategies.Here,we reported nonmuscle myosin IIA(MYH9)-mediated regulation of IAV infection.MYH9 depletion caused a profound inhibition of IAV infection by reducing viral attachment and internalization in human lung epithelial cells.Surprisingly,overexpression of MYH9 also led to a significant reduction in viral productive infection.Interestingly,overexpression of MYH9 retained viral attachment,internalization,or uncoating,but suppressed the viral ribonucleoprotein(vRNP)activity in a minigenome system.Further analyses found that excess MYH9 might interrupt the formation of vRNP by interacting with the viral nucleoprotein(NP)and result in the reduction of the completed vRNP in the nucleus,thereby inhibiting subsequent viral RNA transcription and replication.Together,we discovered that MYH9 can interact with IAV NP protein and engage in the regulation of vRNP complexes,thereby involving viral replication.These findings enlighten new mechanistic insights into the complicated interface of host-IAV interactions,ultimately making it an attractive target for the generation of antiviral drugs.Jian Chen Jian Liu Zhilu Chen Daobin Feng Cuisong Zhu Jun Fan Shuye Zhang Xiaoyan Zhang Jianqing Xu 2023Virologica Sinica2023,38,1:0
8Extraction Process Optimization and Anti-oxidant Activity of Total Flavonoids in Dendrobium nobile Lindl.显示文摘[Objectives]To improve the yield of flavonoids from Dendrobium nobile Lindl.[Methods]On the basis of four single-factor experiments of solid/liquid ratio,extraction temperature,extraction time and ethanol volume fraction,the ultrasonic-assisted extraction process of total flavonoids from D.nobile Lindl.was optimized using quadratic general rotary unitized design,and the in-vitro anti-oxidant activity of the flavonoids extracted was evaluated preliminarily.[Results]The optimized extraction process for flavonoids in D.nobile Lindl.was as follows:extraction time 10 min,solid/liquid ratio 1∶15(g∶mL),extraction temperature 75℃and ethanol volume fraction 90%.According to the revised optimal process,6 repeated experiments were carried out,and the average yield of flavonoids was(0.5515±0.0004)mg/g,consistent with the predicted value of the model.The results of anti-oxidation experiment show that the flavonoids extracted from D.nobile Lindl.have better scavenging effect on ABTS and DPPH free radicals,and the scavenging activity is proportional to the mass concentration of the extract.[Conclusions]The optimized extraction process for flavonoids from D.nobile Lindl.is reasonable and feasible,and the flavonoids in D.nobile Lindl.have strong antioxidant activity.Huashan GAO Minghui CHEN Guang QI Ke ZHANG Yanjiao LI Zhonglin WANG Zhilu ZHANG Weishuang TONG 2020Medicinal Plant2020,11,4:0
9cDNA-SCoT Analysis of Differential Expressed Genes in Allium tuberosum Induced by Botrytis cinerea显示文摘[Objectives]To investigate the molecular mechanism of gene differential expression in A.tuberosum under Botrytis cinerea stress.[Methods]The leaf of A.tuberosum was inoculated with B.cinerea to induce resistance,then leaf samples were taken at 2,4,8,12 and 24 h,respectively,after inoculation,and used for cDNA-SCoT analysis to detect the gene differential expressions.[Results]More than 800 bands with length of 100-1800 bp were obtained using 60 SCoT primers.A total of 40 differentially expressed EST sequences were screened out,and 18 non-redundant ESTs with high quality were obtained by cluster analyses of the ESTs sequencing.The results of BlastN showed that the differential genes of A.tuberosum mainly related to disease defense,signal transduction,and protein metabolism.[Conclusions]Further analysis of gene function indicated that brassinosteroid biosynthesis-like proteins,NBS-LRR type disease resistance protein,jasmonic acid induced protein,abscisic stress ripening protein may be involved in the process of the incompatible interaction between the A.tuberosum and B.cinerea.Minghui CHEN Zhilu ZHANG Yanjiao LI Pengqiang YAO Shiping CHENG Ganqing ZHAO 2020Asian Agricultural Research2020,12,10:0
10Responses of plant^(15)N natural abundance and isotopic fractionation to N addition reflect the N status of a temperate steppe in China显示文摘Aims Elevated anthropogenic nitrogen(N)deposition could alter N status in temperate steppe.However,threshold observations of N status change from N limit to N saturation by far are not conclusive in these ecosystems.Research on the natural abundance of ^(15)N( δ^(15)N)could greatly help provide integrated information about ecosystem N status.The goal of this study was to investigate the suitability of measurements of δ^(15)N of major ecosystem N pools and several key species,plant ^(15)N fractionation,together with key vegetation and soil indicators in response to N fertilization as a tool to identify the N status in a temperate steppe in Inner Mongolia.Methods We carried out a N addition experiment during 2011-14 on a Stipa krylovii steppe in Inner Mongolia,Northern China.We investigated the response of several key N transformation processes,vegetation and soil properties to N addition.Aboveground biomass and below-ground biomass(BGB) δ^(15)N,root and foliar δ^(15)N of three dominant species(Artemisia frigida,S.krylovii and Leymus chinensis), δ^(15)N of soil total N and soil KCl-extractable NO_(3)^(−)-N were determined.The responses of isotope fractionation during plant N uptake and reallo-cation to N addition were also determined.Important Findings Our results suggest that the N addition rate of 5g N m^(−2) yr^(−1) could be regarded as threshold of early N saturation in this S.krylovii steppe as indicated by an increase in plant fraction-ation and a decrease in plant δ^(15)N.When N input rate is>10 g N m^(-2) yr^(-1),increased N deposition can lead to an apparent reduction in species richness and BGB as well as an increase in NO_(3)^(−)in extractable soil pools<30-cm soil profile.With N addition,S.krylovii and A.frigida undergo earlier N status shift from N limitation toward N excess compared with L.chinen-sis,contributing to L.chinensis out-competing other species.Overall,this study provides a better understanding of N status change in temperate steppe based on isotope evidence and several other functional variables and contributes to predicting the responses of temperate steppe to future global N deposition scenario.Zhilu Sheng Yongmei Huang Kejian He Narigele Borjigin Hanyue Yang Huiying Chen Engui Li Xia Xu Lei Duan 2019Journal of Plant Ecology2019,12,3:0

12A“built-up”composite film with synergistic functionalities on Mg-2Zn-1Mn bioresorbable stents improves corrosion control effects and biocompatibility显示文摘Control of premature corrosion of magnesium(Mg)alloy bioresorbable stents(BRS)is frequently achieved by the addition of rare earth elements.However,limited long-term experience with these elements causes concerns for clinical application and alternative methods of corrosion control are sought after.Herein,we report a“built-up”composite film consisting of a bottom layer of MgF2 conversion coating,a sandwich layer of a poly(1,3-trimethylene carbonate)(PTMC)and 3-aminopropyl triethoxysilane(APTES)co-spray coating(PA)and on top a layer of poly(lactic-co-glycolic acid)(PLGA)ultrasonic spray coating to decorate the rare earth element-free Mg-2Zn-1Mn(ZM21)BRS for tailoring both corrosion resistance and biological functions.The developed“built-up”composite film shows synergistic functionalities,allowing the compression and expansion of the coated ZM21 BRS on an angioplasty balloon without cracking or peeling.Of special importance is that the synergistic corrosion control effects of the“built-up”composite film allow for maintaining the mechanical integrity of stents for up to 3 months,where complete biodegradation and no foreign matter residue were observed about half a year after implantation in rabbit iliac arteries.Moreover,the functionalized ZM21 BRS accomplished re-endothelialization within one month.Zhenglong Dou Shuiling Chen Jiacheng Wang Li Xia Manfred F.Maitz Qiufen Tu Wentai Zhang Zhilu Yang Nan Huang 2023Bioactive Materials2023,,7:0
13Cloning and Expression Analysis of TcREMORIN Gene Related to Anthracnose Resistance in Taxus chinesis显示文摘[Objective]The paper was to obtain a pathogen-inducible responsive gene TcREMORIN from Taxus chinesis.[Method]Total RNA was extracted by Trizol reagent,and cDNA was synthesized using TaKaRa reverse transcription kit and Invitrogen M-MLV Reverse Transcriptase kit.The TcREMORIN gene was amplified by qRT-PCR.[Result]The full length of TcREMORIN was 778 bp with a 501 bp open reading frame(ORF)which encoded 166 amino acids.Bioinformatics analysis indicated that TcREMORIN possessed a conserved domain of remorin family genes and located probably in cell plasma membrane.The results of real-time quantitative PCR assay revealed that the target gene was induced by anthracnose of T.chinesis and its expression level was up-regulated by PEG,NaCl,H2O2 and 4℃stresses.[Conclusion]The TcREMORIN gene may play a critical role in plant-pathogen interaction.Chen Minghui Li Yanjiao Zhang Zhilu Tong Weishuang 2020Plant Diseases and Pests2020,11,5:0
