| 1 | The apparent contact angle of water droplet on the micro-structured hydrophobic surface显示文摘The apparent contact angle of Cassie-Baxter state water droplets can be calculated by the existing theoretical formula, but due to the defects of the micro-structured hydrophobic surface and some inevitable tiny disturbances in the experiment, Cassie-Baxter state water droplets will appear partly in Wenzel state, that is, the mixed state water droplets. In this paper, apparent contact angles of Cassie-Baxter state and mixed state water droplets on micro-structured hydrophobic surfaces are compared. The research shows that if the projected area fraction of water-solid F in the Cassie-Baxter formula is replaced by the local projected area fraction of water-solid F′, the apparent contact angles of water droplets in both Cassie-Baxter state and the mixed state can be calculated. Further experimental results indicate that the contact state of water droplets nearby the outermost three-phase contact line plays a more important role in determining the apparent contact angle. | 2 | Efficacy and advantages of modified Traditional Chinese Medicine treatments based on'kidney reinforcing'for chronic aplastic ane-mia:a randomized controlled clinical trial显示文摘OBJECTIVE:To compare the efficacy of modified treatments based on 'kidney reinforcing' in the management of chronic aplastic anemia(CAA),and explore their advantages and specialties.METHODS:One hundred and eleven patients with CAA were randomly divided into three groups:kidney reinforcing alone(KA),'kidney reinforcing and Qi tonifying'(KQ),and 'kidney reinforcing and blood circulation invigorating'(KC).Normal and positive control groups were also formed.All patients were treated for 6 months(two courses).Hemograms,Traditional Chinese Medicine(TCM) syndrome scores,and therapeutic effects were assessed,and changes in T-lymphocyte subsets,regulatory T cells and cytokines were detected.RESULTS:The KQ and KC groups had lower TCM syndrome scores than the positive control group after 6 months(P < 0.05).The KQ group had a higher overall efficacy than the positive control group after 3 months(P < 0.05),while platelet counts increased in the KC group after 6 months(P < 0.05).CD3+ T-lymphocyte ratios decreased only in the KQ group,while CD3 + CD4 + CD8-Tlymphocytes increased only in the KC group after 6 months(P <0.05).Levels of interferon-γ,tumor necrosis fac-tor-α,interleukin(IL)-2 and IL-6 decreased and levels of IL-4 and IL-10 increased in all treated groups after 6 months.Levels of IL-6 in the KQ and KC groups were lower than those in the positive control group(P < 0.05).CONCLUSION:Treatments based on kidney reinforcing have a rebalancing effect on cytotoxic and T helper cells,and regulate expression of interferon-γ,IL-2,IL-6 and IL-4.KQ may be more effective in treating CAA,and KC may have an advantage in platelet recovery. | Wu Dijiong Shen Yiping Ye Baodong Fang Bingmu Lin Shengyun Chen Zhilu Jiang Huifang Feng Changwei He Lüyuan Gao Yanting Liu Yonglin Liu Yonghua Zhu Jiajia Wu Liqiang Shao Keding Zhou Yuhong | 2016 | Journal of Traditional Chinese Medicine2016,36,4: | 6 |
| 3 | Organellar genome assembly methods and comparative analysis of horticultural plants显示文摘Although organellar genomes(including chloroplast and mitochondrial genomes)are smaller than nuclear genomes in size and gene number,organellar genomes are very important for the investigation of plant evolution and molecular ecology mechanisms.Few studies have focused on the organellar genomes of horticultural plants.Approximately 1193 chloroplast genomes and 199 mitochondrial genomes of land plants are available in the National Center for Biotechnology Information(NCBI),of which only 39 are from horticultural plants.In this paper,we report an innovative and efficient method for high-quality horticultural organellar genome assembly from next-generation sequencing(NGS)data.Sequencing reads were first assembled by Newbler,Amos,and Minimus software with default parameters.The remaining gaps were then filled through BLASTN search and PCR.The complete DNA sequence was corrected based on Illumina sequencing data using BWA(Burrows–Wheeler Alignment tool)software.The advantage of this approach is that there is no need to isolate organellar DNA from total DNA during sample preparation.Using this procedure,the complete mitochondrial and chloroplast genomes of an ornamental plant,Salix suchowensis,and a fruit tree,Ziziphus jujuba,were identified.This study shows that horticultural plants have similar mitochondrial and chloroplast sequence organization to other seed plants.Most horticultural plants demonstrate a slight bias toward A+T rich features in the mitochondrial genome.In addition,a phylogenetic analysis of 39 horticultural plants based on 15 protein-coding genes showed that some mitochondrial genes are horizontally transferred from chloroplast DNA.Our study will provide an important reference for organellar genome assembly in other horticultural plants.Furthermore,phylogenetic analysis of the organellar genomes of horticultural plants could accurately clarify the unanticipated relationships among these plants. | Xuelin Wang Feng Cheng Dekai Rohlsen Changwei Bi Chunyan Wang Yiqing Xu Suyun Wei Qiaolin Ye Tongming Yin Ning Ye | 2018 | Horticulture Research2018,5,1: | 4 |
| 4 | Numerieal Prediction of Storm Surge in the Qingdao Area Under the Impact of Climate Change显示文摘A typhoon-induced storm surge simulation system was developed for the Qingdao area, including a typhoon diagnostic model for the generation of wind and pressure fields and a 2D Advanced Circulation(ADCIRC) model for simulating the associated storm surge with a 200 m resolution along the Qingdao coastline. The system was validated by an extreme surge event Typhoon Mamie(8509) and the parameters of Typhoon Mamie were used to investigate the sensitivity of typhoon paths to Qingdao storm surges with four selected paths: the paths of Typhoons Mamie(8509), Opal, 3921 and 2413, the selection being made according to their relative position to Qingdao. Experiments based on the Typhoon Mamie(8509) storm surge were also conducted to study the possible influences of future climate changes, including the sea level rise and sea surface temperature(SST) rise, on storm surges along the Qingdao coast. Storm surge conditions under both present day and future(the end of the 21 st century) climate scenarios associated with the four selected paths were simulated. The results show that with the same intensity, when typhoons follow the paths of 3921 and 2413, they would lead to the most serious disasters in different areas of Qingdao. Sea level and SST affect storm surges in different ways: sea level rise affects storm surge mainly through its influence on the tide amplitude, while the increased SST has direct impact on the intensity of the surges. | 9 | Effect of upper contact line on sliding behavior of water droplet on superhydrophobic surface显示文摘With a low surface energy, high apparent contact angle(>150°) and low sliding angle(<5°), superhy- drophobic surface has recently been attracting a great deal of attention. The true factor determining the sliding angle still remains unclear. In this paper, various superhydrophobic silicon surfaces with pillars are fabricated by photolithography and hydrophobized with octadecyltrichlorosilane (OTS). Relations between sliding angles and micro-structured surfaces are being investigated in detail with 10 mg water droplets in C-B state and mixed state. Experimental pictures and data show that the sliding angle is independent of pillar heights from 20 to 80 μm, increasing the space between pillars causes decrease in sliding angle, and increasing the side lengths of pillars causes increase in sliding angle. Moreover, the sliding angle is irrelevant to the state of interfacial contact area of water-solid and lower contact line. | 13 | Identification and analysis of a minority fraction in a U87 cell line Do side-population cells represent bona fide stem cell-like cancer cells?显示文摘BACKGROUND:Overwhelming evidence suggests that tumor bulks are comprised of differentiated tumor cells and cancer stem cells(CSCs).The stem cell-like side-population(SP) cells account for a minor fraction of the total tumor cells,yet are apparently the cells capable of tumor initiation,growth, maintenance,and recurrence. OBJECTIVE:To identify potential stem cell-like cancer cells in a U87 human brain glioma cell line on the basis of dye efflux,clone formation,and multi-drug resistance capacity. DESIGN,TIME AND SETTING:The cellular and molecular biology experiment was performed at the Laboratory of Shanghai Institute of Hematology and Laboratory of Shanghai Institute of Endocrinology in Ruijin Hospital;in vivo contrast observational animal trial was performed at Experimental Animal Center,School of Medicine,Shanghai Jiao Tong University from June 2007 to May 2008. MATERIALS:The U87 cell line was provided by the Shanghai Institute of Cancer Research, Chinese Academy of Science;DMEM/F12(1:1) and fetal bovine serum were purchased from Gibco, Invitrogen,USA;human recombinant basic fibroblast growth factors were purchased from BD Bioscience,USA;Hoechst 33342,Verapamil,and methyl thiazolyl tetrazolium were purchased from Sigma,USA;phycoerythrin-labeled anti-human-CD133 was purchased from Milteny Biotec, Germany;SYBR(?) PrimeScript^(TM) RT-PCR kit was purchased from TaKaRa Biotechnology,Dalian, China. METHODS:Monolayer cultured cells were harvested by 0.25%Trypsin-EDTA and suspended at a 1×10~6/mL dilution in PBS containing 2%FBS,and were stained with Hoechst 33342 dye,either alone or in combination with Verapamil.Following fluorescence-activated cell sorting,SP and non-SP subsets were cultivated with serum-containing(DMEM plus 10%fetal bovine serum) or serum-free culture medium[DMEM/F12(1:1) + 1×B27 supplement + 10 ng/mL basic fibroblast growth factors + 1×L-glutamine]to determine growth characteristics in vitro.Finally,single free U87 cells and subsets(SP or non-SP cells) were subcutaneously injected into the backs of 5-week-old nude mice for in vivo tumorigenicity. MAIN OUTCOME MEASURES:Cell morphology and clonogenicity were observed under inverted microscope;SP phenotype and fluorescent antibody labeling were analyzed by MoFlo^(TM) flow cytometry;ABC transporter mRNA expression was evaluated by semi-quantitative real-time RT-PCR;efflux capacity for anti-neoplastic drugs from the U87 cell line and subsets was measured with the MTT assay,then detected by enzyme-linked immunosorbent assay at a wavelength of 490 nm;in vivo tumorigenicity in immunodeficient nude mice was evaluated by diameter size. RESULTS:During in vitro passages,human U87 cells maintained a stable SP fraction profile and exhibited the ability to form neurosphere-like clones.SP cell proliferation decreased compared with non-treated U87 cells.CD133 expression was reduced in the SP and non-SP cells.Freshly sorted SP fractions expressed higher levels of ABC drug transporter genes,and exhibited increased potential for cytotoxic drug resistance.The in vivo malignancy of U87 cells was largely dependent on non-SP cells in nude mice,and tumors that formed from the non-SP fraction developed faster and larger compared with tumors from the SP fraction.
