| 1 | Quantitative analysis of Pb adsorption on sulfhydryl-modified biochar显示文摘Biochar is an effective absorbent for remediating heavy metal contaminated soil,but functional optimization is still needed to improve its performance in field application.Here,we characterized the physical structures and surface chemical properties of raw wood biochar and palm biochar(WB and PB)and the corresponding sulfhydryl-modified biochar(SWB and SPB).Their adsorption capacity for Pb was evaluated by combining thermodynamic and kinetic adsorption at 0.01 mol/L KCl and corresponding model simulation.The results demonstrated successful grafting of sulfhydryl groups onto the biochar,which dramatically reduced the specific surface area(SSA)and pore volume of biochar.The pKa in the surface complexation model(SCM)indicated similar proton affinity between sulfhydryl groups and original functional groups on the biochar.SCM could satisfactorily fit the Pb adsorption behaviors,and model analysis revealed that Pb tended to be adsorbed on low-proton affin-ity sites at low pH,but high-proton affinity sites became dominant in Pb adsorption with increasing pH and adsorbed almost all Pb ions at pH>7.0.Besides,the Pb adsorption density of SWB and SPB was improved by 8.86 and 3.64 folds relative to that of WB and PB,respectively.Over 90% of initially added Pb ions were removed in 1440 and 720 min by raw and sulfhydryl-modified biochar,respectively,indicating that sulfhydryl modification accelerated the Pb adsorption of biochar.These results suggest that site density,SSA and pore structure of biochar play crucial roles in heavy metal adsorption,and sulfhydryl modification may improve the performance of biochar in remediating heavy metal contaminated soil. | Juan Xiong Mengge Zhou Chenchen Qu Daohai Yu Chang Chen Mingxia Wang Wenfeng Tan | 2021 | Biochar2021,3,1: | 5 |
| 2 | Prevalence and Phylogenetic Analysis of Crimean-Congo Hemorrhagic Fever Virus in Ticks from Different Ecosystems in Xinjiang,China显示文摘The Crimean-Congo hemorrhagic fever virus(CCHFV),a member of the genus Orthonairovirus and family Nairoviridae,is transmitted by ticks and causes severe hemorrhagic disease in humans.To study the epidemiology of CCHFV in different ecosystems in Xinjiang,China,a total of 58,932 ticks were collected from Tarim Basin,Junggar Basin,Tianshan Mountain,and Altai Mountain from 2014 to 2017.Hyalomma asiaticum asiaticum was the dominant tick species in Tarim and Junggar basins,whereas Dermacentor nuttalli and Hyalomma detritum were found in Tianshan Mountain and Altai Mountain,respectively.Reverse transcription-polymerase chain reaction of the CCHFV small(S)genome segment was used for the molecular detection.The CCHFV-positive percentage was 5.26%,6.85%,1.94%,and 5.56% in Tarim Basin,Junggar Basin,Tianshan Mountain,and Altai Mountain,respectively.Sequences of the S segment were used for phylogenetic analysis and the results showed that the newly identified CCHFV strains belonged to two clades.Our study confirms that H.asiaticum asiaticum is the major vector of CCHFV in desert habitats which is consistent with previous studies,and also suggests that H.detritum and D.nuttalli are emerging vectors for CCHFV in Xinjiang.Moreover,this study reports the presence of CCHFV in the mountain habitat of Xinjiang for the first time,suggesting that future surveillance of CCHFV should also include mountainous areas. | Abulimiti Moming Xihong Yue Shu Shen Chenchen Chang Cheng Wang Tao Luo Yanfang Zhang Rong Guo Zhihong Hu Yujiang Zhang Fei Deng Surong Sun | 2018 | Virologica Sinica2018,33,1: | 5 |
| 3 | Abnormal meiosis in fertile and sterile triploid cyprinid fish显示文摘Meiosis is the key process for producing mature gametes. A natural fertile triploid Carassius auratus population(3nDTCC) and an artificially derived sterile triploid crucian carp(3nCC) have been previously observed, providing suitable model organisms for investigating meiosis characteristics in triploid fish. In the present study, the microstructures and ultrastructures of spermatogenesis were studied in these fishes. Td T-mediated d UTP nick end labeling detection was performed to investigate the apoptosis of spermatocytes. Fluorescence in situ hybridization was employed to trace chromatin pairing. In addition, the m RNA expressions of cell cycle-related genes(i.e., cell division control 2 and cell cycle protein B) were determined by quantitative realtime polymerase chain reaction to illustrate the molecular mechanism of abnormal meiosis in the 3nCC. The results showed that the 3nCC undergoes an irregular prophase I, with the chromosomes distributed in a unipolar radial manner and exhibiting partial pairing, hindered metaphase I, and degenerated cells in the subsequent stages. Meanwhile, the 3nDTCC presented a relatively regular meiotic prophase I with complete conjugate chromosome pairs and chromosomes distributed along the karyotheca,which were presented as a ring structure by slicing. Only the spreads with 130–150 irregular chromosomes can be easily detected in the 3nDTCC, suggesting that it may undergo an abnormal metaphase I. | 4 | Effects of plant diversity on greenhouse gas emissions in microcosms simulating vertical constructed wetlands with high ammonium loading显示文摘Wastewater with relatively high nitrogen concentrations is a major source of nitrous oxide(N_2O) and methane(CH_4) emissions and exerts multiple stresses on the environment.Studies have shown that plant diversity plays an important role in ecosystem functioning.However, the effects of plant species diversity on CH_4 and N_2O emissions under high ammonium(NH_4^+-N) loading rates remain unclear. In this study, a microcosm experiment simulating vertical constructed wetlands supplied with high NH_4^+-N water levels was established. The treatments included four species richness levels(1, 2, 3, 4) and 15 species compositions. There was no significant relationship between species richness and N_2O emissions. However, N_2O emissions were significantly reduced by specific plant species composition. Notably, the communities with the presence of Rumex japonicus L. reduced N_2O emissions by 62% compared to communities without this species. This reduction in N_2O emissions may have been a result of decreased N concentrations and increased plant biomass. CH_4 emissions did not respond to plant species richness or species identity.Overall, plant species identity surpassed species richness in lowering N_2O emissions from constructed wetlands with high NH_4^+-N water. | 6 | An improved hybrid bream derived from a hybrid lineage of Megalobrama amblycephala(♀)×Culter alburnus(♂)显示文摘Distant hybridization is an important technique in fish genetic breeding.In this study,based on the establishment of an allodiploid fish lineage(BT,2n=48,F_(1)–F_(6))derived from distant hybridization between female Megalobrama amblycephala(BSB,2n=48)and male Culter alburnus(TC,2n=48),and the backcross progeny(BTB,2n=48)derived by backcrossing female F_(1) of BT to male BSB,an improved hybrid bream(BTBB,2n=48)was obtained by backcrossing BTB(♀)to BSB(♂).Moreover,the morphological and genetic characteristics of BTBB individuals were investigated;BTBB was similar to BSB in appearance but had a higher body height than BSB.The study results regarding chromosome numbers and DNA content indicated that BTBB is a diploid hybrid fish.The 5S rDNA and Hox gene of BTBB were inherited from the original parents.Gonadal development in BTBB was normal.On the other hand,BTBB had a faster growth rate,higher muscle protein level,and lower muscle carbohydrate level than BSB.Hence,bisexual fertile BTBB is promoted and can be applied as a high-quality fish,and it can also be used as a new fish germplasm resource to develop high-quality fish further.Thus,this study is of great significance for fish genetic breeding. | Dingbin Gong Min Tao Lihui Xu Fangzhou Hu Zehong Wei Shi Wang Yude Wang Qingfeng Liu Chang Wu Kaikun Luo Chenchen Tang Rong Zhou Chun Zhang Yuequn Wang Shaojun Liu | 2022 | Science China(Life Sciences)2022,65,6: | 0 |
| 7 | Metabolic modulation of acetaminophen-induced hepatotoxicity by osteopontin显示文摘Induction of osteopontin(OPN),a well-known pro-inflammatory molecule,has been observed in acetaminophen(APAP)-induced hepatotoxicity.However,the precise cell source for OPN induction and its role during APAP-induced hepatotoxicity has not been fully explored.By employing a hepatotoxic mouse model induced by APAP overdose,we demonstrate that both serum and hepatic OPN levels were significantly elevated in response to APAP treatment.Our in vivo and in vitro studies clearly indicated that the induced expression of hepatic OPN was mainly located in necrosis areas and produced by dying or dead hepatocytes.Functional experiments showed that OPN deficiency protected against the APAP-induced liver injury by inhibiting the toxic APAP metabolism via reducing the expression of the cytochrome P450 family 2 subfamily E member 1(CYP2E1).Interestingly,this inhibition of CYP2E1 expression did not occur in unfasted Opn−/−mice,but was significant in fasted Opn−/−mice and maintained for 2hours after APAP challenge in fasted Opn−/−mice.In addition,despite the early protective role of OPN deficiency on APAP-induced hepatotoxicity,OPN deficiency retarded injury resolution by sensitizing hepatocytes to apoptosis and impairing liver regeneration.Finally,we demonstrated that a siRNA-mediated transient hepatic Opn knockdown could sufficiently and significantly protect animals from APAP-induced hepatotoxicity and death.In conclusion,this study clearly defines the cell source of OPN induction in response to APAP treatment,provides a novel insight into the metabolic role of OPN to APAP overdose,and suggests an Opn-targeted therapeutic strategy for the treatment or prevention of APAP-induced hepatotoxicity. | Yankai Wen Chenchen Wang Jinyang Gu Chang Yu Kaixia Wang Xuehua Sun Yun Sun Hailong Wu Ying Tong Qiang Xia Xiaoni Kong | 2019 | Cellular & Molecular Immunology2019,16,5: | 0 |
