| 1 | Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally.Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans.Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed.Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever,cough,and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation.One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients,with 99.8%to 99.9%nucleotide identities among the isolates.These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity,but is in a separate clade.Moreover,these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However,the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor.Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans. | Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang | 2020 | Chinese Medical Journal2020,,9: | 99 |
| 2 | Evaluation of Tubal Patency with Transvaginal Three-dimensional Hysterosalpingo-contrast Sonography显示文摘Objective To investigate diagnostic efficacy of transvaginal three-dimensional hysterosalpingo-contrast sonography(3D-Hy Co Sy) in assessing tubal patency with chromolaporoscopy. Methods A total of 157 infertile women underwent 3D-Hy Co Sy to evaluate tubal patency. Among these patients, 39 patients were also examined by chromolaporoscopy. The concordance of the two clinical assessment methods was analyzed by the Kappa coefficient test. Results Among the 306 oviducts examined by 3D-Hy Co Sy, 99(32.4%) were patent, 126(41.2%) partially obstructed, and 81(26.5%) completely obstructed. Diagnostic results with 3D-Hy Co Sy were not statistically different from those obtained in the 39 women(78 oviducts) who also underwent chromolaporoscopy, and the two methods showed a high concordance(κ=0.747, P=0.000). The 3D-Hy Co Sy procedure had a sensitivity of 84.8%(28/33), a specificity of 96.2%(25/26), and positive and negative predictive values of 93.3%(28/30) and 86.2%(25/29) respectively. | 3 | Secretory/releasing proteome-based identification of plasma biomarkers in HBV-associated hepatocellular carcinoma显示文摘For successful therapy, hepatocellular carcinoma (HCC) must be detected at an early stage. Herein, we used a proteomic approach to analyze the secretory/releasing proteome of HCC tissues to identify plasma biomarkers. Serum-free conditioned media (CM) were collected from primary cultures of cancerous tissues and surrounding noncancerous tissues. Proteomic analysis of the CM proteins permitted the identification of 1365 proteins. The enriched molecular functions and biological processes of the CM proteins, such as hydrolase activity and catabolic processes, were consistent with the liver being the most important metabolic organ. Moreover, 19% of the proteins were characterized as extracellular or membrane-bound. For validation, secretory proteins involved in transforming growth factor-β signaling pathways were validated in plasma samples. Alphafetoprotein (AFP), metalloproteinase (MMP)1, osteopontin (OPN), and pregnancy-specific beta-1-glycoprotein (PSG)9 were significantly increased in HCC patients. The overall performance of MMP1 and OPN in the diagnosis of HCC remained greater than that of AFP. In addition, this study represents the first report of MMP1 as a biomarker with a higher sensitivity and specificity than AFP. Thus, this study provides a valuable resource of the HCC secretome with the potential to investigate serological biomarkers. | 7 | Computer-aided ionic liquid design for alkane/cycloalkane extractive distillation process显示文摘A computer-aided ionic liquid design(CAILD) study is presented for the frequently encountered alkane/cycloalkane separations in petrochemical industry. Exhaustive experimental data are first collected to extend the UNIFAC-IL model for this system, where the proximity effect in alkanes and cycloalkanes is considered specifically by defining distinct groups. The thermodynamic performances of a large number of ILs for 4 different alkane/cycloalkane systems are then compared to select a representative example of such separations. By applying n-heptane/methylcyclohexane extractive distillation as a case study, the CAILD task is cast as a mixed-integer nonlinear programming(MINLP) problem based on the obtained task-specific UNIFAC-IL model and two semi-empirical models for IL physical properties. | 9 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 10 | The Variation of Stone Cell Content in 236 Germplasms of Sand Pear(Pyrus pyrifolia)and Identification of Related Candidate Genes显示文摘Stone cells have been described to substantially influence pear fruit quality,as lignin and cellulose are the main components of stone cells.However,there are limited studies on the relationship between the variation and molecular basis of stone cells,lignin and cellulose content among different pear varieties.Here,to reveal the variation of stone cell content within different cultivated species,we collected 236 germplasms of sand pear(Pyrus pyrifolia)at 50 days after flower blooming(DAFB),the key stage of stone cell formation.In our results,we measured the content of stone cells,lignin and cellulose and found that these contents ranged from2.82%to 29.00%,8.84%to 55.30%and 11.52%to 30.55%,respectively.Further analysis showed that the variation coefficient of stone cell,lignin and cellulose content was 39.10%,28.03%and 16.71%,respectively.Additionally,a significant correlation between stone cell,lignin and cellulose content were detected,and the correlation coefficient between the contents of stone cell and lignin(0.912)was higher than between the contents of stone cell and cellulose(0.796).Moreover,the average lignin content(29.73%)was higher than the average cellulose content(18.03%)in stone cells in pear fruits,indicating that lignin is the main component of stone cell in pears.Finally,on the basic of the transcriptome data,we identified 10 transcription factors belonging to bHLH,ERF,MYB,and NAC transcript families,which might be involved in lignin formation in stone cells.qRT-PCR experiments verified coincident trends between expression of candidate genes and stone cell content.This research laid foundation for future studies on genetic variation of stone cells in pear fruits and provided important gene resources for stone cell regulation. | Jiaying Zhang Jiaming Li Cheng Xue Runze Wang Mingyue Zhang Kaijie Qi Jing Fan Hongju Hu Shaoling Zhang Jun Wu | 2021 | Horticultural Plant Journal2021,7,2: | 3 |

| 17 | A chromosome-level genome assembly of rugged rose (Rosa rugosa) provides insights into its evolution, ecology, and floral characteristics显示文摘Rosa rugosa,commonly known as rugged rose,is a perennial ornamental shrub.It produces beautiful flowers with a mild fragrance and colorful seed pods.Unlike many other cultivated roses,R.rugosa adapts to a wide range of habitat types and harsh environmental conditions such as salinity,alkaline,shade,drought,high humidity,and frigid temperatures.Here,we produced and analyzed a high-quality genome sequence for R.rugosa to understand its ecology,floral characteristics and evolution.PacBio HiFi reads were initially used to construct the draft genome of R.rugosa,and then Hi-C sequencing was applied to assemble the contigs into 7 chromosomes.We obtained a 382.6Mb genome encoding 39,704 protein-coding genes.The genome of R.rugosa appears to be conserved with no additional whole-genome duplication after the gamma whole-genome triplication(WGT),which occurred~100 million years ago in the ancestor of core eudicots.Based on a comparative analysis of the high-quality genome assembly of R.rugosa and other high-quality Rosaceae genomes,we found a unique large inverted segment in the Chinese rose R.chinensis and a retroposition in strawberry caused by post-WGT events.We also found that floral development-and stress response signaling-related gene modules were retained after the WGT.Two MADS-box genes involved in floral development and the stress-related transcription factors DREB2A-INTERACTING PROTEIN 2(DRIP2)and PEPTIDE TRANSPORTER 3(PTR3)were found to be positively selected in evolution,which may have contributed to the unique ability of this plant to adapt to harsh environments.In summary,the high-quality genome sequence of R.rugosa provides a map for genetic studies and molecular breeding of this plant and enables comparative genomic studies of Rosa in the near future. | Fei Chen Liyao Su Shuaiya Hu Jia-Yu Xue Hui Liu Guanhua Liu Yifan Jiang Jianke Du Yushan Qiao Yannan Fan Huan Liu Qi Yang Wenjie Lu Zhu-Qing Shao Jian Zhang Liangsheng Zhang Feng Chen Zong-Ming(Max)Cheng | 2021 | Horticulture Research2021,8,1: | 2 |

| 19 | Measurement of the neutron total cross section of carbon at the Back-n white neutron beam of CSNS显示文摘To verify the performance of the neutron total cross-sectional spectrometer, the neutron total cross section of carbon is initially measured in the energy range of 1 eV to 20 MeV using the time-of-flight method. The measurement is performed at the Back-n white neutron source with a 76-m time-of-flight path using the China Spallation Neutron Source. A multilayer fast fission chamber with 235U and 238U is employed as the neutron detector. The diameter and thickness of the natural graphite sample are 70 mm and 40 mm, respectively. Signal waveforms are collected using a data acquisition system. Off-line data processing was used to obtain the neutron time-of-flight spectra and transmissions. The uncertainty of the counting statistics is generally approximately 3% for each bin in the energy range of 1–20 MeV. It is determined that the results for the neutron total cross section of carbon obtained using ^235U cells are in good agreement with the results obtained using 238U cells within limits of statistical uncertainty. Macroinitiators prepared by the esterification between poly(ethylene oxide) (PEG) and 2,2-dichloroacetyl chloride were utilized to initiate the polymerization of 6-[4-(4-ethoxyphenylazo)phenoxy]hexyl methacrylate (M6C). The resulting macroinitiators and block copolymers were characterized by 1H NMR, gel permeation chromatography (GPC). Polarizing optical microscopy (POM) and differential scanning calorimetry (DSC) preliminarily revealed the liquid crystalline property of these block copolymers.
