| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | A probiotic treatment containing Lactobacillus,Bifidobacterium and Enterococcus improves IBS symptoms in an open label trial显示文摘Objective: To evaluate the efficacy and safety of live combined Bifidobacterium, Lactobacillus and Enterococcus capsules in treatment of irritable bowel syndrome. Methods: Eighty-five patients [male 32, female 53; age (45.31±11.72) years] were given live combined Bifidobacterium, Lactobacillus and Enterococcus capsules 1260 mg/d t.i.d.×4 weeks. Syndrome scales were used to evaluate the efficacy in gastrointestinal syndrome. Fecal flora was also measured before and after the treatment. Six bacteria were cultured and the colony forming units were counted in stool. SPSS was used for data analysis. Results: Seventy-four patients finished the follow-up. No side-effect was found. For treatment of irritable bowel syndrome, the effective rate of live combined Bifidobacterium, Lactobacillus and Enterococcus capsules was 56.8% in the second week, 74.3% in the fourth week and 73.0% in the sixth week. Single symptom was improved, especially in abdominal pain and stool character. The probiotica con- taining live combined Bifidobacterium, Lactobacillus and Enterococcus could increase bifidobacterium count (P<0.01) and lac- tobacillus count (P<0.05); decrease bacteroides count (P<0.05) and enterococci count (P<0.01); No obvious changes were ob- served in clostridium difficile colonitis and enterobacteriaceae (P>0.05). Conclusion: The result of the study indicated that the administration of live combined Bifidobacterium, Lactobacillus and Enterococcus improved the symptom of irritable bowel syndrome and that there was a gradual increase of this effect. Thereafter conditions remained stable for 2 weeks. | 5 | Effects of nanobubble and hydrodynamic parameters on coarse quartz flotation显示文摘Quartz, the second most abundant mineral in the earth's crust, is a gangue mineral in practically every flotation process. Coarse quartz flotation has been a long standing problem in various mineral processing plants to reduce milling cost and increase valuable mineral recovery. Based on this, the effects of nanobubbles(NBs) and hydrodynamic parameters on coarse quartz particle flotation were systematically investigated. Mechanical flotation experiments were carried out using the 7 cm and 9 cm diameter impellers in order to produce different hydrodynamic conditions. 900–1300 rpm impeller speeds were used for the 7 cm diameter impeller and 554–786 rpm for the 9 cm diameter impeller. The results show that the presence of NBs increased the flotation recovery of à425 + 106 lm quartz by up to 21%. For the7 cm diameter impeller, the maximum flotation recoveries of 86.4% and 98% were obtained in the absence and presence of NBs at Reynolds number(Re) of 81,000 and 66,000, respectively. For the 9 cm diameter impeller, the maximum recoveries of 86.3% and 97.5% were obtained in the absence and presence of NBs at Re of 90,000 and 75,000, respectively. | 7 | Prognostic values of the integrated model incorporating the volume of metastatic regional cervical lymph node and pretreatment serum Epstein-Barr virus DNA copy number in predicting distant metastasis in patients with N1 nasopharyngeal carcinoma显示文摘Background: According to the 7 th edition of the American Joint Committee on Cancer(AJCC) staging system, over50% of patients with nasopharyngeal carcinoma(NPC) have N1 disease at initial diagnosis. However, patients with N1 NPC are relatively under-researched, and the metastasis risk of this group is not well-stratified. This study aimed to evaluate the prognostic values of gross tumor volume of metastatic regional lymph node(GTVnd) and pretreatment serum copy number of Epstein-Barr virus(EBV) DNA in predicting distant metastasis of patients with N1 NPC, and to develop an integrated prognostic model that incorporates GTVnd and EBV DNA copy number for this group of patients.Methods: The medical records of 787 newly diagnosed patients with nonmetastatic, histologically proven N1 NPC who were treated at Sun Yat-sen University Cancer Center between November 2009 and February 2012 were analyzed. Computed tomography-derived GTVnd was measured using the summation-of-area technique. Blood samples were collected before treatment to quantify plasma EBV DNA. The receiver operating characteristic(ROC) curve analysis was used to evaluate the cut-off point for GTVnd, and the area under the ROC curve was used to assess the predicted validity of GTVnd. The survival rates were assessed by Kaplan-Meier analysis, and the survival curves were compared using a log-rank test. Multivariate analysis was conducted using the Cox proportional hazard regression model.Results: The 5-year distant metastasis-free survival(DMFS) rates for patients with GTVnd > 18.9 vs.≤ 18.9 mL were82.2% vs. 93.2%(P < 0.001), and for patients with EBV DNA copy number > 4000 vs. < 4000 copies/mL were 83.5% vs.93.9%(P < 0.001). After adjusting for GTVnd, EBV DNA copy number, and T category in the Cox regression model, both GTVnd > 18.9 mL and EBV DNA copy number > 4000 copies/mL were significantly associated with poor prognosis(both P < 0.05). According to combination of GTVnd and EBV DNA copy number, all patients were divided into low-,moderate-, and high-risk groups, with the 5-year DMFS rates of 96.1,87.4, and 73.8%, respectively(P < 0.001). Multivariate analysis confirmed the prognostic value of this model for distant metastatic risk stratification(hazard ratio [HR],4.17; 95% confidence interval [CI] 2.34-7.59; P < 0.001).Conclusions: GTVnd and serum EBV DNA copy number are independent prognostic factors for predicting distant metastasis in NPC patients with N1 disease. | 9 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 12 | Biofilm Disruption Utilizing α/β Chimeric Polypeptide Molecular Brushes显示文摘Gram-negative bacteria can cause serious infections and are well known problems in biomedical practices. Biofilms of gramnegative bacteria are notorious for their frequently encountered resistance toward antibiotics. We demonstrate that α/β chimeric polypeptide molecular brush (α/β CPMB) exerts potent activities against antibiotic-resistant gram-negative bacteria. MTT viability assay, bacterial colony counting, and live/dead staining all indicate that α/β CPMB not only inhibits biofilm formation of gram-negative Pseudomonas aeruginosa and Acinetobacter baumannii, but also effectively disrupts mature biofllms that are highly resistant to one of the most active antibiotics-colistin. | 13 | Age-specific differences in non-cardiac comorbidities among elderly patients hospitalized with heart failure: a special focus on young-old, old-old, and oldest-old显示文摘Background:Despite the growing epidemic of heart failure(HF),there is limited data available to systematically compare noncardiac comorbidities in the young-old,old-old,and oldest-old patients hospitalized for HF.The precise differences will add valuable information for better management of HF in elderly patients.Methods:A total of 1053 patients aged 65 years or older hospitalized with HF were included in this study.Patients were compared among three age groups:(1)young-old:65 to 74 years,(2)old-old:75 to 84 years,and(3)oldest-old:≥85 years.Clinical details of presentation,comorbidities,and prescribed medications were recorded.Results:The mean age was 76.7 years and 12.7%were 85 years or older.Most elderly patients with HF(97.5%)had at least one of the non-cardiac comorbidities.The patterns of common non-cardiac comorbidities were different between the young-old and oldestold group.The three most common non-cardiac comorbidities were anemia(53.6%),hyperlipidemia(45.9%),and diabetes(42.4%)in the young-old group,while anemia(73.1%),infection(58.2%),and chronic kidney disease(44.0%)in the oldest-old group.Polypharmacy was observed in 93.0%elderly patients with HF.Additionally,29.2%patients were diagnosed with infection,and 67.0%patients were prescribed antibiotics.However,60.4%patients were diagnosed with anemia with only 8.9%of them receiving iron repletion.Conclusions:Non-cardiac comorbidities are nearly universal in three groups but obviously differ by age,and inappropriate medications are very common in elderly patients with HF.Further treatment strategies should be focused on providing optimal medications for age-specific non-cardiac conditions. | Meng-Xi Yang Hui An Xue-Qiang Fan Li-Yuan Tao Qiang Tu Li Qin Li-Fang Zhang Dong-Ping Feng Yu Wang Li Sun Si Gao Wen-Zhuo Guan Jin-Gang Zheng Jing-Yi Ren | 2019 | Chinese Medical Journal2019,,24: | 4 |

| 19 | First genetic assessment of brackish water polychaete Tylorrhynchus heterochaetus: mitochondrial CO/sequences reveal strong genetic differentiation and population expansion in samples collected from southeast China and north Vietnam显示文摘Tylorrhynchus heterochaetus is a widespread benthic polychaete worm found in coastal brackish waters of the west Pacific.It has high ecological and economic value as a biomarker of water quality and as a high-quality feed in aquaculture and fisheries and is considered a delicacy in some areas of Asia.However,it has experienced a marked reduction in recent years due to overexploitation as well as changes in the environment and climate.Here,to comprehensively understand its genetic background and thus provide insights for better conservation and utilization of this species,we assessed the genetic variability and demographic history of T.heterochaetus individuals sampled from eight locations along the coasts of southeast China and north Vietnam based on mitochondrial cytochrome c oxidase I(COI)sequences.We observed high haplotype diversity(Hd),with an average of 0.926,but relatively low nucleotide diversity(π),with a mean of 0.032 across all samples.A total of 94 polymorphic sites and 85 haplotypes were identified among 320 individuals.The pairwise genetic distances among haplotypes ranged from 0.001 to 0.067,with the high intraspecific divergence possibly reflecting geographic isolation and gene pool fragmentation.Significant genetic structures were revealed among the studied locations;specifically,the eight locations could be treated as six genetically different populations based on pairwiseΦST results(0.026–0.951,P<0.01).A significant pattern of isolation-by-distance was detected between the genetic and geographic distances(r=0.873,P=0.001).Three geographic lineages were defined based on phylogenetic tree and network analyses of COI haplotypes.AMOVA results indicated that genetic variations mainly occurred among the three lineages(89.96%).Tests of neutrality and mismatch distribution suggested that T.heterochaetus underwent recent population expansion.These results provide the first report on the genetic status of T.heterochaetus and will be valuable for the management of genetic resources and better understanding of the ecology and evolution in this species. | Xing-Han Chen Sen Yang Wei Yang Yuan-Yuan Si Rui-Wen Xu Bin Fan Le Wang Zi-Ning Meng | 2020 | Zoological Research2020,41,1: | 3 |
| 20 | Fault Analysis on a New Block Cipher DBlock with at Most Two Fault Injections显示文摘DBlock is a new family of block ciphers proposed by Wu et al. in Science China in 2015, which consists of three variants specified as DBlock-128/192/256.DBlock-n employs a 20-round Feistel-type structure with n-bit block size and n-bit key size. We propose the first fault analysis on DBlock and show that no more than 2 pairs of correct/faulty ciphertexts are needed to retrieve the master key. In the attack, a byte-oriented fault is injected in round 16, and three properties including differential distribution of the Sbox, bijection nature of the linear function and Feistel-type key scheduling are fully utilized to distinguish between the correct and wrong keys. A fault position guessing strategy based on known intermediates is adopted, which efficiently makes the known-fault attack apply to the random fault model. The experimental results show that, with a pair of ciphertexts, 11.820-bit exhaustive search is needed to derive the whole 128-bit key on average.
