| 2 | Inactivation of SACE_3446, a TetR family transcriptional regulator, stimulates erythromycin production in Saccharopolyspora erythraea显示文摘Erythromycin A is a widely used antibiotic produced by Saccharopolyspora erythraea;however,its biosynthetic cluster lacks a regulatory gene,limiting the yield enhancement via regulation engineering of S.erythraea.Herein,six TetR family transcriptional regulators(TFRs)belonging to three genomic context types were individually inactivated in S.erythraea A226,and one of them,SACE_3446,was proved to play a negative role in regulating erythromycin biosynthesis.EMSA and qRT-PCR analysis revealed that SACE_3446 covering intact N-terminal DNA binding domain specifically bound to the promoter regions of erythromycin biosynthetic gene eryAI,the resistant gene ermE and the adjacent gene SACE_3447(encoding a longchain fatty-acid CoA ligase),and repressed their transcription.Furthermore,we explored the interaction relationships of SACE_3446 and previously identified TFRs(SACE_3986 and SACE_7301)associated with erythromycin production.Given demonstrated relatively independent regulation mode of SACE_3446 and SACE_3986 in erythromycin biosynthesis,we individually and concomitantly inactivated them in an industrial S.erythraea WB.Compared with WB,the WBΔ3446 and WBΔ3446Δ3986 mutants respectively displayed 36%and 65%yield enhancement of erythromycin A,following significantly elevated transcription of eryAI and ermE.When cultured in a 5 L fermentor,erythromycin A ofWBΔ3446 and WBΔ3446Δ3986 successively reached 4095 mg/L and 4670 mg/L with 23%and 41%production improvement relative to WB.The strategy reported here will be useful to improve antibiotics production in other industrial actinomycete. | Hang Wu Yansheng Wang Li Yuan Yongrong Mao Weiwei Wang Lin Zhu Panpan Wu Chengzhang Fu Rolf Muller David T.Weaver Lixin Zhang Buchang Zhang | 2016 | Synthetic and Systems Biotechnology2016,1,1: | 7 |
| 3 | Design and simulation of an innovative cylinder fabricated by selective laser melting显示文摘Additive Manufacturing(AM) processing has attracted wide attention due to its advantages, e.g., rapid prototyping and free design. In aeronautic and astronautic engineering, it is especially attractive to apply AM processing, especially Selective Laser Melting(SLM), into primary structures that always occupy most of the whole mass of aircraft or spacecraft. In this paper, an innovative lattice sandwich structure that is suitable to be manufactured with SLM is adopted to design the typical cylinder under axial compression. Firstly, the mechanical properties of pyramidal lattice are investigated, and the bearing capacity of the lattice sandwich cylinder is given analytically for each failure mode. Secondly, a composite cylinder is re-designed in the form of lattice sandwich structure, which sizes are ultimately obtained through optimization. Finally, finite element model of the innovative cylinder is established and several kinds of imperfection are introduced to simulate the defects caused by SLM. Numerical result shows that the lattice sandwich design can obviously enhance the bearing capacity of cylindrical shell even though the original one is designed with composite material. | 4 | Tissue engineering using 3D printed nano-bioactive glass loaded with NELL1 gene for repairing alveolar bone defects显示文摘The purposes of this study were to construct a novel tissue engineered bone composed of 3Dprinted bioactive glass block/chitosan nanoparticles(BD/CSn)composites loaded with Nel-like Type I molecular-1 DNA(pDNA-NELL1)and/or bone marrow mesenchymal stem cells(BMSCs),and study their osteogenic activities by repairing bone defects in rhesus monkeys.CSn with NELL1 gene plasmid and rhesus monkey BMSCs were composited with a BD scaffold to prepare the tissue-engineered bone.Four adult female rhesus monkeys with 10-to 12-years old and 5-7 kg in weight were used in animal experiments.The first and second premolar teeth from four regions of each monkey were removed to form bone defects with size of 10×10×5 mm,which were then implanted with above-mentioned tissue engineered bone.At 12 weeks after the implantation,gross observations,X-ray and micro-CT observations revealed that the new bone was extremely close to normal bone in mass,density,hardness,and structure.The bony cortex was smooth and closely connected to the surrounding normal bone.Histological observations revealed moderate inflammation in the repair area,and the new bone tissues were similar to normal ones.In conclusion,tissue engineered bone of this study exhibited good osteoconductivity for promoting the formation of new alveolar bone tissue,and NELL1 gene played a promotional role in bone regeneration. | Jing Zhang Yang Chen Jing Xu Jingjing Wang Chengzhang Li Liyan Wang | 2018 | Regenerative Biomaterials2018,5,4: | 6 |
| 5 | Pt3Ag alloy wavy nanowires as highly effective electrocatalysts for ethanol oxidation reaction显示文摘Direct ethanol fuel cell(DEFC)has received tremendous research interests because of the more convenient storage and transportation of ethanol vs.compressed hydrogen.However,the electrocatalytic ethanol oxidation reaction typically requires precious metal catalysts and is plagued with relatively high over potential and low mass activity.Here we report the synthesis of Pt3Ag alloy wavy nanowires via a particle attachment mechanism in a facile solvothermal process.Transmission microscopy studies and elemental analyses show highly wavy nanowire structures with an average diameter of 4.6±1.0 nm and uniform Pt3Ag alloy formation.Electrocatalytic studies demonstrate that the resulting alloy nanowires can function as highly effective electrocatalysts for ethanol oxidation reactions(EOR)with ultrahigh specific activity of 28.0 mA/cm^2 and mass activity of 6.1 A/mg,far exceeding that of the commercial Pt/carbon samples(1.10 A/mg).The improved electrocatalytic activity may be partly attributed to partial electron transfer from Ag to Pt in the Pt3Ag alloy,which weakens CO binding and the CO poisoning effect.The one-dimensional nanowire morphology also contributes to favorable charge transport properties that are critical for extracting charge from catalytic active sites to external circuits.The chronoamperometry studies demonstrate considerably improved stability for long term operation compared with the commercial Pt/C samples,making the Pt3Ag wavy nanowires an attractive electrocatalyst for EOR. | Xiaoyang Fu Chengzhang Wan Aixin Zhang Zipeng Zhao Huaixun Huyan Xiaoqing Pan Shuaijing Du Xiangfeng Duan Yu Huang | 2020 | Nano Research2020,13,5: | 5 |
| 6 | Catalytic effect of Ni@rGO on the hydrogen storage properties of MgH2显示文摘Uniform-uispersed Ni nanoparticics(NPs)anchored on reduced graphene oxide(Ni@rGO)catalyzed MgH2(MH-Ni@rGO)has been fabricated by mechanical milling.The effects of milling time and Ni loading amount on the hydrogen storage properties of MgH2 have been investigated.The initial hydrogen desorption temperature of MgH2 catalyzed by 10 wt.%Ni4@rGO6 for milling 5 h is significantly decreased from 251℃ to 190℃.The composite can absorb 5.0 wt.%hydrogen in 20 min at 100℃,while it can desorb 6.1 wt.%within 15 min at 300℃.Through the investigation of the phase transformation and dehydrogenation kinetics during hydrogen ab/desorption cycles,we found that the in-situ formed Mg2Ni/Mg2NiH4 exhibited better catalytic effect than Ni.When Ni loading amount is 45 wt.%,the rGO in Ni@rGO catalysts can prevent the reaction of Ni and Mg due to the strong interaction between rGO and Ni NPs. | Pengyang Yao Ying Jiang Yang Liu Chengzhang Wu Kuo-Chih Chou Tao Lyu Qian Li | 2020 | Journal of Magnesium and Alloys2020,8,2: | 4 |
| 7 | High Potential Columnar Nanocrystalline AlN Films Deposited by RF Reactive Magnetron Sputtering显示文摘Columnar nanocrystalline aluminum nitride(cnc-AlN) thin films with(002) orientation and uniform texture have been deposited successfully on large silicon wafers by RF reactive magnetron sputtering.At the optimum sputtering parameters, the deposited cnc-AlN thin films show a c-axis preferred orientation with a crystallite size of about 28 nm and surface roughness(RMS) of about 1.29 nm. The cnc-AlN thin films were well transparent with an optical band gap about 4.8 e V, and the residual compressive stress and the defect density in the film have been revealed by Ramon spectroscopy. | 8 | Ultrathin wavy Rh nanowires as highly effective electrocatalysts for methanol oxidation reaction with ultrahigh ECSA显示文摘Direct methanol fuel cells (DMFCs) have received tremendous research interests because of the facile storage of liquid methanol vs.hydrogen.However,the DMFC today is severely plagued by the poor kinetics and rather high overpotential in methanol oxidation reaction (MOR).Here we report the investigation of the ultrathin Rh wavy nanowires as a highly effective MOR electrocatalyst.We show that ultrathin wavy Rh nanowires can be robustly synthesized with 2-3 nm diameters.Electrochemical studies show a current peak at the potential of 0.61 V vs.reversible hydrogen electrode (RHE),considerably lower than that of Pt based catalysts (~ 0.8-0.9 V vs.RHE).Importantly,with ultrathin diameters and favorable charge transport,the Rh nanowires catalysts exhibit an ultrahigh electrochemically active surface area determined from CO-stripping (ECSAco) of 144.2 m2/g,far exceeding that of the commercial Rh black samples (20 m2/g).Together,the Rh nanowire catalysts deliver a mass activity of 722 mA/mg at 0.61 V,considerably higher than many previously reported electrocatalysts at the same potential.The chronoamperometry studies also demonstrate good stability and CO-tolerance compared with the Rh black control sample,making ultrathin Rh wavy nanowires an attractive electrocatalyst for MOR. | Xiaoyang Fu Zipeng Zhao Chengzhang Wan Yiliu Wang Zheng Fan Frank Song Bocheng Cao Mufan Li Wang Xue Yu Huang Xiangfeng Duan | 2019 | Nano Research2019,12,1: | 3 |
| 9 | Preparation of composite poly(ether block amide)membrane for CO_2 capture显示文摘In this study, a poly(ether block amide)(Pebax 1657) composite membrane applied for CO2 capture was prepared by coating Pebax 1657 solution on polyacrylonitrile(PAN) ultrafiltration membrane. Ethanol/water mixture was used as the solvent of Pebax and the effects of ethanol/water mass ratios and Pebax concentration on the permeation properties of composite membrane were studied. To enhance the composite membrane permeance, the gutter layer, made from reactive amino silicone crosslinking with polydimethylsiloxane(PDMS), was designed. The influence of crosslinking degree of the gutter layer on membrane performance was investigated. As a result, a Pebax/aminoPDMS/PAN multilayer membrane with hexane resistance was developed, showing CO2 permeance of 350 GPU and CO2/N2 selectivity over50. The blend of polyethylene glycol dimethyl ether(PEG-DME) with Pebax as coating material was studied to further improve the membrane performance. The framework principally involves two phases,namely video analysis and crime modeling phases. In video analysis a concept based approach for video event detection is used to detect crime indicator events. Based on the extracted indicators with other related variables, a fuzzy inference system capable of learning is constructed in the second phase. | 12 | An Ensemble Retinal Vessel Segmentation Based on Supervised Learning in Fundus Images显示文摘An ensemble method based on supervised learning for segmenting the retinal vessels in color fundus images is proposed on the basis of previous work of Zhu et al. For each pixel, a 36 dimensional feature vector is extracted, including local features, morphological transformation with multi-scale and multi-orientation, and divergence of vector field which is firstly used to extract feature of retinal image pixels. Then the feature vector is used as input data set to train the weak classifiers by the Classification and regression tree(CART). Finally, an Ada Boost classifier is constructed by iteratively training for the retinal vessels segmentation. The experimental results on the public Digital retinal images for vessel extraction(DRIVE)database demonstrate that the proposed method is efficient and robust on the fundus images with lesions when compared with the other methods. Meanwhile, the proposed method also exhibits high robustness on a new Retinal images for screening(RIS) database. The average accuracy,sensitivity, and specificity of improved method are 0.9535,0.8319 and 0.9607, respectively. To capture the intrinsic data characteristicsfrom heterogeneous aspects, we first evaluate training data by multiple criteria, each ofwhich is reformulated from a spectral clustering. Then, we integrate the multipleevaluation scores generated by the multiple criteria through the proposed multiplespectral clustering aggregation (MSCA) method. Finally, we use the unified score as theindicator of poisoning attack samples.
