| 2 | 5-Hydroxymethylome in Circulating Cell-free DNA as A Potential Biomarker for Non-small-cell Lung Cancer显示文摘Non-small-cell lung cancer(NSCLC), the most common type of lung cancer accounting for 85% of the cases, is often diagnosed at advanced stages owing to the lack of efficient early diagnostic tools. 5-Hydroxymethylcytosine(5 hmC) signatures in circulating cell-free DNA(cfDNA) that carries the cancer-specific epigenetic patterns may represent the valuable biomarkers for discriminating tumor and healthy individuals, and thus could be potentially useful for NSCLC diagnosis. Here,we employed a sensitive and reliable method to map genome-wide 5 hmC in the cfDNA of Chinese NSCLC patients and detected a significant 5 hmC gain in both the gene bodies and promoter regions in the blood samples from tumor patients compared with healthy controls. Specifically, we identified six potential biomarkers from 66 patients and 67 healthy controls(mean decrease accuracy>3.2, P < 3.68E-19) using machine-learning-based tumor classifiers with high accuracy. Thus, the unique signature of 5 hmC in tumor patient's cfDNA identified in our study may provide valuable information in facilitating the development of new diagnostic and therapeutic modalities for NSCLC. | Ji Zhang Xiao Han Chunchun Gao Yurong Xing Zheng Qi Ruijuan Liu Yueqin Wang Xiaojian Zhang Yun-Gui Yang Xiangnan Li Baofa Sun Xin Tian | 2018 | Genomics, Proteomics & Bioinformatics2018,16,3: | 7 |
| 3 | Film mulching treatment inf luences dry matter and yield of oil flax in semi-arid area of China显示文摘A field study was set up to compare dry matter accumulation and grain yield of 3 different types of film mulching, namely FFC(full film muched on tiny ridges covered with soil), PFC(full film muched covered with soil), and PF(full film muched with no cover). The experiment was laid out in a complete random design with 3 replications at northwest(34°40′N, 105°06′E) of China. This study determined that(1) film mulching treatment increased leaf area and dry matter accumulation of oil flax in semi-arid areas;(2) accumulation of dry matter under PFC treatment was markedly higher than other treatments except anthesis stage;(3) average grain yield of oil flax under film mulching treatment was 52.67%-60.33% higher than flat plot without mulched or ridge(CK) treatment;(4) grain yield of oil flax was substantially correlated with plant height, grain number and 1,000 kernels weight. This study demonstrated that film mulching has significant effects on crop grain yield improvement in the northwest of China. | Zhengjun Cui Bing Wu Yuhong Gao Chunchun Li Yongquan Ta Zhibing He Junyi Niu | 2018 | Oil Crop Science2018,3,3: | 0 |
| 4 | Effects of Different Extinction for Morphine-CPP on Hippocampal EEG Power Spectrum in Mice显示文摘The relationship between hippocampal electroencephalogram(EEG)power spectrum and the extinction of addiction memory was investigated.Forty KM mice(Kunming mice)that had successfully established morphine(MOR)-conditioned place preference(CPP)were divided into four groups:saline-training extinction(SAL-TE),SAL-natural extinction(SAL-NE),MORTE,MOR-NE,for extinction treatment and EEG recording in the dorsal and ventral hippocampus(DH/VH).Results show that the CPP score of MOR-TE significantly decreased and the total,βandγbands power spectrum of MOR-TE was suppressed.Notably, the total power from the VH was more correlated with the extinction method, and the TE total power was always lower than NE in SAL or MOR group.Naloxone enhanced the EEG total power and all bands(DH)orβandγbands(VH)power in the SAL group. However,in the MOR-TE group, onlyβandγbands power in VH increased(P<0.05).The results suggest that TE can promote the extinction of opiate addictive memory more effectively than NE,and the extinction of MOR-CPP may have a stronger correlation with changes of VH-EEG power spectrum. | LI Chunchun GAO Jingyao WANG Xiaoqin WANG Gongwu CAO Jun | 2022 | Wuhan University Journal of Natural Sciences2022,27,3: | 0 |
| 5 | Differential Transcriptomic Landscapes of SARS-CoV-2 Variants in Multiple Organs from Infected Rhesus Macaques显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)caused the persistent coronavirus disease 2019(COVID-19)pandemic,which has resulted in millions of deaths worldwide and brought an enormous public health and global economic burden. The recurring global wave of infections has been exacerbated by growing variants of SARS-CoV-2.In this study, the virological characteristics of the original SARS-CoV-2 strain and its variants of concern(VOCs;including Alpha,Beta, and Delta)in vitro,as well as differential transcriptomic landscapes in multiple organs(lung, right ventricle, blood, cerebral cortex, and cerebellum)from the infected rhesus macaques, were elucidated. The original strain of SARS-CoV-2 caused a stronger innate immune response in host cells, and its VOCs markedly increased the levels of subgenomic RNAs,such as N,Orf9b,Orf6,and Orf7ab,which are known as the innate immune antagonists and the inhibitors of antiviral factors.Intriguingly, the original SARS-CoV-2 strain and Alpha variant induced larger alteration of RNA abundance in tissues of rhesus monkeys than Beta and Delta variants did. Moreover,a hyperinflammatory state and active immune response were shown in the right ventricles of rhesus monkeys by the up-regulation of inflammation-and immune-related RNAs.Furthermore, peripheral blood may mediate signaling transmission among tissues to coordinate the molecular changes in the infected individuals.Collectively, these data provide insights into the pathogenesis of COVID-19 at the early stage of infection by the original SARS-CoV-2 strain and its VOCs. | Tingfu Du Chunchun Gao Shuaiyao Lu Qianlan Liu Yun Yang Wenhai Yu Wenjie Li Yong Qiao Sun Cong Tang Junbin Wang Jiahong Gao Yong Zhang Fangyu Luo Ying Yang Yun-Gui Yang Xiaozhong Peng | 2023 | Genomics, Proteomics & Bioinformatics2023,21,5: | 0 |
