| 1 | Effects of applying lactic acid bacteria to the fermentation on a mixture of corn steep liquor and air-dried rice straw显示文摘This study was to determine the fermentation quality of a mixture of corn steep liquor(CSL)(178 g/kg wet basis) and air-dried rice straw(356 g/kg wet basis) after being treated with inoculants of different types of lactic acid bacteria(LAB). The treatments included the addition of no LAB additive(control),which was deionized water; homo-fermentative LAB alone(^(ho)LAB), which was Lactobacillus plantarum alone), and a mixture of homo-fermentative and hetero-fermentative LAB(^(he+ho)LAB), which were L. plantarum, Lactobacillus casei, and Lactobacillus buchneri. The results showed that the inoculation of the mixture of CSL and air-dried rice straw with ^(he+ho)LAB significantly increased the concentration of acetic acid and lactic acid compared with the control(P < 0.05). The addition of ^(he+ho)LAB effectively inhibited the growth of yeast in the silage. The concentration of total lactic acid bacteria in the ^(he+ho)LAB-treated silage was significant higher than those obtained in other groups(P < 0.05). The duration of the aerobic stability of the silages increased from 56 h to >372 h. The control group was the first to spoil, whereas the silage treated with ^(he+ho)LAB remained stable throughout the 372 h period of monitoring. SCM-198, an alkaloid active component of Leonurus japonicas Houtt, has been reported to have anti-oxidative activity. However, the specific mechanisms of SCM-198 in the prevention of endometrial damage remain unknown. In the present study, we assessed the effect of SCM-198 on hydrogen peroxide (H2O2Hnduced oxidative injury in ESCs. ESCs were pretreated with SCM-198 for 4 h and then challenged with H2O2.Morphology ch a nges, apoptosis rate, and intracellular ROS producti on were measured to assess the level of oxidative injury. Flow cytometry and western blot analysis were performed to detect the expression levels of Bax, Bcl-2, active-caspase-3, and mitogen-activated protein kinases pathways. Classic inflammation cytokines were measured by real-time polymerase chain reactions. Our results showed that SCM-198 attenuated apoptosis and ROS generation of ESCs induced by H2O2. H2O2 induced the apparent apoptotic characteristics, including fragmentation of DNA, upregulation of Bax/Bcl2, activation of caspase-3, and secretion of inflammation cytokines, which were all ameliorated by SCM-198. Furthermore, H2O2-induced apoptosis-related ERK1/2 pathway activation was restrained by SCM-198 pretreatme nt. | 7 | Lipids of sulfate-reducing bacteria and sulfur-oxidizing bacteria found in the Dongsheng uranium deposit显示文摘U-bearing sandstones from the Dongsheng deposit in Ordos Basin contain abundant C15-C18 fatty acids. The fatty acids may have been derived from modern and ancient organisms including organisms from the intervals of U mineralization. A certain amount of i15:0, a15:0, a17:0 fatty acids coexist with small amounts of i17:1ω7c and 10me16:0, characteristic biomarkers of Desulfovibrio and Desulfobacter sp., respectively. This indicates the existence of sulfate-reducing bacteria (SRB) in the sandstones. The presence of sulfur-oxidizing bacteria (SOB), such as Beggiatoa and Thioploca, is indicated by significant amounts of 16:1ω7c and 18:1ω7c fatty acids. The existence of the SRB in the deposit, as inferred from the fatty acids, is consistent with results from fossilized microorganisms and isotopic compositions of ore-stage pyrite. This suggests that the environment may have been favorable for the SRB to grow since ore formation (9.8-22 Ma). The bacteria may have degraded hydrocarbons directly, or indirectly utilized hydrocarbons degraded by oxic microbes in the deposits. This process may have produced 12C-rich calcite and prominent baseline humps of unresolved complex mixtures (UCM), and 25-demethylated hopanes and tricyclic terpanes. The existence of sulfur-oxidizing bacteria and sulfate-reducing bacteria in the deposit may have resulted in bacterial sulfate reduction to sulfide, re-oxidization of the sulfide to sulfate and subsequent reduction of the sulfate to sulfide. | 8 | Fine Mapping of RppP25, a Southern Rust Resistance Gene in Maize显示文摘Southern rust (Puccinia polysora Underw.) is a major disease that can cause severe yield losses in maize (Zea mays L.). In our previous study, a major gene RppP25 that confers resistance to southern rust was identified in inbred line P25. Here, we report the fine mapping and candidate gene analysis of RppP25 from the near-isogenic line F939, which harbors RppP25 in the genetic background of the susceptible inbred line F349. The inheritance of resistance to southern rust was investigated in the BC1F1 and BC3F1 populations, which were derived from a cross between F939 and F349 (as the recurrent parent). The 1:1 segregation ratio of resistance to susceptible plants in these two populations indicated that the resistance is controlled by a single dominant gene. Ten markers, including three simple sequence repeat (SSR) markers and seven insertion/deletion (InDel) markers, were developed in the RppP25 region. RppP25 was delimited to an interval between P091 and M271, with an estimated length of 40 kb based on the physical map of B73. In this region, a candidate gene was identified that was predicted to encode a putative nucleotide-binding site leucine-rich repeat (NBS-LRR) protein. Methods: Radioimmunoassay was used to test the level of serum Leptin,and the activity of EMCA was determined chemically in 38 pregnant women with hypertensive disorder complicating pregnancy and 36 normotensive pregnant women. Results: The level of serum Leptin in hypertensive disorder complicating pregnancy(gestational hypertension: 13.76 ± 3.46 ng/ml; preeclampsia:15.76 ± 5.47 ng/ml; eclampsia: 18.32 ± 6.38 ng/ml)was significantly higher than that in normotensive pregnant women(11.33 ± 2.93 ng/ml),respectively. The average EMCA activity of patients with hypertensive disorder complicating pregnancy(gestational hypertension:1.65 ± 0.24 μmol·pi/mg·h ; preeclampsia:1.37 ± 0.19 μ mol·pi/mg·h; eclampsia:1.12 ± 0.14 μ mol·pi/mg·h) was significantly lower than that of normotensive pregnant women(1.83 ± 0.38 μ mol·pi/mg·h),respectively. There was a negative correlation between the level of serum Leptin and the activity of RMCA in hypertensive disorder complicating pregnancy(r = -0.63). Conclusion: Inhibition of EMCA activity of erythrocyte in hypertensive disorder complicating pregnancy may increase cytoplasmic free calcium, which contributes to the development of hypertensive disorder complicating pregnancy. | 10 | Dynamic blood single-cell immune responses in patients with COVID-19显示文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19. | Lulin Huang Yi Shi Bo Gong Li Jiang Zhixin Zhang Xiaoqi Liu Jialiang Yang Yongquan He Zhilin Jiang Ling Zhong Juan Tang Chunfang You Qi Jiang Bo Long Tao Zeng Mei Luo Fanwei Zeng Fanxin Zeng Shuqiang Wang Xingxiang Yang Zhenglin Yang | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 4 |

| 12 | Dynamic Analysis of Supply and Demand Coupling of Ecosystem Services in Loess Hilly Region:A Case Study of Lanzhou,China显示文摘The relationship between the supply and demand for ecosystem services(ESs)is a key issue for the rational allocation of natural resources and optimisation of sustainable development capacity.This paper investigateed the dynamic evolution features of supply and demand of four ESs in Lanzhou of China,namely,water supply,food supply,carbon fixation and soil retention services.The crosssectional data of 2005 and 2017 were used for calculating ESs value and its supply and demand through ArcGIS software,InVEST model,elastic coefficient model and coupling coordination model.Results showed that:1)from 2005 to 2017,the supply of water supply services increased,the demand of soil retention services decreased,and the supply and demand of food supply and carbon fixation services increased.The high-value areas of service supply were mainly distributed in the rocky mountain areas in the southeast and northwest with high vegetation coverage,while the high-value areas of demand were mainly distributed in the urban areas and surrounding areas with high population density.2)There were five different types of coupling relations.Water supply service was dominated by a negative coupling type D,which means that the decrease in demand for ESs has had a positive response on the supply of ESs.Negative coupling type C was the main type of food supply and carbon fixation services,which means that the increase in demand for ESs has had a negative response on the supply of ESs.All three services were supplemented by a positive coupling type A,which means that the increase in demand for ESs has had a positive response on the supply of ESs.Soil retention service generally exhibits a positive coupling type B,which means that the decrease in demand for ESs has had a negative response on the supply of ESs.3)Over the past 12 yr,the coordination degree of supply and demand of water supply,food supply and soil retention services decreased,and the coordination degree of carbon fixation service increased.Various types of ES had a low degree of coupling and coordination,showing different characteristics of temporal and spatial evolution.The areas with imbalanced ESs supply and demand were mainly distributed in urban areas dominated by construction land.The research results are valuable to the optimisation of urban and rural ecological environments and the sustainable development of territory space under the framework of ecological civilisation,including similar ecologically vulnerable areas in other developing countries. | LI Pengjie LIU Chunfang LIU Licheng WANG Weiting | 2021 | Chinese Geographical Science2021,31,2: | 3 |

| 15 | A New Microsurgical Technique to Correct Retinal Detachment显示文摘Purpose: To evaluate the efficacy of a new technique to repair retinal detachments (RD) under the microscope. Methods : Thirty-six consecutive patients (36 eyes) who presented to our clinic with rhegmatogenous RD without severe proliferative vitreoretinopathy ( ≤C1) were included. The sutures for buckling and/or encircling bands were preplaced according to the preoperative location of the breaks using a three-mirror contact lens. Drainage of subretinal fluid, retinal cryotherapy, buckling, locating the retinal breaks, and intravitreal gases injection were performed under surgical microscopy. The surgical effects were compared with those in 37 consecutive patients with rhegmatogenous RD who underwent surgery under binocular indirect ophthalmoscopy. Results: The simultaneous intraoperative observation of fundus details and the sclera through the microscope was excellent in all cases. The effect of retinal cryotherapy was clearly visible. Mild opacity of the refractive media did not interfere with observing cryotherapy and locating the breaks. Retinal reattachment was obtained in 31 eyes (86%) during the primary surgery and in three eyes after a second surgery (94% total). The best-corrected visual acuity was <0.1 in 6 eyes (16.7%) , 0.1-0.4 in 15 eyes (41.7%) and ≥ 0.5 in 15 eyes (41.7%). The results were similar to that of RD surgery performed under indirect ophthalmoscopy.
