| 2 | A rapid reporter assay for recombinant human brain natriuretic peptide(rhBNP)by GloSensor technology显示文摘Accurate determination of biological activity is essential in quality control of recombinant human brain natriuretic peptide(rhBNP). In previous study, we successfully developed a genetically modified cell line 293GCAC3-based ELISA assay for rhBNP. But ELISA procedure is still tedious, so this study was aimed to develop a rapid and simple bioassay for rhBNP using GloSensor technology, which provides a platform of flexible luciferase-based biosensors for real-time detection of signaling events in live cells, including cGMP production. A reporter cell line 293GCAGlo-G1 was constructed by transfecting pGloSensor^(TM) 40 F plasmid into 293GCAC3. The reporter assay based on 293GCAGlo-G1 showed high precision with intraassay CV being 8.3% and inter-assay CV being 14.1%; high accuracy with 80%, 100% and 120% recovery rate being 99.2%, 102.4% and 99.0% respectively; and great linearity with R^2 of linear fitting equation being 0.99. Besides, no significant difference was found in test results of reporter assay and 293GCAC3-based ELISA assay(paired t test, p=0.630). | 3 | The R2R3-type MYB transcription factor MdMYB90-like is responsible for the enhanced skin color of an apple bud sport mutant显示文摘The anthocyanin content in apple skin determines its red coloration,as seen in a Fuji apple mutant.Comparative RNA-seq analysis was performed to determine differentially expressed genes at different fruit development stages between the wild-type and the skin color mutant.A novel R2R3-MYB transcription factor,MdMYB90-like,was uncovered as the key regulatory gene for enhanced coloration in the mutant.The expression of MdMYB90-like was 21.3 times higher in the mutant.MdMYB90-like regulates anthocyanin biosynthesis directly through the activation of anthocyanin biosynthesis genes and indirectly through the activation of other transcription factors that activate anthocyanin biosynthesis.MdMYB90-like bound to the promoters of both structural genes(MdCHS and MdUFGT)and other transcription factor genes(MdMYB1 and MdbHLH3)in the yeast one-hybrid system,electrophoretic mobility shift assay,and dual-luciferase assay.Transgenic analysis showed that MdMYB90-like was localized in the nucleus,and its overexpression induced the expression of other anthocyanin-related genes,including MdCHS,MdCHI,MdANS,MdUFGT,MdbHLH3,and MdMYB1.The mutant had reduced levels of DNA methylation in two regions(−1183 to−988 and−2018 to−1778)of the MdMYB90-like gene promoter,which might explain the enhanced expression of the gene and the increased anthocyanin content in the mutant apple skin. | Chao Sun Chunming Wang Wang Zhang Shuai Liu Weiyao Wang Xinyi Yu Tao Song Maxwell Yu Weichang Yu Shenchun Qu | 2021 | Horticulture Research2021,8,1: | 6 |
| 4 | Bioaugmentation of DDT-contaminated soil by dissemination of the catabolic plasmid pDOD显示文摘A plasmid transfer-mediated bioaugmentation method for the enhancement of dichlorodiphenyltrichloroethane(DDT) degradation in soil was developed using the catabolic plasmid p DOD from Sphingobacterium sp. D-6. The p DOD plasmid could be transferred to soil bacteria, such as members of Cellulomonas, to form DDT degraders and thus accelerate DDT degradation. The transfer efficiency of p DOD was affected by the donor, temperature,moisture, and soil type. Approximately 50.7% of the DDT in the contaminated field was removed 210 days after the application of Escherichia coli TG I(p DOD-gfp). | 5 | Genome of a citrus rootstock and global DNA demethylation caused by heterografting显示文摘Grafting is an ancient technique used for plant propagation and improvement in horticultural crops for at least 1,500 years.Citrus plants,with a seed-to-seed cycle of 5–15 years,are among the fruit crops that were probably domesticated by grafting.Poncirus trifoliata,a widely used citrus rootstock,can promote early flowering,strengthen stress tolerance,and improve fruit quality via scion–rootstock interactions.Here,we report its genome assembly using PacBio sequencing.We obtained a final genome of 303 Mb with a contig N50 size of 1.17Mb and annotated 25,680 protein-coding genes.DNA methylome and transcriptome analyses indicated that the strong adaptability of P.trifoliata is likely attributable to its special epigenetic modification and expression pattern of resistance-related genes.Heterografting by using sweet orange as scion and P.trifoliata as rootstock and autografting using sweet orange as both scion and rootstock were performed to investigate the genetic effects of the rootstock.Single-base methylome analysis indicated that P.trifoliata as a rootstock caused DNA demethylation and a reduction in 24-nt small RNAs(sRNAs)in scions compared to the level observed with autografting,implying the involvement of sRNA-mediated graft-transmissible epigenetic modifications in citrus grafting.Taken together,the assembled genome for the citrus rootstock and the analysis of graft-induced epigenetic modifications provide global insights into the genetic effects of rootstock–scion interactions and grafting biology. | Yue Huang Yuantao Xu Xiaolin Jiang Huiwen Yu Huihui Jia Chunming Tan Gang Hu Yibo Hu Muhammad Junaid Rao Xiuxin Deng Qiang Xu | 2021 | Horticulture Research2021,8,1: | 4 |

| 9 | Carbendazim induces a temporary change in soil bacterial community structure显示文摘The effect of carbendazim applications on the diversity and structure of a soil bacterial community was studied under field conditions using temperature gradient gel electrophoresis (TGGE) and partial sequence analysis of PCR-amplified 16S rRNA gene. After four successive introductions of carbendazim at a level of 0.94 kg active ingredient (a.i.)/ha, the genetic diversity (expressed as Shannon index, H ) decreased from 1.43 in the control to 1.29 in treated soil. This harmful effect seems to increase with the concentration of carbendazim. The value of H in the soil treated with carbendazim at 4.70 kg a.i./ha was reduced to 1.05 (P 0.05). The structure of soil bacterial community was also affected after four repeated applications of carbendazim at levels of 0.94, 1.88 and 4.70 kg a.i./ha, as seen in the relative intensities of the individual band. However, the bacterial community in carbendazim-treated soil recovered to that in the control 360 d after the first treatment. | 10 | Autonomic responses to blast overpressure can be elicited by exclusively exposing the ear in rats显示文摘Blast overpressure has become an increasing cause of brain injuries in both military and civilian populations. Though blast's direct effects on the cochlea and vestibular organs are active areas of study, little attention has been given to the ear's contribution to the overall spectrum of blast injury. Acute autonomic responses to blast exposure, including bradycardia and hypotension, can cause hypoxia and contribute to blast-induced neurotrauma. Existing literature suggests that these autonomic responses are elicited through blast impacting the thorax and lungs. We hypothesize that the unprotected ear also provides a vulnerable locus for blast to cause autonomic responses. We designed a blast generator that delivers controlled overpressure waves into the ear canal without impacting surrounding tissues in order to study the ear's specific contribution to blast injury. Anesthetized adult rats' left ears were exposed to a single blast wave ranging from 0 to 110 PSI(0-758 kPa). Blast exposed rats exhibited decreased heart rates and blood pressures with increased blast intensity, similar to results gathered using shock tubes and whole-body exposure in the literature. While rats exposed to blasts below 50 PSI(345 kPa) exhibited increased respiratory rate with increased blast intensity, some rats exposed to blasts higher than 50 PSI(345 kPa) stopped breathing immediately and ultimately died. These autonomic responses were significantly reduced in vagally denervated rats, again similar to whole-body exposure literature. | 11 | Characteristics of the quasi-16-day wave in the mesosphere and lower thermosphere region as revealed by meteor radar,Aura satellite,and MERRA2 reanalysis data from 2008 to 2017显示文摘This study presents an analysis of the quasi-16-day wave(Q16DW)at three stations in the middle latitudes by using a meteor radar chain in conjunction with Aura Microwave Limb Sounder temperature data and MERRA2(Modern-Era Retrospective Analysis for Research and Applications,Version 2)reanalysis data from 2008 to 2017.The radar chain consists of three meteor radar stations located at Mohe(MH,53.5°N,122.3°E),Beijing(BJ,40.3°N,116.2°E),and Wuhan(WH,30.5°N,114.6°E).The Q16DW wave exhibits similar seasonal variation in the neutral wind and temperature,and the Q16DW amplitude is generally strong during winter and weak around summer.The Q16DW at BJ was found to have secondary enhancement around September in the zonal wind,which is rarely reported at similar latitudes.The latitudinal variations of the Q16DW in the neutral wind and temperature are quite different.The Q16DW at BJ is the most prominent in both neutral wind components among the three stations and the Q16DW amplitudes at MH and WH are comparable,whereas the wave amplitude in temperature decreases with decreasing latitude.The quasi-geostrophic refractive index squared at the three stations in the period from 2008 to 2017 was revealed.The results indicate that the Q16DW in the mesosphere and lower thermosphere(MLT)at MH has a limited contribution from the lower atmosphere.Around March and October,the Q16DW in the troposphere at BJ can propagate upward into the MLT region,whereas at WH,the contribution to the Q16DW in the MLT region is largely from the mesosphere. | Yun Gong Zheng Ma Chun Li XieDong Lv ShaoDong Zhang QiHou Zhou ChunMing Huang KaiMing Huang You Yu GuoZhu Li | 2020 | Earth and Planetary Physics2020,4,3: | 2 |

| 14 | Traumatic brain injury induced by exposure to blast overpressure via ear canal显示文摘Exposure to explosive shockwave often leads to blast-induced traumatic brain injury in military and civilian populations.Unprotected ears are most often damaged following exposure to blasts.Although there is an association between tympanic membrane perforation and TBI in blast exposure victims,little is known about how and to what extent blast energy is transmitted to the central nervous system via the external ear canal.The present study investigated whether exposure to blasts directed through the ear canal causes brain injury in LongEvans rats.Animals were exposed to a single blast(0–30 pounds per square inch(psi))through the ear canal,and brain injury was evaluated by histological and behavioral outcomes at multiple time-points.Blast exposure not only caused tympanic membrane perforation but also produced substantial neuropathological changes in the brain,including increased expression of c-Fos,induction of a profound chronic neuroinflammatory response,and apoptosis of neurons.The blast-induced injury was not limited only to the brainstem most proximal to the source of the blast,but also affected the forebrain including the hippocampus,amygdala and the habenula,which are all involved in cognitive functions.Indeed,the animals exhibited long-term neurological deficits,including signs of anxiety in open field tests 2 months following blast exposure,and impaired learning and memory in an 8-arm maze 12 months following blast exposure.These results suggest that the unprotected ear canal provides a locus for blast waves to cause TBI.This study was approved by the Institutional Animal Care and Use Committee at the University of Mississippi Medical Center(Animal protocol#0932 E,approval date:September 30,2016 and 0932 F,approval date:September 27,2019). | Yang Ou Brad AClifton Jinghui Li David Sandlin Na Li Li Wu Chunming Zhang Tianwen Chen Jun Huang Yue Yu Jerome Allison Fan Fan Richard JRoman James Shaffery Wu Zhou Yi Pang Hong Zhu | 2022 | Neural Regeneration Research2022,17,1: | 1 |
