1Chemical composition and source apportionment of the ambient PM_(2.5)in Hangzhou,China显示文摘To identify and apportion the sources of the ambient PM_(2.5) in the urban area of Hangzhou,China,PM_(2.5)samples were collected at three sites in the city from April 2004 to March 2005.Water-soluble ions,metal elements,and total carbon(TC) in PM_(2.5) samples were analyzed.The results indicated that the 24-h mean concentrations of PM_(2.5) ranged from 17.1 to 267.0 μg/m^3,with an annual average value of 108.2 μg/m^3.Moreover,the seasonal mean values for PM_(2.5) in spring,summer,autumn,and winter were 116,73.1,114.2,and 136.0 μg/m^3.respectively.According to the Chinese ambient quality standard,at least 70% of the monitoring data exceeded the limit value.The total contribution of water-soluble ions,including F^-,Cl^-,NO_3^-,SO_4^(2-).NH_4^+,K^+,and Na^+.to PM_(2.5) mass varied from 32.3% to 36.7%.SO_4^(2-),NO_3^-,and NH_4^+were the main constituents of the ions,with contributions to PM_(2.5) varying from 14.1% to 14.7%,6.0%to 7.8%,and 6.4% to 7.7%,respectively.In addition,the annual mean mass fraction of TC in PM_(2.5) was27.8%.The annual average total contribution of the group of elements of Zn,Pb,Cu,Mn,Cr,Ni,Se,Mo.Cd,Sb,and Ag to the aerosol was in the range of 1.7-2.0%.Furthermore,positive matrix factorization was applied to analyze the PM_(2.5) data collected from the central area,and five factors were identified.The factor contributions to PM_(2.5) mass were 12.8%,31.9%,10.1%,17.2%,and 27.9%,respectively.Iron/steel manufacturing and secondary aerosol were the main sources for the fine particles.These findings may have significance for controlling the atmospheric contamination in the city.Gang Liu Jiuhai Li Dan Wu Hui Xu 2015Particuology2015,13,1:26
2A Transcriptional Network Promotes Anthocyanin Biosynthesis in Tomato Flesh显示文摘Dietary anthocyanins are important health-promoting antioxidants that make a major contribution to the quality of fruits. It is intriguing that most tomato cultivars do not produce anthocyanins in fruit. However, the purple tomato variety Indigo Rose, which has the dominant Aft locus combined with the recessive atv locus from wild tomato species, exhibits light-dependent anthocyanin accumulation in the fruit skin. Here, we report that Aft encodes a functional anthocyanin activator named SlAN2-like, while atv encodes a nonfunctional version of the anthocyanin repressor SlMYBATV. The expression of SlAN2-like is responsive to light, and the functional SlAN2-like can activate the expression of both anthocyanin biosynthetic genes and their regulatory genes, suggesting that SlAN2-like acts as a master regulator in the activation of anthocyanin biosynthesis. We further showed that cultivated tomatoes contain nonfunctional alleles of SlAN2-like and therefore fail to produce anthocyanins. Consistently, expression of a functional SlAN2-like gene driven by the fruit-specific promoter in a tomato cultivar led to the activation of the entire anthocyanin biosynthesis pathway and high-level accumulation of anthocyanins in both the peel and flesh. 4Simultaneous extraction of DNA and RNA from Escherichia coli BL 21 based on silica-coated magnetic nanoparticles显示文摘The extraction of nucleic acid is recognized as one of the most essential steps in molecular biology for initiating other downstream applications such as sequencing, amplification, hybridization, and cloning. Many commercial kits and methods are currently available that allow the extraction of only one type of nucleic acids-DNA or RNA. However, in parallel clinical detection of several diseases, a method for simultaneous extraction of both DNA and RNA from the same source is needed in such cases. In this study, a method for simultaneous extraction of DNA and RNA from bacteria based on magnetic nanoparticles(MNPs) was described. Lysis buffers were prepared to help the nucleic acid released and adsorbed to MNPs. Then, two washing buffers were used to remove the contamination of proteins and carbohydrates. The nucleic acids were finally eluted by Deoxyribonuclease(DNase) and Ribonucleases(RNase) free water. Different factors which might affect the purification of the nucleic acid were investigated, and the quantity and quality parameters of the nucleic acid were also recorded. The DNA and RNA extracted from bacteria were then respectively subjected to polymerase chain reaction(PCR) and reverse transcription PCR(RT-PCR) to further confirm its quality.
