| 1 | The single nucleotide polymorphisms of the chicken myostatin gene are associated with skeletal muscle and adipose growth显示文摘Myostatin, a new member of the TGF-β superfamily, is predominantly expressed in skeletal muscle cells and functions as a negative regulator of skeletal muscle growth in animals. Recently, we have reported three single nucleotide polymorphisms (SNPs) in the chicken myostatin gene. Herein, we investigate the association of those SNPs with the production traits in a F2 chicken line derived from Broilers crossing to Silky with the least square analysis. The results show that the BB and AA genotypes are strongly associated with abdominal fat weight (AFW), abdominal fat percentage (AFP), and birth weight (BW) (P < 0.05). Breast muscle percentage (BMP) of the AA type is higher than that of the AB type. The breast muscle weight and breast muscle percentages of F2 individuals have significant difference between CC and DD genotypes (P < 0.05). Breast muscle weight (BMW) of EF birds is higher than that of EE birds (P < 0.05). | 2 | Spontaneous apoptosis of cells in therapeutic stem cell preparation exert immunomodulatory effects through release of phosphatidylserine显示文摘Mesenchymal stem cell(MSC)-mediated immunomodulation has been harnessed for the treatment of human diseases,but its underlying mechanism has not been fully understood.Dead cells,including apoptotic cells have immunomodulatory properties.It has been repeatedly reported that the proportion of nonviable MSCs in a MSC therapeutic preparation varied from 5-50%in the ongoing clinical trials.It is conceivable that the nonviable cells in a MSC therapeutic preparation may play a role in the therapeutic effects of MSCs.We found that the MSC therapeutic preparation in the present study had about 5%dead MSCs(DMSCs),characterized by apoptotic cells.Namely,1×10^(6) MSCs in the preparation contained about 5×10^(4) DMSCs.We found that the treatment with even 5×10^(4) DMSCs alone had the equal therapeutic effects as with 1×10^(6) MSCs.This protective effect of the dead MSCs alone was confirmed in four mouse models,including concanavalin A(ConA)-and carbon tetrachloride(CCI4)-induced acute liver injury,LPS-induced lung injury and spinal cord injury.We also found that the infused MSCs died by apoptosis in vivo.Furthermore,the therapeutic effect was attributed to the elevated level of phosphatidylserine(PS)upon the injection of MSCs or DMSCs.The direct administration of PS liposomes(PSLs)mimic apoptotic cell fragments also exerted the protective effects as MSCs and DMSCs.The Mer tyrosine kinase(MerTK)deficiency or the knockout of chemokine receptor C-C motif chemokine receptor 2(CCR2)reversed these protective effects of MSCs or DMSCs.These results revealed that DMSCs alone in the therapeutic stem cell preparation or the apoptotic cells induced in vivo may exert the same immunomodulatory property as the'living MSCs preparation'through releasing PS,which was further recognized by MerTK and participated in modulating immune cells. | Xuemei He Weiqi Hong Jingyun Yang Hong Lei Tianqi Lu Cai He Zhenfei Bi Xiangyu Pan Yu Liu Lunzhi Dai Wei Wang Canhua Huang Hongxin Deng Xiawei Wei | 2021 | Signal Transduction and Targeted Therapy2021,6,8: | 6 |
| 3 | T cell-depleting nanoparticles ameliorate bone loss by reducing activated T cells and regulating the Treg/Th17 balance显示文摘Estrogen deficiency is one of the most frequent causes of osteoporosis in postmenopausal women.Under chronic inflammatory conditions caused by estrogen deficiency,activated T cells contribute to elevated levels of proinflammatory cytokines,impaired osteogenic differentiation capabilities of bone marrow mesenchymal stem cells(BMMSCs),and disturbed regulatory T cell(Treg)/Th17 cell balance.However,therapeutic strategies that re-establish immune homeostasis in this disorder have not been well developed.Here,we produced T cell-depleting nanoparticles(TDNs)that ameliorated the osteopenia phenotype and rescued the osteogenic deficiency of BMMSCs in ovariectomized(OVX)mice.TDNs consist of monocyte chemotactic protein-1(MCP-1)-encapsulated mesoporous silica nanoparticles as the core and Fas-ligand(FasL)as the corona.We showed that the delicate design of the TDNs enables rapid release of MCP-1 to recruit activated T cells and then induces their apoptosis through the conjugated FasL both in vitro and in vivo.Apoptotic signals recognized by macrophages help skew the Treg/Th17 cell balance and create an immune tolerant state,further attenuating the osteogenic deficiency of BMMSCs and the osteopenia phenotype.Mechanistically,we found that the therapeutic effects of TDNs were partially mediated by apoptotic T cell-derived extracellular vesicles(ApoEVs),which promoted macrophage transformation towards the M2 phenotype.These findings demonstrate that TDNs may represent a promising strategy for treating osteoporosis and other immune disorders. | Xiaoshan Yang Fuxing Zhou Pingyun Yuan Geng Dou Xuemei Liu Siying Liu Xiangdong Wang Ronghua Jin Yan Dong Jun Zhou Yajie Lv Zhihong Deng Shiyu Liu Xin Chen Ying Han Yan Jin | 2021 | Bioactive Materials2021,6,10: | 4 |

| 5 | Spirulina platensis aqueous extracts ameliorate colonic mucosal damage and modulate gut microbiota disorder in mice with ulcerative colitis by inhibiting inflammation and oxidative stress显示文摘Ulcerative colitis(UC)is a chronic and recurrent inflammatory bowel disease(IBD)that has become a major gastroenterologic problem during recent decades.Numerous complicating factors are involved in UC development such as oxidative stress,inflammation,and microbiota disorder.These factors exacerbate damage to the intestinal mucosal barrier.Spirulina platensis is a commercial alga with various biological activity that is widely used as a functional ingredient in food and beverage products.However,there have been few studies on the treatment of UC using S.platensis aqueous extracts(SP),and the underlying mechanism of action of SP against UC has not yet been elucidated.Herein,we aimed to investigate the modulatory effect of SP on microbiota disorders in UC mice and clarify the underlying mechanisms by which SP alleviates damage to the intestinal mucosal barrier.Dextran sulfate sodium(DSS)was used to establish a normal human colonic epithelial cell(NCM460)injury model and UC animal model.The mitochondrial membrane potential assay 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)and staining with Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI)and Hoechst 33258 were carried out to determine the effects of SP on the NCM460 cell injury model.Moreover,hematoxylin and eosin(H&E)staining,transmission electron microscopy(TEM),enzyme-linked immunosorbent assay(ELISA),quantitative real-time polymerase chain reaction(qPCR),western blot,and 16S ribosomal DNA(rDNA)sequencing were used to explore the effects and underlying mechanisms of action of SP on UC in C57BL/6 mice.In vitro studies showed that SP alleviated DSS-induced NCM460 cell injury.SP also significantly reduced the excessive generation of intracellular reactive oxygen species(ROS)and prevented mitochondrial membrane potential reduction after DSS challenge.In vivo studies indicated that SP administration could alleviate the severity of DSS-induced colonic mucosal damage compared with the control group.Inhibition of inflammation and oxidative stress was associated with increases in the activity of antioxidant enzymes and the expression of tight junction proteins(TJs)post-SP treatment.SP improved gut microbiota disorder mainly by increasing antioxidant enzyme activity and the expression of TJs in the colon.Our findings demonstrate that the protective effect of SP against UC is based on its inhibition of pro-inflammatory cytokine overproduction,inhibition of DSS-induced ROS production,and enhanced expression of antioxidant enzymes and TJs in the colonic mucosal barrier. | Jian WANG Liqian SU Lun ZHANG Jiali ZENG Qingru CHEN Rui DENG Ziyan WANG Weidong KUANG Xiaobao JIN Shuiqing GUI Yinghua XU Xuemei LU | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,6: | 3 |
| 6 | Single nucleotide polymorphism analysis of the encoding region of EX-FABP gene and its association with fattiness trait in chicken显示文摘Fattiness is an important parameter to estimate meat quality, which has high heritability. In this experiment, F2 chickens derived from Broilers crossing to Silky were used to study the effect of extracellular fatty acid inding protein (EX-FABP) gene on abdominal fat accumulation. Five exons and partial introns of the gene were amplified by six pairs of primers, and then single nucleotide polymorphisms (SNPs) were detected by single strand conformation polymorphism (SSCP) and subsequently confirmed by sequencing. There were four nucleotides variations found, T-C at 659, C-T at 924, CA-TG at 968 and C-T at 1985 respectively. The result of least square analysis suggests that the chickens with HH genotype defined by the third pair of primers have a lower abdominal fat weight and abdominal fat percentage than the chickens with other genotypes (GG, GI, GH and II). | 7 | Infrared-transmission spectra and hydrogen content of hydrogenated amorphous silicon显示文摘In this paper, two kinds of methods of calculating the hydrogen content of a-Si:H thin film by means of the wagging mode and the stretching modes of infrared-transmission spectra , are investigated. The reason for the difference in these two calculation results is analyzed. If the contents of SiH2 and (SiH2)n are indicated in terms of a structure factor F=(I840+I880)/I2000, it is shown that the calculation results obtained from the two different methods are almost equal when the refractive index n is approximately 3.4 or the fitting thickness is between 0.71 and 0.89 mm in the case of a small F. It is shown that the ways of fabrication of thin film can influence silicon-hydrogen bonding configuration of a-Si:H film, and different ways of fabrication can lead to different contents of SiH2 and (SiH2)n. The uniformity of the thin film with a big F is bad. In this case, there is great difference between the thickness measured by the SurfCom408A surface profile apparatus and the thickness obtained by fitting the fringes; and the hydrogen contents of a-Si:H films obtained by means of the wagging mode and the stretching modes are different, too. | 8 | Genome-wide profiling of RNA editing sites in sheep显示文摘Background: The widely observed RNA-DNA differences(RDDs) have been found to be due to nucleotide alteration by RNA editing. Canonical RNA editing(i.e., A-to-I and C-to-U editing) mediated by the adenosine deaminases acting on RNA(ADAR) family and apolipoprotein B mRNA editing catalytic polypeptide-like(APOBEC)family during the transcriptional process is considered common and essential for the development of an individual.To date, an increasing number of RNA editing sites have been reported in human, rodents, and some farm animals;however, genome-wide detection of RNA editing events in sheep has not been reported. The aim of this study was to identify RNA editing events in sheep by comparing the RNA-seq and DNA-seq data from three biological replicates of the kidney and spleen tissues.Results: A total of 607 and 994 common edited sites within the three biological replicates were identified in the ovine kidney and spleen, respectively. Many of the RDDs were specific to an individual. The RNA editing-related genes identified in the present study might be evolved for specific biological functions in sheep, such as structural constituent of the cytoskeleton and microtubule-based processes. Furthermore, the edited sites found in the ovine BLCAP and NEIL1 genes are in line with those in previous reports on the porcine and human homologs, suggesting the existence of evolutionarily conserved RNA editing sites and they may play an important role in the structure and function of genes.Conclusions: Our study is the first to investigate RNA editing events in sheep. We screened out 607 and 994 RNA editing sites in three biological replicates of the ovine kidney and spleen and annotated 164 and 247 genes in the kidney and spleen, respectively. The gene function and conservation analysis of these RNA editing-related genes suggest that RNA editing is associated with important gene function in sheep. | 10 | Molecular cloning and characterization of the endothelin 3 gene in black bone sheep显示文摘Background: Black bone sheep was first discovered in Yunnan province of China in 1970, with unique black pigmentation on the body and internal organs. Endothelin 3(EDN3) has been known as a key gene causing hyperpigmentation in black bone chicken, the Silky fowl.Methods: In this study, EDN3 was employed as a candidate gene for regulating black color pigmentation. First,EDN3 was cloned from sheep to obtain the full-length cDNA by using the rapid amplification of cDNA ends(RACE).Genomic EDN3 was screened and a total of thirty predicted single nucleotide polymorphisms(SNPs) were genotyped for allele and genotype frequency analysis in a case-control study involving two black bone sheep populations. Genomic copy number analysis of EDN3 in sheep was conducted to measure the variation in copy number. EDN3 expression levels were observed among the groups in adult liver, lymph node, and kidney tissues, as well as embryo kidney samples. Also, among the tissues of black bone and non-black bone sheep.Results: The size of the full-length cDNA was 1,578 bp, which included 426 bp of 5′-untranslated region(5′-UTR),an open reading frame(ORF) of 639 bp encoding a protein of 212 amino acids, and a 3′-UTR of 513 bp. Genotype and allele frequencies of all the discovered SNPs were found insignificantly different in black bone and non-black bone sheep(P > 0.05). Genomic copy number analysis of EDN3 in sheep revealed no significant difference between the two sheep groups. No significant variations were found in the adult liver and kidney embryo samples. However,the expression in lymph node and kidney tissue was significantly higher in black bone sheep than that in non-black bone sheep(P < 0.05). | 11 | In vitro assay of cytoskeleton nanomechanics as a tool for screening potential anticancer effects of natural plant extract, tubeimoside I on human hepatoma (HepG2) cells显示文摘Cytoskeleton nanomechanics characterizes cancer cell's physical behaviors such as how it spread and invade. For anticancer drug, cytoskeleton nanomechanics may be a target to inhibit invasiveness and metastasis of cancer cells. Therefore, in vitro assay of cytoskeleton nanomechanics may be used to evaluate the effects of potential anticancer drug on various cancer types. Here, we investigated the effects of tubeimoside I (TBMS I) on human hepatoma (HepG2) cells by using optical magnetic twisting cytometry, a well-established technique for measuring nanomechanics of the Factin cytoskeleton. TBMS I is a natural compound extracted from a traditional Chinese herbal medicine, and is reported with antitumor effect. In this study, we demonstrated that the cytoskeleton stiffness (G) of HepG2 cells was affected by TBMS I. G′ exhibited a typical power law with respect to the loading frequency (f), i.e. G^f . The magnitude of G′ and the value of exponent (α) of the HepG2 cells decreased consistently with the increase of concentration for TBMS I exposure. In addition, the HepG2 cells responded to TBMS I much faster than the normal liver (L-02) cells. Such alteration of cytoskeleton nanomechanics induced by TBMS I was reported for the first time, which was further corroborated by assays of Factin cytoskeleton structure and cell migration. | 12 | Constraints on a scalar-tensor theory with an intermediate-range force by binary pulsars显示文摘Searching for an intermediate-range force has been considerable interests in gravity experiments.In this paper,aiming at a scalartensor theory with an intermediate-range force,we have derived the metric and equations of motion (EOMs) in the first post-Newtonian (1PN) approximation for general matter without specific equation of state and N point masses firstly.Subsequently,the secular periastron precession ω of binary pulsars in harmonic coordinates is given.After that,ω˙ of four binary pulsars data (PSR B1913+16,PSR B1534+12,PSR J0737-3039 and PSR B2127+11C) have been used to constrain the intermediate-range force,namely,the parameters α and λ.α and λ respectively represent the strength of the intermediate-range force coupling and its length scale.The limits from four binary pulsars data are respectively λ=(4.95±0.02)×10 8 m and α=(2.30±0.01)×10 8 if β=1,where β is a parameter like standard parametrized post-Newtonian parameter β PPN.When three degrees of freedom (α,λ and β≡β 1) in 1σ confidence level are considered,it yields α=(4.21±0.01)×10 4,λ=(4.51±0.01)×10 7 m and β=(3.30±0.01)×10 3.Through our research on the scalar-tensor theory with the intermediate-range force,it shows that the parameter α is directly related to the parameter γ (α=(1 γ)/(1+ γ)).Thus,this presents the constraints on 1 γ by binary pulsars which is about 10 4 for three degrees of freedom. | DENG XueMei | 2011 | Science China(Physics,Mechanics & Astronomy)2011,54,11: | 1 |

| 20 | Construction of a dual fluorescence whole-cell biosensor to detect N-acyl homoserine lactones显示文摘Detection of N-acyl homoserine lactones(AHLs) is useful for understanding quorum sensing(QS) behaviors, including biofilm formation, virulence and metabolism. For detecting AHLs and indicating the host cells in situ, we constructed the plasmid pUCGMA2T1-4 to make a dual fluorescent wholecell biosensor based on the AhlI/R AHL system of Pseudomonas syringae pv. syringae B728a. The plasmid contains three components: constitutively expressed PnptII::gfp for indicating host cells, PahlI::mcherry that produces red fluorescence in response to AHL, and the ahlR gene that encodes an AHL regulatory protein. Meanwhile, two copies of T1-4(four tandem copies of a transcriptional terminator) were added into the plasmid to reduce background. The results showed that when the plasmid was placed into Escherichia coli, the dual fluorescence whole-cell biosensor was able to respond with red fluorescence within 6 hr to 5 × 10-8–1 × 10-5mol/L of 3OC6-HSL. Bright green fluorescence indicated the host cells. Furthermore, when the plasmid was transferred to wildtype Pseudomonas PhTA125(an AHL-producing bacterium), it also showed both green and red fluorescence.
