| 1 | Systemic immune-inflammation index for predicting prognosis of colorectal cancer显示文摘AIM To investigate the clinical significance of preoperative systemic immune-inflammation index(SII) in patients with colorectal cancer(CRC). METHODS A retrospective analysis of 1383 cases with CRC was performed following radical surgery. SII was calculated with the formula SII =(P × N)/L, where P, N, and L refer to peripheral platelet, neutrophil, and lymphocyte counts, respectively. The clinicopathological features and follow-up data were evaluated to compare SII with other systemic inflammation-based prognostic indices such as the neutrophil-lymphocyte ratio(NLR) and platelet-lymphocyte ratio(PLR) in patients with CRC.RESULTS The optimal cut-off point for SII was defined as 340. The overall survival(OS) and disease-free survival(DFS) were better in patients with low NLR, PLR, and SII(P < 0.05). The SII was an independent predictor of OS and DFS in multivariate analysis. The area under the receiver-operating characteristics(ROC) curve for SII(0.707) was larger than those for NLR(0.602) and PLR(0.566). In contrast to NLR and PLR, SII could effectively discriminate between the TNM subgroups. CONCLUSION SII is a more powerful tool for predicting survival outcome in patients with CRC. | 3 | Colorectal cancer lymph node staining by activated carbon nanoparticles suspension in vivo or methylene blue in vitro显示文摘AIM:To investigate whether activated carbon nanoparticles suspension(ACNS) or methylene blue(MB) can increase the detected number of lymph nodes in colorectal cancer.METHODS:Sixty-seven of 72 colorectal cancer patients treated at our hospital fulfilled the inclusion criteria of the study which was conducted from December 2010 to February 2012.Seven patients refused to participate.Eventually,60 patients were included,and randomly assigned to three groups(20 in each group):ACNS group(group A),MB group(group B) and non-stained conventional surgical group(group C).In group A,patients received subserosal injection of 1 mL ACNS in a 4-quadrant region around the mass.In group B,the main artery of specimen was identified and isolated after the specimen was removed,and 2 mL MB was slowly injected into the isolated,stretched and fixed vessel.In group C,no ACNS and MB were injected.All the mesentery lymph nodes were isolated and removed systematically by visually inspecting and palpating the adipose tissue.RESULTS:No difference was observed among the three groups in age,gender,tumor location,tumor diameter,T-stage,degree of differentiation,postoperative complications and peritoneal drainage retention time.The total number of detected lymph nodes was 535,476 and 223 in the three groups,respectively.The mean number of detected lymph nodes per patient was significantly higher in group A than in group C(26.8 ± 8.4 vs 12.2 ± 3.2,P < 0.001).Similarly,there were significantly more lymph nodes detected in group B than in group C(23.8 ± 6.9 vs 12.2 ± 3.2,P < 0.001).However,there was no significant difference between group A and group B.There were 50,46 and 32 metastatic lymph nodes dissected in 13 patients of group A,10 patients of group B and 11 patients of group C,without significant differences among the three groups.Eleven of the 60 patients had insufficient number of detected lymph nodes(< 12).Only one patient with T 4a rectal cancer had 10 lymph nodes detected in group B,the other 10 patients were all from group C.Based on the different diameter categories,the number of detected lymph nodes in groups A and B was significantly higher than in group C.However,there was no statistically significant difference between group A and group B.The metastatic lymph nodes were not significant different among the three groups.Similarly,tumor location,T stage and tumor differentiation did not affect the staining results.Body mass index was a minor influencing factor in the two different staining methods.The stained lymph nodes can easily be identified from the mesenteric adipose tissues,and the staining time for lymph nodes was not significantly different compared with unstained group.None of the patients in groups A and B had drug-related complications.CONCLUSION:Both activated carbon nanoparticles suspension in vivo and methylene blue in vitro can be used as tracers to increase the detected number of lymph nodes in colorectal cancer. | Hong-Ke Cai Hai-Fei He Wei Tian Mei-Qi Zhou Yue Hu Yong-Chuan Deng | 2012 | World Journal of Gastroenterology2012,18,42: | 39 |

| 7 | Targeting PI3K/AKT signaling for treatment of idiopathic pulmonary fibrosis显示文摘Idiopathic pulmonary fibrosis(IPF) is a chronic progressive fibrotic interstitial pneumonia with unknown causes. The incidence rate increases year by year and the prognosis is poor without cure.Recently, phosphatidylinositol 3-kinase(PI3 K)/protein kinase B(PKB/AKT) signaling pathway can be considered as a master regulator for IPF. The contribution of the PI3 K/AKT in fibrotic processes is increasingly prominent, with PI3 K/AKT inhibitors currently under clinical evaluation in IPF. Therefore,PI3 K/AKT represents a critical signaling node during fibrogenesis with potential implications for the development of novel anti-fibrotic strategies. This review epitomizes the progress that is being made in understanding the complex interpretation of the cause of IPF, and demonstrates that PI3 K/AKT can directly participate to the greatest extent in the formation of IPF or cooperate with other pathways to promote the development of fibrosis. | 8 | (-)-Epigallocatechin-3-gallate inhibits growth of gastric cancer by reducing VEGF production and angiogenesis显示文摘AIM: To investigate the effect of (-)-epigallocatechin- 3-gallate (EGCG) on growth of gastric cancer and its possible mechanism. METHODS: Heterotopic tumors were induced by subcutaneously injection of SGC-7901 cells in nude mice. Tumor growth was measured by calipers in two dimensions. Tumor angiogenesis was determined with tumor microvessel density (MVD) by immunohistology. Vascular endothelial growth factor (VEGF) protein level and activation of signal transducer and activator of transcription 3 (Stat3) were examined by Western blotting. VEGF mRNA expression was determined by RT-PCR and VEGF release in tumor culture medium by ELISA. VEGF-induced cell proliferation was studied by MTT assay, cell migration by gelatin modified Boyden chamber (Transwell) and in vitro angiogenesis by endothelial tube formation in Matrigel. RESULTS: Intraperitoneal injection of EGCG inhibited the growth of gastric cancer by 60.4%. MVD in tumor tissues treated with EGCG was markedly reduced. EGCG treatment reduced VEGF protein level in vitro and in vivo. Secretion and mRNA expression of VEGF in tumor cells were also suppressed by EGCG in a dose-dependent manner. This inhibitory effect was associated with reduced activation of Stat3, but EGCG treatment did not change the total Stat3 expression. EGCG also inhibited VEGF-induced endothelial cell proliferation, migration and tube formation. | 9 | Prognostic value of systemic immune.inflammation index in patients with gastric cancer显示文摘Background:Inflammation-based indexes have been used to predict survival and recurrence in cancer patients.Systemic immune-inflammation index(Sll) was reported to be associated with prognosis in some malignant tumors.In the present study,we aimed to explore the association between Sll and the prognosis of patients with gastric cancer.Methods:We retrospectively analyzed data from 444 gastric cancer patients who underwent gastrectomy at the First Affiliated Hospital of Sun Yat-sen University between January 1994 and December 2005.Preoperative Sll was calculated.The Chi square test or Fisher's exact test was used to determine the relationship between preoperative Sll and clinicopathologic characteristics.Overall survival(OS) rates were estimated using the Kaplan-Meier method,and the effect of Sll on OS was analyzed using the Cox proportional hazards model.Receiver operating characteristic(ROC)curves were used to compare the predictive ability of Sll,NLR,and PLR.Results:Sll equal to or higher than 660 was significantly associated with old age,large tumor size,unfavorable Borrmann classification,advanced tumor invasion,lymph node metastasis,distant metastasis,advanced TNM stage,and high carcino-embryonic antigen level,high neutrophil-lymphocyte ratio,and high platelet-lymphocyte ratio(all P<0.05).High Sll was significantly associated with unfavorable prognosis(P<0.001) and Sll was an independent predictor for OS(P=0.015).Subgroups analysis further showed significant associations between high Sll and short OS in stage Ⅰ,Ⅱ,Ⅲ subgroups(all P<0.05).Sll was superior to NLR and PLR for predicting OS in patients with gastric cancer.Conclusion:Preoperative Sll level is an independent prognostic factor for OS in patients with gastric cancer. | Kang Wang Feiyu Diao Zhijun Ye Xinhua Zhang Ertao Zhai Hui Ren Tong Li Hui Wu Yulong He Shirong Cai Jianhui Chen | 2017 | Chinese Journal of Cancer2017,36,9: | 30 |
| 10 | Denitrification Losses and N_2O Emissions from Nitrogen Fertilizer Applied to a Vegetable Field显示文摘A field experiment was conducted on Chinese cabbage (Brassica campestris L. ssp. pekinensis (Lour.) Olsson) in a Nanjing suburb in 2003. The experiment included 4 treatments in a randomized complete block design with 3 replicates: zero chemical fertilizer N (CK); urea at rates of 300 kg N ha-1 (U300) and 600 kg N ha-1 (U600), both as basal and two topdressings; and polymer-coated urea at a rate of 180 kg N ha-1 (PCU180) as a basal application. The acetylene inhibition technique was used to measure denitrification (N2+N2O) from intact soil cores and N2O emissions in the absence of acetylene. Results showed that compared to CK total denitrification losses were significantly greater (P≤0.05) in the PCU180, U300, and U600 treatments, while N2O emissions in the U300 and U600 treatments were significantly higher (P≤0.05) than CK. In the U300 and U600 treatments peaks of denitrification and N2O emission were usually observed after N application. In the polymer-coated urea treatment (PCU180) during the period 20 to 40 days after transplanting, higher denitrification rates and N2O fluxes occurred. Compared with urea, polymer-coated urea did not show any effect on reducing denitrification losses and N2O emissions in terms of percentage of applied N. As temperature gradually decreased from transplanting to harvest, denitrification rates and N2O emissions tended to decrease. | 11 | Basic Soil Productivity of Spring Maize in Black Soil Under Long-Term Fertilization Based on DSSAT Model显示文摘Increasing basic farmland soil productivity has significance in reducing fertilizer application and maintaining high yield of crops. In this study, we defined that the basic soil productivity(BSP) is the production capacity of a farmland soil with its own physical and chemical properties for a specific crop season under local environment and field management. Based on 22-yr(1990-2011) long-term experimental data on black soil(Typic hapludoll) in Gongzhuling, Jilin Province, Northeast China, the decision support system for an agro-technology transfer(DSSAT)-CERES-Maize model was applied to simulate the yield by BSP of spring maize(Zea mays L.) to examine the effects of long-term fertilization on changes of BSP and explore the mechanisms of BSP increasing. Five treatments were examined:(1) no-fertilization control(control);(2) chemical nitrogen, phosphorus, and potassium(NPK);(3) NPK plus farmyard manure(NPKM);(4) 1.5 time of NPKM(1.5NPKM) and(5) NPK plus straw(NPKS). Results showed that after 22-yr fertilization, the yield by BSP of spring maize significantly increased 78.0, 101.2, and 69.4% under the NPKM, 1.5NPKM and NPKS, respectively, compared to the initial value(in 1992), but not significant under NPK(26.9% increase) and the control(8.9% decrease). The contribution percentage of BSP showed a significant rising trend(P<0.05) under 1.5NPKM. The average contribution percentage of BSP among fertilizations ranged from 74.4 to 84.7%, and ranked as 1.5NPKM>NPKM>NPK≈NPKS, indicating that organic manure combined with chemical fertilizers(1.5NPKM and NPKM) could more effectively increase BSP compared with the inorganic fertilizer application alone(NPK) in the black soil. This study showed that soil organic matter(SOM) was the key factor among various fertility factors that could affect BSP in the black soil, and total N, total P and/or available P also played important role in BSP increasing. Compared with the chemical fertilization, a balanced chemical plus manure or straw fertilization(NPKM or NPKS) not only increased the concentrations of soil nutrient, but also improved the soil physical properties, and structure and diversity of soil microbial population, resulting in an iincrease of BSP. | 12 | Construction of a cucumber genetic linkage map with SRAP markers and location of the genes for lateral branch traits显示文摘Using SRAP (sequence-related amplified polymorphism) markers a genetic linkage map of cucumber was constructed with a population consisting of 138 F2 individuals derived from a cross of the two cucumber lines, S06 and S52. In the survey of parental polymorphisms with 182 primer combinations, 64 polymorphism-revealing primer pairs were screened out, which generated totally 108 polymorphic bands with an average of 1.7 bands per primer pair and at most 6 bands from one primer pair. The constructed molecular linkage map included 92 loci, distributed in seven linkage groups and spanning 1164.2 cM in length with an average genetic distance of 12.6 cM between two neighboring loci. Based on this linkage map, the quantitative trait loci (QTL) for the lateral branch number (lbn) and the lateral branch average length (lbl) in cucumber were identified by QTLMapper1.6. A major QTL lbn1 located between ME11SA4B and ME5EM5 in LG2 could explain 10.63% of the total variation with its positively effecting allele from S06. | 13 | Cloud point extraction coupled with HPLC-UV for the determination of phthalate esters in environmental water samples显示文摘A method based on cloud point extraction was developed to determine phthalate esters including di-ethyl-phthalate (DEP), di-(2-ethylhexyl)-phthalate (DEHP) and di-cyclohexyl-phthalate (DCP) in environmental water samples using high-performance liquid chromatography separation and ultraviolet detection (HPLC-UV). The non-ionic surfactant Triton X-114 was chosen as extraction solvent. The parameters affecting extraction effciency, such as concentrations of Triton X-114 and Na2SO4, equilibration temperature, equilibration time and centrifugation time were evaluated and optimized. Under the optimum conditions, the method can achieve preconcentration factors of 35, 88, 111 and detection of limits of 2.0, 3.8, 1.0 ngml for DEP, DEHP and DCP in 10-ml water sample, respectively. | 16 | (-)-Epigallocatechin-3-gallate inhibits VEGF expression induced by IL-6 via Stat3 in gastric cancer显示文摘AIM: To demonstrate that (-)-Epigallocatechin-3-gallate (EGCG) inhibits vascular endothelial growth factor (VEGF) expression and angiogenesis induced by interleukin-6 (IL-6) via suppressing signal transducer and activator of transcription 3 (Stat3) activity in gastric cancer. METHODS: Human gastric cancer (AGS) cells were treated with IL-6 (50 ng/mL) and EGCG at different concentrations. VEGF, total Stat3 and activated Stat3 protein levels in the cell lyses were examined by Western blotting, VEGF protein level in the conditionedmedium was measured by enzyme-linked immunosorbent assay, and the level of VEGF mRNA was evaluated by reverse transcription polymerase chain reaction (RTPCR). Stat3 nuclear translocation was determined by Western blotting with nuclear extract, and Stat3-DNA binding activity was examined with Chromatin immunoprecipitation (ChIP) assay. IL-6 induced endothelial cell proliferation was measured with 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyl tetrazoliumbromide assay, in vitro angiogenesis was determined with endothelial cell tube formation assay in Matrigel, and IL-6-induced angiogenesis in vitro was measured with Matrigel plug assay. RESULTS: There was a basal expression and secretion of VEGF in AGS cells. After stimulation with IL-6, VEGF expression was apparently up-regulated and a 2.4-fold increase was observed. VEGF secretion in the conditioned medium was also increased by 2.8 folds. When treated with EGCG, VEGF expression and secretion were dose-dependently decreased. IL-6 also increased VEGF mRNA expression by 3.1 folds. EGCG treatment suppressed VEGF mRNA expression in a dose-dependent manner. EGCG dose-dependently inhibited Stat3 activation induced by IL-6, but did not change the total Stat3 expression. When treated with EGCG or AG490, VEGF expressions were reduced to the level or an even lower level in the tumor cells not stimulated with IL-6. However, PD98059 and LY294002 did not change VEGF expression induced by IL-6. EGCG inhibited Stat3 nucleus translocation, and Stat3-DNA binding activity was also markedly decreased by EGCG. Furthermore, EGCG inhibited IL-6 induced vascular endothelial cell proliferation and tube formation in vitro and angiogenesis in vitro . | 19 | Endoscopic Surgery versus External Ventricular Drainage Surgery for Severe Intraventricular Hemorrhage显示文摘The efficacy and applied value of endoscopic hematoma evacuation vs. external ventricular drainage (EVD) in the treatment of severe ventficular hemorrhage (IVH) were explored and compared.From Jan.2015 to Dec.2016,the clinical data of 42 cases of IVH were retrospectively analyzed,including 18 patients undergoing endoscopic hematoma evacuation (group A),and 24patients receiving EVD (group B).The hematoma clearance rate was calculated by 3D Slicer software,and complications and outcomes were compared between the two groups.There were no significant differences in age,sex and Graeb score between groups A and B (P>0.05).The hematoma clearance rate was 70.81%±27.64% in group A and 48.72%±36.58% in group B with a statistically significant difference (P<0.05).The operative time in groups A and B was 72.45±25.26 min and 28.54±15.27min,respectively (P<0.05).The Glasgow Coma Scale (GCS) score increased from 9.28±2.72 at baseline to 11.83±2.91 at 1 week postoperatively in group A,and from 8.25±2.62 at baseline to 10.79±4.12 at 1 week postoperatively in group B (P<0.05).The length of hospital stay was 12.67±5.97 days in group A and 17.33±8.91 days in group B with a statistically significant difference (P<0.05).The GOS scores at 6 months after surgery were 3.83±1.12 in group A,and 2.75±1.23 in group B (P<0.05).
