| 1 | The role of the gut microbiome and its metabolites in metabolic diseases显示文摘It is well known that an unhealthy lifestyle is a major risk factor for metabolic diseases,while in recent years,accumulating evidence has demonstrated that the gut microbiome and its metabolites also play a crucial role in the onset and development of many metabolic diseases,including obesity,type 2 diabetes,nonalcoholic fatty liver disease,cardiovascular disease and so on.Numerous microorganisms dwell in the gastrointestinal tract,which is a key interface for energy acquisition and can metabolize dietary nutrients into many bioactive substances,thus acting as a link between the gut microbiome and its host.The gut microbiome is shaped by host genetics,immune responses and dietary factors.The metabolic and immune potential of the gut microbiome determines its significance in host health and diseases.Therefore,targeting the gut microbiome and relevant metabolic pathways would be effective therapeutic treatments for many metabolic diseases in the near future.This review will summarize information about the role of the gut microbiome in organism metabolism and the relationship between gut microbiome-derived metabolites and the pathogenesis of many metabolic diseases.Furthermore,recent advances in improving metabolic diseases by regulating the gut microbiome will be discussed. | Jiayu Wu Kai Wang Xuemei Wang Yanli Pang Changtao Jiang | 2021 | Protein & Cell2021,12,5: | 40 |
| 2 | Holocene vegetational and climatic variation in westerly-dominated areas of Central Asia inferred from the Sayram Lake in northern Xinjiang,China显示文摘Changes in the vegetation and climate of the westerly-dominated areas in Central Asia during the Holocene were interpreted using pollen-assemblages and charcoal data from a 300-cm-long sediment core of the Sayram Lake,northern Xinjiang.Accele-rator Mass Spectrometry(AMS) radiocarbon dating methods were applied to bulk organic matter of the samples.Artemisia spp./Chenopodiaceae ratios and results from principal component analysis were used to infer that the lake basin was dominated by desert vegetation before ca.9.6 cal.ka BP,which suggests a warm and dry climate in the early Holocene.Desert steppe/steppe expanded during 9.6-5.5 cal.ka BP,indicating a remarkable increase both in the precipitation and temperature during the mid-Holocene.Desert vegetation dominated between 6.5 and 5.5 cal.ka BP,marking an extreme warmer and drier interval.The steppe/meadow steppe recovered,and temperatures decreased from 5.5 cal.ka BP in the late Holocene,as indicated by the increased abundance of Artemisia and the development of meadows.Holocene temperatures and moisture variations in the Sayram Lake areas were similar to those of adjacent areas.This consistency implies that solar radiation was the main driving factor for regional temperature changes,and that the effect of temperature variations was significant on regional changes in humidity.The evolution of climate and environment in the Sayram Lake areas,which were characterized as dry in the early Holocene and relatively humid in the middle-late Holocene,are clearly different from those in monsoonal areas.Dry conditions in the early Holocene in the Sayram Lake areas were closely related to decreased water vapor advection.These conditions were a result of reduced westerly wind speeds and less evaporation upstream,which in turn were caused by seasonal changes in solar radiation superimposed by strong evaporation following warming and drying local climate. | JIANG QingFeng JI JunFeng SHEN Ji Ryo MATSUMOTO TONG GuoBang QIAN Peng REN XueMei YAN DeZhi | 2013 | Science China Earth Sciences2013,56,3: | 32 |
| 3 | Ablation of gut microbiota alleviates obesity-induced hepatic steatosis and glucose intolerance by modulating bile acid metabolism in hamsters显示文摘Since metabolic process differs between humans and mice, studies were performed in hamsters, which are generally considered to be a more appropriate animal model for studies of obesityrelated metabolic disorders. The modulation of gut microbiota, bile acids and the farnesoid X receptor(FXR) axis is correlated with obesity-induced insulin resistance and hepatic steatosis in mice. However,the interactions among the gut microbiota, bile acids and FXR in metabolic disorders remained largely unexplored in hamsters. In the current study, hamsters fed a 60% high-fat diet(HFD) were administeredvehicle or an antibiotic cocktail by gavage twice a week for four weeks. Antibiotic treatment alleviated HFD-induced glucose intolerance, hepatic steatosis and inflammation accompanied with decreased hepatic lipogenesis and elevated thermogenesis in subcutaneous white adipose tissue(sWAT). In the livers of antibiotic-treated hamsters, cytochrome P450 family 7 subfamily B member 1(CYP7 B1) in the alternative bile acid synthesis pathway was upregulated, contributing to a more hydrophilic bile acid profile with increased tauro-β-muricholic acid(TβMCA). The intestinal FXR signaling was suppressed but remained unchanged in the liver. | 4 | Effects of dietary Clostridium butyricum supplementation on growth performance,intestinal development,and immune response of weaned piglets challenged with lipopolysaccharide显示文摘Background: Weanling pigs, with immature immune system and physiological function, usually experience postweaning diarrhea. This study determined the effects of dietary Clostridium butyricum supplementation on growth performance, diarrhea, and immunity of weaned pigs challenged with lipopolysaccharide(LPS).Methods: In Experiment(Exp.) 1,144 weaned piglets were weaned at 21 d and randomly assigned to six groups,with six replicates per group and four pigs per replicate, receiving a control diet(CON) or diet supplemented with antibiotics(AB) or C. butyricum(CB)(0.1%, 0.2%, 0.4%, or 0.8%), respectively. All diets in Exp. 1 were a highly digestible basal diet, with 3,000 mg/kg zinc oxide supplied in the first 2 wk only. In Exp. 2, 180 piglets were weaned at 21 d and randomly assigned to five groups, with six replicates per group and six pigs per replicate, receiving CON, AB, or CB(0.2%, 0.4%, or 0.6%) diets. The digestibility of diets was lower than those in Exp. 1, and did not include zinc oxide. At 36 d of Exp. 2, 12 piglets were selected from each of the CON and 0.4% CB groups, six piglets were intraperitoneally injected with LPS(50 μg/kg body weight) and the other six piglets with normal saline;animals were killed at 4 h after injection to collect blood, intestine, and digesta samples for biochemical analysis.Results: In Exp. 1, CB and AB diets had no effect on growth performance of piglets. In Exp. 2, 0.4% CB decreased feed-gain ratio(P < 0.1), diarrhea score(P < 0.05), and increased duodenal, jejunal, and ileal villus height and jejunal villus height/crypt depth(P < 0.05). The 0.4% CB decreased the plasma tumor necrosis factor(TNF) α(P < 0.05) but increased ileal mucosa IL-10 and TLR2 mRNA expression(P < 0.05). Furthermore, 0.4% CB altered the microbial profile, with Bacillus and Ruminococcaceae UGG-003 at genus level and Lactobacillus casei and Parasutterella secunda at species level were higher than CON in colonic content(P < 0.05).Conclusions: Dietary C. butyricum supplementation had positive effects on growth of weaned piglets with less digestible diets. There was a tendency to reduce the feed-gain ratio, which could reduce feed costs in pig production. | 7 | Rumen microbiome structure and metabolites activity in dairy cows with clinical and subclinical mastitis显示文摘Background:Due to the high prevalence and complex etiology,bovine mastitis(BM)is one of the most important diseases to compromise dairy cow health and milk quality.The shift in milk compositions has been widely investigated during mastitis,but recent studies suggested that gastrointestinal microorganism also has a crucial effect on the inflammation of other peripheral tissues and organs,including the mammary gland.However,research focused on the variation of rumen inner-environment during mastitis is still limited.Therefore,the ruminal microbial profiles,metabolites,and milk compositions in cows with different udder health conditions were compared in the present study.Furthermore,the correlations between udder health status and ruminal conditions were investigated.Based on the somatic cell counts(SCC),California mastitis test(CMT)parameters and clinical symptoms of mastitis,60 lactating Holstein dairy cows with similar body conditions(excepted for the udder health condition)were randomly divided into 3 groups(n=20 per group)including the healthy(H)group,the subclinical mastitis(SM)group and the clinical mastitis(CM)group.Lactation performance and rumen fermentation parameters were recorded.And rumen microbiota and metabolites were also analyzed via 16S rRNA amplicon sequencing and untargeted metabolomics,respectively.Results:As the degree of mastitis increased,rumen lactic acid(LA)(P<0.01),acetate,propionate,butyrate,valerate(P<0.001),and total volatile fatty acids(TVFAs)(P<0.01)concentrations were significantly decreased.In the rumen of CM cows,the significantly increased bacteria related to intestinal and oral inflammation,such as Lachnospiraceae(FDR-adjusted P=0.039),Moraxella(FDR-adjusted P=0.011)and Neisseriaceae(FDR-adjusted P=0.036),etc.,were accompanied by a significant increase in 12-oxo-20-dihydroxy-leukotriene B4(FDR-adjusted P=5.97×10^(−9))and 10beta-hydroxy-6beta-isobutyrylfuranoeremophilane(FDR-adjusted P=3.88×10^(−10)).Meanwhile,in the rumen of SM cows,the Ruminiclostridium_9(FDR-adjusted P=0.042)and Enterorhabdus(FDR-adjusted P=0.043)were increased along with increasing methenamine(FDR-adjusted P=6.95×10^(−6)),5-hydroxymethyl-2-furancarboxaldehyde(5-HMF)(FDR-adjusted P=2.02×10^(−6))and 6-methoxymellein(FDR-adjusted P=2.57×10^(−5)).The short-chain fatty acids(SCFAs)-producing bacteria and probiotics in rumen,including Prevoterotoella_1(FDRadjusted P=0.045)and Bifidobacterium(FDR-adjusted P=0.035),etc.,were significantly reduced,with decreasing 2-phenylbutyric acid(2-PBA)(FDR-adjusted P=4.37×10^(−6)).Conclusion:The results indicated that there was a significant shift in the ruminal microflora and metabolites associated with inflammation and immune responses during CM.Moreover,in the rumen of cows affected by SM,the relative abundance of several opportunistic pathogens and the level of metabolites which could produce antibacterial compounds or had a competitive inhibitory effect were all increased. | Yue Wang Xuemei Nan Yiguang Zhao Linshu Jiang Mengling Wang Hui Wang Fan Zhang Fuguang Xue Dengke Hua Jun Liu Junhu Yao Benhai Xiong | 2021 | Journal of Animal Science and Biotechnology2021,12,3: | 6 |

| 10 | Bivalent Metal Ions Tethered Fluorescent Gold Nanoparticles as a Reusable Peroxidase Mimic Nanozyme显示文摘Nanozymes are a class of mimic enzymes that have both unique properties of nanomaterials and catalytic functions.They are widely used in medicine,food,agriculture,and environment due to their high catalytic efficiency,stability,economy,and large-scale preparation.In this work,a simple,gentle method was applied to synthesize adenosine phosphates(AXP)templated gold nanoparticles(Au NPs)with small size,good optical properties,and peroxidase-like activities.In the presence of hydrogen peroxide(H_(2)O_(2)),the peroxidase substrate 3,3′,5,5′-tetramethylbenzidine(TMB)could be oxidized to form blue-colored oxidized TMB(oxTMB),which demonstrates that the AuAXP NPs have peroxidase-like activities.Inter-estingly,the introduction of bivalent metal ions into AuAXP NPs allows their reusability.Among them,after three rounds of recycling,the catalytic activity of Mg@AuAMP could still reach 92.69%compared to the original level,which showed excellent reusability.In addition,the glutathione(GSH)detection was realized with a linear detection range of 0-200μM due to the GSH-induced loss of peroxidase-like activities.Overall,this work provides a new idea of recycling for biosensor and catalytic analysis. | Liu Liu Hui Jiang Xuemei Wang | 2019 | Journal of Analysis and Testing2019,3,3: | 4 |

| 13 | Relationships between tree-ring cell features of Pinus koraiensis and climate factors in the Changbai Mountains,Northeastern China显示文摘Anatomical characteristics have been proven useful for extracting climatic signals. To examine the climatic signals recorded by tree-ring cell features in the Changbai Mountains, we measured cell number and cell lumen diameter, in addition to ring widths, of Korean pine(Pinus koraiensis) tree rings at sites of varied elevation,and we developed chronologies of cell number(CN), mean lumen diameter(MLD), maximum lumen diameter(MAXLD) and tree-ring width(TRW). The chronologies were correlated with climatic factors monthly mean temperature and the sum of precipitation. As shown by our analysis, the cell parameter chronologies were suitable for dendroclimatology studies. CN and TRW shared relatively similar climatic signals which differed from MLD andMAXLD, and growth-climate relationships were elevationdependent, as shown by the following findings:(1) at each elevation, MLD and MAXLD recorded different monthly climatic signals from those recorded by TRW for the same climatic factors; and(2) MLD and MAXLD recorded climatic factors that were absent from TRW at lower and middle elevations. | 14 | Construction and analysis of a plant transformation binary vector pBDGG harboring a bi-directional promoter fusing dual visible reporter genes显示文摘The constitutive promoter of cauliflower mosaic virus 35S (CaMV 35S) is a polar unidirectional promoter and is widely used in plant genetic engineering. In the present study, the unidirectional CaMV 35S promoter has been modified to a bi-directional promoter by fusing its minimal promoter element to the 5′end of CaMV 35S promoter in the opposite orientation. To qualitatively and quantitatively esti-mate its bi-directional transcriptional function and activity, two visible reporter genes, gusA (β-glucuronidase, GUS) and gfp (green fluo-rescent protein, GFP), were fused to the two ends of the promoter in bi-orientations ending with NOS terminator sequences, respectively. Stable expression of gusA and gfp genes in transgenic tobacco (Nicotiana tabacum L.) was visulized by histochemically staining for GUS and fluorescence microscopic observation under UV for GFP in transgenic plants. The expression of two reporter genes showed that the constructed bi-directional promoter did have the bi-directional transcriptional function in both expected orientations. The quantitative estimation of GUS and GFP were determined on a HITACHI F1000 Fluorescence Spectrophotometer with various wavelengths of excita-tion and emission. The GUS activity varied from 8 to 250 pmol 4-MU / min / mg protein and the GFP content varied from 0.9 to 1.8 μg / mg protein in various lines of transgenic tobacco plants. When AD occurs, the relevant amount of zinc ions in brain considerably changes. In this contribution, we have explored the possibility of in vivo rapid fluorescence imaging of AD through accurate targeting biomarker of zinc gluconate. In this review, we summarize the most recent advances in the synthesis and application of biomedical polymers, and discuss the comprehensive understanding of their property-function relationship for corresponding biomedical applications. In particular, a few burgeoning bioactive polymers, such as peptide/biomembrane/microorganism/cell-based biomedical polymers, are also introduced and highlighted as the emerging biomaterials for cancer precision therapy. Furthermore, the foreseeable challenges and outlook of the development of more efficient, healthier and safer biomedical polymers are discussed.
