1Spontaneous apoptosis of cells in therapeutic stem cell preparation exert immunomodulatory effects through release of phosphatidylserine显示文摘Mesenchymal stem cell(MSC)-mediated immunomodulation has been harnessed for the treatment of human diseases,but its underlying mechanism has not been fully understood.Dead cells,including apoptotic cells have immunomodulatory properties.It has been repeatedly reported that the proportion of nonviable MSCs in a MSC therapeutic preparation varied from 5-50%in the ongoing clinical trials.It is conceivable that the nonviable cells in a MSC therapeutic preparation may play a role in the therapeutic effects of MSCs.We found that the MSC therapeutic preparation in the present study had about 5%dead MSCs(DMSCs),characterized by apoptotic cells.Namely,1×10^(6) MSCs in the preparation contained about 5×10^(4) DMSCs.We found that the treatment with even 5×10^(4) DMSCs alone had the equal therapeutic effects as with 1×10^(6) MSCs.This protective effect of the dead MSCs alone was confirmed in four mouse models,including concanavalin A(ConA)-and carbon tetrachloride(CCI4)-induced acute liver injury,LPS-induced lung injury and spinal cord injury.We also found that the infused MSCs died by apoptosis in vivo.Furthermore,the therapeutic effect was attributed to the elevated level of phosphatidylserine(PS)upon the injection of MSCs or DMSCs.The direct administration of PS liposomes(PSLs)mimic apoptotic cell fragments also exerted the protective effects as MSCs and DMSCs.The Mer tyrosine kinase(MerTK)deficiency or the knockout of chemokine receptor C-C motif chemokine receptor 2(CCR2)reversed these protective effects of MSCs or DMSCs.These results revealed that DMSCs alone in the therapeutic stem cell preparation or the apoptotic cells induced in vivo may exert the same immunomodulatory property as the'living MSCs preparation'through releasing PS,which was further recognized by MerTK and participated in modulating immune cells.Xuemei He Weiqi Hong Jingyun Yang Hong Lei Tianqi Lu Cai He Zhenfei Bi Xiangyu Pan Yu Liu Lunzhi Dai Wei Wang Canhua Huang Hongxin Deng Xiawei Wei 2021Signal Transduction and Targeted Therapy2021,6,8:6
2The Arabidopsis catalase triple mutant reveals important roles of catalases and peroxisome-derived signaling in plant development显示文摘Hydrogen peroxide(H_2O_2)is generated in many metabolic processes.As a signaling molecule,H_2O_2plays important roles in plant growth and development,as well as environmental stress response.In Arabidopsis,there are three catalase genes,CAT1,CAT2,and CAT3.The encoded catalases are predominately peroxisomal proteins and are critical for scavenging H_2O_2.Since CAT1 and CAT3 are linked on chromosome 1,it has been almost impossible to generate cat1/3 and cat1/2/3 mutants by traditional genetic tools.In this study,we constructed cat1/3 double mutants and cat1/2/3 triple mutants by CRISPR/Cas9 to investigate the role of catalases.The cat1/2/3 triple mutants displayed severe redox disturbance and growth defects under physiological conditions compared with wild-type and the cat2/3 double mutants.Transcriptome analysis showed a more profound transcriptional response in the cat1/2/3 triple mutants compared to the cat2/3 mutants.These differentially expressed genes are involved in plant growth regulation as well as abiotic and biotic stress responses.In addition,expression of OXI1(OXIDATIVE SIGNAL INDUCIBLE 1)and several MAPK cascade genes were changed dramatically in the catalase triple mutant,suggesting that H_2O_2produced in peroxisomes could serve as a peroxisomal retrograde signal.Tong Su Pingping Wang Huijuan Li Yiwu Zhao Yao Lu Peng Dai Tianqi Ren Xiaofeng Wang Xuezhi Li Qun Shao Dazhong Zhao Yanxiu Zhao Changle Ma 2018Journal of Integrative Plant Biology2018,60,7:3
3A Bifunctional-Modulated Conformal Li/Mn-Rich Layered Cathode for Fast-Charging,High Volumetric Density and Durable Li-Ion Full Cells显示文摘Lithium-and manganese-rich(LMR)layered cathode materials hold the great promise in designing the next-generation high energy density lithium ion batteries.However,due to the severe surface phase transformation and structure collapse,stabilizing LMR to suppress capacity fade has been a critical challenge.Here,a bifunctional strategy that integrates the advantages of surface modification and structural design is proposed to address the above issues.A model compound Li_(1.2)Mn_(0.54)Ni_(0.13)Co_(0.13)O_(2)(MNC)with semi-hollow microsphere structure is synthesized,of which the surface is modified by surface-treated layer and graphene/car-bon nanotube dual layers.The unique structure design enabled high tap density(2.1 g cm^(−3))and bidirectional ion diffusion pathways.The dual surface coatings covalent bonded with MNC via C-O-M linkage greatly improves charge transfer efficiency and mitigates electrode degradation.Owing to the synergistic effect,the obtained MNC cathode is highly conformal with durable structure integrity,exhibiting high volumetric energy density(2234 Wh L^(−1))and predominant capacitive behavior.The assembled full cell,with nanograph-ite as the anode,reveals an energy density of 526.5 Wh kg^(−1),good rate performance(70.3%retention at 20 C)and long cycle life(1000 cycles).The strategy presented in this work may shed light on designing other high-performance energy devices.Zedong Zhao Minqiang Sun Tianqi Wu Jiajia Zhang Peng Wang Long Zhang Chongyang Yang Chengxin Peng Hongbin Lu 2021Nano-Micro Letters2021,13,8:2

5Comparative Study on Coastal Depth Inversion Based on Multi-source Remote Sensing Data显示文摘Coastal depth is an important research focus of coastal waters and is also a key factor in coastal environment. Dongluo Island in South China Sea was taken as a typical study area. The band ratio model was established by using measured points and three multispectral images of Landsat-8, SPOT-6(Systeme Probatoire d'Observation de la Terre, No.6) and WorldView-2. The band ratio model with the highest accuracy is selected for the depth inversion respectively. The results show that the accuracy of SPOT-6 image is the highest in the inversion of coastal depth. Meanwhile, analyzing the error of inversion from different depth ranges, the accuracy of the inversion is lower in the range of 0–5 m because of the influence of human activities. The inversion accuracy of 5–10 m is the highest, and the inversion error increases with the increase of water depth in the range of 5–20 m for the three kinds of satellite images. There is no linear relationship between the accuracy of remote sensing water depth inversion and spatial resolution of remote sensing data, and it is affected by performance and parameters of sensor. 6Ammonia promotes the proliferation of bone marrow-derived mesenchymal stem cells by regulating the Akt/mTOR/S6k pathway显示文摘Ammonia plays an important role in cellular metabolism.However,ammonia is considered a toxic product.In bone marrow-derived mesenchymal stem cells,multipotent stem cells with high expression of glutamine synthetase(GS)in bone marrow,ammonia and glutamate can be converted to glutamine via glutamine synthetase activity to support the proliferation of MSCs.As a major nutritional amino acid for biosynthesis,glutamine can activate the Akt/mTOR/S6k pathway to stimulate cell proliferation.The activation of mTOR can promote cell entry into S phase,thereby enhancing DNA synthesis and cell proliferation.Our studies demonstrated that mesenchymal stem cells can convert the toxic waste product ammonia into nutritional glutamine via GS activity.Then,the Akt/mTOR/S6k pathway is activated to promote bone marrow-derived mesenchymal stem cell proliferation.These results suggest a new therapeutic strategy and potential target for the treatment of diseases involving hyperammonemia.Yu Liu Xiangxian Zhang Wei Wang Ting Liu Jun Ren Siyuan Chen Tianqi Lu Yan Tie Xia Yuan Fei Mo Jingyun Yang Yuquan Wei Xiawei Wei 2022Bone Research2022,10,4:1
7A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 8Dual interfacial engineering for efficient Cs_(2)AgBiBr_(6) based solar cells显示文摘The emerging lead-free halide double perovskite solar cells have attracted widespread attentions due to their long-term stability and non-toxicity, but suffer from the low device performance. One efficiencylimiting factor is the improper contacts between the halide double perovskite and anode/cathode electrodes. Here, we improve the efficiency and stability of the bismuth-halide double perovskite based solar cells by a synergistic interface design for both electron and hole transport layers(ETL/HTL). The results show that the modification of the TiO_2 ETL with a thin hydrophobic C60 layer and replacement of the lithium-doped small molecule HTL with an un-doped conjugated polymer lead to higher surface quality of perovskite film and better energy-level alignment at the contacts. As a result, the optimized device shows reduced trap density, suppressed charge recombination and enhanced charge extraction, leading to an increase of 69% in device efficiency. In addition, the device also exhibits superior stability in ambient environment, heat stress and light bias after interface optimization. 10Nanoparticle enhanced combination therapy for stem-like progenitors defined by single-cell transcriptomics in chemotherapy-resistant osteosarcoma显示文摘The adaptation of osteosarcoma cells to therapeutic pressure impedes the efficacy of chemotherapy for osteosarcoma.However,the characteristics and cellular organization of therapy-resistant cells in osteosarcoma tumors remain elusive.Here,we utilized single-cell transcriptomics to systematically map the cell-type-specific gene expression in a chemotherapy-resistant osteosarcoma tumor.Our data demonstrated the VEGFR2-JMJD3-abundant subsets as quiescent stem-like cells,thereby establishing the hierarchy of therapy-resistant actively cycling progenitor pools(JMJD3-abundant)in osteosarcoma.VEGFR2 inhibitor and JMJD3 inhibitor synergistically impeded osteosarcoma cell propagation and tumor growth.Although osteosarcoma cells are predisposed to apoptosis induced by the synergistic therapy through activation of the CHOP pro-apoptotic factor via the endoplasmic reticulum(ER)stress,the stem-like/progenitor cells exhibit an adaptive response,leading to their survival.Reduction in cellular glutathione levels in stem-like/progenitor cells caused by the treatment with a glutathione synthesis inhibitor increases ER stress-induced apoptosis.Importantly,the marked therapeutic improvement of synergistic therapy against stem-like/progenitor cells was achieved by using glutathione-scavenging nanoparticles,which can load and release the drug pair effectively.Overall,our study provides a framework for understanding glutathione signaling as one of the therapeutic vulnerabilities of stem-like/progenitor cells.Broadly,these findings revealed a promising arsenal by encapsulating glutathione-scavenging nanoparticles with co-targeting VEGFR2 and JMJD3 to eradicate chemotherapy-resistant osteosarcoma.Li Wang Xiaojia Huang Xinru You Tianqi Yi Bing Lu Jiali Liu Guohao Lu Minglin Ma Changye Zou Jun Wu Wei Zhao 2020Signal Transduction and Targeted Therapy2020,5,1:1

12Inactivated SARS-CoV-2 induces acute respiratory distress syndrome in human A CE2-transgenic mice显示文摘The development of animal models for COVID-19 is essential for basic research and drug/vaccine screening.Previously reported COVID-19 animal models need to be established under a high biosafety level condition for the utilization of live SARS-CoV-2,which greatly limits its application in routine research.Here,we gen erate a mouse model of COVID-19 un der a gen eral laboratory condition that captures multiple characteristics of SARS-CoV-2-induced acute respiratory distress syndrome(ARDS)observed in huma ns.Briefly,human ACE2-tra nsge nic(MCE2)mice were in tratracheally in stilled with the formaldehyde-inactivated SARS-CoV-2,resulting in a rapid weight loss and detrimental changes in lung structure and function.The pulmonary pathologic changes were characterized by diffuse alveolar damage with pulmonary consolidation,hemorrhage,necrotic debris,and hyaline membrane formation.The production of fatal cytokines(IL-β,TNF-α,and IL-6)and the infiltration of activated neutrophils,inflammatory monocyte-macrophages,and T cells in the lung were also determined,suggesting the activation of an adaptive immune response.Therapeutic strategies,such as dexamethasone or passive antibody therapy,could effectively ameliorate the disease progression in this model.Therefore,the established mouse model for SARS-CoV-2-induced ARDS in the current study may provide a robust tool for researchers in the standard open laboratory to investigate the pathological mechanisms or develop new therapeutic strategies for COVID-19 and ARDS.Zhenfei Bi Weiqi Hong Haiying Que Cai He Wenyan Ren Jingyun Yang Tianqi Lu Li Chen Shuaiyao Lu Xiaozhong Peng Xiawei Wei 2022Signal Transduction and Targeted Therapy2022,7,1:0
13Graph Theory Based N-1 Transmission Contingency Selection and Its Application in Security-constrained Unit Commitment显示文摘Contingency analysis is an important building block in the stability and reliability analysis of power grid operations. However, due to the large number of transmission lines,in practice only a limited number of contingencies could be evaluated. This paper proposes a graph theory based N-1 contingency selection method to effectively identify the most critical contingencies, which can be used in security-constrained unit commitment(SCUC) problems to derive secure and economic operation decisions of the power grid. Specifically, the power flow transferring path identification algorithm and the vulnerability index are put forward to rank individual contingencies according to potential transmission line overloads. Effectiveness of the proposed N-1 contingency selection method is quantified by applying the corresponding SCUC solution to the full N-1 security evaluation, i. e., quantifying total post-contingency generation-load imbalance in all N-1 contingencies. Numerical results on several benchmark IEEE systems, including5-bus, 24-bus, and 118-bus systems, show effectiveness of the proposed method. 14Parallel numerical simulation on CO2 geologic storage in Ordos Basin, China显示文摘The authors developed a 3-D numerical injection model on parallel TOUGH2-FLAC3 D based on the site-specific stratigraphic information in Ordos Basin to simulate the Shenhua CO_2 geological storage( CGS)demonstration project injection process for three years and forecast CO_2 migration and layers displacement after injection for seven years. The results suggested that CO_2 migration did not stop after three years of consecutive injection,but migration rate was slowing down with time. As a result,displacement near injection well increased with injection and decreased slowly after injection. The maximum displacement of surface center occurred near the end of injection,which was only 1. 24 cm. 17A case of diffuse large B-cell lymphoma with interstitial pneumonia显示文摘This report involves a 54-year-old female patient diagnosed with diffuse large B-cell lymphoma who developed interstitial pneumonia(IP)during treatment.The patient presented to the ward with enlarged lymph nodes in the neck and was treated with the standard regimen,which included rituximab,cyclophosphamide,doxorubicin liposomes,vincristine,and prednisone(R-CDOP regimen).After 3 cycles,the treatment was assessed as effective.However,following the 4th cycle,the patient experience shortness of breath after physical activity.A repeat lung computer tomography indicated IP,which completely recovered after receiving“full coverage”treatment.Subsequently,the patient underwent 2 cycles of cyclophosphamide,doxorubicin liposomes,vincristine,and prednisone(CDOP),followed by local radiotherapy.Currently,the patient is now being followed up with regular reviews.Ge Song Changxi Zhou Shuxia Wang Tianqi Tao Weiping Guan Xuan Wu Ping Zhu Bo Yang Xuechun Lu 2023Cancer Pathogenesis and Therapy2023,1,3:0
