| 1 | Anti-apoptotic effects of aspirin following cerebral ischemia-reperfusion injury in rats显示文摘BACKGROUND:The pharmacological effects of aspirin on apoptosis are complex.The underlying mechanisms have not been properly defined.OBJECTIVE:To observe the effect of different doses of aspirin on brain cell apoptosis following focal cerebral ischemia-reperfusion injury(CIRI) in rats.DESING,TIME AND SETTING:A randomized,controlled,animal experiment,performed at the School of Medicine and Pharmaceutics,Jiangnan University between June and October 2006.MATERIALS:Twenty-six male,adult,Sprague Dawley rats(grade II),weighing 240-290 g,were obtained from Shanghai Experimental Animal Center,Chinese Academy of Sciences.Aspirin was provided by Sigma(USA).METHODS:The rats were randomly divided into four groups:sham-operation(SO),CIRI + vehicle,CIRI + aspirin(6 mg/kg),and CIRI + aspirin(60 mg/kg).Rats in the lesion groups were intragastrically administrated saline,aspirin(6 mg/kg),or aspirin(60 mg/kg),respectively.MAIN OUTCOME MEASURES:The number of pyramidal neurons with normal appearance in the cerebral cortex at 2-4 mm from the midline;apoptotic cell death as measured by TUNEL;Bcl-2 and Bax protein localization was determined by immunohistochemistry;malondialdehyde(MDA) and super oxidation(SOD) content were determined by biochemistry method;adenosine triphosphate(ATP) content measured by capillary electrophoresis.RESULTS:Following CIRI,the following parameters were altered compared with sham-operated animals:the number of neurons with normal appearance was significantly reduced in the cerebral cortex;the number of apoptotic cells increased;Bax protein expression was enhanced;and the ratio between Bcl-2 and Bax decreased.In addition,MDA content increased significantly,whereas ATP content decreased(P < 0.01).Aspirin ameliorated the loss of healthy pyramidal neurons.Both 6 and 60 mg/kg aspirin increased the ratio between Bcl-2 and Bax,with no significant difference between the treatment groups.In addition,60 mg/kg aspirin decreased MDA content and increased ATP levels.However,6 mg/kg aspirin did not have the same effect.CONCLUSION:Aspirin reduced the number of apoptotic cells following CIRI.These results suggest that the neuroprotective mechanism of aspirin could be related to elevated Bcl-2 protein levels or decreased Bax protein expression.The increase in the ratio of Bcl-2 to Bax appears to be a common anti-apoptotic mechanism of aspirin. | Liying Qiu Bin Du Ying Li Hongbin Fan Zhiyong Yang | 2008 | Neural Regeneration Research2008,3,9: | 4 |
| 2 | Changes in neuronal apoptosis and apoptosis modulatory factors in cerebral ischemia/reperfusion显示文摘BACKGROUND: The high concentration of glutamate release is the main cause for neuronal cell death. The relationship between glutamate level and apoptosis during ischemia/reperfusion injury is still unclear. OBJECTIVE: To observe the neuronal apoptosis at 24 and 72 hours following cerebral ischemia/reperfusion in rats,and analyze the possible influencing factors. DESIGN: A randomized controlled animal experiment. SETTING: School of Medicine,Southern Yangtze University. MATERIALS: Totally 30 male adult Sprague Dawley (SD) rats of clean grade,weighing 240-290 g,were obtained from Shanghai Experimental Animal Center,Chinese Academy of Sciences. The rats were randomly divided into sham-operated group (n=10) and model group (n=20). Each group was observed at 24 and 72 hours after ischemia/reperfusion,5 rats at each time point in the sham-operated group,whereas 12 at 24 hours and 8 at 72 hours in the model group. Kits for determining apoptosis and Bcl-2 were bought from Wuhan Boster Biological Technology,Co.,Ltd.; Kit for calcineurin from Nanjing Jiancheng Bioengineering Institute. METHODS: The experiment was carried out in the Functional Scientific Research Room of Southern Yangtze University from June to October in 2006. ① Right middle cerebral artery was occluded by inserting a thread through internal carotid artery (ICA). The surgical process for the sham-operated rats was the same as that in the model group except a nylon suture inserted the ICA. According to Longa five-degree standard,the neurological deficit evaluation of rats was evaluated after surgery,and grades 1-3 were taken as successful model establishment. The blood was recirculated by withdrawing the nylon filament under anesthesia at 2 hours after ischemia in successful rat models. ②After reperfusion,the brain tissue was quickly removed at 24 or 72 hours and the slices were obtained from optic chiasma to funnel manubrium. The changes of the number of apoptotic cells were observed using the terminal deoxynucleotidyl transferase mediated dUTP-biotin nick-end labeling method. The expressions of Bcl-2 protein were determined with immunohistochemical staining. The activity of calcineurin was determined by the inorganic phosphorus method. The content of excitatory amino acid was detected by high performance liquid chromatography. MAIN OUTCOME MEASURES: ① Glutanate content in brain tissue; ② Conditions of apoptosis; ③ Calcineurin activity in brain tissue; ④ Bcl-2 expression in brain tissue. RESULTS: Totally 30 SD rats were used,5 died and the other 25 were involved in the analysis of results. ① Changes of apoptosis: There were 0-3 apoptotic cells in the sham-operated group. In the model group,the numbers of apoptotic cells were obviously increased at 24 and 72 hours of reperfusion (P < 0.01),and it was markedly reduced at 72 hours as compared with 24 hours (P < 0.01). ② Changes of glutanate content: The glutamate contents at 24 and 72 hours of reperfusion in the model group were obviously higher than those in the sham-operated group (P < 0.01); In the model group,it was obviously increased at 24 hours as compared with 72 hours (P < 0.01). ③ Changes of Bcl-2 protein: In the model group,the Bcl-2 protein expression had no obvious changes at 24 hours of reperfusion,and it was obviously enhanced at 72 hours,which was obviously different from that in the sham-operated group and that at 24 hours (P < 0.01). ④ Changes of calcinerin activity: In the model group,the activity of calcineurin in brain tissue had no obvious changes at 24 hours of reperfusion; The activity of calcineurin at 72 hours was obviously higher than that in the sham-operated group and that at 24 hours (P < 0.01). | 3 | Cholecystokinin octapeptide antagonizes apoptosis in human retinal pigment epithelial cells显示文摘Although cholecystokinin octapeptide-8 is important for neurological function, its neuroprotective properties remain unclear. We speculated that cholecystokinin octapeptide-8 can protect human retinal pigment epithelial cells against oxidative injury. In this study, retinal pigment epithelial cells were treated with peroxynitrite to induce oxidative stress. Peroxynitrite triggered apoptosis in these cells, and increased the expression of Fas-associated death domain, Bax, caspase-8 and Bcl-2. These changes were suppressed by treatment with cholecystokinin octapeptide-8. | 13 | Highly stable aqueous phase black phosphorus quantum dots with enhanced fluorescence property显示文摘Black phosphorus quantum dots(BPQDs)are a kind of outstanding optical material due to the tunable band structure.However,their fluorescence property and stability are obviously weakened in the aqueous phase,which extremely restricts the development of BPQDs.Here,we propose a new method to prepare stable BPQDs in an aqueous solution directly with high absolute fluorescence quantum yield.Aqueous phase BPQDs are synthesized by liquid exfoliation and hydrothermal with NH_(2)-polyethylene glycol(PEG)-NH_(2) as the modification agent to protect the BPQDs,which have high stability more than six months.In addition,NH_(2)-PEG-NH_(2) is also a surface passivation agent to enhance the emission of BPQDs by forming P-N bonds,which is confirmed by an absolute fluorescent quantum yield of 11.5%.Moreover,BPQDs show excellent resistance to a strong acid environment and high ionic strengths except for Fe^(3+).Therefore,the BPQDs are a kind of highly selective and linear response fluorescence probe for Fe^(3+).Considering the good biocompatibility of BPQDs,they are employed as cell fluorescent label probes and show excellent fluorescence imaging contrast.Based on the unique structural stability and fluorescence performance in the aqueous phase,BPQDs are potential candidates for Fe^(3+)detection and optical bio-imaging. | Yingli Ma Xiaoguang Xu Xiaoshuang Zhang Mayifei Rong Liying Lu Yan Li Wenhao Dai Hongwu Du Yong Jiang | 2022 | Journal of Materials Science & Technology2022,,21: | 0 |
| 14 | Short-Term Prediction of Photovoltaic Power Based on Fusion Device Feature-Transfer显示文摘To attain the goal of carbon peaking and carbon neutralization,the inevitable choice is the open sharing of power data and connection to the grid of high-permeability renewable energy.However,this approach is hindered by the lack of training data for predicting new grid-connected PV power stations.To overcome this problem,this work uses open and shared power data as input for a short-term PV-power-prediction model based on feature transfer learning to facilitate the generalization of the PV-power-prediction model to multiple PV-power stations.The proposed model integrates a structure model,heat-dissipation conditions,and the loss coefficients of PV modules.Clear-Sky entropy,characterizes seasonal and weather data features,describes the main meteorological characteristics at the PV power station.Taking gate recurrent unit neural networks as the framework,the open and shared PV-power data as the source-domain training label,and a small quantity of power data from a new grid-connected PV power station as the target-domain training label,the neural network hidden layer is shared between the target domain and the source domain.The fully connected layer is established in the target domain,and the regularization constraint is introduced to fine-tune and suppress the overfitting in feature transfer.The prediction of PV power is completed by using the actual power data of PV power stations.The average measures of the normalized root mean square error(NRMSE),the normalized mean absolute percentage error(NMAPE),and the normalized maximum absolute percentage error(NLAE)for the model decrease by 15%,12%,and 35%,respectively,which reflects a much greater adaptability than is possible with other methods.These results show that the proposed method is highly generalizable to different types of PV devices and operating environments that offer insufficient training data. | Zhongyao Du Xiaoying Chen Hao Wang Xuheng Wang Yu Deng Liying Sun | 2022 | Energy Engineering2022,119,4: | 0 |

| 16 | Application of biofl oc technology in recirculation A rtemia culture system显示文摘Biofl oc technology(BFT)improves water quality,and productivity of the farmed species through converting ammonium nitrogen to microbial protein,stabilizing microbial community,and reducing the production cost.In this study,a small-scale biofl oc development unit was designed in combination of recirculation system(RAS)for Artemia culture.Artemia growth,water quality,and microbial composition of biofl ocs in RAS were studied in comparison with in-situ batch culture(Glu).Glucose was added in RAS and Glu at C/N ratio of 10.The cultures without glucose addition,but with 50%daily water renewal(WRe)and without water renewal(NWRe)were considered as the controls.Arte mia were cultured at 25℃ and salinity 30 for 24 days and fed formulated feed.The results showed that compared to the controls,Glu signifi cantly improved the Artemia biomass,increased the biofl oc volume,and reduced the content of total ammonia nitrogen(TAN),nitrite nitrogen(NO_(2)-N)and nitrate nitrogen(NO_(3)-N)in water column(P<0.05).In addition,RAS had similar results with Glu.High throughput sequencing analysis on biofl oc microbial composition demonstrated that glucose supplement shaped the microbial community structure,and increased proportion of potential probiotic bacteria and suppressed pathogenic bacteria growth.Furthermore,we analyzed the relationship between the microbial composition of biofl oc and environmental factors.Canonical correspondence analysis(CCA)indicated that inorganic nitrogen in culture water had great impact on biofl oc microbial composition in NWRe and WRe,whilst the dissolved organic carbon(DOC)modifi ed the microbial community in Glu and RAS.This study shows the advantages of BFT in Artemia culture and provides practical information for applying BFT-RAS in indoor Artemia culture. | Xuejiao LIANG Chi ZHANG Dongdong DU Meirong GAO Liying SUI | 2022 | Journal of Oceanology and Limnology2022,40,4: | 0 |
| 17 | Transcriptomes of Litopenaeus vannamei reveal modulation of antioxidant system induced by dietary archaeal carotenoids显示文摘Oxidative stress induced by factors such as ammonia nitrogen has become a major issue in shrimp farming.The effects of carotenoids on the growth and antioxidant capability of Litopenaeus vannamei juveniles were investigated in this study using dietary archaeal carotenoids supplementation.For four weeks,shrimp were given diets containing 0 mg/kg(Ctrl)and 55.98 mg/kg(Car)archaeal carotenoids.Dietary archaeal carotenoids significantly enhanced the astaxanthin content in shrimp muscles and carapaces,as well as the superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activity(P<0.05).The malonaldehyde(MDA)content in Car group significantly decreased(P<0.05).The transcriptome analysis was conducted to determine the molecular processes in response to archaeal carotenoids supplementation.A total of 1536 differentially expressed genes(DEGs)were detected,including 538 upregulated DEGs and 998 downregulated DEGs.GO functional enrichment analysis between Ctrl and Car indicated that 26 GO terms including extracellular region,metabolic process,and proteolysis were enriched.The KEGG pathway enrichment analysis revealed that the amino sugar and nucleotide sugar metabolism,cysteine and methionine metabolism,glycine serine and threonine metabolism,and amino acid biosynthesis were enriched.Archaeal carotenoids influenced the expression of several important genes involved in reactive oxygen species(ROS)generation,Nrf2 signaling,and antioxidant enzymes.Seven DEGs were chosen to confirm the RNA-Seq data using qRT-PCR.The genes and pathways discovered in this work assist to elucidate the molecular processes through which archaeal carotenoid enhances L.vannamei antioxidative system. | Wei XIE Guoru DU Honggang DENG Yingchao MA Meirong GAO Hu DUAN Sung YIK YEONG Liying SUI | 2023 | Journal of Oceanology and Limnology2023,41,5: | 0 |
| 18 | Large-scale preparation of high-performance boron nitride/aramid nanofiber dielectric composites显示文摘Dielectric polymers featuring high thermal conductivity,excellent mechanical,and stable dielectric properties over a broad temperature range have attracted extensive scientific attention.In this work,a large-scale,layered film was fabricated using blade-coating approach,which integrated aramid nanofibers(ANFs)and boron nitride nanosheets(BNNSs)through a typical solgel transformation procedure.The as-prepared film with 20 wt.%BNNS displays high thermal conductivity(14.03 W·m^(−1)·K^(−1)),103-fold higher than pure ANF film,attributing to massive continuous thermal conduction pathway between BNNSs so as to facilitate fast phonon transmission.The film boasts excellent mechanical properties(stress 97.14±5.17 MPa,strain 19.36±0.35%),high degradation temperature(~542℃),a moderate dielectric constant(~6.9 at 104 Hz),together with low dielectric loss(~0.026 at 104 Hz).Meanwhile,the film reveals high breakdown voltage(310 MV·m^(−1))and volume resistivity(1013Ω·cm).Notably,these dielectric properties remain largely unchanged over a wide temperature range(25 to 200℃). | Liying Su Xiaoyan Ma Jiale Zhou Xiangchen Liu Fanglin Du Chao Teng | 2022 | Nano Research2022,15,9: | 0 |
| 19 | Nanog reporter system in mouse embryonic stem cells based on highly efficient BAC homologous recombination显示文摘Nanog is a novel transcription factor specifically expressed in mouse embryonic stem cells (mES cells). It has been reported that Nanog plays an essential role in maintaining multi-potency of ES cells. The expression of Nanog is very sensitive to ES cells differentiation, making Nanog one of the best markers to indicate the status of ES cells. In this study, we developed an efficient method to construct Nanog promoter driven EGFP reporter system based on the BAC homologous recombination. We further generated a Nanog-EGFP reporter mES cell line. This reporter mES cell line exhibited features similar to those of normal mES cells, and the EGFP reporter efficiently reflected the expression of Nanog, indi- cating the differentiation status of mES cells. We achieved a reliable experimental reporter system to research self-renewal and differentiation of mES cells.
