| 1 | Component analysis of Chinese medicine and advances in fuming-washing therapy for knee osteoarthritis via unsupervised data mining methods显示文摘OBJECTIVE: To analyze the component law of Chinese medicines in fuming-washing therapy for knee osteoarthritis(KOA), and develop new fuming-washing prescriptions for KOA through unsupervised data mining methods.METHODS: Chinese medicine recipes for fuming-washing therapy for KOA were collected and recorded in a database. The correlation coefficient among herbs, core combinations of herbs, andnew prescriptions were analyzed using modified mutual information, complex system entropy cluster, and unsupervised hierarchical clustering, respectively.RESULTS: Based on analysis of 345 Chinese medicine recipes for fuming-washing therapy, 68 herbs occurred frequently, 33 herb pairs occurred frequently, and 12 core combinations were found.Five new fuming-washing recipes for KOA were developed.CONCLUSION: Chinese medicines for fuming-washing therapy of KOA mainly consist of wind-dampness-dispelling and cold-dispersing herbs, blood-activating and stasis-resolving herbs,and wind-dampness-dispelling and heat-clearing herbs. The treatment of fuming-washing therapy for KOA also includes dispelling wind-dampness and dispersing cold, activating blood and resolving stasis, and dispelling wind-dampness and clearing heat. | 4 | MiR-30a Positively Regulates the Inflammatory Response of Microglia in Experimental Autoimmune Encephalomyelitis显示文摘Multiple sclerosis(MS) is a classical inflammatory demyelinating disease of the central nervous system(CNS). Microglia are the main resident immune cells in the CNS and are closely associated with the pathogenesis of MS.In the present study, we found that mi R-30 a was highly expressed in jellyfish-like microglia in chronic active lesions of MS patients, as well as in the microglia of mice with experimental autoimmune encephalomyelitis(EAE) at the chronic phase. In vitro, the conditioned supernatant of mouse microglia overexpressing miR-30 a promoted the apoptosis of oligodendrocyte precursor cells(OPCs), and inhibited OPC differentiation. In vivo, overexpressing miR-30 a in transplanted microglia exacerbated the progression of EAE.Overexpression and knock-down experiments in primary cultured mouse microglia showed that mi R-30 a increased the expression of IL-1 b and i NOS, which are pro-inflammatory, while inhibiting the expression of Ym-1 and CD206.Mechanistically, mi R-30 a inhibited the expression of Ppargc1 b, which is the co-activator of peroxisome proliferator-activated receptor gamma, resulting in pro-inflammatory effects. | 12 | Regulation of Survivin and CDK4 by Epstein-Barr virus encoded latent mem-brane protein 1 in nasopharyngeal carcinoma cell lines显示文摘Latent membrane protein 1 (LMP1), an important protein encoded by Epstein Barr virus (EBV), has been implied to link with the pathogenesis of nasopharyngeal carcinoma (NPC). Its dual effects of increasing cell proliferation and inhibiting cell apoptosis have been confirmed. In this study, we showed that the expression of Survivin and CDK4 protein in CNE-LMP1, a LMP1 positive NPC epithelial cell line, is higher than in LMP1 negative NPC epithelial cell line- CNE1, and the expression is LMP1 dosage-dependent. Although it was reported that Survivin specifically expressed in cell cycle G2/M phase, our studies suggested that LMP1 could promote the expression of Survivin in G0/G1, S and G2/ M phase. It also showed that Survivin and CDK4 could be accumulated more in the nuclei triggered by LMP1. More interestingly, Survivin and CDK4 could form a protein complex in the nuclei of CNE-LMP1 rather than in that of CNE1, which demonstrated that the interaction between these two proteins could be promoted by LMP1. | 13 | Synergetic effects of aqueous extracts of Fuzi(Radix Aconiti Lateralis Preparata) and Tubeimu(Rhizoma Bolbostemmatis) on MDA-MB-231 and SKBR3 cells显示文摘OBJECTIVE: To test the synergistic effects of the aqueous extract of Tubeimu(Rhizoma Bolbostemmatis) and Fuzi(Radix Aconiti Lateralis Preparata) on MDA-MB-231 and SKBR3 breast cancer cells.METHODS: A combined index was created for the effects of Tubeimu(Rhizoma Bolbostemmatis) and Fuzi(Radix Aconiti Lateralis Preparata) extracts. Cell proliferation was performed by trypan blue exclusion and 3-(4,5-dimethylthiazol-2-yl)-5-(3- carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium(MTS) assays. Flow cytometry was used to assess cell cycle distribution and apoptosis. Cell migration was determined by wound-healing and transwell assays. Confocal microscopy was used to detect E-cadherin and actin filaments.RESULTS: The aqueous extract from Tubeimu(Rhizoma Bolbostemmatis) and Fuzi(Radix Aconiti Lateralis Preparata) exerted synergetic effects on the growth of MDA-MB-231 cells and G1 phase arrest.When exposed to extracts at concentrations of 62.5∶62.5 and 62.5∶31.3 μg/m L, the combination index was 0.83 and 0.74, respectively. Interestingly, 62.5∶31.3 μg/m L of combined drugs enhanced the inhibitory effect of Tubeimu(Rhizoma Bolbostemmatis)on the migration of SKBR3 cells and reduced the stimulative effect of Fuzi(Radix Aconiti Lateralis Preparata)(P < 0.01), in which cells showed an increased expression of E-cadherin and reorganization of actin filaments(P < 0.001). 62.5∶62.5 μg/m L extract also synergistically induced apoptosis of MDA-MB-231 cells(P < 0.05 and P < 0.001). | 14 | Expression level of beta-catenin is associated with prognosis of esophageal carcinoma显示文摘AIM: To examine the association of beta-catenin with the clinicopathologic features and prognosis of esophageal squamous cell carcinoma (ESCC). METHODS: Beta-catenin mRNA expression level in 40 ESCC patients (28 males and 12 females, age range 38-82 years, median 60 years) was analyzed by real- time PCR. Beta-catenin mRNA expression levels in tumor cells were categorized as weaker (level 1) or equal to or stronger (level 2) than those in endothelial cells. We examined the correlation between the beta-catenin expression and the clinicopathological factors and prognosis of ESCC patients. RESULTS: Level 2 beta-catenin expression was found in 29 patients. ESCC with level 2 expression had a higher rate of lymphnode metastasis (0.0776 ± 0.0369 vs 0.3413 ± 0.1803, P < 0.001) and deeper tumor invasion (0.0751 ± 0.0356 vs 0.3667 ± 0.1928, P < 0.001), and a poorer survival rate (P = 0.0024) than ESCC with level 1 expression. | 17 | MHD and kinetic analysis of flow bursts in the Earth's plasma sheet显示文摘In an attempt to study the flow bursts in the Earth’s plasma sheet we select an event that took place on August 7,2004 in the expansion phase of a substorm,using data from the geomagnetic index,solar wind data,plasma and magnetic field observations from C1 Cluster satellite(the Cluster mission has 4 satellites)and from Double Star TC-1 satellite.In MHD approach,TC-1 firstly observed the tailward flow,then the earthward,and finally the flow alternated in two directions.C1 firstly observed the earthward plasma flow,and then the tailward plasma flow.Before flow bursts are observed by TC-1 and C1,there are disturbances in local entropy with their tailward local entropy larger than those of the earthward.The kinetic features of the plasma flow observed by C1 are similar to those in MHD.However,kinetic characteristics of the plasma flow observed by TC-1 are far more than the description in MHD.The inadequacy mainly exists in two cases:(i)the firstly enhanced tailward flows given in MHD are found without significant increase of the energetic tailward flux;(ii)the almost stagnant flow in MHD is composed of the enhanced energetic ion flux in both earthward and tailward directions.The earthward flow burst observed by TC-1 might be multiple overshoots and rebounds.The earthward flow burst observed by C1 might be simply rebounded in the near-Earth.The pulsation observed by C1 is earlier than that observed by TC-1 with the former intensity less than that of the latter.After the energetic ion flux in the tailward direction is significantly enhanced,the power spectrum intensity of the ULF wave commences to increase obviously,which may suggest that the stream instability is closely correlated with ULF pulsations. | MA YuDuan CAO JinBin FU HuiShan REME H. Cx32 and Cx43 mRNA expression was detected by real-time polymerase chain reaction(PCR). Cx32 and Cx43 promoter CpG island methylation status was determined by methylation-specific PCR(MSP), bisulfite PCR sequencing(BSP) and MassArray methods. RESULTS: The relative mRNA expression levels in the gastric mucosa of patients with NGM, NAG, CAG, IM, DYS and GC were 0.146 ± 0.011, 0.133 ± 0.026, 0.107 ± 0.035, 0.039 ± 0.032, 0.037 ± 0.01 and 0.03 ± 0.011 for Cx32; and 0.667 ± 0.057, 0.644 ± 0.051, 0.624 ± 0.049, 0.555 ± 0.067, 0.536 ± 0.058 and 0.245 ± 0.121 for Cx43, respectively, which were gradually decreasing and significantly different(GC vs NGM: P < 0.001 for Cx32, P < 0.001 for Cx43). The promoter methylation levels in the gastric mucosa from NGM to GC stages by MSP were 38.8% ± 9.0%, 43.1% ± 9.4%, 56.5% ± 3.1%, 64.4% ± 9.7%, 72.5% ± 4.2% and 79.6% ± 6.8% for Cx32; and 49.0% ± 3.9%, 58.1% ± 5.0%, 66.5% ± 7.9%, 74.0% ± 8.8%, 78.3% ± 3.6% and 88.7% ± 6.2% for Cx43, respectively, which were gradually increasing and significantly different(P = 0.039, P = 0.019). The promoter methylation levels by BSP and MassArray exhibited similar trends. | 20 | An antibody against Siglec-15 promotes bone formation and fracture healing by increasing TRAP^(+)mononuclear cells and PDGF-BB secretion显示文摘Osteoporosis(OP)is a common age-related disease characterized by a deterioration of bone mass and structure that predisposes patients to fragility fractures.Pharmaceutical therapies that promote anabolic bone formation in OP patients and OP-induced fracture are needed.We investigated whether a neutralizing antibody against Siglec-15 can simultaneously inhibit bone resorption and stimulate bone formation.We found that the multinucleation of osteoclasts was inhibited in SIGLEC-15 conditional knockout mice and mice undergoing Siglec-15 neutralizing antibody treatment.The secretion of platelet-derived growth factor-BB(PDGF-BB),the number of tartrate-resistant acid phosphatase-positive(TRAP+)mononuclear cells,and bone formation were significantly increased in the SIGLEC-15 conditional knockout mice and antibody-treated mice.The anabolic effect of the Siglec-15 neutralizing antibody on bone formation was blunted in mice with Pdgfb deleted in TRAP-1'cells.These findings showed that the anabolic effect of the Siglec-15 neutralizing antibody was mediated by elevating PDGF-BB production of TRAP4 mononuclear cells.To test the therapeutic potential of the Siglec-15 neutralizing antibody,we injected the antibody in an ovariectomy-induced osteoporotic mouse model,which mimics postmenopausal osteoporosis in women,and in two fracture healing models because fracture is the most serious health consequence of osteoporosis.The Siglec-15 neutralizing antibody effectively reduced bone resorption and stimulated bone formation in estrogen deficiency-induced osteoporosis.Of note,the Siglec-15 neutralizing antibody promoted intramembranous and endochondral ossification at the damaged area of cortical bone in fracture healing mouse models.Thus,the Siglec-15 neutralizing antibody shows significant translational potential as a novel therapy for OP and bone fracture. | Gehua Zhen Yang Dan Ruomei Wang Ce Dou Qiaoyue Guo Melissa Zarr Linda N.Liu Lieping Chen Ruoxian Deng Yusheng Li Zengwu Shao Xu Cao | 2021 | Bone Research2021,9,4: | 6 |
