1Neonatal ketamine exposure-induced hippocampal neuroapoptosis in the developing brain impairs adult spatial learning ability显示文摘Ketamine exposure can lead to selective neuroapoptosis in the developing brain.p66ShcA,the cellular adapter protein expressed selectively in immature neurons,is a known pro-apoptotic molecule that triggers neuroapoptosis when activated.Sprague-Dawley rats at postnatal day 7 were subcutaneously injected in the neck with ketamine 20 mg/kg,six times at 2-hour intervals.At 0,1,3,and 6 hours after final injection,western blot assay was used to detect the expression of cleaved caspase-3,p66ShcA,and phosphorylated p66ShcA.We found that the expression of activated p66ShcA and caspase-3 increased after ketamine exposure and peaked at 3 hours.The same procedure was performed on a different group of rats.At the age of 4 weeks,spatial learning and memory abilities were tested with the Morris water maze.Latency to find the hidden platform for these rats was longer than it was for control rats,although the residence time in the target quadrant was similar.These findings indicate that ketamine exposure resulted in p66ShcA being activated in the course of an apoptotic cascade during the neonatal period.This may have contributed to the deficit in spatial learning and memory that persisted into adulthood.The experimental protocol was approved by the Institutional Animal Care and Use Committee at the University of Texas at Arlington,USA (approval No.A13.008) on January 22,2013.Dan Lyu Ning Tang Andrew W. 3Transport of Asian surface pollutants to the global stratosphere from the Tibetan Plateau region during the Asian summer monsoon显示文摘Due to its surrounding strong and deep Asian summer monsoon(ASM)circulation and active surface pollutant emissions,surface pollutants are transported to the stratosphere from the Tibetan Plateau region,which may have critical impacts on global climate through chemical,microphysical and radiative processes.This article reviews major recent advances in research regarding troposphere-stratosphere transport from the region of the Tibetan Plateau.Since the discovery of the total ozone valley over the Tibetan Plateau in summer from satellite observations in the early 1990s,new satellite-borne instruments have become operational and have provided significant new information on atmospheric composition.In addition,in situ measurements and model simulations are used to investigate deep convection and the ASM anticyclone,surface sources and pathways,atmospheric chemical transformations and the impact on global climate.Also challenges are discussed for further understanding critical questions on microphysics and microchemistry in clouds during the pathway to the global stratosphere over the Tibetan Plateau.Jianchun Bian Dan Li Zhixuan Bai Qian Li Daren Lyu Xiuji Zhou 2020National Science Review2020,7,3:5

7Iron-Imprinted Single-Atomic Site Catalyst-Based Nanoprobe for Detection of Hydrogen Peroxide in Living Cells显示文摘Fe-based single-atomic site catalysts(SASCs),with the natural metalloproteases-like active site structure,have attracted widespread attention in biocatalysis and biosensing.Precisely,controlling the isolated single-atom Fe-N-C active site structure is crucial to improve the SASCs’performance.In this work,we use a facile ion-imprinting method(IIM)to synthesize isolated Fe-N-C single-atomic site catalysts(IIM-Fe-SASC).With this method,the ion-imprinting process can precisely control ion at the atomic level and form numerous well-defined single-atomic Fe-N-C sites.The IIM-Fe-SASC shows better peroxidase-like activities than that of non-imprinted references.Due to its excellent properties,IIM-Fe-SASC is an ideal nanoprobe used in the colorimetric biosensing of hydrogen peroxide(H_(2)O_(2)).Using IIM-Fe-SASC as the nanoprobe,in situ detection of H_(2)O_(2)generated from MDA-MB-231 cells has been successfully demonstrated with satisfactory sensitivity and specificity.This work opens a novel and easy route in designing advanced SASC and provides a sensitive tool for intracellular H_(2)O_(2)detection.Zhaoyuan Lyu Shichao Ding Maoyu Wang Xiaoqing Pan Zhenxing Feng Hangyu Tian Chengzhou Zhu Dan Du Yuehe Lin 2021Nano-Micro Letters2021,13,9:2

10Analysis of cyclin E co-expression genes reveals nuclear transcription factor Y subunit alpha is an oncogene in gastric cancer显示文摘Objective: To explore genes potentially co-expressed with cyclin E in gastric cancer and discover possible targets for gastric cancer treatment. Methods: The Cancer Genome Atlas (TCGA) stomach adenocarcinoma sequencing data were used to predict genes co-expressed with cyclin E. Co-expression genes predicted by cBioPortal online analysis with Pearson correlation coefficient ≥0.4 were analyzed by gene ontology (GO) enrichment annotation using the PANTHER online platform (Ver. 7). Interactions between proteins encoded by these genes were analyzed using the STRING online platform (Ver. 10.5) and Cytoscape software (Ver. 3.5.1). Genes displaying a high degree of connection were analyzed by transcription factor enrichment prediction using FunRich software (Ver. 3). The significant transcription factor and cyclin E expression levels and their impact on gastric cancer progression were analyzed by Western blotting and Kaplan-Meier survival curve analysis. Results: After filtering the co-expression gene prediction results, 78 predicted genes that included 73 protein coding genes and 5 non-coding genes with Pearson correlation coefficient ≥0.4 were selected. The expressions of the genes were considered to be correlated with cyclin E expression. Among the 78 genes co-expressed with cyclin E, 19 genes at the central of the regulatory network associated with cyclin E were discovered. Nuclear transcription factor Y subunit alpha (NF-YA) was identified as a significant transcription factor associated with cyclin E co-expressing genes. Analysis of specimen donors’ clinical records revealed that high expression of NF-YA tended to be associated with increased cyclin E expression. The expression of both was associated with progression of gastric cancer. Western blotting results showed that compared with normal tissues, NF-YA and cyclin E were highly expressed in tumor tissues (P < 0.001). Survival curve analysis clearly demonstrated relatively poor overall survival of gastric cancer patients with high cyclin E or high NF-YA expression level, compared to patients with low cyclin E or NF-YA expression (P < 0.05). Conclusions: NF-YA may promote gastric cancer progression by increasing the transcription of cyclin E and other cell cycle regulatory genes. 15Taxonomic revision of Amolops chunganensis(Pope,1929)(Amphibia: Anura) and description of a new species from southwestern China, with discussion on Amolops monticola group and assignment of species groups of the genus Amolops显示文摘Amolops chunganensis is a species complex and reported widely from eastern,southern,and southwestern China.Based on molecular data of 19 populations of A.chunganensis sensu lato from China,including the population from Mt.Wuyi(type locality),we recognize A.chunganensis sensu stricto and provide an expanded description based on the topotypic specimens.Combining morphological and molecular data,we describe a new species,Amolops chaochin sp.nov.,from southwestern China,which was previously identified as A.chunganensis.The new species is distinguished from all other species in the A.monticola group by:(1)moderate body size,SVL 35.3-39.2 mm in males(n=7),and 50.5-54.4 mm in females(n=7);(2)distinct tympanum,larger than half of eye diameter;(3)small tooth-like projection on anteromedial edge of mandible;(4)circummarginal groove on all fingers;(5)white tubercles on dorsal side of posterior body in both sexes;(6)distinct tubercles on dorsal thigh and white spinose tubercles on dorsal tibia in both sexes;(7)white tubercles on posterior region of tympanum in males;(8)toe webbing reaching disk by dermal fringe on inner side of toe II;(9)vomerine teeth present;(10)transverse bands on dorsal limbs;(11)external vocal sacs present in males.We further reviewed the assignment of Amolops groups,with an overall revision of membership and diagnosis of all species groups.Ke Jiang Jin-Long Ren Zhi-Tong Lyu Dan Wang Zeng Wang Ke Lv Jia-Wei Wu Jia-Tang Li 2021Zoological Research2021,42,5:1

18Unconventional energetics of small vacancy clusters in BCC high-entropy alloy Nb_(0.75)ZrTiV_(0.5)显示文摘The stability of small vacancy clusters including divacancy,trivacancy and tetravacancy has been studied in body-centered cubic high-entropy alloy Nb_(0.75)ZrTiV_(0.5) in structures of random solid solution and short-range order by first-principles calculations and molecular dynamics simulations.Different from conventional body-centered cubic metals,the tightly bound configurations have a lower structural stability and are not preferred energetically in the studied high-entropy alloy.Instability of vacancy configurations leads to vacancy-atom exchanges that favor less compact configurations.The formation energy of small vacancy clusters is much smaller than its constituent elements of Nb and V due to the large structural adjustment induced by severe local lattice distortion.The difference in local lattice distortion and elemental arrangement in the vacancy neighborhood leads to significant site-to-site variation in vacancy cluster energy and configuration.The formation energy has a strong correlation with the local energy state of the vacancy configuration and the extent of structural relaxation.Compared to random solid solution,the structure of short-range order has a higher stability for the most compact cluster configurations and tends to have higher vacancy cluster formation energy.According to classical molecular dynamics simulations of cluster diffusion at high temperature,the studied high-entropy alloy has a higher probability of cluster dissociation compared to Nb and V.The unconventional energetics of small vacancy clusters is expected to have a profound impact on their generation,diffusion,dissociation,coalescence,as well as the defect microstructure evolution during irradiation.Tan Shi Xi Qiu Yundi Zhou Sixin Lyu Jing Li Dan Sun Qing Peng Yong Xin Chenyang Lu 2023Journal of Materials Science & Technology2023,,15:0

20Tracking Cryptosporidium in urban wastewater treatment plants in a cold region:Occurrence,species and infectivity显示文摘This study investigated the occurrence,species,infectivity and removal efficiency of Cryptosporidium spp.across typical wastewater treatment train.Samples from different process units were collected seasonally and synchronously from four wastewater treatment plants(WWTPs)in Northeastern China.Live Cryptosporidium oocysts were identified in most samples from both influent(97.50%)and effluent(90.00%)wastewaters of the four WWTPs,at an average density of 26.34 and 4.15 oocysts/L,respectively.The overall removal efficiency was 84.25%,and oocysts were mainly removed(62.01%)by the modified secondary sedimentation process.Ten Cryptosporidium species were identified in the effluent samples.C.andersoni,C.bovis,and C.ryanae were the three most prevalent species.Oocyst viability assays indicated no reduction of excystation rate during the primary and secondary wastewater treatments(varied in the range of 63.08%–68.50%),but the excystation rate declined to 52.21%in the effluent after disinfection.Notably,the Cryptosporidium oocysts showed higher infection intensity in the cold season(winter and spring)than that in summer and autumn.The influences of environmental temperature on virulence factors of Cryptosporidium were further examined.It was observed that more extracellular secretory proteins were bound on the oocyst surface and several virulence genes were expressed relatively strongly at low temperatures,both of which could facilitate oocyst adhesion,invasion,and host immune evasion.This research is of considerable interest since it serves as an important step towards more accurate panoramic recognition of Cryptosporidium risk reduction in WWTPs,and especially highlights the potential health risk associated with Cryptosporidium in cold regions/seasons.Dan Xiao Zhaofeng Lyu Shiheng Chen Yang Huo Wei Fan Mingxin Huo 2022Frontiers of Environmental Science & Engineering2022,16,9:0
