| 1 | Developing the Science Product Algorithm Testbed for Chinese Next-Generation Geostationary Meteorological Satellites:Fengyun-4 Series显示文摘Fengyun-4A(FY-4A), the first of the Chinese next-generation geostationary meteorological satellites, launched in2016, offers several advances over the FY-2: more spectral bands, faster imaging, and infrared hyperspectral measurements. To support the major objective of developing the prototypes of FY-4 science algorithms, two science product algorithm testbeds for imagers and sounders have been developed by the scientists in the FY-4 Algorithm Working Group(AWG). Both testbeds, written in FORTRAN and C programming languages for Linux or UNIX systems, have been tested successfully by using Intel/g compilers. Some important FY-4 science products, including cloud mask, cloud properties, and temperature profiles, have been retrieved successfully through using a proxy imager, Himawari-8/Advanced Himawari Imager(AHI), and sounder data, obtained from the Atmospheric Infra Red Sounder, thus demonstrating their robustness. Since 2005, La_2Hf_2O_7 was fabricated into transparent ceramics and much more attentions were paid on A_2B_2O_7 transparent ceramics for new applications. In this review, the development of A_2B_2O_7 system transparent ceramics was described. The structure characteristics, powder synthesis method, and sintering techniques of the final A_2B_2O_7 transparent ceramics were summarized. After that, the mostly reported A_2Hf_2O_7, A_2Zr_2O_7, and A_2Ti_2O_7 system transparent ceramics were systematically introduced. | 5 | A Phytophthora capsid Effector Targets ACD11 Binding Partners that Regulate ROS-Mediated Defense Response in Arabidopsis显示文摘Reactive oxygen species (ROS) play a vital role in plant immune response, but the genes involved in the regulation of ROS are scantily reported. Phytophthora pathogens produce a large number of effectors to promote infection, but the modes of action adopted are largely unknown. Here, we report that RxLR207 could activate ROS-mediated cell death in Nicotiana benthamiana and was essential for virulence of P. capsici. We found that this effector targeted BPA1 (binding partner of ACD11) and four members of BPLs (BPAl-Like proteins)in Arabidopsis, and the bpa1 and bpl mutants had enhanced ROS accumulation and cell death under biotic or abiotic stresses. Furthermore, we showed that BPA1 and several BPLs functioned redundantly in plant immunity to P. capsici. We discovered that BPA1 and all six BPLs interacted with ACD11, and stabilization of ACD11 was impaired in the bpa 1, bpl2, bpl3, and bpl4 mutants. RxLR207 could promote the degradation of BPA1, BPL1, BPL2, and BPL4 to disrupt ACD11 stabilization in a 26S proteasome-dependent manner. | 7 | Preclinical efficacy against acute myeloid leukaemia of SH1573, a novel mutant IDH2 inhibitor approved for clinical trials in China显示文摘Acute myeloid leukaemia(AML) is the most common form of acute leukaemia in adults,with increasing incidence with age and a generally poor prognosis.Almost 20% of AML patients express mutant isocitrate dehydrogenase 2(mIDH2),which leads to the accumulation of the carcinogenic metabolite 2-hydroxyglutarate(2-HG),resulting in poor prognosis.Thus,global institutions have been working to develop mIDH2 inhibitors.SH1573 is a novel mIDH2 inhibitor that we independently designed and synthesised.We have conducted a comprehensive study on its pharmacodynamics,pharmacokinetics and safety.First,SH1573 exhibited a strong selective inhibition of mIDH2 R140 Q protein,which could effectively reduce the production of 2-HG in cell lines,serum and tumors of an animal model.It could also promote the differentiation of mutant AML cell lines and granulocytes in PDX models.Then,it was confirmed that SH1573 possessed characteristics of high bioavailability,good metabolic stability and wide tissue distribution.Finally,toxicological data showed that SH1573 had no effects on the respiratory system,cardiovascular system and nervous system,and was genetically safe.This research successfully promoted the approval of SH1573 for clinical trials(CTR20200247).All experiments demonstrated that,as a potential drug against mIDH2 R140 Q acute myeloid leukaemia,SH1573 was effective and safe. | Zhiqiang Wang Zhibo Zhang Yong Li Li Sun Dezhen Peng Danyu Du Xian Zhang Luwei Han Liwen Zhao Ligong Lu Hongzhi Du Shengtao Yuan Meixiao Zhan | 2021 | Acta Pharmaceutica Sinica B2021,11,6: | 3 |

| 9 | Environmental behaviors of phoxim with two formulations in bamboo forest under soil surface mulching显示文摘Phoxim(emulsifiable concentrate(EC) and granules(G)) has been widely used in bamboo forests. The persistence and magnitude of phoxim residues in the crop and soil must be investigated to ensure human and environmental safety. The environmental behaviors of the two formulations were investigated in a bamboo forest under soil surface mulching conditions(CP) and non-covered cultivation conditions(NCP). The half-lives of phoxim in soil under the two conditions in soil were 4.1–6.2 days(EC) and 31.5–49.5 days(G),respectively. Phoxim in EC could be leached from the topsoil into the subsoil. A minimized leaching effect was observed for G under NCP. Inversely, an enhanced leaching effect was observed for G under CP. The G formulation resulted in more parent compound(in bamboo shoots) and metabolite(in soil) residues of phoxim than in the case of EC, especially under CP conditions. In addition, the intensity and duration of the formulation effect on soil p H adjustment from G were more obvious than that from EC. Results showed that the environmental behaviors(distribution, degradation, residue) of phoxim in the bamboo forest were significantly influenced by the type of formulation. The prolongation effect from phoxim G might cause persistence and long-term environmental risk. | 12 | Sequence Diversity and Enzyme Activity of Ferric-Chelate Reductase LeFRO1 in Tomato显示文摘Ferric-chelate reductase which functions in the reduction of ferric to ferrous iron on root surface is a critical protein for iron homeostasis in strategy I plants.LeFRO1 is a major ferric-chelate reductase involved in iron uptake in tomato.To identify the natural variations of LeFRO1 and to assess their effect on the ferric-chelate reductase activity,we cloned the coding sequences of LeFRO1 from 16 tomato varieties collected from different regions,and detected three types of LeFRO1(LeFR01^(MM),LeFRO1^(Ailsa) and LeFRO1^(Monita)) with five amino acid variations at the positions 21,24,112,195 and 582.Enzyme activity assay revealed that the three types of LeFRO1 possessed different ferric-chelate reductase activity(LeFRO1^(Ailsa)>LeFRO1^(MM)>LeFRO1^(Monita)).The 112th amino acid residue Ala of LeFRO1 is critical for maintaining the high activity of ferric-chelate reductase,because modification of this amino acid resulted in a significant reduction of enzyme activity.Further,we showed that the combination of the amino acid residue Ile at the site 24 with Lys at the site 582 played a positive role in the enzyme activity of LeFRO1.In conclusion,the findings are helpful to understand the natural adaptation mechanisms of plants to iron-limiting stress,and may provide new knowledge to select and manipulate LeFRO1 for improving the iron deficiency tolerance in tomato. | Danyu Kong Chunlin Chen Huilan Wu Ye Li Junming Li Hong-Qing Ling | 2013 | Journal of Genetics and Genomics2013,40,11: | 2 |
