1Colorectal cancer and 18FDG-PET/CT:What about adding the T to the N parameter in loco-regional staging?显示文摘AIM:To evaluate whether FDG-positron emission tomography(PET)/computed tomography(CT)may be an accurate technique in the assessment of the T stage in patients with colorectal cancer.METHODS:Thirty four consecutive patients(20 men and 14 women;mean age:63 years)with a histologically proven diagnosis of colorectal adenocarcinoma and scheduled for surgery in our hospital were enrolled in this study.All patients underwent FDG-PET/CT preoperatively.The primary tumor site and extent were evaluated on PET/CT images.Colorectal wall invasion was analysed according to a modified T classification that considers only three stages(≤T2,T3,T4).Assessment of accuracy was carried out using 95%confidence intervals for T.RESULTS:Thirty five/37(94.6%)adenocarcinomas were identified and correctly located on PET/CT images.PET/CT correctly staged the T of 33/35 lesions identified showing an accuracy of 94.3%(95%CI:87%-100%).All T1,T3 and T4 lesions were correctly staged,while two T2 neoplasms were overstated as T3.CONCLUSION:Our data suggest that FDG-PET/CT may be an accurate modality for identifying primary tumor and defining its local extent in patients with colorectal cancer.Pier Paolo Mainenti Delfina Iodice Sabrina Segreto Giovanni Storto Mario Magliulo Giovanni Domenico De Palma Marco Salvatore Leonardo Pace 2011World Journal of Gastroenterology2011,17,11:5
2HASTY modulates miRNA biogenesis by linking pri-miRNA transcription and processing显示文摘Post-transcriptional gene silencing mediated by microRNAs(miRNAs)modulates numerous developmental and stress response pathways.For the last two decades,HASTY(HST),the ortholog of human EXPORTIN 5,was considered to be a candidate protein that exports plant miRNAs from the nucleus to the cytoplasm.Here,we report that HST functions in the miRNA pathway independent of its cargo-exporting activity in Arabidopsis.We found that Arabidopsis mutants with impaired HST shuttling exhibit normal subcellular distribution of miRNAs.Interestingly,protein-protein interaction and microscopy assays showed that HST directly interacts with the microprocessor core component DCL1 through its N-terminal domain.Moreover,mass spectrometry analysis revealed that HST also interacts independently of its N-terminal domain with the mediator complex subunit MED37.Further experiments revealed that HST could act as a scaffold to facilitate the recruitment of DCL1 to genomic MIRNA loci by stabilizing the DCL1-MED37 complex,which in turn promotes the transcription and proper processing of primary miRNA transcripts(primiRNAs).Taken together,these results suggest that HST is likely associated with the formation of the miRNA biogenesis complex at MIRNA genes,promoting the transcription and processing of primiRNAs rather than the direct export of processed miRNAs from the nucleus.Damian A.Cambiagno Axel J.Giudicatti Agustin L.Arce Delfina Gagliardi Lei Li Wei Yuan Derek S.Lundberg Detlef Weigel Pablo A.Manavella 2021Molecular Plant2021,14,3:5
3CURLY LEAF Regulates MicroRNA Activity by Controlling ARGONAUTE 1 Degradation in Plants显示文摘CURLY LEAF ( CLF) encodes the methyltransferase subunit of the Polycomb Repressor Complex 2 (PRC2), which regulates the expression of target genes through H3K27 trimethylation. We isolated a new CLF mutant allele ( clf-78) using a genetic screen designed to identify microRNA (miRNA) deficient mutants. CLF mutant plants showed impaired miRNA activity caused by increased ubiquitination and enhanced degradation of ARGONAUTE 1 (AGO1) in specific tissues. Such CLF-mediated AGO1 regulation was evident when plants were exposed to UV radiation, which caused increased susceptibility of clf mutants to some UV-induced responses. Furthermore, we showed that CLF directly regulates FBW2, which in turn triggers AGO1 degradation in the clf mutants. Interestingly, AGO1 bound to a target appeared particularly prone to degradation in the mutant plants, a process that was exacerbated when the complex bound a non-cleavable target. 9Intimate partner violence:A loop of abuse,depression and victimization显示文摘Intimate partner violence has been recognized as a serious public health issue.Exposure to violence contributes to the genesis of,and exacerbates,mental health conditions,and existing mental health problems increase vulnerability to partner violence,a loop that imprisons victims and perpetuates the abuse.A recently described phenomenon is when male violence against females occurs within intimate relationships during youth,and it is termed adolescent or teen dating violence.In this narrative review,factors associated with intimate partner violence and consequences of exposure of children to parental domestic violence are discussed,along with possible intensification of violence against women with the spread of coronavirus disease 2019 pandemic and subsequent lockdown.Intervention programs with a multicomponent approach involving many health care settings and research have a pivotal role in developing additional strategies for addressing violence and to provide tailored interventions to victims.Prevention policy with a particular attention on healthy child and adolescent development is mandatory in the struggle against all forms of violence.Marianna Mazza Giuseppe Marano Angela Gonsalez del Castillo Daniela Chieffo Laura Monti Delfina Janiri Lorenzo Moccia Gabriele Sani 2021World Journal of Psychiatry2021,11,6:1

13C-reactive protein is differentially modulated by co-existing infections,vitamin deficiencies and maternal factors in pregnant and lactating indigenous Panamanian women显示文摘Background:The usefulness of C-reactive protein(CRP)as a non-specific marker of inflammation during pregnancy and lactation is unclear in impoverished populations where co-existing infections and vitamin deficiencies are common.Methods:This cross-sectional study in Panama recruited 120 pregnant and 99 lactating Ngäbe-Buglewomen from 14 communities in rural Panama.Obstetric history,indoor wood smoke exposure,fieldwork,BMI,vitamins A,B_(12),D,and folic acid,and inflammation markers(CRP,neutrophil/lymphocyte ratio(NLR),plateletcrit and cytokines)were measured.Multiple regressions explored both associations of CRP with other inflammatory markers and associations of CRP and elevated CRP based on trimester-specific cut-offs with maternal factors,infections and vitamin deficiencies.Results:CRP was higher in pregnancy(51.4±4.7 nmol/L)than lactation(27.8±3.5 nmol/L)and was elevated above trimester specific cut-offs in 21%of pregnant and 30%of lactating women.Vitamin deficiencies were common(vitamin A 29.6%;vitamin D 68.5%;vitamin B1268%;folic acid 25.5%)and over 50%of women had two or more concurrent deficiencies as well as multiple infections.Multiple regression models highlighted differences in variables associated with CRP between pregnancy and lactation.In pregnancy,CRP was positively associated with greater indoor wood smoke exposure,caries and hookworm and negatively associated with Ascaris and vaginal Lactobacillus and Bacteroides/Gardnerella scores.Consistent with this,greater wood smoke exposure,caries as well as higher diplococcal infection score increased the odds of trimester-elevated CRP concentrations whereas longer gestational age lowered the likelihood of a trimester-elevated CRP.During lactation,folic acid deficiency was associated with higher CRP whereas parity,number of eosinophils and Mobiluncus score were associated with lower CRP.Also,a higher BMI and Trichomonas vaginalis score increased the likelihood of an elevated CRP whereas higher parity and number of eosinophils were associated with lower likelihood of an elevated CRP.Conclusions:Infections both raise and lower CRP concentrations in pregnant and lactating mothers.Only folic acid deficiency during lactation was associated with higher CRP concentrations.Caution is required when interpreting CRP concentrations in pregnant and lactating women who have co-existing nutrient deficiencies and multiple infections.Doris Gonzalez-Fernandez Emerita del Carmen Pons Delfina Rueda Odalis Teresa Sinisterra Enrique Murillo Marilyn E.Scott Kristine G.Koski 2017Infectious Diseases of Poverty2017,6,1:1
