2A deep learning-integrated micro-CT image analysis pipeline for quantifying rice lodging resistance-related traits显示文摘Lodging is a common problemin rice,reducing its yield andmechanical harvesting efficiency.Rice architecture is a key aspect of its domestication and a major factor that limits its high productivity.The ideal rice culm structure,includingmajor_axis_culm,minor axis_culm,andwall thickness_culm,is critical for improving lodging resistance.However,the traditionalmethod ofmeasuring rice culms is destructive,time consuming,and labor intensive.In this study,we used a high-throughput micro-CT-RGB imaging system and deep learning(SegNet)todevelopa high-throughputmicro-CTimageanalysis pipelinethatcanextract 24 riceculmmorphological traits and lodging resistance-related traits.When manual and automatic measurements were compared at themature stage,the mean absolute percentage errors for major_axis_culm,minor_axis_culm,andwall_thickness_culmin 104 indica rice accessionswere 6.03%,5.60%,and 9.85%,respectively,and the R^(2) valueswere 0.799,0.818,and 0.623.We also builtmodels of bending stress using culmtraits at the mature and tillering stages,and the R^(2) values were 0.722 and 0.544,respectively.The modeling results indicated that this method can quantify lodging resistance nondestructively,even at an early growth stage.In addition,we also evaluated the relationships of bending stress toshoot dryweight,culm density,and drought-related traits and found that plants with greater resistance to bending stress had slightly higher biomass,culm density,and culm area but poorer drought resistance.In conclusion,we developed a deep learning-integrated micro-CT image analysis pipeline to accurately quantify the phenotypic traits of rice culms in4.6 min per plant;this pipeline will assist in future high-throughput screening of large rice populations for lodging resistance.Di Wu Dan Wu Hui Feng Lingfeng Duan Guoxing Dai Xiao Liu Kang Wang Peng Yang Guoxing Chen Alan P.Gay John H.Doonan Zhiyou Niu Lizhong Xiong Wanneng Yang 2021Plant Communications2021,2,2:7
3Hepatitis B virus X protein enhances hepatocarcinogenesis by depressing the targeting of NUSAP1 mRNA by miR-18b显示文摘Objective: The aim of this study was to investigate the underlying mechanism whereby HBx modulates the targeting of NUSAP1 by miR-18b to enhance hepatocarcinogenesis.Methods: We employed an integrated approach of bioinformatics analysis and molecular experiments in hepatoma cells, HBV transgenic mice, and clinical liver cancer tissues to investigate the role of HBx-regulated miR-18b in the development of liver cancer.Results: In this study, we report that the HBx-mediated tumor suppressor miR-18b modulates hepatocarcinogenesis during the host-HBV interaction. The expression levels of miR-18b were lower in clinical HBV-positive liver cancer tissues and liver tissues of HBV-transgenic mice. Interestingly, HBx inhibited miR-18b expression by inducing the methylation of CpG islands in its promoter. Accordingly, we tested the hypothesis that HBx enhanced hepatocarcinogenesis by increasing the expression of target genes of miR-18b. Moreover, we identified nucleolar spindle-associated protein 1(NUSAP1) as one of the target genes of miR-18b.NUSAP1 was expressed at high levels in liver cancer tissues. Interestingly, HBx up-regulated NUSAP1 by suppressing miR-18b.Functionally, miR-18b significantly inhibited the proliferation of hepatoma cells by depressing NUSAP1 levels in vivo and in vitro.Conclusions: Thus, we conclude that the targeting of NUSAP1 mRNA by the tumor suppressor miR-18b is controlled by HBxmodulated promoter methylation during the host-virus interaction, leading to hepatocarcinogenesis. 11Finite Element Analysis for In-Plane Crushing Behaviour of Aluminium Honeycombs显示文摘A number of finite element simulations were performed to analyze the in-plane crushing behaviour of aluminium honeycombs and the results are presented and discussed. The simulations include both X1 and X2 cases. All the analyses are quasi-static, and can be divided into three groups, which are designed to investigate the effects of cell size, foil thickness and yield stress of the foil material, respectively, on the structural response of honeycombs. The result indicates that these factors can significantly affect the plateau stresses of honeycomb cellular structures in both directions, and the plateau stresses in X2 direction are slightly smaller than those in X1 direction. The simulation results were further compared with published theoretical predictions and show higher values. 13High-throughput volumetric reconstruction for 3D wheat plant architecture studies显示文摘For many tller crops,the plant archit ecture(PA),including the plant fresh weight,plant height,number of tllrs,tller angle and stem diameter,sigificantly afects the grain yield.In this study,we propose a method based on volumetric reconstruction for high-throughput three-dimensional(3D)wheat PA studies.The proposed methodology involves plant volumetric reconst ruction from multiple images,plant model processing and phenotypic parameter estimation and analysis.This study was performed on 80 Triticum aestium plants,and the results were analyzed.Comparing the automated measurements with manual measurements,the mean absolute per-centage error(MAPE)in the plant height and the plant fresh weight was 2.71%(1.08cm with an average plant height of 40.07cm)and 10.06%(1.41g with an average plant fresh weight of 14.06 g),respectively.The root mean square error(RMSE)was 137 cm and 1.79g for the plant height and plant fresh weight,respectively.The correlation cofficients were 0.95 and 0.96 for the plant height and plant fresh weight,respectively.Additionally,the proposed methodology,in-cluding plant reconstruction,model processing and trait ext raction,required only approximately 20s on average per plant using parallel computing on a graphics processing unit(GPU),dem-onstrating that the methodology would be valuable for a high-throughput phenotyping platform.Wei Fang Hui Feng Wanneng Yang Lingfeng Duan Guoxing Chen Lizhong Xiong Qian Liu 2016Journal of Innovative Optical Health Sciences2016,9,5:1

17Syn-Faulting Calcite Ages:Constraint for the Late Mesozoic Deformation of the Ningzhen Mountain,Eastern China显示文摘It is difficult to date a brittle fault because of the absence of effective dating methods.Recently,calcite U-Pb dating has drawn the attention of many researchers and has been successfully applied to constrain the age of brittle deformation.The South China Block(SCB)has experienced multiphase deformations characterized by widespread brittle faults and folds,which has led to significant debate on the Mesozoic dynamic mechanism and deformational phases.A syn-faulting calcite vein that occurred in the Permian limestones of the Ningzhen Mountain region was analyzed using U-Pb dating and geochemistry techniques in this paper.The U-Pb dating results display multiphase activities with two mixed ages of 109.9±5.8 Ma(MSWD=46,n=231)and 117.2±2.4 Ma(MSWD=3.6,n=150).The age of 117.2±2.4 Ma likely represents the timing of the brittle faults.Trace elements and rare earth elements data indicate that these fault-related calcites are mainly derived from host limestones and unrelated to Cretaceous magmatic activity.These faults formed in a tectonic reactive period and dormant time of the adakitic magma in the Lower Yangtze region.Jinbao Su Hongbing Tan Yuexing Feng Guoxing Qin 2021Journal of Earth Science2021,32,6:0
18Seismic stability of expansive soil slopes reinforced by anchor cables using a modified horizontal slice method显示文摘Earthquake-induced slope failures are common occurrences in engineering practice and pre-stressed anchor cables are an effective technique in maintaining slope stability,especially in areas that are prone to earthquakes.Furthermore,the soil at typical engineering sites also exhibit unsaturated features.Explicit considerations of these factors in slope stability estimations are crucial in producing accurate results.In this study,the seismic responses of expansive soil slopes stabilized by anchor cables is studied in the realm of kinematic limit analysis.A modified horizontal slice method is proposed to semi-analytically formulate the energy balance equation.An illustrative slope is studied to demonstrate the influences of suction,seismic excitations and anchor cables on the slope stability.The results indicate that the stabilizing effect of soil suction relates strongly to the seismic excitation and presents a sine shape as the seismic wave propagates.In higher and steeper slopes,the stabilizing effect of suction is more evident.The critical slip surface tends to be much more shallow as the seismic wave approaches the peak and vice versa.Wang Long Chen Guoxing Hu Wei Zhou Enquan Feng Jianxue Huang Anping 2024Earthquake Engineering and Engineering Vibration2024,23,2:0
19Identification of binding domains and key amino acids involved in the interaction between BmLARK and G4 structure in the BmPOUM2 promoter in Bombyx mori显示文摘It has been found that the non-B form DNA structures,like G-quadruplex(G4)and i-motif,are involved in many important biological processes.Our previous study showed that the silkworm transcription factor BmLARK binds to the G4 structure in the promoter of the transcription factor BmPOUM2 and regulates its promoter activity.However,the binding mechanism between BmLARK and BmPOUM2 G4 structure remains unclear.In this study,binding domains and key amino acid residues involved in the interaction between BmLARK and BmPOUM2 G4 were studied.The electrophoretic mobility shift assay results indicated that the two RNA-recognition motifs(RRM)of BmLARK are simultaneously required for the binding with the G4 structure.Either RRM1 or RRM2 alone could not bind with the G4 structure.The zinc-finger motif was not involved in the binding.A series of mutant proteins with specific amino acid mutations were expressed and used to identify the key amino acid residues involving the interaction.The results indicated thatβsheets,especially theβ1 andβ3 sheets,in the RRM domains of BmLARK played critical roles in the binding with the G4 structure.Several amino acid mutations of RRM1/2 in ribonucleoprotein domain 1(RNP1)(motif inβ3 strand)and RNP2(motif inβ1 strand)caused loss of binding ability,indicating that these amino acids are the key sites for the binding.All the results suggest that RRM domains,particularly their the RNP1 and RNP2 motifs,play important roles not only in RNA recognition,but also in the G4 structure binding.Yuling Peng Kangkang Niu Guoxing Yu Mingxi Zheng Qiulan Wei Qisheng Song Qili Feng 2021Insect Science2021,28,4:0
20Surface modification of γ-Al_2O_3 by sodium ions显示文摘1H/23Na double resonance NMR techniques were employed to study the modification of surface hydroxyls on Y-Al2O3 by sodium ions. 1H-23Na cross polarization (CP) experiment can resolve three kinds of sodium cations that are closely associated with surface hydroxyl groups, while the signal of deposited salt-Na2CO3, which is not connected to surface hydroxyl groups, is completely suppressed. 1H{23Na} spin echo double resonance experiments reveal the surface modification in more detail. At low Na+ coverage (5%, 10%), the acidic hydroxyl groups are preferentially coordinated with sodium ions, while both the acidic and the basic hydroxyl groups are accessible for sodium ions at highcoordination that causes proton of the OH groups to be easily dissolved, which is evidenced by the fact that with calcination temperature of the catalysts increasing, the acidic hydroxyl groups are preferentially removed and only the basic hydroxyl groups remain when the calcination temperature is raised to 600℃.DENG Feng, YUE Yong, DU Youru, YE Chaohui, KONG Yuhua, WANG Guoxing & LI Xiaoding1.
