1Production of Two Elite Glutinous Rice Varieties by Editing Wx Gene显示文摘The waxy gene(Wx) in rice, which encodes the granule bound starch synthase enzyme, is responsible for amylose synthesis. Glutinous(sticky) rice has little or no amylose that can be used in various applications, such as brewing. In this study, knockout of the Wx gene with CRISPR/Cas9 technology was conducted in two elite japonica rice lines, Huaidao 5(HD5) and Suken 118(SK118), aiming to develop elite sticky rice varieties. We achieved six homozygous T_0 plants with more than 200 bp deletion in the Wx gene, as well as 36 wx-HD5 and 18 wx-SK118 homozygous transgene-free plants in the T_1 generation. The seeds of all the mutants were white and opaque, similar to those of sticky rice, and contained only 2.6%–3.2% amylose. Results of scanning electron microscopy showed that the quality of rice did not change. 2Geniposide, the component of the Chinese herbal formula Tongluojiunao, protects amyloid-β peptide(1–42)-mediated death of hippocampal neurons via the non-classical estrogen signaling pathway显示文摘Tongluojiunao(TLJN) is an herbal medicine consisting of two main components, geniposide and ginsenoside Rg1. TLJN has been shown to protect primary cultured hippocampal neurons. However, its mechanism of action remains unclear. In the present study, primary cultured hippocampal neurons treated with Aβ1–42(10 μmol/L) significantly increased the release of lactate dehydrogenase, which was markedly reduced by TLJN(2 μL/mL), specifically by the component geniposide(26 μmol/L), but not ginsenoside Rg1(2.5 μmol/L). The estrogen receptor inhibitor, ICI182780(1 μmol/L), did not block TLJN- or geniposide-mediated decrease of lactate dehydrogenase under Aβ1–42-exposed conditions. However, the phosphatidyl inositol 3-kinase or mitogen-activated protein kinase pathway inhibitor, LY294002(50 μmol/L) or U0126(10 μmol/L), respectively blo cked the decrease of lactate dehydrogenase mediated by TLJN or geniposide. 5Interaction of arterial proteoglycans with low density lipoproteins(LDLs):From theory to promising therapeutic approaches显示文摘Although atherosclerosis is a multifactorial process,proteoglycans mediated lipoprotein(LDL)retention at the subendothelial space is a necessary and sufficient event in provoking lesion initiation.Proteoglycans(PGs)are usually composed of one core protein backbone with one or more glycosaminoglycan chains(GAGs)covalently linked,mainly include perlecan,biglycan,versican,and decorin.The interaction between LDL and proteoglycans is apparently mediated by the basic amino acids in apoB-100,the moiety of LDL,electrostatic interacting with the negatively charged GAGs(sulfate or carbohydrate groups)of proteoglycans or though some bridge molecules like sphingomyelinase(SMase)or lipoprotein lipase(LpL).In the later section,we collate the promising therapeutic approaches that have been proposed up to now,targeting LDL-PGs interaction.It should be concluded that previous studies on interaction between LDL and PGs mainly focused on perlecan,biglycan,decorin,and versican that all located in the extracellular matrix(ECM),future studies should pay more attention to the endothelial surface glycocalyx and its interaction with LDLs,seeking promising therapeutic targets more specifically.Hongyan Kang Jinyan Lu Jiali Yang Yubo Fan Xiaoyan Deng 2019Medicine in Novel Technology and Devices2019,,3:1

12Completely positive tensors in the complex field显示文摘In this paper, we introduce the complex completely positive tensor, which has a symmetric complex decomposition with all real and imaginary parts of the decomposition vectors being non-negative. Some properties of the complex completely positive tensor are given. A semidefinite algorithm is also proposed for checking whether a complex tensor is complex completely positive or not. We give a characterization of PP-matrices. A semidefinite algorithm is presented for checking whether a matrix is partially positive or not. Its properties are studied. 16Subspace choices for the Celis-Dennis-Tapia problem显示文摘Grapiglia et al.(2013) proved subspace properties for the Celis-Dennis-Tapia(CDT) problem. If a subspace with lower dimension is appropriately chosen to satisfy subspace properties, then one can solve the CDT problem in that subspace so that the computational cost can be reduced. We show how to find subspaces that satisfy subspace properties for the CDT problem, by using the eigendecomposition of the Hessian matrix of the objection function. The dimensions of the subspaces are investigated. 17Insights into varicella-zoster virus assembly from the B-and C-capsid at near-atomic resolution structures显示文摘Varicella-zoster is a highly communicable virus that can be transmitted through the airborne route.About one quarter of people are infected with this virus.Previous studies have described the structure of A-capsid and a blurred reconstruction of the C-capsid with icosahedral symmetry.In this study,we have determined the more precise detailed structures of the varicella-zoster virus(VZV)B-and C-capsid in icosahedral symmetry using a combination of block-based reconstruction and symmetry relaxation strategies.In addition,we are reporting structural details of the portal vertex reconstructions in five-fold symmetry and portal reconstructions in twelve-fold symmetry.The structures unveil the basis for the high thermal stability of the VZV capsid.The conformational flexibility of structural elements of the capsid plays a role in the assembly of the capsid and drives processes critical for the viral life cycle.The results of the study open up new avenues for the development of drugs against a highly prevalent and contagious pathogen.Lei Cao Nan Wang Zhe Lv Wenyuan Chen Zhonghao Chen Lifei Song Xueyan Sha Guiqiang Wang Yaling Hu Xiaojun Lian Guoliang Cui Jinyan Fan Yaru Quan Hongrong Liu Hai Hou Xiangxi Wang 2024hLife2024,2,2:0
