1A Novel Mitogen-Activated Protein Kinase Gene in Maize (Zea mays),ZmMPK3,is Involved in Response to Diverse Environmental Cues显示文摘In search for components of mitogen-activated protein kinase (MAPK) cascades in maize (Zea mays) involved in response to abscisic acid (ABA) stimulus,a novel MAPK gene,ZmMPK3,from ABA-treated maize leaves cDNA was isolated and characterized.The full length of the ZmMPK3 gene is 1 520 bp and encodes a 376 amino acid protein with a predicted molecular mass of 43.5 kD and a pI of 5.83.ZmMPK3 contains all 11 MAPK conserved subdomains and the phosphorylation motif TEY.Amino acid sequence alignment revealed that ZmMPK3 shared high identity with group-A MAPK in plants.A time course (30–360 min) experiment using a variety of signal molecules and stresses revealed that the transcripts level of ZmMPK3 accumulated markedly and rapidly when maize seedlings were subjected to exogenous signaling molecules: ABA,H2O2,jasmonic acid and salicylic acid,various abiotic stimuli such as cold,drought,ultraviolet light,salinity,heavy metal and mechanical wounding.Its transcription was also found to be tissue-specific regulated.Here,we show that ABA and H2O2 induced a significant increase in the ZmMPK3 activity using immunoprecipitation and in-gel kinase assay.Furthermore,the results showed that the ZmMPK3 protein is localized mainly to the nucleus.These results suggest that the ZmMPK3 may play an important role in response to environmental stresses.Jinxiang Wang Haidong Ding Aying Zhang Fangfang Ma Jianmei Cao Mingyi Jiang 2010Journal of Integrative Plant Biology2010,52,5:16
2Variation in cis-regulation of a NAC transcription factor contributes to drought tolerance in wheat显示文摘Drought is a major environmental factor limiting wheat production worldwide,and developing drought-tolerant cultivars is a central challenge for wheat breeders globally.Therefore,it is important to identify genetic components determining drought tolerance in wheat.In this study,we identified a wheat NAC gene(TaNAC071-A)that is tightly associated with drought tolerance by a genome-wide association study.Knockdown of TaNAC071-A in wheat attenuated plant drought tolerance,whereas its overexpression significantly enhanced drought tolerance through improved water-use efficiency and increased expression of stress-responsive genes.This heightened water-saving mechanism mitigated the yield loss caused by water deficit.Further candidate gene association analysis showed that a 108-bp insertion in the promoter of TaNAC071-A alters its expression level and contributes to variation in drought tolerance among wheat accessions.This insertion contains two MYB cis-regulatory elements(CREs)that can be directly bound by the MYB transcription activator,TaMYBL1,thereby leading to increased TaNAC071-A expression and plant drought tolerance.Importantly,introgression of this 108-bp insertion allele,TaNAC071-AIn-693,into drought-sensitive cultivars could improve their drought tolerance,demonstrating that it is a valuable genetic resource for wheat breeding.Taken together,our findings highlight a major breakthrough in determining the genetic basis underlying phenotypic variation in wheat drought tolerance and showcase the potential of exploiting CRE-containing indels for improving important agronomical traits.Hude Mao Shumin Li Bin Chen Chao Jian Fangming Mei Yifang Zhang Fangfang Li Nan Chen Tian Li Linying Du Li Ding Zhongxue Wang Xinxiu Cheng Xiaojing Wang Zhensheng Kang 2022Molecular Plant2022,15,2:9
3Mechanism underlying the retarded nuclear translocation of androgen receptor splice variants显示文摘As shown in our previous study, two alternatively spliced androgen receptor(AR) variants, which are exclusively expressed in the granulosa cells of patients with polycystic ovary syndrome, exhibit retarded nuclear translocation compared with wild-type AR. However, researchers have not yet determined whether these abnormalities correlate with heat shock protein 90(HSP90)and importin α(the former is a generally accepted co-chaperone of AR, and the latter is a component of classical nuclear import complexes). Here, these two variants were mainly retained in cytoplasm with HSP90 and importin α in the presence of dihydrotestosterone(DHT), and their levels in nucleus were significantly reduced, according to the immunofluorescence staining. The binding affinity of two AR variants for importin α was consistently decreased, while it was increased in WT-AR following DHT stimulation, leading to reduced nuclear import, particularly for the insertion-AR(Ins-AR). However, the binding affinities of two AR variants for HSP90 were increased in the absence of DHT compared with WT-AR, which functioned to maintain spatial structural stability, particularly for the deletion-AR(Del-AR). 4Physiological performance of three calcifying green macroalgae Halimeda species in response to altered seawater temperatures显示文摘The effects of seawater temperature on the physiological performance of three Halimeda species were studied for a period of 28 d.Five treatments were established for Halimeda cylindracea,Halimeda opuntia and Halimeda lacunalis,in triplicate aquaria representing a factorial temperature with 24°C,28°C,32°C,34°C and 36°C,respectively.The average Fv/Fm of these species ranged from 0.732 to 0.756 between 24°C and 32°C but declined sharply between 34°C(0.457±0.035)and 36°C(0.122±0.014).Calcification was highest at 28°C,with net calcification rates(Gnet)of(20.082±2.482)mg/(g·d),(12.825±1.623)mg/(g·d)and(6.411±1.029)mg/(g·d)for H.cylindracea,H.opuntia and H.lacunalis,respectively.Between 24°C and 32°C,the specific growth rate(SGR)of H.lacunalis(0.079%–0.110%d–1)was lower than that of H.cylindracea(0.652%–1.644%d–1)and H.opuntia(0.360%–1.527%d–1).Three Halimeda species gradually bleached at 36°C during the study period.Malondialdehyde(MDA)and proline levels in tissues of the three Halimeda were higher in 34–36°C than those in 24–32°C.The results indicate that seawater temperature with range of 24–32°C could benefit the growth and calcification of these Halimeda species,however,extreme temperatures above 34°C have negative impacts.The measured physiological parameters also revealed that H.cylindracea and H.opuntia displayed broader temperature tolerance than H.lacunalis.Zhangliang Wei Jiahao Mo Ruiping Huang Qunju Hu Chao Long Dewen Ding Fangfang Yang Lijuan Long 2020Acta Oceanologica Sinica2020,39,2:2

20Light-Programmed Bistate Colloidal Actuation Based on Photothermal Active Plasmonic Substrate显示文摘Active particles have been regarded as the key models to mimic and understand the complex systems of nature.Although chemical and field-powered active particles have received wide attentions,lightprogrammed actuation with long-range interaction and high throughput remains elusive.Here,we utilize photothermal active plasmonic substrate made of porous anodic aluminum oxide filled with Au nanoparticles and poly(N-isopropylacrylamide)(PNIPAM)to optically oscillate silica beads with robust reversibility.The thermal gradient generated by the laser beam incurs the phase change of PNIPAM,producing gradient of surface forces and large volume changes within the complex system.The dynamic evolution of phase change and water diffusion in PNIPAM films result in bistate locomotion of silica beads,which can be programmed by modulating the laser beam.This light-programmed bistate colloidal actuation provides promising opportunity to control and mimic the natural complex systems.Fangfang Deng Juntao Chen Junxiang Xiang Yong Li Yan Qiao Ze Liu Tao Ding 2023Research2023,,1:0
