2Effects of dietary Clostridium butyricum supplementation on growth performance,intestinal development,and immune response of weaned piglets challenged with lipopolysaccharide显示文摘Background: Weanling pigs, with immature immune system and physiological function, usually experience postweaning diarrhea. This study determined the effects of dietary Clostridium butyricum supplementation on growth performance, diarrhea, and immunity of weaned pigs challenged with lipopolysaccharide(LPS).Methods: In Experiment(Exp.) 1,144 weaned piglets were weaned at 21 d and randomly assigned to six groups,with six replicates per group and four pigs per replicate, receiving a control diet(CON) or diet supplemented with antibiotics(AB) or C. butyricum(CB)(0.1%, 0.2%, 0.4%, or 0.8%), respectively. All diets in Exp. 1 were a highly digestible basal diet, with 3,000 mg/kg zinc oxide supplied in the first 2 wk only. In Exp. 2, 180 piglets were weaned at 21 d and randomly assigned to five groups, with six replicates per group and six pigs per replicate, receiving CON, AB, or CB(0.2%, 0.4%, or 0.6%) diets. The digestibility of diets was lower than those in Exp. 1, and did not include zinc oxide. At 36 d of Exp. 2, 12 piglets were selected from each of the CON and 0.4% CB groups, six piglets were intraperitoneally injected with LPS(50 μg/kg body weight) and the other six piglets with normal saline;animals were killed at 4 h after injection to collect blood, intestine, and digesta samples for biochemical analysis.Results: In Exp. 1, CB and AB diets had no effect on growth performance of piglets. In Exp. 2, 0.4% CB decreased feed-gain ratio(P < 0.1), diarrhea score(P < 0.05), and increased duodenal, jejunal, and ileal villus height and jejunal villus height/crypt depth(P < 0.05). The 0.4% CB decreased the plasma tumor necrosis factor(TNF) α(P < 0.05) but increased ileal mucosa IL-10 and TLR2 mRNA expression(P < 0.05). Furthermore, 0.4% CB altered the microbial profile, with Bacillus and Ruminococcaceae UGG-003 at genus level and Lactobacillus casei and Parasutterella secunda at species level were higher than CON in colonic content(P < 0.05).Conclusions: Dietary C. butyricum supplementation had positive effects on growth of weaned piglets with less digestible diets. There was a tendency to reduce the feed-gain ratio, which could reduce feed costs in pig production. 3Effects of dietary supplementation with essential oils and protease on growth performance,antioxidation,inflammation and intestinal function of weaned pigs显示文摘This experiment evaluated the impacts of essential oils(EO)and protease(PRO),independently or in combination,on growth performance,antioxidation,inflammation and intestinal function of weaned pigs.One hundred and sixty weaned pigs(21 d of age,BW of 6.74±0.20 kg)were randomly divided into 4 treatments with 8 replicate pens of 5 pigs per pen.Dietary treatments included the following:1)control diet(CON),2)CON with 300 mg/kg essential oils(EO),3)CON with 500 mg/kg protease(PRO),4)CON with 300 mg/kg essential oil and 500 mg/kg protease(EO + PRO).On d 8,one pig from each penwas selected for sampling.The remaining pigs were fed for an additional week and growth performance was monitored during this period.Dietary treatments had no marked effects(P>0.05)on the growth performance of pigs.However,pigs receiving EO diet had higher(P<0.05)serumglutathione peroxidase(GSH-Px)activity,and tended to decrease(P=0.063)serum concentration of tumor necrosis factor-a(TNF-a).In addition,pigs receiving EO diet had higher(P<0.05)abundances of phylum Actinobacteria,and genera Bifidobacterium,and lower(P<0.05)phylum Bacteroidetes and genera Alloprevotella in colonic digesta.Pigs receiving PRO diet decreased(P<0.05)the serum concentration of malondialdehyde(MDA)and diamine oxidase activity,increased(P<0.05)the villus height and the ratio of villus height to crypt depth in duodenum,increased sucrase activity in jejunal mucosa,and also increased the abundance of phylum Actinobacteria in colonic digesta.Furthermore,the synergistic effects of EO and PROwas observed(P<0.05)for pigs with decreasing serum TNF-a concentration and increasing serum GSH-Px activity.Collectively,the results indicated that dietary supplementation of EO and PRO had no significant effects on growth performance of weaned pigs.EO diet appeared to improve antioxidant activity and intestinal microbiota,while PRO diet improved intestinal morphology and digestive enzyme activity,and there was a synergistic effect of EO and PRO on reducing inflammatory parameters in weaned pigs.Xie Peng Qiang Zhou Cheng Wu Jian Zhao Quan Tan Ying He Liang Hu Zhengfeng Fang Yan Lin Shengyu Xu Bin Feng Jian Li Yong Zhuo Chris Van Ginneken Xuemei Jiang De Wu Lianqiang Che 2022Animal Nutrition2022,,2:8

6Clinicopathological analysis of metaplastic meningioma: report of 15 cases in Huashan Hospital显示文摘Objective: Metaplastic meningioma is a rare subtype of benign meningiomas, classified as WHO grade I with well prognosis. Here we presented our experiences on 15 cases of metaplastic meningioma, to investigate the clinicopathological features, therapies and prognosis of these cases. Methods: 15 patients underwent surgical treatment for intracranial metaplastic meningioma between 2001 and 2010 at Neurosurgery Department of Huashan Hospital, Shanghai, China. The clinical data, radiological manifestation, treatment strategy, pathological findings and prognosis of all patients were analyzed retrospectively. Results: Among the 15 cases (10 males and 5 females), the age ranged from 22 to 74 years old (the mean age was 50.67-year old). The clinical manifestations include headache, dizziness, seizure attack, vision decrease, and weakness of bilateral lower limbs. All the patients received surgical treatment, combined with radiotherapy in some cases. In the follow-up period, recurrence occurred in 2 cases, of which 1 patient died of other system complications. Conclusions: Metaplastic meningiomas are characterized by focal or widespread mesenchymal differentiation with formation of bone, cartilage, fat, and xanthomatous tissue elements. Surgical removal is the optimal therapy, and the overall prognosis is well. 9Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research.Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao 2021Cancer Biology & Medicine2021,18,1:5

11Inclusion of purified dietary fiber during gestation improved the reproductive performance of sows显示文摘Background: This study aimed to investigate the impacts of guar gum and cellulose as the source of dietary fiber during gestation on the reproductive performance of sows.Methods: A total of 210 sows(parities 3–6) were randomly allocated into six diets(n = 35) throughout gestation to feed graded levels of dietary fiber(DF), including a corn-soybean meal-based control diet with no wheat bran inclusion(CON, 12.5% DF), a wheat bran-rich diet(DF1, 17.4% DF), and another 4 diets(DF2, 17.7% DF;DF3, 18.1%DF;DF4, 18.4% DF;DF5, 18.8% DF) in which wheat bran were equally substituted by 1%, 2%, 3% and 4% purified FIBER MIX(guar gum and cellulose, 1:4). All sows received similar DE and other nutrients throughout gestation.Results: DF treatment during gestation resulted in normal fecal score(1 to 5 with 1 = dry and 5 = watery) in sows compared with those received the CON diet(P < 0.05). The number of total born piglets had a tendency to be affected by dietary treatment(P = 0.07), and correlation analysis revealed a linear response of total born to dietary fiber levels during gestation(P < 0.01). Sows received the DF2, DF3, and DF5 diets during gestation had a greater ADFI during lactation compared with those in the CON group(P < 0.05) without affecting the daily body weight gain of suckling piglets. Gut microbiota compositions were dramatically changed by the gestation stage and some of those were changed by DF inclusion. 12Dietary energy intake affects fetal survival and development during early and middle pregnancy in Large White and Meishan gilts显示文摘This experiment was designed to determine the effects of variations in dietary energy intake on reproductive performance and gene expression of luteal and endometrium tissues in Large White(LW)and Meishan(MS) gilts during early and middle pregnancy. After insemination, 32 LW gilts were assigned to high and low(HE_Land LE_L, 14.23 and 12.56 MJ DE/kg, respectively) diet treatment groups,while 32 MS gilts were allocated to HE_M and LE_M(12.56 and 10.88 MJ DE/kg) groups. Gilts were slaughtered on days 35, 55 and 90 of gestation. The fetal survival and luteal progesterone(P_4) concentration in the HE_Lgroup were higher on day 35 but lower on day 90 of gestation compared with the LE_L group(P < 0.05) for LW gilts. However, fetal survival and luteal P_4 concentration on day 35 of gestation were greater(P < 0.05) in the LE_M group than in the HE_M group for MS gilts, but no significant difference in mid-gestation was showed. The fetal weights of both breeds were higher for the high energy diets compared with the respective control group on day 90 of gestation(P < 0.05). In addition,the m RNA levels of P_4 synthesis-related proteins had correlated with luteal P_4 concentration in both breeds. Further, endometrial levels of uteroferrin(ACP5), retinol-binding protein 4(RBP_4) and secreted phosphoprotein 1(SPP1) m RNA were upregulated in the HE_Lgroup on day 35 of gestation but ACP5 and SPP1 were downregulated on day 55 of gestation compared with the LE_Lgroup(P < 0.05) for LW gilts. In MS gilts, diet only affected the expression of SPP1(P < 0.05). Our results revealed the differential sensitivity of LW and MS breeds to variations in dietary energy intake. For LW gilts, the HE_Lgroup improved fetal survival on day 35 but a sustained high energy diet decreased fetal survival on day 90 of gestation. The differences in dietary energy intake did not influence fetal survival on day 90 of gestation but the higher energy diet did increase fetal weight in the MS breed compared with the lower energy intake diet. 13Human embryonic stem cells-derived endothelial cell therapy facilitates kidney regeneration by stimulating renal resident stem cell proliferation in acute kidney injury显示文摘Endothelial cell therapy has been implicated to enhance tissue regeneration and vascularization in ischemic kidney. However, no published study has yet examined direct effects of endothelial cell treatment in kidney recovery. This study investigated the therapeutic efficacy of endothelial cells in a mouse model with acute kidney injury (AKI). Thus, human embryonic stem cells-derived endothelial cells (hESC-ECs) labeled with a reporter system encoding a double fusion reporter gene for firefly luciferase (Fluc) and green fluorescent protein (GFP) were characterized by Fluc imaging and immunofluoresence staining. Cultured hESC-ECs (1×106) were injected into ischemic kidney shortly after AKI. Survival of the transplanted hESC-ECs was monitored in vivo from day 1 to 14 after endothelial cell transplantation and potential impact of hESC-EC treatment on renal regeneration was assessed by histological analyses. We report that a substantial level of bioluminescence activity was detected 24 h after hESC-EC injection followed by a gradual decline from 1 to 14 d. Human ESC-ECs markedly accelerated kidney cell proliferation in response to ischaemia-induced damage, indicated by an elevated number of BrdU+ cells. Co-expression of Sca-1, a kidney stem cell proliferation marker, and BrdU further suggested that the observed stimulation in renal cell regeneration was, at least in part, due to increased proliferation of renal resident stem cells especially within the medullary cords and arteriole. Differentiation of hESC-ECs to smooth muscle cells was also observed at an early stage of kidney recovery. 15Establishment of a diagnostic model of coronary heart disease in elderly patients with diabetes mellitus based on machine learning algorithms显示文摘OBJECTIVE To establish a prediction model of coronary heart disease(CHD)in elderly patients with diabetes mellitus(DM)based on machine learning(ML)algorithms.METHODS Based on the Medical Big Data Research Centre of Chinese PLA General Hospital in Beijing,China,we identified a cohort of elderly inpatients(≥60 years),including 10,533 patients with DM complicated with CHD and 12,634 patients with DM without CHD,from January 2008 to December 2017.We collected demographic characteristics and clinical data.After selecting the important features,we established five ML models,including extreme gradient boosting(XGBoost),random forest(RF),decision tree(DT),adaptive boosting(Adaboost)and logistic regression(LR).We compared the receiver operating characteristic curves,area under the curve(AUC)and other relevant parameters of different models and determined the optimal classification model.The model was then applied to 7447 elderly patients with DM admitted from January 2018 to December 2019 to further validate the performance of the model.RESULTS Fifteen features were selected and included in the ML model.The classification precision in the test set of the XGBoost,RF,DT,Adaboost and LR models was 0.778,0.789,0.753,0.750 and 0.689,respectively;and the AUCs of the subjects were 0.851,0.845,0.823,0.833 and 0.731,respectively.Applying the XGBoost model with optimal performance to a newly recruited dataset for validation,the diagnostic sensitivity,specificity,precision,and AUC were 0.792,0.808,0.748 and 0.880,respectively.CONCLUSIONS The XGBoost model established in the present study had certain predictive value for elderly patients with DM complicated with CHD.Hu XU Wen-Zhe CAO Yong-Yi BAI Jing DONG He-Bin CHE Po BAI Jian-Dong WANG Feng CAO Li FAN 2022Journal of Geriatric Cardiology2022,19,6:3
16Radiomics signature:A potential biomarker forβ-arrestin1 phosphorylation prediction in hepatocellular carcinoma显示文摘BACKGROUND The phosphorylation status ofβ-arrestin1 influences its function as a signal strongly related to sorafenib resistance.This retrospective study aimed to develop and validate radiomics-based models for predictingβ-arrestin1 phosphorylation in hepatocellular carcinoma(HCC)using whole-lesion radiomics and visual imaging features on preoperative contrast-enhanced computed tomography(CT)images.AIM To develop and validate radiomics-based models for predictingβ-arrestin1 phosphorylation in HCC using radiomics with contrast-enhanced CT.METHODS Ninety-nine HCC patients(training cohort:n=69;validation cohort:n=30)receiving systemic sorafenib treatment after surgery were enrolled in this retrospective study.Three-dimensional whole-lesion regions of interest were manually delineated along the tumor margins on portal venous CT images.Radiomics features were generated and selected to build a radiomics score using logistic regression analysis.Imaging features were evaluated by two radiologists independently.All these features were combined to establish clinico-radiological(CR)and clinico-radiological-radiomics(CRR)models by using multivariable logistic regression analysis.The diagnostic performance and clinical usefulness of the models were measured by receiver operating characteristic and decision curves,and the area under the curve(AUC)was determined.Their association with prognosis was evaluated using the Kaplan-Meier method.RESULTS Four radiomics features were selected to construct the radiomics score.In the multivariate analysis,alanine aminotransferase level,tumor size and tumor margin on portal venous phase images were found to be significant independent factors for predictingβ-arrestin1 phosphorylation-positive HCC and were included in the CR model.The CRR model integrating the radiomics score with clinico-radiological risk factors showed better discriminative performance(AUC=0.898,95%CI,0.820 to 0.977)than the CR model(AUC=0.794,95%CI,0.686 to 0.901;P=0.011),with increased clinical usefulness confirmed in both the training and validation cohorts using decision curve analysis.The risk ofβ-arrestin1 phosphorylation predicted by the CRR model was significantly associated with overall survival in the training and validation cohorts(log-rank test,P<0.05).CONCLUSION The radiomics signature is a reliable tool for evaluatingβ-arrestin1 phosphorylation which has prognostic significance for HCC patients,providing the potential to better identify patients who would benefit from sorafenib treatment.Feng Che Qing Xu Qian Li Zi-Xing Huang Cai-Wei Yang Li Ye Wang Yi Wei Yu-Jun Shi Bin Song 2022World Journal of Gastroenterology2022,28,14:3

18全颗粒饲料对奶牛生产性能的影响显示文摘饲料中的中性洗涤纤维(NDF)含量和饲料颗粒直径是影响瘤胃功能的重要因素。试验评估了以颗粒饲料为主的奶牛瘤胃健康情况、NDF消化率和生产性能。选取8头荷斯坦母牛(336±30 d,体重346±35 kg),随机分配到交叉重复试验组中,奶牛安置于牛栏,自由采食。试验期为3周,前两周是适应期,最后一周为数据收集期。各组饲粮原料相同,但物理形式不同,分为全混合日粮(TMR)和颗粒料(直径=8 mm)。两组间物理有效NDF(pe NDF)不同,TMR组为39.8%,颗粒料组为11.8%。在试验过程中,干物质采食量(DMI)、饮水量、瘤胃反刍时间、瘤胃温度和pH情况均记录。3周后收集粪样,确定潜在的可消化NDF总消化率(pdNDF)。每阶段结束后计算平均日增重和饲料转化率。颗粒料组的DMI、DMI/体重和饮水量更高。而两组间平均日增重和饲料转化率无显著差异。颗粒料组的瘤胃反刍时间为241 min·d-1低于TMR组的507 min·d-1。饲料的不同物理外形对瘤胃温度、瘤胃pH无显著影响。TMR组的pdNDF总消化率为90.25%高于颗粒料组的86.82%。试验结果表明,动物易接受完整的颗粒饲料,DMI较高,且反刍时间减少,但瘤胃pH无显著变化。TMR组的pdNDF消化率显著增加。尽管两组奶牛在DMI和纤维消化方面表现不同,但总体表现相似,两组饲料的保存时间与其物理形式有关。在短期内,提供能满足NDF需求的完全颗粒饲料可改善瘤胃健康状况和动物生产性能,而较长时间的数据则需要进一步试验。Wang J Yang M Cao M Lin Y Che L Q Duraipandiyan V Al-Dhabi N A Fang Z F Xu S Y Feng B Liu G Wu D Bonfante E Palmonari A Mammi L Canestrari G Fustini M Formigoni A 2016饲料博览2016,0,12:2

20Experimental and Numerical Study on Co-combustion Behaviors and NO_(x) Emission Characteristics of Semi-coke and Coal in a Tangentially Fired Utility Boiler显示文摘The utilization of powdery semi-coke as a power fuel in pulverized coal-fired power plants has become a new and potential technique to consume the excess powdery semi-coke.The characteristic of low volatile results in poor combustion performance and high NO_(x) emission,and to co-fire with bituminous coal is a practical strategy to address this problem.However,the co-combustion characteristics and the inherent interaction between semi-coke and coal remain insufficiently understood.In addition,the influences of secondary air arrangement,the boiler operation load,and the fuel type on co-combustion process are still unclear,which is urgent to be further explored.In the present study,experiments and numerical simulations were jointly utilized to inquire into the co-combustion behaviors and NO_(x) emission features of semi-coke and coal.The results demonstrated that the'out-furnace method'was a suitable choice for small-capacity boiler when the proportion of semi-coke was 33%,due to the limited combinations of the semi-coke injection position.It was recommended that semi-coke was preferred to be injected from the middle layers of the furnace under the'in-furnace method'to improve the overall co-combustion performance.The critical value of the separated over fire air ratio in this study was 27.5%,over which a slight drop of carbon content in fly ash could come about.Moreover,the elevation in the proportion of separated over fire air gave rise to the significant decline of NO_(x) concentration.The constricted secondary air arrangement was preferred to be employed due to the high boiler efficiency.The separated over fire air and the surrounding air needed to maintain a wide-open degree to prevent the increase of NO_(x) emissions and the coking of nozzles.For the load reduction regulation method adopted in this study,the NO_(x) concentration first rose and then dropped,while the burnout ratio decreased obviously as the operation load was reduced.Different combinations of coal and semi-coke generated significant influences on co-combustion behaviors within the furnace.The NO_(x )generated by high-volatile fuel (bituminous coal) combustion was mainly affected by volatile-N,while the NO_(x )generated by low-volatile fuel (semi-coke) was mainly impacted by char-N.This study is of guiding significance for the efficient and clean utilization and beneficial to the large-scale application of powder semi-coke in power plants.WANG Chang’an FENG Qinqin JIN Liyan WANG Zhichao YAO Wei WANGPengqian DU Yongbo ZHANG Xilai YANG Zhongcan CHE Defu 2021Journal of Thermal Science2021,30,4:2
