| 1 | Immune Responses to the Attenuated Mycoplasma hyopneumoniae 168 Strain Vaccine by Intrapulmonic Immunization in Piglets显示文摘To investigate the immune responses to the attenuated Mycoplasma hyopneumoniae 168 strain vaccine, 8-15 d old piglets were immunized with M. hyopneumoniae 168 strain vaccine by intrapulmonic route. And the specific IgG antibody in serum, lymphoproliferation, IFNγ, and specific secretory IgA (SIgA) antibody in bronchoalveolar lavage fluid were detected on 30 and 60 d post-immunization (DPI), respectively. On 60 DPI, all the pigs except for those in health control group were challenged with a field M. hyopneumoniae strain JS. Necropsy was performed on 30 d post-challenge (DPC). The results showed that IFNγ and specific SIgA were stimulated on surface of respiratory tract after immunization. And peripheral blood mononuclear cells could also be proliferated about 1.81 and 2.12 fold on 30 and 60 DPI when stimulated by M. hyopneumoniae protein in vitro. However, no serum IgG antibody against M. hyopneumoniae was detected during the whole immune phage. After challenge, vaccinated pigs were observed with only very slight histological lesion in individual lobes. None of vaccinated pigs showed any clinical signs. While the unvaccinated pigs from challenge control group showed varying degrees of clinical sign and severe macroscopical lesion of mycoplasmal pneumonia of swine (MPS). | 4 | Deep learning applied to two-dimensional color Doppler flow imaging ultrasound images significantly improves diagnostic performance in the classification of breast masses:a multicenter study显示文摘Background:The current deep learning diagnosis of breast masses is mainly reflected by the diagnosis of benign and malignant lesions.In China,breast masses are divided into four categories according to the treatment method:inflammatory masses,adenosis,benign tumors,and malignant tumors.These categorizations are important for guiding clinical treatment.In this study,we aimed to develop a convolutional neural network(CNN)for classification of these four breast mass types using ultrasound(US)images.Methods:Taking breast biopsy or pathological examinations as the reference standard,CNNs were used to establish models for the four-way classification of 3623 breast cancer patients from 13 centers.The patients were randomly divided into training and test groups(n=1810 vs.n=1813).Separate models were created for two-dimensional(2D)images only,2D and color Doppler flow imaging(2D-CDFI),and 2D-CDFI and pulsed wave Doppler(2D-CDFI-PW)images.The performance of these three models was compared using sensitivity,specificity,area under receiver operating characteristic curve(AUC),positive(PPV)and negative predictive values(NPV),positive(LR+)and negative likelihood ratios(LR-),and the performance of the 2D model was further compared between masses of different sizes with above statistical indicators,between images from different hospitals with AUC,and with the performance of 37 radiologists.Results:The accuracies of the 2D,2D-CDFI,and 2D-CDFI-PW models on the test set were 87.9%,89.2%,and 88.7%,respectively.The AUCs for classification of benign tumors,malignant tumors,inflammatory masses,and adenosis were 0.90,0.91,0.90,and 0.89,respectively(95%confidence intervals[CIs],0.87-0.91,0.89-0.92,0.87-0.91,and 0.86-0.90).The 2D-CDFI model showed better accuracy(89.2%)on the test set than the 2D(87.9%)and 2D-CDFI-PW(88.7%)models.The 2D model showed accuracy of 81.7%on breast masses≤1 cm and 82.3%on breast masses>1 cm;there was a significant difference between the two groups(P<0.001).The accuracy of the CNN classifications for the test set(89.2%)was significantly higher than that of all the radiologists(30%).Conclusions:The CNN may have high accuracy for classification of US images of breast masses and perform significantly better than human radiologists.Trial registration:Chictr.org,ChiCTR1900021375;http://gffzz62a03ee25e6f4a77hw9n6qopf9bf96pq6.ffgz.tsg.suse.edu.cn/showproj.aspx?proj=33139. | Teng-Fei Yu Wen He Cong-Gui Gan Ming-Chang Zhao Qiang Zhu Wei Zhang Hui Wang Yu-Kun Luo Fang Nie Li-Jun Yuan Yong Wang Yan-Li Guo Jian-Jun Yuan Li-Tao Ruan Yi-Cheng Wang Rui-Fang Zhang Hong-Xia Zhang Bin Ning Hai-Man Song Shuai Zheng Yi Li Yang Guang | 2021 | Chinese Medical Journal2021,,4: | 11 |

| 7 | Comparison of sentinel lymph node detection performances using blue dye in conjunction with indocyanine green or radioisotope in breast cancer patients: a prospective singlecenter randomized study显示文摘Objective:This randomized study aimed to compare the clinical efficacy between the novel dual tracer composed of indocyanine green(ICG)and blue dye(BD)and the conventional dual tracer composed of radioisotope and BD for sentinel lymph node(SLN)mapping in patients with breast cancer.Methods:This study enrolled 471 clinically lymph node-negative patients with primary breast cancer.All patients underwent mastectomy,and those undergoing sentinel lymph node biopsy(SLNB)were randomized to receive blue dye plus radioisotope(RB group)or BD plus ICG(IB group).The detection performances on SLN identification rate,positive SLN counts,detection sensitivity,and false-negative rate were compared between the two groups.Results:In the IB group,97%(194/200)of the patients who underwent the ICG and BD dual tracer injection showed fluorescentpositive lymphatic vessels within 2–5 min.The identification rate of SLNs was comparable between the IB group(99.0%,198/200)and the RB group(99.6%,270/271)(P=0.79).No significant differences were observed in the identification rate of metastatic SLNs(22.5%vs.22.9%,P>0.05,RB group vs.IB group,the same below),positive SLN counts(3.72±2.28 vs.3.91±2.13,P>0.05),positive metastatic SLN counts(0.38±0.84 vs.0.34±0.78,P>0.05),SLNB detection sensitivity(94.4%vs.92.5%,P>0.05),or false-negative rate(5.6%vs.7.5%,P>0.05)between the two groups.Conclusions:ICG can be used as a promising alternative tracer for radioisotope in SLN mapping,and when it is combined with BD in lymphangiography,it offers comparable detection sensitivity compared to the conventional lymphatic mapping strategies that are widely used in clinical practice. | Long Yuan Xiaowei Qi Yi Zhang Xinhua Yang Fan Zhang Linjun Fan Li Chen Kongyong Zhang Ling Zhong Yanling Li Sijie Gan Wenying Fu Jun Jiang | 2018 | Cancer Biology & Medicine2018,15,4: | 9 |
| 8 | Involvement of the Mitochondrion-dependent and the Endoplasmic Reticulum Stress-signaling Pathways in Isoliquiritigenin-induced Apoptosis of HeLa Cell显示文摘Objective Isoliquiritigenin(ISL),a licorice chalconoid,is considered to be a bioactive agent with chemopreventive potential.This study investigates the mechanisms involved in ISL-induced apoptosis in human cervical carcinoma HeLa cells.Methods Cell viability was evaluated using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide(MTT)assay.Apoptosis was determined by flow cytometry using an Annexin V-FITC Apoptosis Detection Kit.The intracellular ROS levels were assessed using a 2,7-dichlorofluorescein probe assay.The mitochondrial membrane potential was measured with the dual-emission potential-sensitive probe 5,5',6,6'-tetra-chloro-1,1',3,3'-tetraethyl-imidacarbocyanine iodide(JC-1).The degradation of poly-ADP-ribose polymerase(PARP)protein,the phosphorylation of PKR-like ER kinase(PERK),the phosphorylation of the α-subunit of eukaryotic initiation factor 2(eIF2α),the expression of the 78 kD glucose-regulated protein(GRP 78),and the activation of caspase-12 were analyzed via western blot analysis.Results ISL significantly inhibited the proliferation,the increase in ROS levels and apoptotic rates of HeLa cells in a concentration-dependent manner.Moreover,ISL induced mitochondrial dysfunction,caspase activation,and PARP cleavage,which displayed features of mitochondria dependent on apoptotic signals.Besides,exposure of HeLa cells to ISL triggered endoplasmic reticulum(ER)stress,as indicated by the increase in p-eIF2α and GRP78 expression,ER stress-dependent apoptosis is caused by the activation of ER-specific caspase-12.Conclusion The findings from our study suggest that ISL-induced oxidative stress causes HeLa cell apoptosis via the mitochondrion-dependent and the ER stress-triggered signaling pathways. | YUAN Xuan ZHANG Bo GAN Lu WANG Zhen Hua YU Ba Cui LIU Liang Liang ZHENG Qiu Sheng WANGZhi Ping | 2013 | Biomedical and Environmental Sciences2013,26,4: | 8 |
| 9 | Elastoplastic Contact Mechanics Model of Rough Surface Based on Fractal Theory显示文摘Because the result of the MB fractal model contradicts with the classical contact mechanics, a revised elastoplastic contact model of a single asperity is developed based on fractal theory. The critical areas of a single asperity are scale dependent, with an increase in the contact load and contact area, a transition from elastic, elastoplastic to full plastic deformation takes place in this order. In considering the size distribution function, analytic expression between the total contact load and the real contact area on the contact surface is obtained. The elastic, elastoplastic and full plastic contact load are obtained by the critical elastic contact area of the biggest asperity and maximun contact area of a single asperity. The results show that a rough surface is firstly in elastic deformation. As the load increases, elastoplastic or full plastic deformation takes place. For constant characteristic length scale G, the slope of load-area relation is proportional to fractal dimension D. For constant fractal dimension D, the slope of load-area relation is inversely proportional to G. For constant D and G, the slope of load-area relation is inversely proportional to property of the material ? , namely with the same load, the material of rough surface is softer, and the total contact area is larger. | 10 | High-Q resonances governed by the quasi-bound states in the continuum in all-dielectric metasurfaces显示文摘The realization of high-Q resonances in a silicon metasurface with various broken-symmetry blocks is reported. Theoretical analysis reveals that the sharp resonances in the metasurfaces originate from symmetry-protected bound in the continuum(BIC) and the magnetic dipole dominates these peculiar states. A smaller size of the defect in the broken-symmetry block gives rise to the resonance with a larger Q factor. Importantly, this relationship can be tuned by changing the structural parameter, resulting from the modulation of the topological configuration of BICs. Consequently, a Q factor of more than 3,000 can be easily achieved by optimizing dimensions of the nanostructure. At this sharp resonance, the intensity of the third harmonic generation signal in the patterned structure can be 368 times larger than that of the flat silicon film. | 12 | Risk factors of interstitial lung diseases in clinically amyopathic dermatomyositis显示文摘Background:Clinically amyopathic dermatomyositis(CADM)is a unique sub-type of idiopathic inflammatory myopathies with a high prevalence of interstitial lung disease(ILD).Poor prognosis of the patients was strongly associated with rapid progressive ILD.The aim of this study was to identify risk factors for prediction of different types of ILD in CADM.Methods:In this study,data of 108 inpatients with CADM were collected,including 87 with ILD.The baseline clinical data and laboratory parameters,including myositis-specific and associated antibodies and tumor-associated antigens were analyzed to identify risk factors for acute or subacute interstitial pneumonitis(A/SIP)and chronic interstitial pneumonitis(CIP).Results:In 87 patients with CADM-ILD,39(36.1%)were A/SIP,and 48(44.4%)were CIP.There were 22(20.4%)patients with asymptomatic ILD who were detected by routine high resolution computed tomography.Cytokeratin-19 fragment(CYFRA21-1)was significantly higher in CADM-ILD than that in CADM patients without ILD;carcinoembryonic antigen and neuron-specific enolase were significantly elevated in A/SIP than that in CIP.Patients with A/SIP had a higher positive rate of anti-melanoma differentiation-associated gene 5(MDA5),while patients with CIP had a higher positive rate of anti PL-12 and anti-Ro-52.Logistic regression analysis indicated that elevation of CYFRA21-1 was a risk factor for ILD,higher titer of anti-MDA5 indicated increased likelihood for A/SIP,and higher titer of anti-Ro-52 was also clearly associated with CIP.Conclusions:This study indicated that the prevalence of ILD was high in CADM.Asymptomatic ILD has been previously underestimated.Anti-MDA5 was a risk factor for the presence of A/SIP,and CYFRA21-1 was a risk factor for ILD. | Yu-Zhou Gan Li-Hua Zhang Lin Ma Feng Sun Yu-Hui Li Yuan An Zhan-Guo Li Hua Ye | 2020 | Chinese Medical Journal2020,,6: | 7 |

| 14 | Comparison of the short-term and long-term outcomes of laparoscopic hysterectomies and of abdominal hysterectomies: a case study of 4,895 patients in the Guangxi Zhuang Autonomous Region, China显示文摘Background: To evaluate the short-term and long-term outcomes after laparoscopic hysterectomy(LH) compared with abdominal hysterectomy(AH) in case of benign gynecological disease. Methods: A multi-center cohort retrospective comparative study of population among 4,895 hysterectomies(3,539 LH vs.1,356 AH) between 2007 and 2013 was involved. Operative time(OT), estimated blood loss(EBL), intra-operative and post-operative complications, passing flatus; days with indwelling catheter, questionnaires covering pelvic floor functions and sexual functions were assessed.Results: The EBL(174.1±157.4 vs. 263.1±183.2 cc, LH and AH groups, respectively), passing flatus(38.7±14.1 vs. 48.1±13.2 hours), days with indwelling catheter(1.5±0.6 vs. 2.2±0.8 days), use of analgesics(6.5% vs. 73.1%), intra-operative complication rate(2.4% vs. 4.1%), post-operative complication rate(2.3% vs. 5.7%), post-operative constipation(12.1% vs. 24.6%), mild and serious stress urinary incontinence(SUI) post-operative(P<0.001; P=0.014), and proportion of Female Sexual Functioning Index(FSFI) total score <26.55 post-operative(P<0.001) of the LH group were significantly less than those of AH group. There wereno significant differences in OT(106.5±34.5 vs. 106.2±40.3 min) between the two groups.Conclusions: LH is a safe and efficient operation for improving patients' long-term quality of life(Qo L), and LH is a cost-effectiveness procedure for treating benign gynecological disease. It remains controversial whether c-Jun N-terminal kinase(JNK) participates in the apoptosis of retinal ganglion cells in glaucoma. This study sought to explore a possible mechanism of action of JNK signaling pathway in glaucoma-induced retinal optic nerve damage. We established a mouse model of chronic ocular hypertension by reducing the aqueous humor followed by photocoagulation using the laser ignition method. Results showed significant pathological changes in the ocular tissues after the injury. Apoptosis of retinal ganglion cells increased with increased intraocular pressure, as did JNK3 m RNA expression in the retina. | 17 | Cholesterol-tuned liposomal membrane rigidity directs tumor penetration and anti-tumor effect显示文摘Recently, liposomes have been widely used in cancer therapeutics, but their anti-tumor effects are suboptimal due to limited tumor penetration. To solve this problem, researchers have made significant efforts to optimize liposomal diameters and potentials, but little attention has been paid to liposomal membrane rigidity. Herein, we sought to demonstrate the effects of cholesterol-tuned liposomal membrane rigidity on tumor penetration and anti-tumor effects. In this study, liposomes composed of hydrogenated soybean phospholipids(HSPC), 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000](DSPE-PEG2000) and different concentrations of cholesterol were prepared. It was revealed that liposomal membrane rigidity decreased with the addition of cholesterol. Moderate cholesterol content conferred excellent diffusivity to liposomes in simulated diffusion medium, while excessive cholesterol limited the diffusion process. We concluded that the differences of the diffusion rates likely stemmed from the alterations in liposomal membrane rigidity, with moderate rigidity leading to improved diffusion. Next, the in vitro tumor penetration and the in vivo anti-tumor effects were analyzed. The results showed that liposomes with moderate rigidity gained excellent tumor penetration and enhanced anti-tumor effects.
