| 1 | Establishment of cell clones with different metastatic potential from the metastatic hepatocellular carcinoma cell line MHCC97显示文摘ALM To establish clone cells with different metastatic potential for the study of metastasis-related mechanisms. METHODS Cloning procedure was performed on parental hepatocellular carcinoma (HCC) cell line MHCC97. andbiological characteristics of the target clones selected by in vivo screening were studied.``RESULTS Two clones with high MHCC97-H and IowMHCC9--L1 metastatic potential were isolated from theparent cell line. Compared with MHCC97-L. MHCC97-H hadsmaller cell size average cell diameter 43 um vs 50 μmand faster in vitro and in vivo growth rate tumor celldoubling time was 34.2 h vs 60.0 h. The main ranges ofchromosomes were 5.5 58 in MHCC97-H and 57 62 inMHCC97-L. Boyden chamber in vitro invasion assay demonstrated that the number of penetrating cells through the artificial basement membrane was 137.5 - 11 .0) cellsfield for MHC_C99--H vs 17.7 - 6.3) field for MHCC97-L.The proportions of cells in GO Gl phase. | 2 | Sorafenib inhibits growth and metastasis of hepatocellular carcinoma by blocking STAT3显示文摘AIM: To investigate the inhibitory role and the underlying mechanisms of sorafenib on signal transducer and activator of transcription 3 (STAT3) activity in hepatocellular carcinoma (HCC).METHODS: Human and rat HCC cell lines were treated with sorafenib. Proliferation and STAT3 dephosphorylation were assessed. Potential molecular mechanisms of STAT3 pathway inhibition by sorafenib were evaluated. In vivo antitumor action and STAT3 inhibition were investigated in an immunocompetent orthotopic rat HCC model.RESULTS: Sorafenib decreased STAT3 phosphorylationat the tyrosine and serine residues (Y705 and S727), but did not affect Janus kinase 2 (JAK2) and phosphatase shatterproof 2 (SHP2), which is associated with growth inhibition in HCC cells. Dephosphorylation of S727 was associated with attenuated extracellular signal-regulated kinase (ERK) phosphorylation, similar to the effects of a mitogen-activated protein kinase (MEK) inhibitor U0126, suggesting that sorafenib induced S727 dephosphorylation by inhibiting MEK/ERK signaling. Meanwhile, sorafenib could also inhibit Akt phosphorylation, and both the phosphatidylinositol-3-kinase (PI3K) inhibitor LY294002 and Akt knockdown resulted in Y705 dephosphorylation, indicating that Y705 dephosphorylation by sorafenib was mediated by inhibiting the PI3K/Akt pathway. | 3 | Application of needle-knife in difficult biliary cannulation for endoscopic retrograde cholangiopancreatography显示文摘BACKGROUND: Getting directly into the common bile duct (CBD) is the most important step for successful therapeutic biliary endoscopy. In 5%-10% of cases, the CBD remains inaccessible, necessitating pre-cut papillotomy or fistulotomy with a needle-knife. The aim of this study was to assess the value of early application of the needle-knife in difficult biliary cannulation for endoscopic retrograde cholangiopancreatography (ERCP). METHODS: Patients with failed biliary cannulation after 10 minutes or guide wire entering the pancreatic tube 3 times were randomly divided into group of needle-knife cut and group of persistent cannulation by standard techniques. The cannulation times, success rates and complication rates were compared between the two groups. RESULTS: A total of 948 therapeutic biliary ERCP procedures were performed between October 2004 and February 2006. Of 91 patients with difficult biliary cannulation, 43 patients underwent needle-knife cut: the cannulation success rate was 90.7%, the mean cannulation time was 5.6 minutes, and the complication rate was 9.3%. The other 48 patients underwent persistent cannulation by standard techniques: the cannulation success rate was 75%, the mean cannulation time was 10.2 minutes, and the complication rate was 14.6%. | 4 | Serum hyaluronic acid, procollagen type Ⅲ and Ⅳ in histological diagnosis of liver fibrosis显示文摘OBJECTIVE: To assess the significance of serum hyaluronic acid (HA), proeollagen type Ⅲ (PCⅢ),collagen type Ⅳ (CⅣ) in the histological diagnosis of liver fibrosis.METHODS: The concentrations of serum HA, PCⅢ, CⅣ in 253 patients with chronic liver diseases weremeasured by radioimmunoassay. Liver biopsies were performed in all patients at the same time. The liverwas pathologically evaluated by a pathologist according to a scoring system. Combined with the results ofliver pathological diagnosis, the accuracy of serum HA, PCⅢ, CⅣ in diagnosing patients with hepaticfibrosis (staging≥S2) or cirrhosis (S4) was assessed using the receiver operating curve (ROC).RESULTS: The cutoff values of serum HA, PCⅢ and CⅣ for identifying patients with hepatic fibrosis(≥S2) or cirrhosis (S4) were determined. The cutoff values of serum HA, PCⅢ and CⅣ for detectingpatients with fibrosis (stage≥S2) were 90μg/L, 90μg/L, 75μg/L, respectively; their sensitivity(Se) was 80.4%, 82%, 63.1%; their specificity (Spe) was 70.2%, 60.8%, 83.8%; their positivepredictive values (PPV) were 86.7%, 83.5%, 90.4%; their negative predictive values (NPV) were59.8%, 58.4%, 48.4%, respectively. The cutoff values for detecting patients with liver cirrhosis were210μg/L for HA, 96.2% for Se, 85.3% for Spe, 65.4% for PPV, 98.8% for NPV; 150μg/L forPCⅢ, 76.4% for Se, 68.7% for Spe, 40.4% for PPV, 91.3% for NPV; 90μg/L for CⅣ, 80% forSe, 75.8% for Spe, 47.8% for PPV, 93.2% for NPV, respectively.CONCLUSIONS: Serum HA, PCⅢ and CⅣ can be determined for an accurate diagnosis of hepaticfibrosis in various stages. | 5 | Putative epidermal stem cell convert into corneal epithelium-like cell under corneal tissue in vitro显示文摘Rhesus putative epidermal stem cells are being investigated for their potential use in regenerative corneal epithelium-like cells, which may provide a practical source of autologous seed cells for the construction of bioengineered corneas. The goal of this study was to investigate the potential of epi-dermal stem cells for trans-differentiation into corneal epithelium-like cells. Rhesus putative epidermal stem cells were isolated by type IV collagen attachment method. Flow cytometry analysis, immuno-histology and RT-PCR were conducted to identify the expression of specific markers (β1, α6 integrin, K15, K1/K10, K3/K12 and CD71) on the isolated rapid attaching cells. The isolated cells were cocultured with human corneal limbal stroma and corneal epithelial cells. After coculture, the expression of the same specific markers was evaluated in order to identify expression difference caused by the coculture conditions. K3/K12 expression was analyzed in coculture cells on day 2, 4, 6, 8 and 10. Putative epi-dermal stem cells in conditioned culture media were used as control. Putative epidermal stem cells were predominant in rapid attaching cells by type IV collagen attachment isolation. Before being co-cultured, the rhesus putative epidermal stem cells expressed K15, α6 and β1 integrin, but no CD71, K1/K10 and K3/K12. After coculture, these cells expressed K3/K12 (a marker of corneal epithelial cells), K15 and β 1 integrin, but no K1/K10. Cells being not coculture converted into terminally differentiated cells expressing K1/K10. | 7 | Endoscopic biliary drainage for biliary obstruction显示文摘OBJECTIVE: To improve the successful rate of endoscopic biliary drainage.METHODS: Three hundred and fifty-two patients with biliary obstruction were given biliary drainage viaduodenoscope from January 1998 to December 2002. 258 patients received endoscopic naso-biliarydrainage (ENBD), 51 endoscopic retrograde biliary drainage (ERBD), and 43 endoscopic metal biliaryendoprothesis (EMBE).RESULTS: Of the 352 patients with biliary obstruction, 337 succeeded in drainage by endoscopy and 15failed. Ten ENBD failed patients were handled successfully by readjustment of the site of the naso-biliarytube. In 3 ERBD failed patients, 2 were given plastic stents with appropriate length and got a fluentdrainage. Percutaneous transhepatic biliary drainage (PTBD) was performed in one patient after failure ofendoscopic management. In 2 patients with failed EMBE, one stent could not exceed the site of tumorstricture and one was obstructed by tumor implantation only one month after EMBE. Placement of anothermetallic stent and a plastic stent through the previous prothesis for each patient ensured a successfuldrainage.CONCLUSIONS: The effect of endoscopic biliary drainage for biliary obstruction is definite. | 8 | Novel assay of competitively differentiated polymerase chain reaction for screening point mutation of hepatitis B virus显示文摘AIM: Point mutation, one of the commonest gene mutations,is the most important molecular pathogenesis of cancer and chronic infection. The commonest methods for detection of point mutation are based on polymerase chain reaction (PCR). These techniques, however, cannot be used in large scale screening since they are neither accurate nor simple.For this reason, this study established a novel method of competitively differentiated PCR (CD-PCR) for screening point mutation in clinical practice.METHODS: Two competitively differentiated primers for mutant-type and wild-type templates respectively with an identically complemented region in 3′ end except for last 2base pairs and a different non-complemented region in 5′end were designed. Thus, competitive amplification might be carried out at a lower annealing temperature at first, and then differentiated amplification at a higher annealing temperature when primers could not combine with initial templates. The amplification was performed in one-tube.The products of CD-PCR were detected using microplate hybridization assay. CD-PCR was evaluated by detecting G1896A variant of hepatitis B virus (HBV) in form of recombinant plasmids and in sera from patients with hepatitis B, and compared with allele-specific PCR (AS-PCR) and competitive AS-PCR.RESULTS: CD-PCR was successfully established. It could clearly distinguish wild-type and mutant-type plasmid DNA of G1896A variant when the amount of plasmid DNA was between 102-108copies/reaction, while for AS-PCR and competitive AS-PCR, the DNA amount was between 102-104copies/reaction. CD-PCR could detect one copy of G1896A variant among 10-100 copies of wild-type plasmid DNA. The specificity of CD-PCR was higher than those of AS-PCR and competitive AS-PCR in the detection of HBV G1896A variant in sera from patients with hepatitis B. CD-PCR was independent of the amount of HBV DNA in serum. HBV G1896A variant was more often found in HBeAg (-) patients with a lower level of detectable viremia than that with a higher level of detectable viremia (P=0.0192).CONCLUSION: CD-PCR is more specific since it is less influenced by the amount of initial templates and the cross amplification between mutant- and wild-type amplified products. It is also simple and time-saving. Thus, CD-PCR might be useful in routine gene typing and point mutation screening. | 9 | Prognostic significance and clinical relevance of Sprouty 2 protein expression in human hepatocellular carcinoma显示文摘BACKGROUND:In vitro experiments and mice models have confirmed the importance of Sprouty 2(Spry2) in inhibiting tumorigenesis and the progression of human cancer.However,the prognostic value of Spry2 in cancer patients remains unknown.This study is aimed to investigate the clinical relevance and prognostic significance of Spry2 expression in patients with hepatocellular carcinoma(HCC).METHODS:With samples from 240 randomly-selected HCC patients who underwent surgery,immunohistochemistry was used to investigate Spry2 expression on tissue microarrays.The correlation of Spry2 expression with survival was estimated by the Kaplan-Meier method and univariate/multivariate Cox proportional hazard regression analysis.Spry2,ERK and phospho-ERK expression in HCC cell lines was detected by Western blotting.RESULTS:Among the patients,86.3%(207 of 240) exhibited down-regulation of Spry2 expression.Patients negative for Spry2 showed poorer survival(P=0.002) and increased recurrence(P=0.003).Multivariate analysis further established Spry2 as an independent predictor of postoperative recurrence in HCC patients(HR=1.47;95% CI,1.02-2.08;P=0.037).Downregulation of Spry2 was associated with highly malignant phenotypes like vascular invasion and advanced tumor stages,and was positively correlated with the metastatic potential of HCC cell lines.CONCLUSION:In the era of molecular targeted therapy,the expression of Spry2 in HCC may have relevant clinical significance and turn out to be a key factor in prognostic assessment and in treatment planning. | Kang Song,Qiang Gao,Jian Zhou,Shuang-Jian Qiu,Xiao-Wu Huang,Xiao-Ying Wang and Jia Fan Liver Cancer Institute, Zhongshan Hospitaland Institute of Biomedical Sciences , Fudan University, Shanghai 200032, China | 2012 | Hepatobiliary & Pancreatic Diseases International2012,11,2: | 4 |

| 11 | LAPAROSCOPIC RADICAL PROSTATECTOMY:INITINAL EXPREIENCE OF 23 CASES IN SUN YAT-SEN UNIVERSITY显示文摘Objective: To report our prelimilary experience of laparoscopic radical prostatectomy (LRP) for clinically localized prostatic cancer.Methods:23 cases with localized prostatic cancer underwent LRP in our institution from October 2000 to August 2003. The median age was 64.5 years old, the median PSA was 13 ng/ml and median Gleason score of biopsy specimens was < 7. The operations were performed according to Montsouris technique. Briefly, operations were carried out transperitonealty, combining posterior and anterior approachs to the prostate, transecting the bladder neck, lateral dissection of the prostate, urcthrovesical anastomosis, completing the operation.Results:No death and no conversion rate were observed in this series of 23 cases of LRP. No re-intervention was needed. The median operating time was 325 minutes (range 270 to 660 min) including the lymphadenectomy phase that was considered necessary in the case whose PSA value was 51ng/ml and pathological reading was negative to thoses lymph nodes; The median intraoperative blood loss was 430 ml(rang 200 to 1100 ml). Postoperative bladder catheterization time was 20 days. Surgical positive margin was found in I case of pathlogical stage of pT 2b and Casodex 50 mg had been used on this case for 3 mon after the PSA value was < 0.2 ng/ml. Postoperation follow-up of 3 to 24 mo. showed no signs of complications such as urethral stricture or urine incontinence. | 13 | Endoscopic sphincterotomy or papillary balloon dilatation for choledocholithiasis显示文摘Objective: To assess the safety and effect of endoscopicsphincterotomy (EST) and endoscopic papillary bal-loon dilatation (EPBD) for choledocholithiasis.Methods: 328 patients with choledocholithiasis weresubjected to EST or EPBD; they included 174 patientswith single stone, 112 patients with two stones and 42patients with three stones (one patient with 20 stones).Patients with stones less than 10 mm in diameter un-derwent EPBD and those with stones larger than 11mm in diameter underwent EST.Results: EST and EPBD succeeded in 323 patients(98.5%), and failed in 5. Stones in 98 patients wereexcluded spontaneously after endoscopic therapy. 207patients were subjected to basket or balloon stone ex-traction. Stones in 14 patients were discharged bybasket lithotripsy. Four patients were given wavelithotripsy. Stones in 22 of the 323 patients were ex-tracted thoroughly after 2-3 times attempts. Totalcomplications were noted in 2.5% of the patients.Hemorrhage from the gastrointestinal tract was seenin one patient, cholangitis in 4 patients, and pancre-atitis in 3 patients. | 15 | Thermal evaluation of the injectable liquid metal bone cement in orthopedic treatment显示文摘The thermophysical properties of bone cement are important parameters for its application in the orthopedic treatment. This article focused on the thermal evaluation of the low-melting-point metal(Bi In Sn alloy), which has been proved to be an excellent bone cement. Firstly, the basic thermophysical properties of Bi In Sn alloys with different melting points were measured.Secondly, 15 fresh porcine femurs placed in the saline bath, bone cements with different melting points and amounts were injected into the bone cavities, respectively. Thermocouples were used to measure the temperature changes of the bone-cement interface and peripheral bone tissue. The possibility of thermal necrosis was evaluated. Moreover, a three-dimensional human knee model was built to numerically assess the effects of thermal parameters, such as melting point and latent heat on tissue temperature distribution. All the experimental and numerical results implied the heat distribution in the tissue depended on the thermal performances of liquid metal bone cement(LMBC). For LMBC of the same melting point, with increased amounts, the damage to the bone tissue is more severe, while for the same amount of different melting point LMBCs, with the higher melting point, which will lead to more serious damage to the tissue. Also, higher latent heat of LMBC has distinct longer solidification process, which may cause irreversible damage to surrounding tissues. | 16 | Liquid metal hydraulics paradigm:Transmission medium and actuation of bimodal signals显示文摘In this article,room temperature gallium-based fluids(RTGFs)with unique thermal and conductive properties are proposed as a transmission fluid for the force carrying medium of hydraulics,which rectify the lack of the multi-functional hydraulic roles of liquid metal(LM).The typical physical properties of RTGFs and comparative conventional hydraulic fluids(commercial hydraulic oil and deionized water),such as thermal stability and rheological characteristics,are evaluated.Experimental and numerical methods,then,were adopted to clarify the force transmission performance of RTGFs and commercial hydraulic oils,as well as the influence of temperature fields on the viscosity of fluids.The results disclosed that the advantages of inherent flame resistance,wide liquid temperature range,and the viscosity changing slightly with temperature made RTGFs potential in finding efficient application in the hydraulics as new working fluids.Finally,for illustration,the rigid and flexible actuators driven by RTGFs were designed as hydraulic fluid and demonstrated their capabilities in grasping objects with various shapes and weights,respectively.And the tunable stiffness of such a flexible actuator was enabled via the solid-liquid phase change of LM.Additionally,a frequency-adjustable antenna was manufactured and showcased owing to the introduced transformable electromagnetic behaviors of LM.Overall,the gallium-based LM fluids with thermoelectrical and mechanical multimodal signal medium would serve as a potential candidate for future complex multifunctional signal transmission systems. | FU JunHeng GAO JianYe QN Peng LI DongDong YU DeHai SUN Peng HE ZhiZhu DENG ZhongShan LIU Jing | 2022 | Science China(Technological Sciences)2022,65,1: | 1 |

The optimum Fe-doped content in terms of activity is approximately 0. 5 %. The increase in photoactivity is probably due to the higher adsorption and the inhibition of electron-hole recombination. The photocatalytic oxidation reaction of nitrite over the Fe-doped TiO2 catalyst follows zero-order kinetics, which is different from that over pure TiO2.
