| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Operating principles and detection characteristics of the Visible and Near-Infrared Imaging Spectrometer in the Chang'e-3显示文摘The Visible and Near-Infrared Imaging Spectrometer(VNIS), using two acousto-optic tunable filters as dispersive components, consists of a VIS/NIR imaging spectrometer(0.45–0.95 μm), a shortwave IR spectrometer(0.9–2.4 μm) and a calibration unit with dust-proofing functionality. The VNIS was utilized to detect the spectrum of the lunar surface and achieve in-orbit calibration, which satisfied the requirements for scientific detection. Mounted at the front of the Yutu rover, lunar objects that are detected with the VNIS with a 45° visual angle to obtain spectra and geometrical data in order to analyze the mineral composition of the lunar surface. After landing successfully on the Moon, the VNIS performed several explorations and calibrations, and obtained several spectral images and spectral reflectance curves of the lunar soil in the region of Mare Imbrium. | 3 | Effect of Morinda officinalis capsule on osteoporosis in ovariectomized rats显示文摘AIM:To explore the therapeutic effects of Morinda officinalis capsules(MOP) on osteoporosis in ovariectomized rats.METHOD:Six-month-old female Sprague–Dawley rats were induced for postmenopausal osteoporosis(PMOP) by bilateral ovariectomy and divided into seven groups as follows:sham-operated group,ovariectomized(OVX) control group,OVX treated with xianlinggubao(XLGB)(270 mg·kg-1·d-1),OVX treated with alendronate sodium(ALN)(3 mg·kg-1·d-1),and OVX treated with Morinda officinalis capsule(MOP) of graded doses(90,270 and 810 mg·kg-1·d-1) groups.Oral treatments were administered daily on the 4th week after ovariectomy and lasted for 12 weeks.The bone mineral density was evaluated by dual-energy X-ray absorptiometry.The tartrate-resistant acid phosphatase(TRAP),alkaline phosphatase(AKP),and osteocalcin(OC) levels in the serum and plasma were determined by standard colorimetric and enzyme immunoassays methods.Bone biomechanical properties and morphological parameters were analyzed by three-point bending test and histomorphometry respectively.RESULTS:Morinda officinalis capsules at all doses were able to significantly prevent the OVX-induced loss of bone mass due to diminishing serum AKP and TRAP levels while elevating OC level in the plasma.Morinda officinalis capsules also enhanced the bone strength and prevented the deterioration of trabecular microarchitecture.CONCLUSION:Morinda officinalis capsules possess potent anti-osteoporotic activity in OVX rats which could be an effective treatment for postmenopausal osteoporosis. | LI Ye Lü Shan-Shan TANG Gui-Ying HOU Min TANG Qing ZHANG Xiao-Na CHEN Wei-Hai CHEN Gang XUE Qiang ZHANG Cong-Cong ZHANG Ji-Fen CHEN Yi XU Xiao-Yu | 2014 | Chinese Journal of Natural Medicines2014,12,3: | 10 |
| 4 | Development of genome-wide insertion/deletion markers in rice based on graphic pipeline platform显示文摘DNA markers play important roles in plant breeding and genetics.The Insertion/Deletion(InDel) marker is one kind of co-dominant DNA markers widely used due to its low cost and high precision.However,the canonical way of searching for InDel markers is time-consuming and laborintensive.We developed an end-to-end computational solution(InDel Markers Development Platform,IMDP) to identify genome-wide InDel markers under a graphic pipeline environment.IMDP constitutes assembled genome sequences alignment pipeline(AGA-pipe) and next-generation resequencing data mapping pipeline(NGS-pipe).With AGA-pipe we are able to identify 12,944 markers between the genome of rice cultivars Nipponbare and 93-11.Using NGS-pipe,we reported 34,794 InDels from re-sequencing data of rice cultivars Wu-Yun-Geng7 and Guang-Lu-Ai4.Combining AGApipe and NGS-pipe,we developed 205,659 InDels in eight japonica and nine indica cultivars and 2,681 InDels showed a subgroup-specific pattern.Polymerase chain reaction(PCR)analysis of subgroup-specific markers indicated that the precision reached 90%(86 of 95).Finally,to make them available to the public,we have integrated the InDels/markers information into a website(Rice InDel Marker Database,RIMD,http://gffzz71b3756633b74a6ehp0qopkou59pf6cxb.ffgz.tsg.suse.edu.cn/).The application of IMDP in rice will facilitate efficiency for development of genome-wide InDel markers,in addition it can be used in other species with reference genome sequences and NGS data. | Yang L Xiao Cui Rui Li Piaopiao Huang Jie Zong Danqing Yao Gang Li Dabing Zhang Zheng Yuan | 2015 | Journal of Integrative Plant Biology2015,57,11: | 9 |

High expression of the BDNF-green fluorescent protein (GFP) gene persists for a long time after transfection into neural stem cells. Nevertheless, little is known about the biological characteristics of BDNF-GFP modified nerve stem cells in vivo and their ability to induce BDNF expression or repair spinal cord injury. In the present study, we transplanted BDNF-GFP transgenic neural stem cells into a hemisection model of rats. Rats with BDNF-GFP stem cells exhibited significantly increased BDNF expression and better locomotor function compared with stem cells alone. | 9 | Effect of oxygen partial pressure and transparent substrates on the structural and optical properties of ZnO thin films and their performance in energy harvesters显示文摘Zinc oxide(ZnO) thin films were deposited onto different substrates — tin-doped indium oxide(ITO)/glass, ITO/polyethylene naphthalate(PEN), ITO/polyethylene terephthalate(PET) — by the radio-frequency(RF) magnetron sputtering method. The effect of various O2/(Ar+O2) gas flow ratios(0, 0.1, 0.2, 0.3, 0.4, 0.5, and 0.6) was studied in detail. ZnO layers deposited onto ITO/PEN and ITO/PET substrates exhibited a stronger c-axis preferred orientation along the(0002) direction compared to ZnO deposited onto ITO/glass. The transmittance spectra of ZnO films showed that the maximum transmittances of ZnO films deposited onto ITO/glass, ITO/PEN, and ITO/PET substrates were 89.2%, 65.0%, and 77.8%, respectively. Scanning electron microscopy(SEM) images of the film surfaces indicated that the grain was uniform. The cross-sectional SEM images showed that the ZnO films were columnar structures whose c-axis was perpendicular to the film surface. | 11 | Structural characteristics of the KPR-CBR triple-junction inferred from gravity and magnetic interpretations,Philippine Sea Plate显示文摘The intersection of the Kyushu-Palau Ridge(KPR)and the Central Basin Rift(CBR)of the West Philippine Basin(WPB)is a relic of a trench-trench-rift(TTR)type triple-junction,which preserves some pivotal information on the cessation of the seafloor spreading of the WPB,the emplacement and disintegration of the proto-Izu-Bonin-Mariana(IBM)Arc,and the transition from initial rifting to steady-state spreading of the Parece Vela Basin(PVB).However,the structural characteristics of this triple-junction have not been thoroughly understood.In this paper,using the newly acquired multi-beam bathymetric,gravity,and magnetic data obtained by the Qingdao Institute of Marine Geology,China Geological Survey,the authors depict the topographic,gravity,and magnetic characteristics of the triple-junction and adjacent region.Calculations including the upward continuations and total horizontal derivatives of gravity anomaly are also performed to highlight the major structural features and discontinuities.Based on these works,the morphological and structural features and their formation mechanisms are analyzed.The results show that the last episode amagmatic extension along the CBR led to the formation of a deep rift valley,which extends eastward and incised the KPR.The morphological and structural fabrics of the KPR near and to the south of the triple-junction are consistent with those of the western PVB,manifesting as a series of NNE-SSW-and N-S-trending ridges and troughs,which were produced by the extensional faults associated with the initial rifting of the PVB.The superposition of the above two reasons induced the prominent discontinuity of the KPR in deep and shallow crustal structures between 15°N‒15°30′N and 13°30′N‒14°N.Combined with previous authors’results,we propose that the stress produced by the early spreading of the PVB transmitted westward and promoted the final stage amagmatic extension of the CBR.The eastward propagation of the CBR destroyed the KPR,of which the magmatism had decayed or ceased at that time.The destruction mechanism of the KPR associated with the rifting of the PVB varies along strike the KPR.Adjacent to the triple-junction,the KPR was destroyed mainly due to the oblique intersection of the PVB rifting center.Whereas south of the triple-junction,the KPR was destroyed by the E-W-directional extensional faulting on its whole width. | Lu-ning Shang Pan-feng Li Run-lin Du Feng-long Bai Gang Hu Wen-chao Lü Xia Li Xi Mei Tian-yu Zhang Hou-zhen Cao Jing-yi Cong Xian-yao Shi | 2021 | China Geology2021,4,4: | 3 |
