| 1 | The alteration in the architecture of a T-DNA insertion rice mutant osmtd1 is caused by up-regulation of Micro RNA156f显示文摘Plant architecture is an important factor for crop production. Some members of micro RNA156(mi R156) and their target genes SQUAMOSA Promoter-Binding Protein-Like(SPL) were identi fi ed to play essential roles in the establishment of plant architecture. However, the roles and regulation of mi R156 is not well understood yet. Here, we identi fi ed a T-DNA insertion mutant Osmtd1(Oryza sativa multi-tillering and dwarf mutant). Osmtd1 produced more tillers and displayed short stature phenotype. We determined that the dramatic morphological changes were caused by a single T-DNA insertion in Osmtd1. Further analysis revealed that the T-DNA insertion was located in the gene Os08g34258 encoding a putative inhibitor I family protein. Os08g34258 was knocked out and Osmi R156 f was signi fi cantly upregulated in Osmtd1. Overexpression of Os08g34258 in Osmtd1 complemented the defects of the mutant architecture, while overexpression of Osmi R156 f in wild-type rice phenocopied Osmtd1. We showed that the expression of Os SPL3, Os SPL12,and Os SPL14 were signi fi cantly downregulated in Osmtd1 or Osmi R156 f overexpressed lines, indicating that Os SPL3,Os SPL12, and Os SPL14 were possibly direct target genes of Osmi R156 f. | 2 | Retinoschisis at Macular Area in Highly Myopic Eye by Optic Coherence Tomography显示文摘Purpose:To improve our understanding of macular schisis in highly myopic eyes. Methods:From 2002.6-2003.12, the highly myopic eyes(>6D) which has macular schisis found by OCT at our hospital were included in the study. Each eye then received further examinations including visual acuity,refractive error and ultra-sonography. Then the results were analyzed to find the characteristic of the entity. Results:During that period, OCT found macular schisis in 38 highly myopic eyes from 36 patients. Their vision was low, and their average axial length and refractive error were 29.6mm and -10.5D respectively. The appearance and the area of the schisis are different among the eyes. But the schisis can be largely divided into two types: inner and outer schisis. They can exit independently or concurrently in the same eye. And the schisis was often accompanied by other macular findings, like retinal detachment, macular hole. Conclusion:Macular schisis in highly myopic eyes is not uncommon in the clinic. | 6 | G protein b_1λ_2 subunits purification and their interaction with adenylyl cyclase显示文摘A preliminary study on the interaction of G protein (guanine triphosphate binding pro- tein) b1g2 subunits and their coupled components in cell signal transduction was conducted in vitro. The insect cell lines, Sf9 (Spodoptera frugiperda) and H5 (Trichoplusia ni ) were used to express the recombinant protein Gb1g2. The cell membrane containing Gb1g2 was isolated through affinity chromatography column with Ni-NTA agarose by FPLC method, and the highly purified protein was obtained. The adenylyl cyclase 2 (AC2) activity assay showed that the purified Gb1g2 could signifi-cantly stimulate AC2 activity. The interaction of b1g2 subunits of G protein with the cytoplasmic tail of various mammalian adenylyl cyclases was monitored by BIAcore technology using NTA sensor chip, which relies on the phenomenon of surface plasmon resonance (SPR). The experiments showed the direct binding of Gb1g2 to the cytoplasmic tail C2 domain of AC2.
