| 2 | Wheat breeding history reveals synergistic selection of pleiotropic genomic sites for plant architecture and grain yield显示文摘Diversity surveys of crop germplasm are important for gaining insights into the genomic basis for plant architecture and grain yield improvement,which is still poorly understood in wheat.In this study,we exome sequenced 287 wheat accessions that were collected in the past 100 years.Population genetics analysis identified that 6.7%of the wheat genome falls within the selective sweeps between landraces and cultivars,which harbors the genes known for yield improvement.These regions were asymmetrically distributed on the A and B subgenomes with regulatory genes being favorably selected.Genome-wide association study(GWAS)identified genomic loci associated with traits for yield potential,and two underlying genes,TaARF12 encoding an auxin response factor and TaDEP1 encoding the G-proteinγ-subunit,were located and characterized to pleiotropically regulate both plant height and grain weight.Elite single-nucleotide haplotypes with increased allele frequency in cultivars relative to the landraces were identified and found to have accumulated over the course of breeding.Interestingly,we found that TaARF12 and TaDEP1 function in epistasis with the classical plant height Rht-1 locus,leading to propose a“Green Revolution”-based working model for historical wheat breeding.Collectively,our study identifies selection signatures that fine-tune the gibberellin pathway during modern wheat breeding and provides a wealth of genomic diversity resources for the wheat research community. | Aili Li Chenyang Hao Zhenyu Wang Shuaifeng Geng Meiling Jia Fang Wang Xiang Han Xingchen Kong Lingjie Yin Shu Tao Zhongyin Deng Ruyi Liao Guoliang Sun Ke Wang Xingguo Ye Chengzhi Jiao Hongfeng Lu Yun Zhou Dengcai Liu Xiangdong Fu Xueyong Zhang Long Mao | 2022 | Molecular Plant2022,15,3: | 4 |
| 3 | Experimental and computational investigations of LaNi(5-x)Alx (x = 0,0.25, 0.5, 0.75 and 1.0) tritium-storage alloys显示文摘Although already scientists in recent years have reported some experimental and theoretical results of LaNi-Al series of tritium-storage alloys, several key aspects remain the subject of considerable debate. In an effort to interpret some of these unknowns, we have performed experimental and theoretical investigations for LaNi_(5-x)Al_x(x = 0, 0.25, 0.5, 0.75 and 1.0) tritium-storage alloys. Firstly, the XRD characterization indicates that the unit cell volumes of LaNi_(5-x)Al_x increase with Al content in alloys. Secondly, the PCisotherm measurement of LaNi_(5-x)Al_xalloys shows that their hydrogen absorption/desorption plateau pressures reduce with the increase of Al content while their plateau widths narrow simultaneously. The deuterium absorption/desorption plateaus have a similar trend to hydrogen's except for their plateaus being higher than hydrogen's. To explain the above experimental findings, a series of calculations based on density functional theory(DFT) and frozen phonon approach have been performed. The results manifest that:(1) the partial substitutions of Al for Ni reduce the hydrogen formation energies of LaNi_(5-x)Al_xH and the number of available interstitial sites, and therefore lead to the absorption/desorption plateau pressures being reduced and the plateau widths being narrowed down at the same experimental temperatures;(2) the covalent interaction between H and Ni is an important factor for estimating the stability of LaNi_(5-x)Al_x-H system;(3) since the calculated enthalpy change H is generally more accurate than the calculated entropy change S with respect to the corresponding experimental value for each LaNi_(5-x)Al_xH(or D), the curves of H vs. hydrogen storage capacity instead of Van't Hoff relation, can be used to predict the experimental plateau pressures of LaNi_(5-x)Al_x-H(D or T) at a given temperature;(4) the hydrogen isotope effect of LaNi_(5-x)Al_x-H(D or T) system can be quantitatively described as a linearity relation between ⊿ZPE + ⊿H^(vib) and 1/√mQ(Q = H, D, T). From the good agreement between the predicted and experimental ln(P_H/P_0) and ln(P_D/P_0), it is deduced that predicting ln(P_T/P_0) of LaNi_(5-x)Al_x T is feasible. | 5 | Detection of the Pandemic H1N1/2009 Influenza A Virus by a Highly Sensitive Quantitative Real-time Reverse-transcription Polymerase Chain Reaction Assay显示文摘A quantitative real time reverse-transcription polymerase chain reaction (qRT-PCR) assay with specific primers recommended by the World Health Organization (WHO) has been widely used successfully for detection and monitoring of the pandemic H1N1/2009 influenza A virus. In this study, we report the design and characterization of a novel set of primers to be used in a qRT-PCR assay for detecting the pandemic H1N1/2009 virus. The newly designed primers target three regions that are highly conserved among the hemagglutinin (HA) genes of the pandemic H1N1/2009 viruses and are different from those targeted by the WHO-recommended primers. The qRT-PCR assays with the newly designed primers are highly specific, and as specific as the WHO-recommended primers for detecting pandemic H1N1/2009 viruses and other influenza viruses including influenza B viruses and influenza A viruses of human, swine, and raccoon dog origin. Furthermore, the qRT-PCR assays with the newly designed primers appeared to be at least 10-fold more sensitive than those with the WHO-recommended primers as the detection limits of the assays with our primers and the WHO-recommended primers were 2.5 and 25 copies of target RNA per reaction, respectively. When tested with 83 clinical samples, 32 were detected to be positive using the qRT-PCR assays with our designed primers, while only 25 were positive by the assays with the WHO-recommended primers. | 16 | The soft glumes of common wheat are sterile-lemmas as determined by the domestication gene Q显示文摘The Q gene in common wheat encodes an APETALA2(AP2) transcription factor that causes the free threshing attribute. Wheat spikelets bearing several florets are subtended by a pair of soft glumes that allow free liberation of seeds. In wild species, the glumes are tough and rigid,making threshing difficult. However, the nature of these 'soft glumes', caused by the domestication allele Q is not clear. Here, we found that over expression of Q in common wheat leads to homeotic florets at glume positions. We provide phenotypic, microscopy, and marker genes evidence to demonstrate that the soft glumes of common wheat are in fact lemma-like organs, or so-called sterile-lemmas. By comparing the structures subtending spikelets in wheat and other crops such as rice and maize, we found that AP2 genes may play conserved functions in grasses by manipulating vestigial structures, such as floret-derived soft glumes in wheat and empty glumes in rice. | 18 | Determination of the predictive factors for diagnostic positivity of nucleic acid amplification tests for diagnosing pulmonary tuberculosis显示文摘Background:Tuberculosis(TB)remains a major threat to human health,and TB diagnostic methods remain unsatisfactory.Nucleic acid amplification tests(NAATs)show higher sensitivity compared with culture for the diagnosis of pulmonary TB(PTB).However,NAATs are expensive and cannot be easily implemented outside major medical centers.To improve the sensitivity of NAATs for PTB diagnosis,we investigated the predictive factors that might optimize NAAT utilization.Methods:A total of 1263 patients with suspected PTB were enrolled for evaluation.The sensitivity,specificity,and accuracy of methods including smear-microbiology,culture of Mtb and NAAT for Mycobacterium tuberculosis(Mtb)detection in sputum and bronchoalveolar lavage fluid samples were compared.Odds ratios and 95%confidence intervals were used to assess variables that might be associated with positive NAAT results for sputum and bronchoalveolar lavage fluid from patients with suspected PTB.Results:NAAT showed higher sensitivity for Mtb detection(61.1%)when compared with smear(9.0%)and Mtb culture(47.8%).We found that an elevated erythrocyte sedimentation rate,the presence of cavities,and positive interferon-��release assay(IGRA)results were indicative of positive Mtb detection by NAAT.Moreover,individuals who had all three of these characteristics showed an 86%diagnostic positivity for PTB from Mtb detection by NAAT.Conclusions:Our study suggests that an elevated erythrocyte sedimentation rate,a positive IGRA result,and the presence of pulmonary cavities are helpful factors for predicting positive Mtb detection by NAAT.Patients with the three positive clinical markers should undergo NAAT for Mtb detection because they are the most likely individuals to be bacteriologically confirmed as having TB. | Xingfang Hou Qinglong Guo Qiao Lin Taosheng Ye Jing Bi Juan Liang Ke Yang Yuan Zhou Juanjuan Zhang Zhihang Liang Xuefeng Zhou Gengwei Zhang Xiangdong Fu Hongjian Zhong Guoliang Zhang | 2022 | Infectious Medicine2022,1,1: | 0 |
