Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China, has become a public health emergency of international concern. The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models. Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs, viral replication, chest X-ray,histopathological changes and immune response. Results:Viral replication of nasopharyngeal swabs, anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema, notably, old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection. | Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin | 2020 | Animal Models and Experimental Medicine2020,3,1: | 22
Sequential infection with H1N1 and SARS-CoV-2 aggravated COVID-19 pathogenesis in a mammalian model, and covaccination as an effective method of prevention of COVID-19 and influenza显示文摘Influenza A virus may circulate simultaneously with the SARS-CoV-2 virus, leading to more serious respiratory diseases during this winter. However, the influence of these viruses on disease outcome when both influenza A and SARS-CoV-2 are present in the host remains unclear.Using a mammalian model, sequential infection was performed in ferrets and in K18-MCE2 mice, with SARS-CoV-2 infection following H1N1.We found that co-infection with H1N1 and SARS-CoV-2 extended the duration of clinical manifestation of COVID-19,and enhanced pulmonary damage, but reduced viral shedding of throat swabs and viral loads in the lungs of ferrets. Moreover, mortality was increased in sequentially infected mice compared with single-infection mice.Compared with singlevaccine inoculation,co-inoculation of PiCoVacc(a SARS-CoV-2 vaccine)and the flu vaccine showed no significant differences in neutralizing antibody titers or virus-specific immune responses.Combined immunization effectively protected K18-MCE2 mice against both H1N1 and SARS-CoV-2 infection. Our findings indicated the development of systematic models of co-infection of H1N1 and SARS-CoV-2,which together notably enhanced pneumonia in ferrets and mice,as well as demonstrated that simultaneous vaccination against HINT and SARS-CoV-2 may be an effective prevention strategy for the coming winter. | Linlin Bao Wei Deng Feifei Qi Qi Lv Zhiqi Song Jiangning Liu Hong Gao Qiang Wei Pin Yu Yanfeng Xu Yajin Qu Fengdi Li Jing Xue Shuran Gong Mingya Liu Guanpeng Wang Shunyi Wang Binbin Zhao Bin Cong Chuan Qin | 2021 | Signal Transduction and Targeted Therapy2021,6,6: | 4
ACE2 expression is regulated by AhR in SARS-CoV-2-infected macaques显示文摘During the current outbreak of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),more than 115 million people have been infected, and 2.5 million have died.1,2 Despite such great harm to human health, the pathogenesis of COVID-19 remains unclear. As the first step in the pathogenetic process, viral entry is mediated by the binding of the SARS-CoV-2 surface spike(S)protein to angiotensin-converting enzyme 2(ACE2)on host cells, such as lung epithelial cells. As an alternative to S protein-blocking strategies, manipulating host cell ACE2 expression may exert an inhibitory effect on SARS-CoV-2 infection. However, the molecular mechanism regulating ACE2 expression remains unclear. | Jiadi Lv Pin Yu Zhenfeng Wang Wei Deng Linlin Bao Jiangning Liu Fengli Li Qiangqiang Zhu Nannan Zhou Qi Lv Guanpeng Wang Shunyi Wang Yabo Zhou Jiangping Song Wei-Min Tong Yuying Liu Chuan Qin Bo Huang | 2021 | Cellular & Molecular Immunology2021,18,5: | 1

Establishment of Indirect ELISA Diagnosis Technique based on the VP1 Protein of Foot and Mouth Disease Virus Serotype A显示文摘The VP1 protein of foot-and-mouth disease virus serotype A was prokaryotically expressed and purified to replace the traditional virus antigen for establishing a fast, safe, effective indirect ELISA method,so as to detecting antibody of foot-and-mouth disease virus serotype A. Western-Blot test showed that the VP1 recombinant protein could be used as detective antigen as it can be specifically recognized by bovine positive serum of FMDV serotype A. By employing matrix titration method, the optimal parameters were obtained as follows: 1 mg / L VP1 protein as coating antigen,Vserum∶ Vblocking solution = 1∶50 dilution for serum and Vsecondary enzyme-linked antibodies∶Vblocking solution = 1∶2 000 for enzyme combined antibodies. The results showed that the sensitivity and specificity of this method were 94. 32% and 99. 09% respectively, the coefficients of variations in intra-assay and inter-assay reproducibility tests was lower than 8%. Compared with liquid phase blocking ELISA kits, the agreement of 201 serum samples reached 92. 54%. The VP1-ELISA method established here is specific, sensitive, stable and simple, which can be used to monitor the antibody level of FMD serotype A. | Lu Qingxia Liu Chang Jin Qianyue Guo Guanpeng Xing Guangxu Liu Yunchao Deng Ruiguang Zhang Gaiping | 2014 | Animal Husbandry and Feed Science2014,6,6: | 1
Vacuum wetting of Ag/TA2 to develop a novel micron porous Ti with significant biocompatibility and antibacterial activity显示文摘Porous Ti with low modulus, excellent bio-corrosion resistance, biocompatibility, and antibacterial activity is highly pursued as advanced implant materials. In this work,a new approach to prepare micron porous structures on the surface layer of a grade 2 commercially-pure Ti(TA2)was proposed, which utilized a simple vacuum wetting process of pure Ag on the surface of TA2.The microstructure, corrosion resistance, biocompatibility, mechanical properties, antibacterial ability, and formation mechanism of the asfabricated porous Ti were studied. The results show that the pores(with average pore sizes of 0.5-5μm)are distributed on the surface layer of the TA2 with a depth of~10μm.In particular,a large number of silver nanoparticles(Ag NPs)form which are dispersed on the porous structures. The formation mechanisms of the porous structures and Ag NPs were elucidated, suggesting that the volatilization/sublimation of Ag in TA2 is crucial. The porous Ti possesses excellent bio-corrosion resistance, surface wettability, biocompatibility, antibacterial activity, and a relatively low elastic modulus of 40-55 GPa,which may have a promising future in the field of orthopedic implants. This work also provides a novel idea for the development of advanced porous Ti materials for orthopedic-related basic research and biomedical applications. | Guanpeng Liu Yulong Li Ming Yan Jicai Feng Jian Cao Min Lei Quanwen Liu Xiaowu Hu Wenqin Wang Xuewen Li | 2022 | Journal of Materials Science & Technology2022,,21: | 0
Synthesis and Property of H_2La_2Ti_3O_(10)/CdS Layered Nanocomposite Photocatalyst显示文摘H2La2Ti3O10/CdS nanocomposite with higher photoactivity was synthesized by a stepwise exchange process from H2La2Ti3O 10 , which was obtained by H+-exchanging reaction of K2La2Ti3O 10 with HCl other than the normally used HNO3. The pillaring process was investigated by XRD, TEM, FT-IR and BET methods. The photocatalytic decomposition of aniline was used as the model system to evaluate the photochemical properties of H2La2Ti3O 10 /CdS, H2La2Ti3O 10 and K2La2Ti3O 10 . It is found that the photoactivity of layered H2La2Ti3O 10 is greatly improved by the intercalation of CdS in the interlayer. In general, the excellent photoactivity of the H2La2Ti3O 10 /CdS nanocomposite might be attributing to the extremely small particle size of incorporated CdS and good contact between CdS and La2Ti3O 10 2- layers. On the other hand, the coupling of two semiconductor particles with different energy gap is useful to achieve effective charge separation. In H2La2Ti3O 10 /CdS, a photo-generated electron can transfer from CdS to the La2Ti3O 10 2- layer, while the holes remain in the CdS particles. This helps to diffuse the electrons and holes before reaching the interface, and the holes and electrons can be effectively captured by the electrolyte in the solution. Simultaneously, the optimal operating condition of photodecomposition aniline was investigated. | Zhang Lili Liu Guanpeng Zhang Weiguang Lu Lude Yang Xujie Wang Xin | 2006 | Journal of Rare Earths2006,24,2: | 0
The K Domain Mediates Homologous and Heterologous Interactions Between FLC and SVP Proteins of Brassica juncea显示文摘The transcription factors FLOWERING LOCUS C(FLC) and SHORT VEGETATIVE PHASE(SVP) can interact to form homologous and heterologous protein complexes that regulate flowering time in Brassica juncea Coss.(Mustard). Previous studies showed that protein interactions were mediated by the K domain, which contains the subdomains K1,K2 and K3. However,it remains unknown how the subdomains mediate the interactions between FLC and SVP. In the present study,we constructed several mutants of subdomains K1–K3 and investigated the mechanisms involved in the heterologous interaction of BjFLC/BjSVP and in the homologous interaction of BjFLC/Bj FLC or BjSVP/BjSVP. Yeast two-hybrid and β-galactosidase activity assays showed that the 19 amino acids of the K1 subdomain in BjSVP and the 17 amino acids of the K1 subdomain in BjFLC were functional subdomains that interact with each other to mediate hetero-dimerization. The heterologous interaction was enhanced by the K2 subdomain of BjSVP protein, but weakened by its interhelical domain L2. The heterologous interaction was also enhanced by the K2 subdomain of BjFLC protein, but weakened by its K3 subdomain. The homologous interaction of BjSVP was mediated by the full K-domain. However, the homologous interaction of BjFLC was regulated only by its K1 and weakened by its K2 and K3 subdomains. The results provided new insights into the interactions between FLC and SVP,which will be valuable for further studies on the molecular regulation mechanisms of the regulation of flowering time in B. juncea and other Brassicaceae. | MA Guanpeng LIU Zhiyu YANG Puli XU Junqiang SONG Ming TANG Qinglin | 2015 | Horticultural Plant Journal2015,1,1: | 0
