1Oleic Acid and Eicosapentaenoic Acid Reverse Palmitic Acid-induced Insulin Resistance in Human HepG2 Cells via the Reactive Oxygen Species/JUN Pathway显示文摘Oleic acid(OA),a monounsaturated fatty acid(MUFA),has previously been shown to reverse saturated fatty acid palmitic acid(PA)-induced hepatic insulin resistance(IR).However,its underlying molecular mechanism is unclear.In addition,previous studies have shown that eicosapentaenoic acid(EPA),aω-3 polyunsaturated fatty acid(PUFA),reverses PA-induced muscle IR,but whether EPA plays the same role in hepatic IR and its possible mechanism involved need to be further clarified.Here,we confirmed that EPA reversed PA-induced IR in HepG2 cells and compared the proteomic changes in HepG2 cells after treatment with different free fatty acids(FFAs).A total of 234 proteins were determined to be differentially expressed after PA+OA treatment.Their functions were mainly related to responses to stress and endogenous stimuli,lipid metabolic process,and protein binding.For PA+EPA treatment,the PA-induced expression changes of 1326 proteins could be reversed by EPA,415 of which were mitochondrial proteins,with most of the functional proteins involved in oxidative phosphorylation(OXPHOS)and tricarboxylic acid(TCA)cycle.Mechanistic studies revealed that the protein encoded by JUN and reactive oxygen species(ROS)play a role in OA-and EPA-reversed PA-induced IR,respectively.EPA and OA alleviated PA-induced abnormal adenosine triphosphate(ATP)production,ROS generation,and calcium(Ca^(2+))content.Importantly,H_(2)O_(2)-activated production of ROS increased the protein expression of JUN,further resulting in IR in HepG2 cells.Taken together,we demonstrate that ROS/JUN is a common response pathway employed by HepG2 cells toward FFA-regulated IR.Yaping Sun Jifeng Wang Xiaojing Guo Nali Zhu Lili Niu Xiang Ding Zhensheng Xie Xiulan Chen Fuquan Yang 2021Genomics, Proteomics & Bioinformatics2021,19,5:6
2Biomechanical properties of acellular sciatic nerves treated with a modified chemical method显示文摘Nerve grafts are able to adapt to surrounding biomechanical environments if the nerve graft itself exhibits appropriate biomechanical properties (load, elastic modulus, etc.). The present study was designed to determine the differences in biomechanical properties between fresh and chemically acellularized sciatic nerve grafts. Two different chemical methods were used to establish acellular nerve grafts. The nerve was chemically extracted in the Sondell method with a combination of Triton X-100 (nonionic detergent) and sodium deoxycholate (anionic detergent), and in the modified method with a combination of Triton X-200 (anionic detergent), sulfobetaine-10 (SB-10, amphoteric detergents), and sulfobetaine-16 (SB-16, amphoteric detergents). Following acellularization, hematoxylin-eosin staining and scanning electron microscopy demonstrated that the effect of acellularization via the modified method was similar to the traditional Sondell method. However, effects of demyelination and nerve fiber tube integrity were superior to the traditional Sondell method. Biomechanical testing showed that peripheral nerve graft treated using the chemical method resulted in decreased biomechanical properties (ultimate load, ultimate stress, ultimate strain, and mechanical work to fracture) compared with fresh nerves, but the differences had no statistical significance (P > 0.05). These results demonstrated no significant effect on biomechanical properties of nerves treated using the chemical method. 3Numerical simulation of spray performance based on the Euler-Lagrange Approach显示文摘Numerical simulations have been carded out to investigate the liquid atomization and spray process using theDiscrete Phase Model of the commercial CFD code combined with the Wall-Film boundary conditions. The effectsof spray parameters on droplets Santer mean diameter (SMD), droplet collision speed, the thickness of liquid-film,the surface temperature and its uniformity were analyzed in the present study. The simulation results andthe experimental data obtained in the available literature agree within 13.8%. The computational results show thatthe spray pressure is the main factor to realize the atomization. 4Sphingosine-1-phosphate,a novel TREM2 ligand,promotes microglial phagocytosis to protect against ischemic brain injury显示文摘The mechanism of sphingosine-1-phosphate(S1P)-mediated phagocytosis remains unknown.Here,we found that S1P or FTY720(an analog of S1P)promoted microglial phagocytosis in stroke independent of S1PRs.First,we used computer simulation of molecular docking to predict that S1P might be a ligand for triggering receptor expressed on myeloid cells 2(TREM2).Next,microscale thermophoresis(MST),surface plasmon resonance(SPR)and liquid chromatography–tandem mass spectrometry(LC–MS/MS)were performed to reveal that S1P was a novel TREM2 ligand.Then,we confirmed the pro-phagocytosis of S1P targeting in Trem2-Dap12 transfected CHO cells and TREM2 knockdown microglia.Point mutation analysis showed that D104 was the critical binding residue.Trem2^(−/−)mice were used to demonstrate the role of S1P-induced phagocytosis targeting on TREM2 in protecting against ischemic brain injury.Finally,further studies revealed that apolipoprotein E(APOE)loaded with S1P was released by microglia and bound to apoptotic neurons via LDL receptor related protein 1B(LRP1B)and thereby induced microglia to phagocytose apoptotic neurons.Overall,the present work reveals for the first time that S1P acts as a novel endogenous ligand of TREM2 to effectively promote microglial phagocytosis.Our findings provide a new lead compound for developing immunomodulator targeting on TREM2.Tengfei Xue Juan Ji Yuqin Sun Xinxin Huang Zhenyu Cai Jin Yang Wei Guo Ruobing Guo Hong Cheng Xiulan Sun 2022Acta Pharmaceutica Sinica B2022,12,4:3
5Experimental and Numerical Study of an 80-kW Zigzag Printed Circuit Heat Exchanger for Supercritical CO_(2)Brayton Cycle显示文摘In this study,an experimental system was built to investigate the global performance of an 80-k W zigzag printed circuit heat exchanger(PCHE).It could meet the requirement of the pre-cooler for the supercritical carbon dioxide(S-CO_(2))Brayton power cycle and the modified effectiveness considering the pinch point is between 61.5%and 79.3%.When the outlet S-CO_(2)temperature is near the pseudo-critical point,the thermo-physical properties have more effects on heat transfer performance compared to flow characteristics.For the local performance,the mass flow rates of both sides have crucial influences on the location where the peak of S-CO_(2)Nusselt number occurs while only the S-CO_(2)flow rate affects the variation of the peak value.In addition,the influence of the radius of curvature on the secondary-flow should not be ignored.In the end,new empirical correlations were proposed considering the drastic variations of the Prandtl number.ZHANG Huzhong CHENG Keyong HUAI Xiulan ZHOU Jingzhi GUO Jiangfeng 2021Journal of Thermal Science2021,30,4:3

8Effects of Chemical Reaction Caused by Cooling Stream on Film Cooling Effectiveness显示文摘With the increase of inlet temperature of gas turbines, the benefits by using the conventional methods are likely to approach their limits. Therefore, it is essential to study novel film cooling methods for surpassing these current limits. Based on the theory of heat transfer enhancement, a film cooling method with chemical reaction by cool- ing stream is proposed. In order to test the feasibility of the proposed method, numerical simulations have been conducted. The classic flat plate structure with a 30 degree hole is used for the simulation. In the present study, the effects of the parameters in relation to the chemical reaction on film cooling effectiveness, such as chemical heat sink, volume changes, and reaction rate, are investigated numerically. The conventional film cooling is also calculated for the comparison. The results show that film cooling effectiveness is improved obviously due to the chemical reaction, and the reaction heat and reaction rate of cooling stream have an important effect on film ef- fectiveness. 12Sprayable and rapidly bondable phenolic-metal coating for versatile oil/water separation显示文摘Phenolic-metal complexation coatings have been discovered to be a universal route for the deposition of multifunctional coatings. However, most complexation coatings have been prepared by the immersion method, which limits their practical large-scale application. Herein, we describe a facile and green engineering strategy that involves spraying phenolic compound and metal ions on substrate to form in-situ complexation coating with different coordination states. The coating is formed within minutes and it can be achieved in large scale by the spray method. The pyrogallol-Fem complexation coating is prepared at pH 7.5, which consists predominantly of biscoordination complexation with a small amount of tris-coordination complexation. It displays that the water contact angle is near zero due to the generation of rough hierarchical structures and massive hydroxyl groups. The superhydrophilic cotton resulting from the deposition of the pyrogallol-Fe^Ⅲ complexation can separate oil/water mixtures and surfactant-stabilized oil-in-water emulsions with high separation efficiency. 15PRMT6 physically associates with nuclear factor Y to regulate photoperiodic flowering in Arabidopsis显示文摘The timing of floral transition is critical for reproductive success in flowering plants.In long-day(LD)plant Arabidopsis,the floral regulator gene FLOWERING LOCUS T(FT)is a major component of the mobile florigen.FT expression is rhythmically activated by CONSTANS(CO),and specifically accumu-lated at dusk of LDs.However;the underlying mechanism of adequate regulation of FT transcription in response to day-length cues to warrant flowering time still remains to be investigated.Here,we identify a homolog of human protein arginine methyltransferases 6(HsPRMT6)in Arabidopsis,and confirm AtPRMT6 physically interacts with three positive regulators of flowering Nuclear Factors YC3(NF-YC3),NF-YC9,and NF-YB3.Further investigations find that AtPRMT6 and its encoding protein accumulate at dusk of LDs.PRMT6-mediated H3 R2me2a modification enhances the promotion of NF-YCs on FT transcription in response to inductive LD signals.Moreover,AtPRMT6 and its homologues proteins AtPRMT4a and AtPRMT4b coordinately inhibit the expression of FLOWERING LOCUS C,a suppressor of FT.Taken together,our study reveals the role of arginine methylation in photoperiodic pathway and how the PRMT6-mediating H3R2me2a system interacts with NF-CO module to dynamically control FT expression and facilitate flowering time.Pingxian Zhang Xiulan Li Yifan Wang Weijun Guo Sadaruddin Chachar Adeel Riaz Yuke Geng Xiaofeng Gu Liwen Yang 2021aBIOTECH2021,2,4:1
