1Utilization of unmodified gold nanoparticles in colorimetric detection显示文摘This review begins with an overview of the appealing properties and various applications of gold nanoparticles, and briefly summarizes recent advances in using unmodified gold nanoparticles to detect different kinds of targets including nucleic acids, proteins, metal ions and small organic molecules. The key point to the unmodified gold nanoparticle-based visual detection assay is to control dispersion and aggregation of colloidal nanoparticles by targets of interest, which usually relies on affinities between gold nanoparticles and targets. The degree of dispersion or aggregation can be visualized through the change of the solution color or the precipitation of nanoparticles from the solution. Thus, the existence of the target molecules can be trans-lated into optical signals and monitored by the naked eye conveniently. 2Recent progress in the application of microfluidic systems and gold nanoparticles in immunoassays显示文摘Immunoassays are useful for many bioassays. Many new techniques and materials are introduced into the immunoassay to improve the efficiency. This paper reviews recent progress in the application of microfluidic systems and gold nanoparticles in immunoassay. The micro/nano technologies and materials can offer good sensitivity, fast detection, cost-effectiveness and easy signal readout. 3Characterization of H3 methylation in regulating oocyte development in cyprinid fish显示文摘Histone post-modifications are important epigenetic markers involved in multiple cellular processes via regulation of gene transcription or remodeling of chromatin structure. Oocyte development is a critical process under rigorous control to prevent the generation of aberrant gametes. However, the regulatory mechanism of oocyte early development is not well-understood due to the tiny size and poor distinguishability of the gonad in juvenile stages. Here, two cyprinid hybrid fishes, a sterile allotriploid fish and a gynogenetic hybrid fish with delayed oocyte development, provided research models to investigate the mechanisms involved. We used cytogenetic and molecular methods to confirm the pachytene arrest of oocytes in allotriploid fish and gynogenetic hybrid fish. On the basis of these developmental differences, we screened 21 different histone H3 modifications by ELISA and found that four modifications(H3 K4 me3, H3 K9 me3, H3 K79 me, and H3 K79 me3) differed significantly in the two cyprinid hybrid fishes. Changes in histone methylation at the three residues(H3 K4, K9, K79) were caused by specific methyltransferases and demethylases. 6Nucleic acids analysis显示文摘Nucleic acids are natural biopolymers of nucleotides that store, encode, transmit and express genetic information, which play central roles in diverse cellular events and diseases in living things. The analysis of nucleic acids and nucleic acids-based analysis have been widely applied in biological studies, clinical diagnosis, environmental analysis, food safety and forensic analysis.During the past decades, the field of nucleic acids analysis has been rapidly advancing with many technological breakthroughs.In this review, we focus on the methods developed for analyzing nucleic acids, nucleic acids-based analysis, device for nucleic acids analysis, and applications of nucleic acids analysis. The representative strategies for the development of new nucleic acids analysis in this field are summarized, and key advantages and possible limitations are discussed. 9An improved hybrid bream derived from a hybrid lineage of Megalobrama amblycephala(♀)×Culter alburnus(♂)显示文摘Distant hybridization is an important technique in fish genetic breeding.In this study,based on the establishment of an allodiploid fish lineage(BT,2n=48,F_(1)–F_(6))derived from distant hybridization between female Megalobrama amblycephala(BSB,2n=48)and male Culter alburnus(TC,2n=48),and the backcross progeny(BTB,2n=48)derived by backcrossing female F_(1) of BT to male BSB,an improved hybrid bream(BTBB,2n=48)was obtained by backcrossing BTB(♀)to BSB(♂).Moreover,the morphological and genetic characteristics of BTBB individuals were investigated;BTBB was similar to BSB in appearance but had a higher body height than BSB.The study results regarding chromosome numbers and DNA content indicated that BTBB is a diploid hybrid fish.The 5S rDNA and Hox gene of BTBB were inherited from the original parents.Gonadal development in BTBB was normal.On the other hand,BTBB had a faster growth rate,higher muscle protein level,and lower muscle carbohydrate level than BSB.Hence,bisexual fertile BTBB is promoted and can be applied as a high-quality fish,and it can also be used as a new fish germplasm resource to develop high-quality fish further.Thus,this study is of great significance for fish genetic breeding.Dingbin Gong Min Tao Lihui Xu Fangzhou Hu Zehong Wei Shi Wang Yude Wang Qingfeng Liu Chang Wu Kaikun Luo Chenchen Tang Rong Zhou Chun Zhang Yuequn Wang Shaojun Liu 2022Science China(Life Sciences)2022,65,6:0
10In-situ monitoring of nitrile-bearing pesticide residues by background-free surface-enhanced Raman spectroscopy显示文摘In-situ monitoring of pesticide residues during crop growth or/and in related products is of great significance in avoiding the abuse of pesticides but remains challenging thus far.In this report,we proposed a background-free surface-enhanced Raman spectroscopy(bf-SERS)platform to non-destructively track the nitrile-bearing pesticide residues in soybean leaves with high sensitivity and selectivity.The outstanding feature of the assay stems from the dramatic Raman enhancement effect of the 50 nm-sized gold nanoparticles(AuNPs)towards the pesticides and simultaneously the background-free Raman signal of the nitrile group in the so-called Raman-silent region(1800-2800 cm^(-1)).This bf-SERS assay was applied to evaluate the penetration effects of nitrile-bearing pesticides and monitor their residues in soybean leaves after rinsing with various solutions,providing a reliable tool for guiding the safe use of nitrile-bearing pesticides in agriculture.Guorui Wu Wenshuai Li Weijun Du Aiqin Yue Jinzhong Zhao Dingbin Liu 2022Chinese Chemical Letters2022,33,1:0
11Direct cytoplasm delivery of gold nanoparticles for real-time apoptosis detection显示文摘Gold nanoparticles(AuNPs)assembled with fluorescent peptides through Au-S bonds(pep-AuNPs)have been widely used in biomolecular detection.However,due to the endo/lysosomal trapping after the nanoprobes enter cells,the direct delivery of AuNP probes into the cytoplasm for real-time imaging remains a difficult barrier for many cytoplasm-targeting agents.Here,we prepare AuNP@gel by wrapping a multi-functional nanogel structure on the surface of a single AuNP probe by in-situ polymerization in order to directly deliver AuNP probes into the cell cytoplasm.Compared with the pep-AuNP probes,which are trapped inside lysosomes for long periods,the AuNP@gel probes use the proton-sponge effect to effectively disrupt endo/lysosomal membranes and remain in the cytoplasm.In addition,the AuNP@gel probes rapidly escape from endo/lysosomes to avoid the complex environment that interferes with the stability of the AuNP probes and the lysosomal-storage trigger the upregulation of oxidative stress into the cells.The nanogel structure enables the AuNP probes to avoid some detrimental effects and to achieve high-fidelity fluorescence signals in the cells.Compared to traditional strategies for lysosomal escape,this one-step in-situ polymerization procedure avoids the complicated modification of additional ligands and is generally applicable to peptide-,DNA-,and polymerlinked AuNP probes.Qiang Li Fengchao Wang Jie Yang Dingbin Liu 2020Nano Research2020,13,3:0

13Using selenium-conjugated polyethylene glycol to enhance the stability of gold nanoparticles in biologically relevant samples显示文摘Using stabilizing agents to maintain the physicochemical properties of colloids in complex environments is crucial for their realworld applications. In this article, we describe how selenium-(Se-) terminated polyethylene glycol(PEG) can serve as a highaffinity stabilizing agent for gold nanoparticles(AuNPs). Compared to Au NPs modified with standard thiolated PEG(S-PEG),Se-PEG-coated Au NPs are much more stable under extreme conditions such as high/low pH, high salt content, and high temperatures. We demonstrate that the Se anchor can prevent the dissociation of PEG ligands from Au NP surfaces in living cells,where a higher concentration of biothiols is usually present. 14Advanced technologies for single-cell in situ protein profiling显示文摘Cells are inherently heterogeneous to achieve a diverse spectrum of biological functions.To understand the underlying protein machinery that achieves these fascinating functions,it is important to develop advanced analytical methods that can profile proteins in their native environment,as protein expression,aggregation,degradation,and regulation define both normal physiology as well as pathogenicity.Genome and transcriptome sequencing have seen major advances at the single-cell levels,but comprehensive proteomic profiling is still challenging.The conventional proteomic methods,such as enzyme-linked immunosorbent assay(ELISA),western blot,and protein chips,can characterize biomarkers of interest.Still,these ensemble techniques are unsuitable for single-cell studies.Increasing evidence has shown the significance of in situ,sensitive,quantitative,and multiplexed profiling of biomarkers in single cells for diagnosis and treatment guidance.Here,we review the recent development of advanced imaging and spectroscopy techniques,including mass cytometry,immunofluorescence,and surfaceenhanced Raman spectrometry(SERS)for single-cell proteomic imaging.We also provide our view on the challenges and the outlook.Wen Zhou Xiaoyue Ni Chen Xie Quli Fan Dingbin Liu 2022Science China Chemistry2022,65,1:0
