1ASAF: altered spontaneous activity fingerprinting in Alzheimer’s disease based on multisite fMRI显示文摘Several monocentric studies have noted alterations in spontaneous brain activity in Alzheimer's disease (AD), although there is no consensus on the altered amplitude of low-frequency fluctuations in AD patients. The main aim of the present study was to identify a reliable and reproducible abnormal brain activity pattern in AD. The amplitude of local brain activity (AM), which can provide fast mapping of spontaneous brain activity across the whole brain, was evaluated based on multisite rs-fMRI data for 688 subjects (215 normal controls (NCs), 221 amnestic mild cognitive impairment (aMCI) 252 AD). Two-sample t-tests were used to detect group differences between AD patients and NCs from the same site. Differences in the AM maps were statistically analyzed via the Stouffer's meta-analysis. Consistent regions of lower spontaneous brain activity in the default mode network and increased activity in the bilateral hippocampus/parahippocampus, thalamus, caudate nucleus, orbital part of the middle frontal gyrus and left fusiform were observed in the AD patients compared with those in NCs. 2Ultra-strong and thermally stable nanocrystalline CrCoNi alloy显示文摘Grain refinement to the nanocrystalline regime is the most effective way to strengthen materials but this often deteriorates the grain-size thermal stability and plasticity. Here we manufactured a nanocrystalline face centred cubic Cr Co Ni medium entropy alloy with columnar grains via magnetron sputtering. Compression of CrCoNi pillars with diameters of ~1 μm revealed a record high yield strength of ~5 GPa for pillars with face centred cubic structures and engineering plastic strain of > 30%. The alloy possessed an outstanding grain-size thermal stability even at 1073 K. Both nanocrystalline grain size and a high density of nanotwins/stacking faults are critical to the exceptional yield strength. Deformation twinning, grains refinement during deformation, grain boundary sliding and random grain orientation all contribute to the large plasticity. 3Magnetic levitation using diamagnetism:Mechanism,applications and prospects显示文摘As a typical contact-free manipulation technique that removes friction and contamination risk,levitation has gradually become a preferred candidate for various applications.Magnetic levitation using diamagnetism,beyond Earnshaw’s theorem,is a kind of passive stable levitation that can be achieved at normal temperatures with no energy input.Appealingly,most seemingly nonmagnetic materials can be levitated in a magnetic field and can stabilize free levitation of magnetic materials.This review focuses on the fundamental principles of magnetic levitation using diamagnetism,with emphasis on its burgeoning applications.The theoretical basis associated with the magnetic levitation using diamagnetism is discussed by elucidating the characteristics of diamagnetic materials,and the key levitation mechanisms are clarified.Afterwards,state-of-the-art applications in various aspects,including sensing and measurement,actuating and micromanipulation,energy harvesting and magnetic gravity compensation,are summarized and compared.Finally,the review concludes with a brief outlook on future perspectives.GAO QiuHua YAN Han ZOU HongXiang LI WenBo PENG ZhiKe MENG Guang ZHANG WenMing 2021Science China(Technological Sciences)2021,64,1:3
4Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2显示文摘Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and glycolysis.Importantly, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. 5MAP3K2 augments Th1 cell differentiation via IL-18 to promote T cell-mediated colitis显示文摘T cell-mediated immunity in the intestine is stringently controlled to ensure proper immunity against pathogenic microbes and to prevent autoimmunity,a known cause of inflammatory bowel disease.However,precisely how T cells regulate intestine immunity remains to be fully understood.In this study,we found that mitogen-activated protein kinase kinase kinase 2(MAP3K2)is required for the CD4^(+)T cell-mediated inflammation in the intestine.Using a T cell transfer colitis model,we found that MAP3K2-deficient naïve CD4^(+)T cells had a dramatically reduced ability to induce colitis compared to wild type T cells.In addition,significantly fewer IFN-γ-but more IL-17A-producing CD4^(+)T cells in the intestines of mice receiving MAP3K2-deficient T cells than in those from mice receiving wild type T cells was observed.Interestingly,under well-defined in vitro differentiation conditions,MAP3K2-deficient naïve T cells were not impaired in their ability to differentiate into Th1,Th17 and Treg.Furthermore,the MAP3K2-regulated colitis severity was mediated by Th1 but not Th17 cells in the intestine.At the molecular level,we showed that MAP3K2-mediated Th1 cell differentiation in the intestine was regulated by IL-18 and required specific JNK activation.Together,our study reveals a novel regulatory role of MAP3K2 in intestinal T cell immunity via the IL-18-MAP3K2-JNK axis and may provide a novel target for intervention in T cell-mediated colitis.Ningbo Wu Dongping Chen Hongxiang Sun Jianmei Tan Yao Zhang Tianyu Zhang Yuheng Han Hongzhi Liu Xinxing Ouyang Xiao-Dong Yang Xiaoyin Niu Jie Zhong Zhengting Wang Bing Su 2021Science China(Life Sciences)2021,64,3:3

7Optimization Transmission Efficiency with Driver Intention for Automotive Continuously Variable Transmission under Slip Mode显示文摘This study proposes and experimentally validates an optimal integrated system to control the automotive continuously variable transmission(CVT)by Model Predictive Control(MPC)to achieve its expected transmission efficiency range.The control system framework consists of top and bottom layers.In the top layer,a driving intention recognition system is designed on the basis of fuzzy control strategy to determine the relationship between the driver intention and CVT target ratio at the corresponding time.In the bottom layer,a new slip state dynamic equation is obtained considering slip characteristics and its related constraints,and a clamping force bench is established.Innovatively,a joint controller based on model predictive control(MPC)is designed taking internal combustion engine torque and slip between the metal belt and pulley as optimization dual targets.A cycle is attained by solving the optimization target to achieve optimum engine torque and the input slip in real-time.Moreover,the new controller provides good robustness.Finally,performance is tested by actual CVT vehicles.Results show that compared with traditional control,the proposed control improves vehicle transmission efficiency by approximately 9.12%-9.35%with high accuracy.Ling Han Hui Zhang Ruoyu Fang Hongxiang Liu 2021Chinese Journal of Mechanical Engineering2021,34,5:1

12Unveiling the grain boundary-related effects on the incipient plasticity and dislocation behavior in nanocrystalline CrCoNi medium-entropy alloy显示文摘The incipient plasticity and dislocation behavior in a nanocrystalline(NC)CrCoNi medium-entropy alloy were systematically investigated in terms of pop-in events during instrumental nano-indentation tests.Quantitative statistical analysis and molecular dynamic simulations were performed to reveal the effects of grain boundaries(GBs)on initial stages of plastic deformation.Multiple pop-in events appeared during loading on the NC CrCoNi.The first pop-in that represents the initial yielding was identified to be controlled by dislocation nucleation,which is in sharp contrast to the continuous elastic-plastic transition mediated by GB mechanisms in NC pure metals.This can be attributed to the sluggish kinetics of the chemically complex GBs(CCGBs)in the NC CrCoNi that hinders diffusive GB activities but facilitates dislocation nucleation.Subsequent pop-ins were also found to be closely related to the extra dragging effects imposed by the CCGBs on dislocation propagation in the NC alloy.Moreover,the extremely small grain sizes and the consequent high-volume fraction of GBs in the NC alloy severely restrict the lengths of dislocation source and the radii of dislocation loop,giving rise to a higher critical stress,smaller activation volume and lower pop-in width as compared with its coarse-grained counterpart.These results provide new insights into the onset of nano-plasticity in concentrated multi-principal element alloys.Shuo Sun Yang Yang Chenxu Han Guixun Sun Yan Chen Hongxiang Zong Jiangjiang Hu Shuang Han Xiaozhou Liao Xiangdong Ding Jianshe Lian 2022Journal of Materials Science & Technology2022,,32:0

14Lipid biomarker composition in surface sediments from the Carlsberg Ridge near the Tianxiu Hydrothermal Field显示文摘Hydrothermal venting has a profound effect on the chemical and biological properties of local and distal seawater and sediments. In this study, lipid biomarkers were analyzed to examine the potential influence of hydrothermal activity on the fate of organic matter(OM) in surface sediments around Tianxiu Hydrothermal Field in the Carlsberg Ridge(CR), Northwest Indian Ocean. By comparing the biomarker distributions of the samples with that of other typical hydrothermal sediments in the mid ocean ridge, it is shown that the location of the samples is not affected by the hydrothermal activity. The relatively low abundances of terrestrial n-alkyl lipids and riverine1,15-C_(32) diol suggested a minor contribution of terrigenous OM to the study area. The bacteria contributed predominantly to sedimentary marine OM;however, other marine source organisms, e.g., eukaryotes(i.e.,phytoplankton and fungi) could not be completely neglected. The marine-originated biomarkers showed significantly variable distributions between the two sediments, suggesting different dynamic physical and biogeochemical processes controlling the fate of marine OM. 15scRNA-seq profiling of neonatal and adult thymus-derived CD4+ T cells by a T cell origin-time tracing model显示文摘It is well documented that the neonatal thymus-derived (neonatal-TD) regulatory T cells (Treg) are essential to prevent lethal autoimmune diseases and allergies, and neonatal and adult thymus possesses distinct output potentials for naïve T cells, including Treg. However, the molecular features and detailed functional differences between neonatal-TD and adult thymus-derived (adult-TD) T cells in terms of their ability to maintain immune homeostasis during long-term environmental influences are still largely unknown, partially due to the lack of appropriate animal models to precisely trace these cells at specific time points. In this study, neonatal-TD and adult-TD CD4+ T cells from the spleen and Peyer's patches were traced for 9 weeks by a T cell origin-time tracing mouse model and analysed by single-cell RNA sequencing. More Treg but fewer naïve T cells were found in neonatal-TD CD4+ T cells from both tissues than those from adult-TD counterparts. Interestingly, the neonatal-TD Treg in both the spleen and Peyer's patches exhibited augmented expression of Foxp3, Gata3, Ctla4, Icos, Il2ra, Tgfb1, and Nrp1, as well as enriched Gene Ontology terms like T cell activation and tolerance induction, indicating an enhanced immunosuppressive function. These results were further confirmed by flow cytometry analysis and in vitro immune suppression assays. Flow cytometry also revealed a significantly higher proportion of neonatal-TD Treg in total Treg than that of adult-TD counterparts, suggesting the longer lifespan of neonatal-TD Treg. To investigate the intrinsic features of neonatal-TD and adult-TD CD4+ T cells, a shortened tracing time was performed. Surprisingly, the neonatal-TD and adult-TD CD4+ T cells had similar proportions of Treg and did not exhibit significant differences in Foxp3, Gata3, Ctla4, Icos, Il2ra, and Tgfb1 expression levels after tracing for 12 days. On the other hand, neonatal-TD Treg present an increased Nrp1 expression level compared with adult-TD counterparts, indicating the enhanced stability. 16Crystals Array via Oriented Nucleation and Growth Induced by Smectic E Mesophase of C7-T-BTBT显示文摘Long-range order crystalline thin films of organic semiconductors have attracted wide attention owing to their high charge carrier mobility.However,uncontrolled crystal nucleation and growth during the thin film drying process cause the formation of grain boundaries,thereby limiting the long-range order.Herein,we achieved the oriented nucleation and growth of organic semiconductors by off-centre spin-coating at the temperature of the smectic E(SmE)liquid crystal mesophase,and then followed by Ostwald ripening during solvent vapour annealing.The thin film of 2-(5-heptylthiophen-2-yl)[1]benzothieno[3,2-b][1]benzothiophne(C7-T-BTBT)blended with 40%(mass fraction)poly(methyl methacrylate)(PMMA)was prepared by off-centrespin-coating at SmE mesophase(170°C),followed by solvent vapour annealing in chloroform for 24 h(chloroform is a good solvent for C7-T-BTBT and PMMA).The C7-T-BTBT molecules grew to rod-like crystals,which were mostly arranged parallel to each other.The crystal growth was perfect and resulted in a single crystal.The average length of the crystals was approximately 87µm.Moreover,the highest charge carrier mobility is 1.62 cm^(2)·V^(−1)·s^(−1) as against that of the film prepared at 25°C(0.06 cm^(2)·V^(−1)·s^(−1)).LI Hongxiang WANG Sichun LIU Xinyu WU Fan ZHANG Qiang YUAN Jian MA Wei HAN Yanchun 2022Chemical Research in Chinese Universities2022,38,4:0
17Regulation of CD8^(+)T memory and exhaustion by the mTOR signals显示文摘CD8^(+)T cells are the key executioners of the adaptive immune arm,which mediates antitumor and antiviral immunity.Naïve CD8^(+)T cells develop in the thymus and are quickly activated in the periphery after encountering a cognate antigen,which induces these cells to proliferate and differentiate into effector cells that fight the initial infection.Simultaneously,a fraction of these cells become long-lived memory CD8^(+)T cells that combat future infections.Notably,the generation and maintenance of memory cells is profoundly affected by various in vivo conditions,such as the mode of primary activation(e.g.,acute vs.chronic immunization)or fluctuations in host metabolic,inflammatory,or aging factors.Therefore,many T cells may be lost or become exhausted and no longer functional.Complicated intracellular signaling pathways,transcription factors,epigenetic modifications,and metabolic processes are involved in this process.Therefore,understanding the cellular and molecular basis for the generation and fate of memory and exhausted CD8^(+)cells is central for harnessing cellular immunity.In this review,we focus on mammalian target of rapamycin(mTOR),particularly signaling mediated by mTOR complex(mTORC)2 in memory and exhausted CD8^(+)T cells at the molecular level.Yao Chen Ziyang Xu Hongxiang Sun Xinxing Ouyang Yuheng Han Haihui Yu Ningbo Wu Yiting Xie Bing Su 2023Cellular & Molecular Immunology2023,20,9:0
