2Tissue level microstructure and mechanical properties of the femoral head in the proximal femur of fracture patients显示文摘This study aims to investigate the regional variations of trabecular morphological parameters and mechanical parameters of the femoral head,as well as to determine the relationship between trabecular morphological and mechanical parameters.Seven femoral heads from patients with fractured proximal femur were scanned using a micro-CT system.Each femoral head was divided into 12 sub-regions according to the trabecular orientation.One 125 mm^3 trabecular cubic model was reconstructed from each sub-region.A total of 81 trabecular models were reconstructed,except three destroyed sub-regions from two femoral heads during the surgery.Trabecular morphological parameters,i.e.trabecular separation(Tb.Sp),trabecular thickness(Tb.Th),specific bone surface(BS/B V),bone volume fraction(BV/TV),structural model index(SMI),and degree of anisotropy(DA) were measured.Micro-finite element analyses were performed for each cube to obtain the apparent Young's modulus and tissue level von Mises stress distribution under 1%compressive strain along three orthogonal directions,respectively.Results revealed significant regional variations in the morphological parameters(P<0.05).Young's moduli along the trabecular orientation were significantly higher than those along the other two directions.In general,trabecular mechanical properties in the medial region were lower than those in the lateral region.Trabecular mechanical parameters along the trabecular orientation were significantly correlated with BS/BV,BV/TV,Tb.Th,and DA.In this study,regional variations of microstructural features and mechanical properties in the femoral head of patients with proximal femur fracture were thoroughly investigated at the tissue level.The results of this study will help to elucidate the mechanism of femoral head fracture for reducing fracture risk and developing treatment strategies for the elderly.Linwei Lü Guangwei Meng He Gong Dong Zhu Jiazi Gao Yubo Fan 2015Acta Mechanica Sinica2015,31,2:4

4Investigations into the perplexing interrelationship of the Genus Takifugu Abe, 1949 (Tetraodontiformes, Tetraodontidae)显示文摘The phylogenetic relationships within the genus Takifugu Abe, 1949 (Tetraodontiformes, Tetraodonti- dae) remain unresolved. Because of the use of Takifugu as model organisms, the resolution of these relationships is crucial for the interpretation of evolutionary trends in biology. Pufferfishes of this ge- nus are comprised of a comparatively small number of species and are mainly distributed along the coastal region of the western part of the Sea of Japan and the coastline of China. Mitochondrial gene sequences were employed to test the phylogenetic hypotheses within the genus. Seventeen species of the genus were examined. Molecular phylogenetic trees were constructed using the maximum parsi- mony, neighbor-joining, maximum likelihood and Bayesian methods. Our hypothesis of internal relationships within the genus differs from previous hypotheses. Our results indicate that (1) the genus Takifugu is a monophyletic assemblage; (2) the genus is divided into 6 subgroups based on the mo- lecular data; and (3) there is low genetic diversity among the species within this genus. In addition, speciation within Takifugu appears to be driven by hybridization and isolation by distribution. 5Global transcriptomic analysis reveals Lnc-ADAMTS9 exerting an essential role in myogenesis through modulating the ERK signaling pathway显示文摘Background:Long non-coding RNAs(lncRNAs)are emerging key regulators involved in a variety of biological processes such as cell differentiation and development.The balance between myogenesis and adipogenesis is crucial for skeletal muscle homeostasis in humans and meat quality in farm animals.The present study aimed to reveal the global transcriptomic profiles of adipogenic(Adi-)and myogenic(Myo-)precursors derived from porcine skeletal muscle and identify lncRNAs involved in the modulation of myogenesis homeostasis in porcine skeletal muscle.Results:In this study,a total of 655 novel individual lncRNAs including differentially expressed 24 lncRNAs,and 755 differentially expressed mRNAs were identified(fold change≥2 or≤0.5 and adjusted P<0.05).Integrated results of Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis accompanied by the variation of intracellular Ca2+concentration highlighted Lnc-ADAMTS9 involved in the modulation of myogenesis homeostasis in porcine skeletal muscle.Although Lnc-ADAMTS9 knock-down did not alter the mRNA expression of ADAMTS9,we demonstrated that Lnc-ADAMTS9 can promote myogenic proliferation and myogenic differentiation of myogenic precursors through inhibiting the ERK/MAPK signaling pathway.Conclusion:We deciphered a comprehensive catalog of mRNAs and lncRNAs that might be involved in the regulation of myogenesis and adipogenesis homeostasis in the skeletal muscle of pigs.The Lnc-ADAMTS9 exerts an essential role in myogenesis through the ERK signaling pathway.Liqi Wang Ting He Xin Zhang Yubo Wang Kai Qiu Ning Jiao Linjuan He Jingdong Yin 2021Journal of Animal Science and Biotechnology2021,12,2:3
6A point-based rendering approach for real-time interaction on mobile devices显示文摘Mobile device is an important interactive platform. Due to the limitation of computation, memory, display area and energy, how to realize the efficient and real-time interaction of 3D models based on mobile devices is an important research topic. Considering features of mobile devices, this paper adopts remote rendering mode and point models, and then, proposes a transmission and rendering approach that could interact in real time. First, improved simplification algorithm based on MLS and display resolution of mobile devices is proposed. Then, a hierarchy selection of point models and a QoS transmission control strategy are given based on interest area of operator, interest degree of object in the virtual environment and rendering error. They can save the energy consumption. Finally, the rendering and interaction of point models are completed on mobile devices. 7Age- and direction-related adaptations of lumbar vertebral trabecular bone with respect to apparent stiffness and tissue level stress distribution显示文摘The objective of this study was to study theage-related adaptation of lumbar vertebral trabecular bone atthe apparent level,as well as the tissue level in three orthog-onal directions.Ninety trabecular specimens were obtainedfrom six normal L4 vertebral bodies of six male cadavers intwo age groups,three aged 62 years and three aged 69 years,and were scanned using a high-resolution micro-computedtomography(micro-CT)system,then converted to micro-finite element models to do micro-finite element analyses.The relationship between apparent stiffness and bone volumefraction,and the tissue level von Mises stress distribution foreach trabecular specimen when compressed separately in thelongitudinal direction,medial-lateral and anterior-posteriordirections(transverse directions)were derived and comparedbetween two age groups.The results showed that at the appar-ent level,trabecular bones from 69-year group had stifferbone structure relative to their volume fractions in all threedirections,and in both age groups,changes in bone volume fraction could explain more variations in apparent stiffnessin the longitudinal direction than the transverse directions;at the tissue level,aging had little effect on the tissue yonMises stress distributions for the compressions in all thethree directions.The novelty of the present study was that itprovided quantitative assessments on the age and direction-related adaptation of Chinese male lumbar vertebral trabec-ular bone from two different levels:stiffness at the apparentlevel and stress distribution at the tissue level.It may helpto understand the failure mechanisms and fracture risks ofvertebral body associated with aging and direction for theprevention of fracture risks in elder individuals.He Gong Yubo Fan Ming Zhang Ling Qin 2009Acta Mechanica Sinica2009,25,1:2
8Comprehensive optimization design of aerodynamic and electromagnetic scattering characteristics of serpentine nozzle显示文摘Comprehensive optimization design of serpentine nozzle with trapezoidal outlet was studied to improve its aerodynamic and electromagnetic scattering performance.Serpentine nozzles with different center offsets and different ratios of the bases of the trapezoidal outlet were generated based on curvature control regulation.Computational Fluid Dynamics(CFD)simulations have been conducted to obtain the flow field in the nozzle,and Forward-Backward Iterative Physical Optics(FBIPO)method was applied to study the electromagnetic scattering characteristics of the nozzle.Guarantee Convergence Particle Swarm Optimization(GCPSO)algorithm based on Radial Basis Function(RBF)neural network was used to optimize the geometry of the nozzle in consideration of its aerodynamic and electromagnetic scattering characteristics.The results show that the GCPSO method based on RBF can be used to optimize the aerodynamic characteristics of the internal flow and the scattering characteristics of the cavity of the serpentine nozzle with irregular outlet.The optimized model has a higher center offset and a lower ratio of the bases of the trapezoidal outlet after optimization compared to the original model.The optimized model leads to a slight change in aerodynamic performance,with a total pressure recovery coefficient increase of 0.31%and a discharge coefficient increase of 0.41%.In addition,the Radar Cross Section(RCS)decreases also by around 83.33%and the overall performance is significantly improved,with a decrease of the optimized objective function by around 38.74%.Yubo HE Qingzhen YANG Xiang GAO 2021Chinese Journal of Aeronautics2021,34,3:2

10Accelerated discovery of a large family of quaternary chalcogenides with very low lattice thermal conductivity显示文摘The development of efficient thermal energy management devices such as thermoelectrics and barrier coatings often relies on compounds having low lattice thermal conductivity(κl).Here,we present the computational discovery of a large family of 628 thermodynamically stable quaternary chalcogenides,AMM′Q_(3)(A=alkali/alkaline earth/post-transition metals;M/M′=transition metals,lanthanides;Q=chalcogens)using high-throughput density functional theory(DFT)calculations.We validate the presence of lowκl in these materials by calculatingκl of several predicted stable compounds using the Peierls–Boltzmann transport equation.Our analysis reveals that the lowκl originates from the presence of either a strong lattice anharmonicity that enhances the phononscatterings or rattler cations that lead to multiple scattering channels in their crystal structures.Our thermoelectric calculations indicate that some of the predicted semiconductors may possess high energy conversion efficiency with their figure-of-merits exceeding 1 near 600 K.Our predictions suggest experimental research opportunities in the synthesis and characterization of these stable,low κ_(l) compounds.Koushik Pal Yi Xia Jiahong Shen Jiangang He Yubo Luo Mercouri G.Kanatzidis Chris Wolverton 2021npj Computational Materials2021,,1:1

13Arginine promotes myogenic differentiation and myotube formation through the elevation of cytoplasmic calcium concentration显示文摘This study aimed to explore the mechanism underlying arginine-promoted myogenesis of myoblasts.C2C12 cells were cultured with a medium containing 0.1,0.4,0.8,or 1.2 mmol/L arginine,respectively.Cell proliferation,viability,differentiation indexes,cytoplasmic Ca^(2+)concentration,and relative mRNA expression levels of myogenic regulatory factors(MRF)and key Ca2+channels were measured in the absence or presence of 2 chemical inhibitors,dantrolene(DAN,10μmol/L)and nisoldipine(NIS,10μmol/L),respectively.Results demonstrated that arginine promoted myogenic differentiation and myotube formation.Compared with the control(0.4 mmol/L arginine),1.2 mmol/L arginine upregulated the relative mRNA expression levels of myogenin(MyoG)and Myomaker at d 2 during myogenic induction(P<0.05).Cytoplasmic Ca^(2+)concentrations were significantly elevated by arginine supplementation at d 2 and 4(P<0.05).Relative mRNA expression levels of Ca^(2+)channels including the type 1 ryanodine recepto r(RyR1)and voltage-gated Ca^(2+)channel(Cav1.1)were upregulated by 1.2 mmol/L arginine during2-d myogenic induction(P<0.01).However,arginine-promoted myogenic potential of myoblasts was remarkably compromised by DAN and NIS,respectively(P<0.05).These findings evidenced that the supplementation of arginine promoted myogenic differentiation and myotube formation through increasing cytoplasmic Ca^(2+)concentration from both extracellular and sarcoplasmic reticulum Ca^(2+).Lu Gong Xin Zhang Kai Qiu Linjuan He Yubo Wang Jingdong Yin 2021Animal Nutrition2021,,4:1
14Dietary supplementation of laminarin improves the reproductive performance of sows and the growth of suckling piglets显示文摘Background Maternal nutrition is essential in keeping a highly efficient production system in the pig industry.Laminarin has been shown to improve antioxidant capacity,reduce the inflammatory response,and favor the homeostasis of intestinal microbiota.However,the effect of dietary supplementation of laminarin on the reproductive performance of sows and the growth of suckling offspring remains unknown.Methods A total of 40 Landrace×Yorkshire multiparous sows on d 85 of gestation,similar in age,body weight(BW),parity and reproductive performance,were randomly divided into four dietary treatments with 10 sows per treatment,receiving a control diet(basal pregnancy or lactating diets)and a basal diet supplemented with 0.025%,0.05%and 0.10%laminarin,respectively.The experiment lasted from d 85 of gestation to d 21 of lactation.Results Laminarin supplementation linearly increased number born alive per litter(P=0.03),average daily feed intake(ADFI,P<0.01),and total milk yield of sows during the lactation of 1–21 d(P=0.02).Furthermore,maternal laminarin supplementation increased the average daily gain(ADG)of piglets while tending to reduce the culling and death rate before weaning.In addition,alterations to the composition of colostrum and milk,as well as to serum inflammatory cytokines and immunoglobulins of sows were observed.The fecal microbiota profile of sows supported the improvement of reproductive performance in sows and the growth performance in suckling offspring.Conclusions Dietary supplementation of laminarin during late pregnancy and lactation could significantly improve reproductive performance of sows and growth performance of piglets.Pengguang Zhang Guoyuan Jiang Chenghong Ma Yubo Wang Enfa Yan Linjuan He Jianxin Guo Xin Zhang Jingdong Yin 2024Journal of Animal Science and Biotechnology2024,15,1:0
15Ryanodine receptor RyR1-mediated elevation of Ca^(2+)concentration is required for the late stage of myogenic differentiation and fusion显示文摘Background:Cytosolic Ca^(2+)plays vital roles in myogenesis and muscle development.As a major Ca^(2+)release channel of endoplasmic reticulum(ER),ryanodine receptor 1(RyR1)key mutations are main causes of severe congenital myopathies.The role of RyR1 in myogenic differentiation has attracted intense research interest but remains unclear.Results:In the present study,both RyR1-knockdown myoblasts and CRISPR/Cas9-based RyR1-knockout myoblasts were employed to explore the role of RyR1 in myogenic differentiation,myotube formation as well as the potential mechanism of RyR1-related myopathies.We observed that RyR1 expression was dramatically increased during the late stage of myogenic differentiation,accompanied by significantly elevated cytoplasmic Ca^(2+)concentration.Inhibition of RyR1 by siRNA-mediated knockdown or chemical inhibitor,dantrolene,significantly reduced cytosolic Ca^(2+)and blocked multinucleated myotube formation.The elevation of cytoplasmic Ca^(2+)concentration can effectively relieve myogenic differentiation stagnation by RyR1 inhibition,demonstrating that RyR1 modulates myogenic differentiation via regulation of Ca^(2+)release channel.However,RyR1-knockout-induced Ca^(2+)leakage led to the severe ER stress and excessive unfolded protein response,and drove myoblasts into apoptosis.Conclusions:Therefore,we concluded that Ca^(2+)release mediated by dramatic increase in RyR1 expression is required for the late stage of myogenic differentiation and fusion.This study contributes to a novel understanding of the role of RyR1 in myogenic differentiation and related congenital myopathies,and provides a potential target for regulation of muscle characteristics and meat quality.Kai Qiu Yubo Wang Doudou Xu Linjuan He Xin Zhang Enfa Yan Lu Wang Jingdong Yin 2022Journal of Animal Science and Biotechnology2022,13,4:0
16Spike protein-based epitopes predicted against SARS-CoV-2 through literature mining显示文摘Background:With the diffusion of SARS-CoV-2 around the world,human health is being threatened.As there is no effective vaccine yet,the development of the vaccine is urgently in progress.Materials and methods:Immunoinformatics methods were applied to predict epitopes from the Spike protein through mining literature associated with B-and T-cell epitopes prediction published or preprinted since the outbreak of the virus till June 1,2020.3D structure of the Spike protein were obtained(PDB ID:6VSB)for prediction of discontinuous B-cell epitopes and localization of epitopes in the hotspot regions.Results:Methods provided by the Immune Epitope Database(IEDB)server were the most frequently used to predict epitopes.Sequence alignment of the epitopes extracted from literature with the Spike protein demonstrated that the epitopes in different studies converged to multiple short hotspot regions.There were three hotspot regions found in RBD of the Spike protein harboring B-cell linear epitopes(‘RQIAPGQTGKIADYNYKLPD’,‘SYGFQPTNGVGYQ’and‘YAWNRKRISNCVA’)predicted to have high antigenicity score.Two T-cell epitopes(‘KPFERDISTEIYQ’and‘NYNYLYRLFR’)predicted to be highly antigenic in the original studies were discovered in the hotspot region.Toxicity and allergenicity analysis confirmed all the five epitopes are of non-toxin,and four of them are of non-allergen.The five epitopes identified in hotspot regions of RBD were found fully exposed based on the 3D structure of the Spike protein.Conclusion:The five epitopes we discovered from literature mining may be potential candidates for diagnostics and vaccine development against SARS-CoV-2.Wendong Li Lin Li Ting Sun Yufei He Guang Liu Zixuan Xiao Yubo Fan Jing Zhang 2020Medicine in Novel Technology and Devices2020,,4:0
17Dietary guanidinoacetic acid supplementation improves water holding capacity and lowers free amino acid concentration of fresh meat in finishing pigs fed with various dietary protein levels显示文摘The current study was carried out to detect the effect of dietary guanidinoacetic acid(GAA)supplementation on carcass characteristics and meat quality in finishing pigs fed different dietary crude protein(CP)levels.Sixty-four barrows with an initial body weight of 73.05±2.34 kg were randomly allocated into 1 of 4 dietary treatments in a 2(100%vs.125%NRC CP level)×2(0 vs.300 mg/kg GAA)factorial arrangement(n=7).The feeding trial lasted for 49 d.GAA supplementation significantly reduced drip loss(P=0.01),free water distribution(T23peak area ratio)(P=0.05)and the concentrations of free alanine,threonine,methionine and isoleucine(P<0.05);but increased total glycine content(P=0.03)in the longissimus dorsi muscle of finishing pigs regardless of the dietary CP levels.Furthermore,primary myogenic cell differentiation system was employed to investigate the influence of inclusion of GAA on free amino acid concentrations in myotubes(n=4)and validate the finding in the animal feeding trial.We found that GAA inclusion in culture medium also decreased intracellular concentrations of free alanine,threonine,methionine,isoleucine,valine and proline in differentiated primary myogenic cells in vitro(P<0.05).Meanwhile,relative to diets with 100%NRC CP level,the intake of diets with 125%NRC CP level improved sarcoplasmic protein solubility,increased the contents of carnosine and total free amino acids as well as flavor amino acids in the longissimus dorsi muscle and decreased backfat thickness at the 6-7th ribs in pigs(P<0.05).In addition,we observed that the impact of dietary GAA supplementation on the last rib fat thickness,shear force,and free lysine content in the longissimus dorsi muscle was dependent on dietary CP levels(P<0.05).Collectively,dietary GAA supplementation can reduce drip loss,decrease the concentrations of free amino acids and flavor amino acids of fresh meat independent of dietary CP levels.Lu Wang Yubo Wang Doudou Xu Linjuan He Xiaoyan Zhu Jingdong Yin 2022Animal Nutrition2022,,4:0
18Low BMI patients with advanced EGFR mutation-positive NSCLC can get a better outcome from metformin plus EGFR-TKI as first-line therapy:A secondary analysis of a phase 2 randomized clinical trial显示文摘Background:The synergistic association between metformin and epidermal growth factor receptor(EGFR)-tyrosine kinase inhibitors(TKIs)has been confirmed in in vitro studies.It is still controversial which patients can benefit from metformin plus EGFR-TKIs treatment.Body mass index(BMI)was proved to be independently associated with prolonged progression-free survival(PFS)and overall survival(OS).This study aimed to in-vestigate whether BMI is associated with the synergistic effect of metformin and EGFR-TKIs in advanced EGFR mutation(EGFR m)-positive non-small cell lung cancer(NSCLC)among nondiabetic Asian population.Methods:We performed a post hoc analysis of a prospective,double-blind phase II randomized clinical trial(COAST,NCT01864681),which enrolled 224 patients without diabetes with treatment-naïve stage IIIB-IV EGFR m NSCLC.We stratified patients into those with a high BMI(≥24 kg/m^(2))and those with a low BMI(<24 kg/m^(2))to allow an analysis of the difference in PFS and OS between the two groups.The PFS and OS were analyzed using Kaplan-Meier curves,and the differences between groups were compared using log-rank test.Results:In the univariate analysis,patients who had a high BMI(n=56)in the gefitinib+metformin group(n=28)did not have a better PFS(8.84 months vs.11.67 months;P=0.351)or OS(15.58 months vs.24.36 months;P=0.095)than those in the gefitinib+placebo group(n=28).Similar results were also observed in the low-BMI groups.Strikingly,in the metformin plus gefitinib group,patients who had low BMI(n=69)showed significantly better OS than those with high BMI(24.89 months[95%CI,20.68 months-not reached]vs.15.58 months[95%CI,13.78-31.53 months];P=0.007),but this difference was not observed in PFS(10.78 months vs.8.84 months;P=0.285).Conclusions:Our study showed that nondiabetic Asian advanced NSCLC patients with EGFR mutations who have low BMI seem to get better OS from metformin plus EGFR-TKI treatment.Rui Han Jianghua Li Yubo Wang Tingting He Jie Zheng Yong He 2023Chinese Medical Journal Pulmonary and Critical Care Medicine2023,1,2:0
19Detection of Malicious PDF Files Using a Two-Stage Machine Learning Algorithm显示文摘Portable document format(PDF)files are increasingly used to launch cyberattacks due to their popularity and increasing number of vulnerabilities.Many solutions have been developed to detect malicious files,but their accuracy decreases rapidly in face of new evasion techniques.We explore how to improve the robustness of classifiers for detecting adversarial attacks in PDF files.Content replacement and the n-gram are implemented to extract robust features using proposed guiding principles.In the two-stage machine learning model,the objects are divided based on their types,and the anomaly detection model is first trained for each type individually.The former detection results are organized into tree-like information structure and treated as inputs to convolutional neural network.Experimental results show that the accuracy of our classifier is nearly 100% and the robustness against evasive samples is excellent.The object features also enable the identification of different vulnerabilities exploited in malicious PDF files.HE Kang ZHU Yuefei HE Yubo LIU Long LU Bin LIN Wei 2020Chinese Journal of Electronics2020,29,6:0
