| 1 | New mutation points in 23S rRNA gene associated with Helicobacter pyloriresistance to clarithromycin in northeast China显示文摘AIM: TO investigate the resistance rate of Helicobacter pylori(H pylori) to clarithromycin, metronidazole, amoxicillin and tetracycline to guide clinical practice, and to study the mechanism of Hpylori resistant to clarithromycin. METHODS: Thirty Hpyloristrains were isolated from the mucosa of peptic ulcer, gastric tumor and chronic gastritis patients, then the minimal inhibitory concentration (MIC) to clarithromycin, metronidazole, amoxicillin and tetracycline was evaluated by E-test method. The sequence analysis of PCR fragments was conducted in 23S rRNA gene of H pylori resistant to clarithromycin to get the resistance mechanism of the bacteria. RESULTS: Among 30 Hpyloristrains, 7 cases were resistant to clarithromycin, 12 to metronidazole, 2 to tetracycline and no strain was found to be resistant to amoxicillin. The resistance rates were 23.3%, 40%, 6.7% and 0%, respectively. Three new mutation points were found to be related to the clarithromycin resistance in Hpyloriisolates, which were G2224A, C2245T and T2289C. CONCLUSION: In northeast China, H pylori shows high resistance to metronidazole, while sensitive to amoxicillin. | 2 | Chromic-P32 phosphate treatment of implanted pancreatic carcinoma: Mechanism involved显示文摘AIM: To study the effects of chromic-P32 phosphate (32p colloids) interstitial administration in Pc-3 implanted pancreatic carcinoma, and investigate its anticancer mechanism.METHODS: Ninety-eight tumor bearing nude mice werekilled at different time points after the injection of 32Pcolloids to the tumor core with observed radioactivity. The light microscopy, transmission electron microscopy (TEM) and immuno-histochemistry and flow cytometry were used to study the rates of tumor cell necrosis, proliferating cell nuclear antigen index, the micro vessel density (MVD). The changes of the biological response to the lymphatic transported 32p colloids in the inguinal lymph node (ILN) were dynamically observed, and the percentage of tumor cell apoptosis, and Apo2.7, caspase-3, Bcl-2, Baxrelated gene expression were observed too.RESULTS: The half-life of effective medication is 13 dafter injection of 32P colloids to the tumor stroma, in 1-6groups, the tumor cell necrosis rates were 20%, 45%,65%, 70%, 95% and 4%, respectively (F= 4.14-105.36, P<0.01). MVD were 38.5±4.0, 28.0±2.9, 17.0±2.9, 8.8±1.5,5.7±2.3 and 65.0±5.2 (t= 11.9-26.1, P<0.01), respectively.Under TEM fairly differentiated Pc-3 cells were found. Thirty days after medication, tumors were shrunk and dried with scabs detached, and those in control group increased in size prominently with plenty of hypodermic blood vessels. In all animals the ILN were enlarged but in medicated animals they appeared later and smaller than those in control group. The extent of irradiative injury in ILN was positively correlated to the dosage of medication. Typical tumor cell apoptosis could be found under TEM inanimals with intra-tumoral injection of low dosed 32P colloids. The peak of apoptosis occurred in 2.96 MBq group and 24 h after irradiation. In the course of irradiationinduced apoptosis, the value of Bcl-2/Bax was down regulated; Apo2.7 and caspase-3 protein expression were prominently increased dose dependently. CONCLUSION: 32p colloids intra-tumor injection having prominent anticancer effectiveness may reveal the ability of promoting cell differentiation. | 3 | Preparation of magnetic polybutylcyanoacrylate nanospheres encapsulated with aclacinomycin A and its effect on gastric tumor显示文摘AIM: To evaluate the effect of aclacinomycin A-loaded magnetic polybutylcyanoacrylate nanoparticles on gastric tumor growth in vivo and in vitro.METHODS: Magnetic polybutylcyanoacrylate (PBCA)nanospheres encapsulated with aclacinomycin A (MPNS-ACM) were prepared by interracial polymerization. Particle size, shape and drug content were examined. Female BABL/c nude mice were implanted with MKN-45 gastric carcinoma tissues subcutaneously to establish human gastric carcinoma model. The mice were randomly divided into 5 groups of 6 each: ACM group (8 mg/kg bin); group of high dosage of MPNS-ACM (8 mg/kg bin); group of low dosage of MPNS-ACM (1.6 mg/kg bin); group of magnetic PBCA nanosphere (MPNS) and control group(normal saline). Magnets (2.5 T) were implanted into the tumor masses in all of the mice one day before the therapy.Above-mentioned drugs were administered intravenously to the mice of every group on the first day and sixth day.When the mice were sacrificed, tumor weight was measured, and the assay of granulocyte- macrophage colony forming-unit (CFU-GM) was performed on semi-solid culture. White blood cell, alanine aminotransferase and creatine were examined. 3-[4-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) was used to examine the viability of MKN-45 cells afger incubation with different concentrations of ACM, MPNS and MPNS-ACM suspension respectively for 48 h.RESULTS: Content of ACM in MPNS-ACM was 12.0% and the average diameter of the particles was 210 nm. The inhibitory rates of ACM (8 mg/kg bin), high dosage of MPNS-ACM (8 mg/kg bin), low dosage of MPNS-ACM (1.6 mg/kg bin)and MPNS on human gastric carcinoma in nude mice were 22.63%, 52.55%, 30.66% and 10.22%, respectively. There was a significant decrease in the number of CFU-GM of bone marrow in ACM group compared with control group,whereas no obvious change was observed in that of the nanosphere groups. | 4 | Prehistoric agriculture development in the Yunnan-Guizhou Plateau, southwest China: Archaeobotanical evidence显示文摘The origin, development and expansion of prehistoric agriculture in East Asia have been widely investigated over the past two decades using archaeobotanical analysis from excavated Neolithic and Bronze Age sites. Research on prehistoric agriculture has predominantly focused in the valleys of the Yellow River and the Yangtze River. Agricultural development during the Neolithic and Bronze Age periods in the Yunnan-Guizhou Plateau of southwest China, an important passageway for human migration into Southeast Asia, still remains unclear. In this paper, based on macrofossil and microfossil analysis and radiocarbon dating at the Shilinggang site, we investigate plant subsistence strategies in the Nujiang River valley during the Bronze Age period. Combined with previous archaeobotanical studies in the Yunnan-Guizhou Plateau, we explore agricultural development processes in this area during the Neolithic and Bronze Age. Our results indicate that rice and foxtail millet were cultivated in Shilinggang around 2500 cal a BP. Three phases of prehistoric agricultural development in the Yunnan-Guizhou Plateau can be identified: rice cultivation from 4800–3900 cal a BP, mixed rice and millet crop(foxtail millet and broomcorn millet) cultivation from 3900–3400 cal a BP, and mixed rice, millet crop and wheat cultivation from 3400–2300 cal a BP. | 6 | Establishment of a cell-based assay to screen regulators for Klotho gene promoter显示文摘AIM: To discover compounds which can regulate Klotho promoter activity. Klotho is an aging suppressor gene. A defect in Klotho gene expression in the mouse results in the phenotype similar to human aging. Recombinant Klotho protein improves age-associated diseases in animal models. It has been proposed that up-regulation of Klotho gene expression may have anti-aging effects, METHODS: Klotho promoter was cloned into a vector containing luciferase gene, and the reporter gene vector was transfected into HEK293 cells to make a stable cell line (HEK293/KL). A model for cellular aging was established by treating HEK293/KL cells with H2O2. These cells were treated with extracts from Traditional Chinese Medicines (TCMs). The luciferase activity was detected to identify compounds that can regulate Klotho promoter. RESULTS:The expression of luciferase in these cells was under control of Klotho promoter and down-regulated after H2O2 treatment The down-regulation of luciferase expression was H2O2 concentration-dependent with an IC50 at approximately 0.006%. This result demonstrated that the Klotho gene promoter was regulated by oxidative stress. Using the cell-based reporter gene assay, we screened natural product extracts for regulation of Klotho gene promoter. Several extracts were identified that could rescue the H2O2 effects and up-regulated Klotho promoter activity. CONCLUSION: A cell -based assay for high-throughput drug screening was established to identify compounds that regulate Klotho promoter activity, and several hits were discovered from natural products.
