2Super-resolution fluorescence-assisted diffraction computational tomography reveals the threedimensional landscape of the cellular organelle interactome显示文摘The emergence of super-resolution(SR)fluorescence microscopy has rejuvenated the search for new cellular substructures.However,SR fluorescence microscopy achieves high contrast at the expense of a holistic view of the interacting partners and surrounding environment.Thus,we developed SR fluorescence-assisted diffraction computational tomography(SR-FACT),which combines label-free three-dimensional optical diffraction tomography(ODT)with two-dimensional fluorescence Hessian structured illumination microscopy.The ODT module is capable of resolving the mitochondria,lipid droplets,the nuclear membrane,chromosomes,the tubular endoplasmic reticulum,and lysosomes.Using dual-mode correlated live-cell imaging for a prolonged period of time,we observed novel subcellular structures named dark-vacuole bodies,the majority of which originate from densely populated perinuclear regions,and intensively interact with organelles such as the mitochondria and the nuclear membrane before ultimately collapsing into the plasma membrane.This work demonstrates the unique capabilities of SR-FACT,which suggests its wide applicability in cell biology in general.Dashan Dong Xiaoshuai Huang Liuju Li Heng Mao Yanquan Mo Guangyi Zhang Zhe Zhang Jiayu Shen Wei Liu Zeming Wu Guanghui Liu Yanmei Liu Hong Yang Qihuang Gong Kebin Shi Liangyi Chen 2020Light(Science & Applications)2020,9,1:7
3Distribution of Microelements and Their Influence on the Corrosion Behavior of Aluminum Foil显示文摘The distribution of microelement Fe, Si, Cu and Mg in the surface layer of aluminum foil annealed at 300℃ and 500℃ were determined by secondary ion mass spectrometer. The corrosion structure produced by electrochemical etching was also observed. It was found that the Mg concentration at external surface was increased exponentially over the fourth degree and promoted by higher annealing temperature, which will increase the number of corrosion pits inside the large grains, and therefore the specific capacity of the foils for electrolytic capacitors. 4Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 6Synaptic non-GluN2B-containing NMDA receptors regulate tyrosine phosphorylation of GluN2B 1472 tyrosine site in rat brain slices显示文摘Activation of N-methyl-D-aspartate receptors(NMDARs)mediates changes in the phosphorylation status of the glutamate receptors themselves.Previous studies have indicated that during synaptic activity,tyrosine kinases(Src and Fyn)or phosphatases(PTPαand STEP)are involved in regulating the phosphorylation of NMDARs.In this study,we used immunoblotting to investigate the role of an NMDAR subpopulation on the phosphorylation level of the GluN2B subunit at the Y1336 and Y1472sites in rat brain slices after NMDA treatment.We found that NMDA stimulation dramatically decreased the phosphorylation level of GluN2B at Y1472 in a dose-and time-dependent manner,but not at Y1336.Extrasynaptic NMDAR activation did not reduce the phosphorylation of GluN2B at Y1472.In addition,ifenprodil,a selective antagonist of GluN2Bcontaining NMDARs,did not abolish the decreased phosphorylation of GluN2B at Y1472 triggered by NMDA.These results suggest that the activation of synaptic GluN2A-containing NMDARs is required for the decreased phosphorylation of GluN2B at Y1472that is induced by NMDA treatment in rat brain slices.Heng Ai Wen Lu Mao Ye Wei Yang 2013Neuroscience Bulletin2013,29,5:4
7Recognition of moyamoya disease and its hemorrhagic risk using deep learning algorithms:sourced from retrospective studies显示文摘Although intracranial hemorrhage in moyamoya disease can occur repeatedly,predicting the disease is difficult.Deep learning algorithms developed in recent years provide a new angle for identifying hidden risk factors,evaluating the weight of different factors,and quantitatively evaluating the risk of intracranial hemorrhage in moyamoya disease.To investigate whether convolutional neural network algorithms can be used to recognize moyamoya disease and predict hemorrhagic episodes,we retrospectively selected 460 adult unilateral hemispheres with moyamoya vasculopathy as positive samples for diagnosis modeling,including 418 hemispheres with moyamoya disease and 42 hemispheres with moyamoya syndromes.Another 500 hemispheres with normal vessel appearance were selected as negative samples.We used deep residual neural network(ResNet-152)algorithms to extract features from raw data obtained from digital subtraction angiography of the internal carotid artery,then trained and validated the model.The accuracy,sensitivity,and specificity of the model in identifying unilateral moyamoya vasculopathy were 97.64±0.87%,96.55±3.44%,and 98.29±0.98%,respectively.The area under the receiver operating characteristic curve was 0.990.We used a combined multi-view conventional neural network algorithm to integrate age,sex,and hemorrhagic factors with features of the digital subtraction angiography.The accuracy of the model in predicting unilateral hemorrhagic risk was 90.69±1.58%and the sensitivity and specificity were 94.12±2.75%and 89.86±3.64%,respectively.The deep learning algorithms we proposed were valuable and might assist in the automatic diagnosis of moyamoya disease and timely recognition of the risk for re-hemorrhage.This study was approved by the Institutional Review Board of Huashan Hospital,Fudan University,China(approved No.2014-278)on January 12,2015.Yu Lei Xin Zhang Wei Ni Heng Yang Jia-Bin Su Bin Xu Liang Chen Jin-Hua Yu Yu-Xiang Gu Ying Mao 2021Neural Regeneration Research2021,16,5:3
8Study on the surface cracking in hot-rolled plates of Fe-23Mn-2Al-V austenitic steel显示文摘The study introduces the chemical composition,mechanical properties,physical properties and production processes of the Fe-23Mn-2Al-V austenitic steel.Investigations were made into the formation of cracks in the hot-rolled Fe-23Mn-2Al-V steel plates,with optical microscopy (OM),scanning electron microscopy (SEM) and energy dispersive spectrum (EDS).The results have shown that intergranular oxidation in the reheating furnace is the main cause of the cracks on the surface of the hot-rolled steel plates.The surface cracking can be largely avoided by controlling the remaining oxygen content in the reheating furnace and improving continuous casting (CC) processes to increase the thickness of fine equiaxed grain layer of the steel slabs.LIAO Hongjun MAO Zhengdong WANG Lintao ZHU Jun LI Yuan ZHANG Heng 2011Baosteel Technical Research2011,5,1:2

12In situ growth of a-few-layered MoS_(2)on CdS nanorod for high efficient photocatalytic H_(2)production显示文摘An ultrathin MoS_(2)was grown on CdS nanorod by a solid state method using sulfur powder as sulfur source for photocatalytic H_(2)production.The characterization result reveals that the ultrathin MoS_(2)nanosheets loaded on CdS has a good contact state.The photoelectrochemical result shows that MoS_(2)not only are beneficial for charge separation,but also works as active sites,thus enhancing photocatalytic activity.Compared with pure CdS,the photocatalytic activity of MoS_(2)loaded CdS was significantly improved.The hydrogen evolution rate on m(MoS_(2)):m(CdS)=1:50(m is mass)reaches 542μmol/h,which is 6 times of that on pure CdS(92μmol/h).This work provides a new design for photocatalysts with high photocatalytic activities and provides a deeper understanding of the effect of MoS_(2)on enhancing photocatalytic activity.Wei CHEN Xiang LIU Shaojie WEI Qianqian HENG Binfen WANG Shilong LIU Li GAO Liqun MAO 2021Frontiers in Energy2021,15,3:1

14Plant metabolomics for studying the effect of two insecticides on comprehensive constituents of Lonicerae Japonicae Flos显示文摘Pesticides'overuse and misuse have been reported to induce ingredient variations in herbal medicine,which is now gaining attention in the medicinal field as a form of alternative medicine.To date,available studies on pesticide-induced ingredient variations of herbal medicine are limited only on a few compounds and remain most others unexamined.In this study,a plant metabolomics-based strategy was performed to systematically explore the effects of two frequently used insecticides on the comprehensive constituents of Lonicerae Japonicae Flos(LJF),the flower buds of Lonicera japonica Thunb..Field trials were designed on a cultivating plot of L.japonica with controls and treatments of imidacloprid(IMI)and compound flonicamid and acetamiprid(CFA).Unbiased metabolite profiling was conducted by ultra-high performance liquid chromatography/quadrupole-Orbitrap mass spectrometer.After data pretreatment by automatic extraction and screening,a data matrix of metabolite features was submitted for statistical analyses.Consequently,29 metabolic markers,including chlorogenic acids,iridoids and organic acid-glucosides were obtained and characterized.The relative quantitative assay was subsequently performed to monitor their variations across flowering developments.This is the first study that systematically explored the insecticide-induced metabolite variations of LJF while taking into account the inherent variability of flowering development.The results were beneficial for holistic quality assessment of LJF and significant for guiding scientific use of pesticides in the large-scale cultivation.PAN Hui-Qin ZHOU Heng MIAO Shui GUO De-An ZHANG Xiao-Li HU Qing MAO Xiu-Hong JI Shen 2021Chinese Journal of Natural Medicines2021,19,1:1

173D Hessian deconvolution of thick light-sheet z-stacks for high-contrast and high-SNR volumetric imaging显示文摘Due to its ability of optical sectioning and low phototoxicity,z-stacking light-sheet microscopy has been the tool of choice for in vivo imaging of the zebrafish brain.To image the zebrafish brain with a large field of view,the thickness of the Gaussian beam inevitably becomes several times greater than the system depth of field(DOF),where the fluorescence distributions outside the DOF will also be collected,blurring the image.In this paper,we propose a 3D deblurring method,aiming to redistribute the measured intensity of each pixel in a light-sheet image to in situ voxels by 3D deconvolution.By introducing a Hessian regularization term to maintain the continuity of the neuron dis-tribution and using a modified stripe removal algorithm,the reconstructed z stack images exhibit high contrast and a high signal-to-noise ratio.These performance characteristics can facilitate subsequent processing,such as 3D neuron registration,segmentation,and recognition.ZHE ZHANG DONGZHOU GOU FAN FENG RUYI ZHENG KE DU HONGRUN YANG GUANGYI ZHANG HUITAO ZHANG LOUIS TAO LIANGYI CHEN HENG MAO 2020Photonics Research2020,8,6:1

19Scientific objectives of the Hot Universe Baryon Surveyor(HUBS) mission显示文摘The Hot Universe Baryon Surveyor(HUBS) is a proposed space-based X-ray telescope for detecting X-ray emissions from the hot gas content in our universe. With its unprecedented spatially-resolved high-resolution spectroscopy and large field of view,the HUBS mission will be uniquely qualified to measure the physical and chemical properties of the hot gas in the interstellar medium, the circumgalactic medium, the intergalactic medium, and the intracluster medium. These measurements will be valuable for two key scientific goals of HUBS, namely to unravel the AGN and stellar feedback physics that governs the formation and evolution of galaxies, and to probe the baryon budget and multi-phase states from galactic to cosmological scales. In addition to these two goals, the HUBS mission will also help us solve some problems in the fields of galaxy clusters, AGNs, difuse X-ray backgrounds, supernova remnants, and compact objects.
